Evaluation of coated QCM for the detection of atmospheric ozone
A coated acoustic wave sensor has been developed to selectively detect atmospheric ozone. The selective detection has been assessed using a variety of coatings: beeswax, gallic acid, indigo carmine, polybutadiene, potassium iodide and sodium nitrite. Polybutadiene was the most sensitive with a limit of detection of 55 ppb. The sensitivity was improved by operating at higher harmonics and was shown to increase linearly with harmonic up to the 11th harmonic. This novel work shows that ozone detection can be improved by operating at the crystals' harmonic frequencies and in conjunction with a suitable flow rate, a potentially highly sensitive and fast response sensor can be created based on acoustic wave technolog

Uncovering regulatory pathways that affect hematopoietic stem cell function using 'genetical genomics'
We combined large-scale mRNA expression analysis and gene mapping to identify genes and loci that control hematopoietic stem cell (HSC) function. We measured mRNA expression levels in purified HSCs isolated from a panel of densely genotyped recombinant inbred mouse strains. We mapped quantitative trait loci (QTLs) associated with variation in expression of thousands of transcripts. By comparing the physical transcript position with the location of the controlling QTL, we identified polymorphic cis-acting stem cell genes. We also identified multiple trans-acting control loci that modify expression of large numbers of genes. These groups of coregulated transcripts identify pathways that specify variation in stem cells. We illustrate this concept with the identification of candidate genes involved with HSC turnover. We compared expression QTLs in HSCs and brain from the same mice and identified both shared and tissue-specific QTLs. Our data are accessible through WebQTL, a web-based interface that allows custom genetic linkage analysis and identification of coregulated transcripts.
The antimicrobial polymer PHMB enters cells and selectively condenses bacterial chromosomes
To combat infection and antimicrobial resistance, it is helpful to elucidate drug mechanism(s) of action. Here we examined how the widely used antimicrobial polyhexamethylene biguanide (PHMB) kills bacteria selectively over host cells. Contrary to the accepted model of microbial membrane disruption by PHMB, we observed cell entry into a range of bacterial species, and treated bacteria displayed cell division arrest and chromosome condensation, suggesting DNA binding as an alternative antimicrobial mechanism. A DNA-level mechanism was confirmed by observations that PHMB formed nanoparticles when mixed with isolated bacterial chromosomal DNA and its effects on growth were suppressed by pairwise combination with the DNA binding ligand Hoechst 33258. PHMB also entered mammalian cells, but was trapped within endosomes and excluded from nuclei. Therefore, PHMB displays differential access to bacterial and mammalian cellular DNA and selectively binds and condenses bacterial chromosomes. Because acquired resistance to PHMB has not been reported, selective chromosome condensation provides an unanticipated paradigm for antimicrobial action that may not succumb to resistance
IFNβ Protects Neurons from Damage in a Murine Model of HIV-1 Associated Brain Injury.
Infection with human immunodeficiency virus-1 (HIV-1) causes brain injury. Type I interferons (IFNα/β) are critical mediators of any anti-viral immune response and IFNβ has been implicated in the temporary control of lentiviral infection in the brain. Here we show that transgenic mice expressing HIV-1 envelope glycoprotein 120 in their central nervous system (HIVgp120tg) mount a transient IFNβ response and provide evidence that IFNβ confers neuronal protection against HIVgp120 toxicity. In cerebrocortical cell cultures, neuroprotection by IFNβ against gp120 toxicity is dependent on IFNα receptor 1 (IFNAR1) and the β-chemokine CCL4, as IFNAR1 deficiency and neutralizing antibodies against CCL4, respectively, abolish the neuroprotective effects. We find in vivo that IFNβ mRNA is significantly increased in HIVgp120tg brains at 1.5, but not 3 or 6 months of age. However, a four-week intranasal IFNβ treatment of HIVgp120tg mice starting at 3.5 months of age increases expression of CCL4 and concomitantly protects neuronal dendrites and pre-synaptic terminals in cortex and hippocampus from gp120-induced damage. Moreover, in vivo and in vitro data suggests astrocytes are a major source of IFNβ-induced CCL4. Altogether, our results suggest exogenous IFNβ as a neuroprotective factor that has potential to ameliorate in vivo HIVgp120-induced brain injury
Nutritional status and structural brain changes in Alzheimer's disease: The NUDAD project
INTRODUCTION: Weight loss is associated with higher mortality and progression of cognitive decline, but its associations with magnetic resonance imaging (MRI) changes related to Alzheimer's disease (AD) are unknown. METHODS: We included 412 patients from the NUDAD project, comprising 129 with AD dementia, 107 with mild cognitive impairment (MCI), and 176 controls. Associations between nutritional status and MRI measures were analyzed using linear regression, adjusted for age, sex, education, cognitive functioning, and cardiovascular risk factors. RESULTS: Lower body mass index (BMI), fat mass (FM), and fat free mass index were associated with higher medial temporal atrophy (MTA) scores. Lower BMI, FM, and waist circumference were associated with more microbleeds. Stratification by diagnosis showed that the observed associations with microbleeds were only significant in MCI. DISCUSSION: Lower indicators of nutritional status were associated with more MTA and microbleeds, with largest effect sizes in MCI
Adiabatic Theory of Electron Detachment from Negative Ions in Two-Color Laser Field
Negative ion detachment in bichromatic laser field is considered within the
adiabatic theory. The latter represents a recent modification of the famous
Keldysh model for multiphoton ionization which makes it quantitatively
reliable. We calculate angular differential detachment rates, partial rates for
particular ATD (Above Threshold Detachment) channels and total detachment rates
for the Hydrogen ion in a bichromatic field with 1:3 frequency ratio and
various phase differences. Reliability of the present, extremely simple
approach is testified by comparison with much more elaborate earlier

The first determination of Generalized Polarizabilities of the proton by a Virtual Compton Scattering experiment
Absolute differential cross sections for the reaction (e+p -> e+p+gamma) have
been measured at a four-momentum transfer with virtuality Q^2=0.33 GeV^2 and
polarization \epsilon = 0.62 in the range 33.6 to 111.5 MeV/c for the momentum
of the outgoing photon in the photon-proton center of mass frame. The
experiment has been performed with the high resolution spectrometers at the
Mainz Microtron MAMI. From the photon angular distributions, two structure
functions which are a linear combination of the generalized polarizabilities

PECAM-Independent Thioglycollate Peritonitis Is Associated With a Locus on Murine Chromosome 2
Background: Previous studies have demonstrated that knockout or inhibition of Platelet/Endothelial Cell Adhesion Molecule (PECAM, CD31) in a number of murine strains results in impaired inflammatory responses, but that no such phenotype is seen in the C57BL/6 (B6) murine background. Methodology/Principal Findings: We have undertaken a quantitative trait locus (QTL) mapping effort between FVB/n (FVB) and B6 mice deficient for PECAM to identify the gene or genes responsible for this unique feature of B6 mice. We have identified a locus on murine chromosome 2 at approximately 35.8 Mb that is strongly associated (LOD score = 9.0) with inflammatory responses in the absence of PECAM. Conclusions/Significance: These data potentiate further study of the diapedesis machinery, as well as potential identification of new components of this machinery. As such, this study is an important step to better understanding the processes of inflammation
