Effects of black tea on body composition and metabolic outcomes related to cardiovascular disease risk: a randomized controlled trial

cardiovascular health. Randomized controlled trials have shown that tea can improve cardiovascular
disease risk factors. We have previously reported a non-caffeine associated beneficial effect of regular
black tea consumption on blood pressure and its variation. Objective: To explore the non-caffeine
associated effects of black tea on body weight and body fat distribution, and cardiovascular disease related
metabolic outcomes. Design: regular tea-drinking men and women (n ¼ 111; BMI 20–35 kg m 2) were
recruited to a randomized controlled double-blind 6 month parallel-designed trial. Participants consumed
3 cups per day of either powdered black tea solids (tea) or a flavonoid-free flavour- and caffeine-matched
placebo (control). Body weight, waist- and hip-circumference, endothelial function and plasma biomarkers
were assessed at baseline, 3 months and 6 months. Results: Compared to control, regular ingestion of
black tea over 3 months inhibited weight gain ( 0.64 kg, p ¼ 0.047) and reduced waist circumference
( 1.88 cm, P ¼ 0.035) and waist-to-hip ratio ( 0.03, P ¼ 0.005). These effects were no longer significant
at 6 months. There were no significant effects observed on fasting glucose, insulin, plasma lipids or
endothelial function. Conclusion: Our study suggests that short-term regular ingestion of black tea over 3
months can improve body weight and body fat distribution, compared to a caffeine-matched control
beverage. However, there was no evidence that these effects were sustained beyond 3 months
The Influence of Mineralization on Intratrabecular Stress and Strain Distribution in Developing Trabecular Bone
The load-transfer pathway in trabecular bone is largely determined by its architecture. However, the influence of variations
in mineralization is not known. The goal of this study was to examine the influence of inhomogeneously distributed degrees
of mineralization (DMB) on intratrabecular stresses and strains. Cubic mandibular condylar bone specimens from fetal and newborn
pigs were used. Finite element models were constructed, in which the element tissue moduli were scaled to the local DMB. Disregarding
the observed distribution of mineralization was associated with an overestimation of average equivalent strain and underestimation
of von Mises equivalent stress. From the surface of trabecular elements towards their core the strain decreased irrespective
of tissue stiffness distribution. This indicates that the trabecular elements were bent during the compression experiment.
Inhomogeneously distributed tissue stiffness resulted in a low stress at the surface that increased towards the core. In contrast,
disregarding this tissue stiffness distribution resulted in high stress at the surface which decreased towards the core. It
was concluded that the increased DMB, together with concurring alterations in architecture, during development leads to a
structure which is able to resist increasing loads without an increase in average deformation, which may lead to damage
Increased insulin sensitivity and diminished pancreatic beta-cell function in DNA repair deficient Ercc1(d/-) mice
Background: Type 2 diabetes (T2DM) is an age-associated disease characterized by hyperglycemia due to insulin resistance and decreased beta-cell function. DNA damage accumulation has been associated with T2DM, but whether DNA damage plays a role in the pathogenesis of the disease is unclear. Here, we used mice deficient for the DNA excision-repair gene Ercc1 to study the impact of persistent endogenous DNA damage accumulation on energy metabolism, glucose homeostasis and beta-cell function. Methods: ERCC1-XPF is an endonuclease required for multiple DNA repair pathways and reduced expression of ERCC1-XPF causes accelerated accumulation of unrepaired endogenous DNA damage and accelerated aging in humans andmice. In this study, energy metabolism, glucose metabolism, beta-cell function and insulin sensitivity were studied in Ercc1(d/-) mice, which model a human progeroid syndrome. Results: Ercc1(d/-) mice displayed suppression of the somatotropic axis and altered energy metabolism. Insulin sensitivitywas increased, whereas, plasma insulin levelswere decreased in Ercc1(d/-) mice. Fasting induced hypoglycemia in Ercc1(d/-) mice, whichwas the result of increased glucose disposal. Ercc1(d/-) mice exhibit a significantly reduced beta-cell area, even compared to control mice of similar weight. Glucose-stimulated insulin secretion in vivo was decreased in Ercc1(d/-) mice. Islets isolated from Ercc1(d/-) mice showed increased DNA damage markers, decreased glucose-stimulated insulin secretion and increased susceptibility to apoptosis. Conclusion: Spontaneous DNA damage accumulation triggers an adaptive response resulting in improved insulin sensitivity. Loss of DNA repair, however, does negatively impacts beta-cell survival and function in Ercc1(d/-) mice. (C) 2021 The Author(s). Published by Elsevier Inc
