Trigonometric Parallaxes of Massive Star Forming Regions: II. Cep A & NGC 7538
We report trigonometric parallaxes for the sources NGC 7538 and Cep A,
corresponding to distances of 2.65 [+0.12/-0.11] kpc and 0.70 [+0.04/-0.04]
kpc, respectively. The distance to NGC 7538 is considerably smaller than its
kinematic distance and places it in the Perseus spiral arm. The distance to Cep
A is also smaller than its kinematic distance and places it in the Local arm or
spur. Combining the distance and proper motions with observed radial velocities
gives the location and full space motion of the star forming regions. We find
significant deviations from circular Galactic orbits for these sources: both
sources show large peculiar motions (> 10 km/s) counter to Galactic rotation

Anti-inflammatory, pro-proliferative and antimicrobial potential of the compounds isolated from Daemonorops draco (Willd.) Blume
Ethno-pharmacological relevance: Daemonorops draco (D. draco) commonly known as âDragon's bloodâ is one of the most used plants by Momok, Anak Dalam and Talang Mamak tribes from Indonesia as a remedy for wound healing. Aim of the study: This study aimed to identify the extract, fractions and compounds responsible for the anti-inflammatory and pro-proliferative activities of the D. draco resin. Additionally, the antimicrobial activity against two bacteria and one yeast species was analysed. Materials and methods: Bio-guided isolation of compounds with anti-inflammatory, pro-proliferative and antimicrobial activities from the D. draco resin was carried out by measuring: the inhibition of NF-ÎșB and activation of Nrf2 in THP-1, HaCaT, NIH-3T3 cells; cell proliferation in NIH-3T3 and HaCaT cells; and the antimicrobial effect on E. coli, S. aureus and C. albicans. Results: Guided isolation by bioassay gave rise to the isolation and characterisation by nuclear magnetic resonance and mass spectrometry of three compounds: 1 (Bexarotene), 2 (Taspine) and 3 (2-hydroxy-1-naphthaldehyde isonicotinoyl hydrazone). All compounds showed NF-ÎșB inhibitory activity with IC50 values of 0.10â0.13, 0.22â0.24 and 3.75â4.78 ÎŒM, respectively, while the positive control, Celastrol, had an IC50 of 7.96 ÎŒM. Likewise, all compounds showed an activating effect of Nrf2 with EC50 values of 5.34â5.43, 163.20â169.20 and 300.82â315.56 nM, respectively, while the positive control, CDDO-Me, had an EC50 of 0.11 nM. In addition, concerning the pro-proliferative activity, compound 1 (IC50 = 8.62â8.71 nM) showed a capacity of 100%, compound 2 (IC50 = 166â171 nM) showed a capacity of 75%, and compound 3 (IC50 = 469â486 nM) showed a capacity of 65%, while FSB 10% (positive control) had a pro-proliferative activity of 100% in the NIH3T3 cell lines (fibroblasts) and HaCaT (keratinocytes). Finally, all the compounds showed antimicrobial activity with MIC values of 0.12â0.16, 0.31â0.39 and 3.96â3.99 ÎŒM, respectively, in S. aureus, E. coli and C. albicans strains, while the positive control, Ofloxacin, had a MIC of 27.65 ÎŒM. Chandra Study of the Cepheus B Star Forming Region: Stellar Populations and the Initial Mass Function
Cepheus B (Cep B) molecular cloud and a portion of the nearby Cep OB3b OB
association, one of the most active regions of star formation within 1 kpc, has
been observed with the ACIS detector on board the Chandra X-ray Observatory. We
detect 431 X-ray sources, of which 89% are confidently identified as clustered
pre-main sequence stars. Two main results are obtained. First, we provide the
best census to date for the stellar population of the region. We identify many
members of two rich stellar clusters: the lightly obscured Cep OB3b
association, and the deeply embedded cluster in Cep B whose existence was
previously traced only by a handful of radio sources and T Tauri stars. Second,
we find a discrepancy between the X-ray Luminosity Functions of the Cep OB3b
and the Orion Nebula Cluster. This may be due to different Initial Mass
Functions of two regions (excess of ~0.3 solar mass stars), or different age
distributions. Several other results are obtained. A diffuse X-ray component
seen in the field is attributed to the integrated emission of unresolved low
mass PMS stars. The X-ray emission from HD 217086 (O7n), the principle ionizing
source of the region, follows the standard model involving many small shocks in
an unmagnetized radiatively accelerated wind. The X-ray source #294 joins a
number of similar superflare PMS stars where long magnetic structures may

Quantifying the importance and location of SARS-CoV-2 transmission events in large metropolitan areas
Detailed characterization of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) transmission across different settings can help design less disruptive interventions. We used real-time, privacy-enhanced mobility data in the New York City, NY and Seattle, WA metropolitan areas to build a detailed agent-based model of SARS-CoV-2 infection to estimate the where, when, and magnitude of transmission events during the pandemicâs first wave. We estimate that only 18% of individuals produce most infections (80%), with about 10% of events that can be considered superspreading events (SSEs). Although mass gatherings present an important risk for SSEs, we estimate that the bulk of transmission occurred in smaller events in settings like workplaces, grocery stores, or food venues. The places most important for transmission change during the pandemic and are different across cities, signaling the large underlying behavioral component underneath them. Our modeling complements case studies and epidemiological data and indicates that real-time tracking of transmission events could help evaluate and define targeted mitigation policies. Perrault syndrome with neurological features in a compound heterozygote for two TWNK mutations : Overlap of TWNK-related recessive disorders

