    Insights into the nutritional properties and molecular basis of biosynthesis of amino acids and vitamins of Gastrodia elata offered by metabolomic and transcriptomic analysis

    Gastrodia elata Blume (GE), a traditional and precious Chinese medicinal material, has been approved as a functional food. However, understanding GE’s nutritional properties and its molecular basis remains limited. Here, metabolomic and transcriptomic analyses were performed on young and mature tubers of G. elata.f.elata (GEEy and GEEm) and G. elata.f.glauca (GEGy and GEGm). A total of 345 metabolites were detected, including 76 different amino acids and their derivatives containing all human essential amino acids (e.g., l-(+)-lysine, l-leucine), 13 vitamins (e.g., nicotinamide, thiamine), and 34 alkaloids (e.g., spermine, choline). GEGm has higher amino acid accumulation than GEEy, GEEm and GEGy, and vitamin contents were also slightly different in all four samples. Implying that GE, especially GEGm, is a kind of excellent complementary food as amino acid nutrition provider. From assembled 21,513 transcripts (genes) based on the transcriptome, we identified many genes that encode enzymes (e.g., pfkA, bglX, tyrAa, lysA, his B, aroA), which are responsible for the biosynthesis of amino acids and enzymes (e.g., nadA, URH1, NAPRT1, punA, rsgA) that related to vitamins metabolism. A total of 16 pairs of the differentially expressed genes (DEG) and differentially accumulated metabolites (DAM) (e.g., gene-tia006709 coding GAPDH and l-(+)-arginine, and gene-tia010180 coding tyrA and l-(+)-arginine) and three DEG-DAM pairs (e.g., gene-tia015379 coding NadA and nicotinate d-ribonucleoside) show significant similar positive or negative correlation based on three, and two comparisons of GEEy vs. GEGy, GEGy vs. GEGm, GEEy vs. GEGy and GEEm vs. GEGm, which involved into amino acid biosynthesis, and nicotinate nicotinamide metabolism, respectively. These results prove that the enzyme coded by these DEG promotes (positive correlation) or inhibits (negative correlation) the biosynthesis of parallel DAM in GE. Overall, the data and corresponding analysis in this study provide new insights into the nutritional properties of GE and the related molecular basis

    Impact Assessment of Pozzolanic Material Coupled with River Bed Aggregate on Expansive Behavior of High Strength Concrete

    High strength concrete is widely used in engineering structures. Present work is an effort for suitability assessment of river bed aggregates in high strength concrete to be used at structural units of Dam (Spillway & Powerhouse) and the vulnerability of placed concrete expansion. Multiple sizes of coarse aggregate (5-20mm, 20-40mm, and 40-80mm) from Beor river bed material have been tested for physical (water absorption, crushing index, soundness, shape, and bulk density), and mineralogical characterization. Concrete Mix Design (CMD) for Spillway has been optimized using Fly ash and river aggregate that achieved the Unconfined Compressional Strength (UCS) up to 32.5 MPa. The accelerated mortar bar test (AMBT) has been introduced to gauge the reactive aggregates used in CMD.. Expansive properties of concrete were observed at the age of 7 days and 28 days that demonstrate more expansion of the specimen with slag rather than the Fly ash. Results of AMBT suggest that a minimum proportion of GGBS (40%) is needed to limit the AMBT expansion to less than 0.1% for crushed river bed aggregate and sand from the Beor source. The petrographical characterization of coarse aggregate shows the presence of deformed quartz in the coarse aggregate, which directly relates to water absorption (Wa), suggesting its suceseptility to Alkali-Silica Reaction (ASR). Modifications in Pozzolanic additives in CMD indicates that 30% mixing of Fly ash can reduce the expansion rate of concrete up to 96.15%

    Digital Twin Concept, Method and Technical Framework for Smart Meters

    Smart meters connect smart grid electricity suppliers and users. Smart meters have become a research hotspot as smart grid applications like demand response, power theft prevention, power quality monitoring, peak valley time of use prices, and peer-to-peer (P2P) energy trading have grown. But, as the carriers of these functions, smart meters have technical problems such as limited computing resources, difficulty in upgrading, and high costs, which to some extent restrict the further development of smart grid applications. To address these issues, this study offers a container-based digital twin (CDT) approach for smart meters, which not only increases the user-facing computing resources of smart meters but also simplifies and lowers the overall cost and technical complexity of meter changes. In order to further validate the effectiveness of this method in real-time applications on the smart grid user side, this article tested and analyzed the communication performance of the digital twin system in three areas: remote application services, peer-to-peer transactions, and real-time user request services. The experimental results show that the CDT method proposed in this paper meets the basic requirements of smart grid user-side applications for real-time communication. The container is deployed in the cloud, and the average time required to complete 100 P2P communications using our smart meter structure is less than 2.4 seconds, while the average time required for existing smart meter structures to complete the same number of P2P communications is 208 seconds.     Effect of Sustainable HRM Practices on Job Performance: Mediating Role of Employee Retention