Toward a generic extraction method for simultaneous determination of pesticides, mycotoxins, plant toxins, and veterinary drugs in feed and food matrixes
A fast and straightforward generic procedure for the simultaneous extraction of various classes of pesticides, mycotoxins, plant toxins, and veterinary drugs in various matrixes has been developed, for subsequent analysis by liquid chromatography with mass spectrometric detection. As a first step, four existing multianalyte procedures and three newly proposed methods were compared for a test set of 172 pesticides, mycotoxins, and plant toxins spiked to a feed matrix. The new procedures, which basically involved extraction/dilution of the sample with water and an acidified organic solvent (methanol, acetonitrile, or acetone), were most promising. The three new generic extraction methods were further tested for applicability to other matrixes (maize, honey, milk, egg, meat). Overall, the best recoveries were obtained for acetone, followed by acetonitrile. With respect to matrix effects, acetonitrile was the most favorable solvent and methanol was the worst. The occurrence of matrix effects decreased for the matrixes in the order of feed > maize > meat > milk > egg > honey. The extraction method selected as the default procedure (water/acetonitrile/1% formic acid) was also evaluated for applicability to multiple classes of veterinary drugs in all six matrixes, with satisfactory results. Finally, the generic extraction procedure was validated for 136 pesticides, 36 natural toxins, and 86 veterinary drugs in compound feed and honey at three levels (0.01, 0.02, and 0.05 mg/kg) using ultraperformance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) for analysis of the extracts. For over 80% of the analytes, recoveries were between 70 and 120% and precision (expressed as relative standard deviation) was mostly in the range of 5-10% (except for feed at 0.01 mg/kg; adequate recoveries for 62% of the analytes). The limits of detection were from <0.01 to 0.05 mg/kg for most analytes, which is usually sufficient to verify compliance of products with legal tolerances. The results clearly demonstrate the feasibility of the generic approach proposed. Application of the method in routine monitoring programs would imply a drastic reduction of both effort and time
The reliability of postural balance measures in single and dual tasking in elderly fallers and non-fallers
BACKGROUND: The purpose of this study was to determine the reliability of a forceplate postural balance protocol in a group of elderly fallers and non-fallers. The measurements were tested in single and dual-task conditions, with and without vision. METHODS: 37 elderly (mean age 73 +/- 6 years) community-dwellers were included in this study. All were tested in a single (two-legged stance) and in a dual-task (two-legged stance while counting backwards aloud in steps of 7's) condition, with and without vision. A forceplate was used for registering postural variables: the maximal and the root-mean-square amplitude in medio-lateral (Max-ML, RMS-ML) and antero-posterior (Max-AP, RMS-AP) direction, mean velocity (MV), and the area of the 95% confidence ellipse (AoE). Reliability of the test protocol was expressed with intraclass correlation coefficients (ICC), with 95% limits of agreement (LoA), and with the smallest detectable difference (SDD). RESULTS: The ICCs for inter-rater reliability and test-retest reliability of the balance variables were r = 0.70-0.89. For the variables Max-AP and RMS-AP the ICCs were r = 0.52-0.74. The SDD values were for variable Max-ML and Max-AP between 0.37 cm and 0.83 cm, for MV between 0.48 cm/s and 1.2 cm/s and for AoE between 1.48 cm2 and 3.75 cm2. The LoA analysis by Bland-Altman plots showed no systematic differences between test-retest measurements. CONCLUSION: The study showed good reliability results for group assessment and no systematic errors of the measurement protocol in measuring postural balance in the elderly in a single-task and dual-task condition
Childhood traumatic experiences and mental health problems in sexually offending and non-sexually offending juveniles
OBJECTIVE: To examine the relationship between a history of childhood abuse and mental health problems in juveniles who sexually offended (JSOs) over and above general offending behavior.