In some cases, patients present with a diversity of neurological signs. To date, mutations in six genes are known to cause Perrault syndrome, but they do not explain all clinically-diagnosed cases. In addition, the number of reported cases and the spectra of mutations are still small to establish conclusive genotype-phenotype correlations. Methods: Affected siblings from family SH19, who presented with features that were suggestive of Perrault syndrome, were subjected to audiological, neurological and gynecological examination. The genetic study included genotyping and haplotype analysis for microsatellite markers close to the genes involved in Perrault syndrome, whole-exome sequencing, and Sanger sequencing of the coding region of the TWNK gene. Results: Three siblings from family SH19 shared similar clinical features: childhood-onset bilateral sensorineural hearing impairment, which progressed to profound deafness in the second decade of life; neurological signs (spinocerebellar ataxia, polyneuropathy), with onset in the fourth decade of life in the two females and at age 20 years in the male; gonadal dysfunction with early cessation of menses in the two females. The genetic study revealed two compound heterozygous pathogenic mutations in the TWNK gene in the three affected subjects: c.85C>T (p.Arg29â), previously reported in a case of hepatocerebral syndrome; and a novel missense mutation, c.1886C>T (p.Ser629Phe). Mutations segregated in the family according to an autosomal recessive inheritance pattern. Conclusions: Our results further illustrate the utility of genetic testing as a tool to confirm a tentative clinical diagnosis of Perrault syndrome. Studies on genotype-phenotype correlation from the hitherto reported cases indicate that patients with Perrault syndrome caused by TWNK mutations will manifest neurological signs in adulthood. Molecular and clinical characterization of novel cases of recessive disorders caused by TWNK mutations is strongly needed to get further insight into the genotype-phenotype correlations of a phenotypic continuum encompassing Perrault syndrome, infantile-onset spinocerebellar ataxia, and hepatocerebral syndrome
Cloud Structure and Physical Conditions in Star-Forming Regions from Optical Observations. II. Analysis
To complement the optical absorption-line survey of diffuse molecular gas in
Paper I, we obtained and analyzed far ultraviolet H and CO data on lines of
sight toward stars in Cep OB2 and Cep OB3. Possible correlations between column
densities of different species for individual velocity components, not total
columns along a line of sight as in the past, were examined and were
interpreted in terms of cloud structure. The analysis reveals that there are
two kinds of CH in diffuse molecular gas: CN-like CH and CH-like CH.
Evidence is provided that CO is also associated with CN in diffuse molecular
clouds. Different species are distributed according to gas density in the
diffuse molecular gas. Both calcium and potassium may be depleted onto grains
in high density gas, but with different dependences on local gas density. Gas
densities for components where CN was detected were inferred from a chemical
model. Analysis of cloud structure indicates that our data are generally
consistent with the large-scale structure suggested by maps of CO
millimeter-wave emission. On small scales, the gas density is seen to vary by
factors greater than 5.0 over scales of 10,000 AU. The relationships
between column densities of CO and CH with that of H along a line of sight
show similar slopes for the gas toward Cep OB2 and OB3, but the CO/H and
CH/H ratios tend to differ which we ascribe to variation in average density

Evaluating the Quality of Research into a Single Prognostic Biomarker: A Systematic Review and Meta-analysis of 83 Studies of C-Reactive Protein in Stable Coronary Artery Disease
Background
Systematic evaluations of the quality of research on a single prognostic biomarker are rare. We sought to evaluate the quality of prognostic research evidence for the association of C-reactive protein (CRP) with fatal and nonfatal events among patients with stable coronary disease.
Methods and Findings
We searched MEDLINE (1966 to 2009) and EMBASE (1980 to 2009) and selected prospective studies of patients with stable coronary disease, reporting a relative risk for the association of CRP with death and nonfatal cardiovascular events. We included 83 studies, reporting 61,684 patients and 6,485 outcome events. No study reported a prespecified statistical analysis protocol; only two studies reported the time elapsed (in months or years) between initial presentation of symptomatic coronary disease and inclusion in the study. Studies reported a median of seven items (of 17) from the REMARK reporting guidelines, with no evidence of change over time.
The pooled relative risk for the top versus bottom third of CRP distribution was 1.97 (95% confidence interval [CI] 1.78â2.17), with substantial heterogeneity (I2 = 79.5). Only 13 studies adjusted for conventional risk factors (age, sex, smoking, obesity, diabetes, and low-density lipoprotein [LDL] cholesterol) and these had a relative risk of 1.65 (95% CI 1.39â1.96), I2 = 33.7. Studies reported ten different ways of comparing CRP values, with weaker relative risks for those based on continuous measures. Adjusting for publication bias (for which there was strong evidence, Egger's p<0.001) using a validated method reduced the relative risk to 1.19 (95% CI 1.13â1.25). Only two studies reported a measure of discrimination (c-statistic). In 20 studies the detection rate for subsequent events could be calculated and was 31% for a 10% false positive rate, and the calculated pooled c-statistic was 0.61 (0.57â0.66).
Conclusion
Multiple types of reporting bias, and publication bias, make the magnitude of any independent association between CRP and prognosis among patients with stable coronary disease sufficiently uncertain that no clinical practice recommendations can be made. Publication of prespecified statistical analytic protocols and prospective registration of studies, among other measures, might help improve the quality of prognostic biomarker research