    The primary objective of this research was to investigate the mediating role of employee retention in the relationship between employee empowerment, high-performance work systems, and job performance in the health sector. A cross-sectional study design was used, and data was collected from the health sector operating in South Punjab, Pakistan. Statistical random sampling method was employed, to a sample of 550 employees the questionnaires was randomly distributed for data collection through a self-administered plan. SPSS software was used and Partial Least Squares Structural Equation Modelling (PLS-SEM) technique was employed for testing of research hypotheses. Current study based its investigation into theory of performance. The findings showed that the relationship between employee empowerment and job performance was insignificant, but the relationship between high-performance work systems and job performance was significant. Additionally, employee retention has a significant mediating effect on the relationship between High Performance Work System, and job performance. The results provide insight for the government, practitioners, and policymakers to better understand the impact of various sustainable human resource management practices on job performance.

    Determinants of Anxiety in Amputees Owed to Traumatic & Non-Traumatic Causes in Quetta

    Objective: To find out the causes and factors of anxiety among amputees suffering from traumatic and non-traumatic causes. Methodology: This descriptive cross-sectional study was conducted in Quetta from the month of May 2018 to July 2018. This study included those clients with amputations as a result of Traumatic & non-traumatic causes and the data was collected from them. This study involved a total of 54 participants. All amputees who visited the three Physical Rehabilitation Centre were included during data collection process. A structured adopted questionnaire using the Hospital Anxiety and Depression Scale (HADS) according to the inclusion & exclusion criteria & then tabularized and analyzed by applying Chi square test. Results: This study involved a total of 54 participants of whom 7(13%) were females and 47(87%) males. Among amputees 36(66.7 %), amputations were due to Traumatic injuries and 18(33.3%) were due to non-traumatic amputations. In traumatic, the majority was due to RTA with 25(46.3 %) and in non-traumatic causes diabetes stood first with 13(24.1%) amputation. Traumatic are more anxious than non-traumatic. There is a significant relationship between marital status and anxiety level (p-value 0.047). Statistically significant relationships between amputation and anxiety level were noticed, p value=0.049. Conclusion: Amputation has a significant association with anxiety level. Traumatic experiences make people feel more anxious than non-traumatic ones. Socio-demographics has association with levels of anxiety. It is recommended to do regular or annual screening of these patients following amputation

    Moderating Role of Organizational Justice between Emotional Exhaustion and Job-related Outcomes: Longitudinal Study at Education Sector

    Previous studies anticipated that there is need of justice in educational institutions in order to protect the academicians’ psychological as well as emotional needs under teaching profession. Therefore, the purpose of current study is to examine the moderating role of organizational justice between the relationship of emotional exhaustion and job related outcomes (Organizational commitment and Organizational loyalty).In order to accomplish this purpose, the survey questionnaire method was used. The study is quantitative, descriptive and longitudinal in nature. 250 respondents were selected in time1 & time2 by applying simple random sampling. Results indicated that emotional exhaustion and employee loyalty are negatively associated with each other (Bt1= -.328; Tr2= -.329). Similarly negative correlation found between emotional exhaustion and commitment (Tr1= -.425; Tr2= -.427).Results in time2 exhibited more understanding about understudy relationships. Moreover, results also enlightened that organizational justice significantly moderate on the relationship of emotional exhaustion and job related outcomes (Organizational commitment and Organizational loyalty)

    Oxidative Stress Diminishing Perspectives of Green and Black Tea Polyphenols: A Mechanistic Approach

    Polyphenols have credentials to tackle the oxidative stress. Oxidative stress is the imbalance between free radicals production and antioxidant enzymes ability to tackle these radicals resulting the onset various metabolic related disorders. Polyphenols based foods have credential as a shield against these glitches mainly owing to their antioxidant potential. In this context, tea polyphenols have gained paramount attention of scientific community as therapeutic agents for the prevention and treatment of various oxidative stress induce maladies owing to their structural diversity, strong antioxidant ability and capacity to modulate various expression involved in the pathogenesis of these maladies. The notable polyphenols are catechins which are mainly present in green tea and further subdivided into various compounds like ECG, EGC, EGCG which has their unique therapeutic potential. The catechins undergo various structural changes and transformed into theaflavins and thearubigins in the process of black tea formation. These are high molecular weight polyphenols and promising candidates in obesity, diabetes and cancer treatment. Mechanistically, these polyphenols ameliorate oxidative stress by trapping the noxious radicals like superoxide and peroxyl, promote the activity of glutathione, suppressing the malondialdehyde (MDA) activity.