METHODS: A sample of 44 JSOs incarcerated in two juvenile detention centers in the Netherlands between May 2008 and March 2014 were examined for childhood abuse history (Childhood Trauma Questionnaire-Short Form) and mental health problems (Massachusetts Youth Screening Instrument-Version 2). Furthermore, the connection between childhood abuse and mental health problems in JSOs was compared to a sample of 44 propensity score matched juveniles who offended non-sexually (non-JSOs).
RESULTS: In JSOs, sexual abuse was related to anger problems, suicidal ideation, and thought disturbance. These associations were significantly stronger in JSOs than in non-JSOs.
CONCLUSIONS: Our results suggest that the relationship between childhood abuse and both internalizing and externalizing mental health problems is of more salience for understanding sexual offending than non-sexual offending, and should, therefore, be an important focus in the assessment and treatment of JSOs
Impact of flavonoid-rich black tea and beetroot juice on postprandial peripheral vascular resistance and glucose homeostasis in obese, insulin-resistant men: a randomized controlled trial.
BACKGROUND: Insulin-stimulated muscle blood flow facilitates plasma glucose disposal after a meal, a mechanism that is impaired in obese, insulin-resistant volunteers. Nitrate- or flavonoid-rich products, through their proposed effects on nitric oxide, may improve postprandial blood flow and, subsequently, glucose disposal. To investigate whether a single dose of nitrate-rich beetroot juice or flavonoid-rich black tea lowers postprandial muscle vascular resistance in obese volunteers and alters postprandial glucose or insulin concentrations. METHOD: In a randomised, controlled, cross-over study, 16 obese, insulin-resistant males consumed 75 g glucose, which was combined with 100 ml black tea, beetroot juice or control (water). Peripheral vascular resistance (VR), calculated as mean arterial pressure divided by blood flow, was assessed in the arm and leg conduit arteries, resistance arteries and muscle microcirculation across 3 h (every 30-min) after the oral glucose load. RESULTS: During control, we found no postprandial response in VR in conduit, resistance and microvessels (all P > 0.05). Black tea decreased VR compared to control in conduit, resistance and microvessels (all P < 0.05). Beetroot juice decreased postprandial VR in resistance vessels, but not in conduit artery and microvessels. Although postprandial glucose response was similar after all interventions, postprandial insulin response was attenuated by ~29 % after tea (P < 0.0005), but not beetroot juice. CONCLUSIONS: A single dose of black tea decreased peripheral VR across upper and lower limbs after a glucose load which was accompanied by a lower insulin response. Future studies in insulin-resistant subjects are warranted to confirm the observed effects and to explore whether long-term regular tea consumption affects glucose homeostasis. Avoidance as a Predictor of the Biological Course of HIV Infection Over a 7-Year Period in Gay Men
The present study prospectively investigated the relation
between avoidance coping and active cognitive and behavioral coping
and the progression of HIV infection over 7 years in 181 gay men.
Findings revealed that for a number of medical and behavioral
factors, (a) avoidance coping predicted a lower rate of decline in
CD4 cells, (b) the proportional hazard (PH) attributable to
avoidance of developing a syncytium-inducing HIV variant was 0.72
(95% confidence interval [CI]: 0.53 − 0.99,

< .05), and (c) the PH attributable to
avoidance of dropping below 200 CD4 cells/μl was 0.66
(95% CI: 0.50 − 0.89,

.01). Avoidance coping was not related to the development of
