Shakespeare (way down) along the Nile: How a pidgin adaptation of Cymbeline gave South Sudan its theatre

    Get PDF
Aim of the paper is to analyse the gradual development of Southern Sudanese theatre, focusing on the development of Juba Arabic in the light of the contemporary translation of Shakespeare’s Cymbeline by the prominent intellectual Joseph Abuk, who is also the father of the South Sudan national anthem. The paper is divided in two parts: in the first one, the authors investigate the history of theatre in Sudan and South Sudan, considering the historical background that influenced it with a focus on the adaptations of Shakespeare’s works in Arab and African countries. The second part is dedicated to the relation between languages and ethnicity in South Sudan, thus offering a brief overview of its socio-linguistic landscape. A major attention is given to Abuk’s adaptation of Shakespeare’s Cymbeline from English into Juba Arabic (Cymbeline: li katib Shakespeare) for the South Sudan Theatre Organization (SSTO). This second part also comprises an appendix, which provides a brief linguistic analysis of some selected parts from the 2012 adaptation, as part of the recorded play Cymbeline (London, June 2012), performed by the South Sudan Theatre Organization (SSTO). The original Abuk’s play script (2012) is yet unpublished, while a full video-recording of the play is available at: https://globeplayer.tv/videos/Cymbelin

With 40% share of the global plastic demand, plastic packaging is currently the major sector in plastic industry (PlasticsEurope, 2019b). The fact that majority of plastic materials are not biodegradable points out the critical importance of plastic packaging waste treatment and its role in reducing their negative environmental footprint. Despite significant improvements in plastic recycling technologies, recycling of plastic packaging waste remains challenging. Intensive energy consumption, high costs and complexity in sorting, cleaning and pre-treatments steps, reprocessing issues, as well as inferior quality of the recycled materials compared to raw plastics are amongst the many impediments associated with recycling of plastic packaging wastes. Enhancing the current recycling rate and up taking the quality of the recycled martials in plastic packaging is achievable through designing for recyclability, improved collection and sorting, advancements in recycling technologies, and more importantly, incorporation of the recyclates into new packaging structures

Signalling and transcriptional regulation of early developmental lineage decisions

Embryonic stem (ES) cells are cell lines isolated from the embryo at a time just prior to implantation into the uterus. In the right cocktail of medium and cytokines, these cell lines can be maintained indefinitely in vitro in a self-renewing state. Initially it was assumed that these cells represented a homogeneous population however, more recently it has been shown that there are a great number of genes that are expressed heterogeneously. ES cell cultures are therefore a mix of different subpopulations, some of which have distinct functional properties including a bias or ‘lineage priming’ towards a particular cell fate. These populations are also dynamic in nature, converting from one state to another with fairly rapid kinetics. The main focus of this thesis was to gain a more in depth understanding of the mechanisms regulating heterogeneity and lineage priming in murine ES cells by asking which signalling pathways play a role in this phenomenon and how the switch between states is regulated at a transcriptional level. These questions were asked using an ES cell line containing a sensitive reporter for the endoderm marker Hex. This reporter, developed by a previous lab member, allowed the identification and separation of a population of ES cells primed towards a primitive endoderm fate. Primarily, I assessed the effect of a defined culture system (2i) on the Hex-expressing population. This culture system contains inhibitors that block FGF signalling and the Wnt pathway component GSK3. Culturing ES cells in 2i has been suggested to generate a more homogeneous culture. Here, I have shown that culturing ES cells or pre-implantation embryos in 2i did not eliminate heterogeneity but maintained them in an early state prior to lineage segregation. When ES cells were cultured in standard serum-containing medium, Hex was expressed in a mutually exclusive manner with the embryonic marker NANOG, while in 2i a subpopulation of cells coexpressed both Hex and NANOG. This population was functionally primed towards extraembryonic endoderm and trophoblast. Furthermore, these ES cells could efficiently contribute to 2-cell embryos in chimaera assays. LIF signalling promoted this population through the JAK/STAT pathway. I then asked how transcription was regulated during the switch between unprimed ES cells to those primed towards a primitive endoderm fate, as well as how regulation changes during further differentiation. To ask this, Hex positive (primed) and negative (unprimed) ES cell populations were sorted as well as a Hex positive differentiated sample. These samples were analysed by GRO-seq to determine the location, density and orientation of RNA-polymerase throughout the genome. Changes in gene expression between primed and unprimed states were regulated primarily through elongation whereas genes upregulated during differentiation were regulated at the point of de novo initiation

    Get PDF

The transcription factor Rreb1 regulates epithelial architecture, invasiveness, and vasculogenesis in early mouse embryos.

Ras-responsive element-binding protein 1 (Rreb1) is a zinc-finger transcription factor acting downstream of RAS signaling. Rreb1 has been implicated in cancer and Noonan-like RASopathies. However, little is known about its role in mammalian non-disease states. Here, we show that Rreb1 is essential for mouse embryonic development. Loss of Rreb1 led to a reduction in the expression of vasculogenic factors, cardiovascular defects, and embryonic lethality. During gastrulation, the absence of Rreb1 also resulted in the upregulation of cytoskeleton-associated genes, a change in the organization of F-ACTIN and adherens junctions within the pluripotent epiblast, and perturbed epithelial architecture. Moreover, Rreb1 mutant cells ectopically exited the epiblast epithelium through the underlying basement membrane, paralleling cell behaviors observed during metastasis. Thus, disentangling the function of Rreb1 in development should shed light on its role in cancer and other diseases involving loss of epithelial integrity

A(s) Guerra(s) dos Meninos: Uma Adaptação, Vários Sentidos

    Get PDF
