    Experimental and analytical study of cracks under biaxial fatigue

    Most mechanical components experience multi-axial cyclic loading conditions during service. Experimental analysis of fatigue cracks under such conditions is not easy and most works tend to focus more on the simpler but less realistic case of uni-axial loading. Consequently, there are many uncertainties related to the load sequence effect that are now well known and are not normally incorporated into the growth models. The current work presents a new methodology for evaluating overload effect in biaxial fatigue cracks. The methodology includes evaluation of mixed-mode (KI and KII) stress intensity factor and the Crack Opening Displacement for samples with and without overload cycle under biaxial loading. The methodology is tested under a range of crack lengths. All crack-tip information is obtained with a hybrid methodology that combines experimental full-field digital image correlation data and Williams' elastic model describing the crack-tip field.Universidad de Málaga.     Some experimental observations of crack-tip mechanics with displacement data

    Estudio de la mecánica en el vértice de la grieta mediante datos de desplazamiento.In the past two decades, crack-tip mechanics has been increasingly studied with full-field techniques. Within these techniques, Digital Image Correlation (DIC) has been most widely used due to its many advantages, to extract important crack-tip information, including Stress Intensity Factor (SIF), Crack Opening Displacement, J-integral, T-stress, closure level, plastic zone size, etc. However, little information is given in the literature about the experimental setup that provides best estimations for the different parameters. The current work aims at understanding how the experimental conditions used in DIC influence the crack-tip information extracted experimentally. The influence of parameters such as magnification factor, size of the images, position of the images with respect the crack-tip and size of the subset used in the correlation is studied. The influence is studied in terms of SIF and T-stress by using Williams’ model. The concept of determination of the K-dominance zone from experimental data has also explored. In this regard, cyclic loading on a fatigue crack in a compact tension (CT) specimen, made of aluminium 2024-T351 alloy, has been applied and the surface deformation ahead of the crack tip has been examined. The comparison between theoretical and experimental values of KI showed that the effect of subset size on the measured KI is negligible compared to the effect of size of the image.Universidad de Málaga.     Nonclassicality and criticality in symmetry-protected magnetic phases

    Quantum and global discord in a spin-1 Heisenberg chain subject to single-ion anisotropy (uniaxial field) are studied using exact diagonalisation and the density matrix renormalisation group (DMRG). We find that these measures of quantum nonclassicality are able to detect the quantum phase transitions confining the symmetry protected Haldane phase and show critical scaling with universal exponents. Moreover, in the case of thermal states, we find that quantum discord can increase with increasing temperature.Comment: 7 pages, 6 figures, Close to published version.     Economic Fluctuations, Child Mortality and Policy Considerations in the Least Developed Countries

    Between 1990 and 2010 child mortality decreased in general terms in the Least Developed Countries (LDCs), although the differences between countries over time are significant. This paper examines the relationship between short-term economic fluctuations and changes in child mortality in the LDCs during the period 1990-2010. Unlike other studies, we consider a large group of LDCs and provide empirical evidence of the asymmetrical effects of variations in Gross Domestic Product per capita on the evolution of child mortality rate in periods of economic recession and expansion. The significance of said effects diminishes when other relevant socio-economic control variables are considered, and some development policy considerations are addressed in order to achieve the Millennium Development Goal 4 target

    Regionalization and connectivity in the Gulf of Cádiz and Alborán Sea implication for marine spatial planning and coastal management

    This work proposes a regionalization of marine waters south of the Iberian Peninsula based on empirical orthogonal functions and GIS analysis. The regions identified are consistent and correspond to hydrological mesoscale and macroscale structures well characterized from physical and biological viewpoints. Although it is difficult to determine the boundaries of a fluid continuously moving, the procedure used recognizes the most frequent patterns and can be used to reference geographically the most likely position of the limit among the regions. Once established the regions, the connectivity among them is addressed by means of geostrophic currents derived from altimetry data. The main geostrophic circulation pattern depict an elevate connectivity in the area that might facilitate the conservation and recovering of species, but also imply a higher vulnerability to negative externalities and highlight the need of models, marine spatial planning, and coastal management approaches that includes the pelagic ecosystem and connectivity of the seas. Furthermore, the results confirm that the cooperation between public authorities at different levels (local, regional, state) as well as among riparian countries is essential for marine spatial planning and prevention of potential risk derived from upcoming marine activities in the framework of the 2020 agenda and Blue Growth strategy.Universidad de Málaga.     Ice morphology modification and solute recovery improvement by heating and annealing during block freeze-concentration of coffee extracts

    Several treatments on ice blocks can be applied during block freeze-concentration to increase the solute recovery from the ice. In the present study, the changes in the ice block’s temperature and the application of annealing during the block freeze-concentration of aqueous coffee extracts were studied. The ice block was subjected to heating and annealing prior to the thawing stage. The effect of coolant temperature during ice block heating (T = -10 and -5 °C) and the application of annealing (+, -) on solute recovery and ice structure morphology was evaluated. The use of annealing during block freeze-concentration modified the ice crystal morphology and increased the solute recovery only when it is applied at the highest temperature. In general, the annealing process increased the size and circularity of the ice crystals, consequently improving the solute recovery. Thus, annealing can be used to increase the solute recovery during block freeze-concentration.Postprint (published version

    QTL analysis of high thermotolerance with superior and downgraded parental yeast strains reveals new minor QTLs and converges on novel causative alleles involved in RNA processing

    Revealing QTLs with a minor effect in complex traits remains difficult. Initial strategies had limited success because of interference by major QTLs and epistasis. New strategies focused on eliminating major QTLs in subsequent mapping experiments. Since genetic analysis of superior segregants from natural diploid strains usually also reveals QTLs linked to the inferior parent, we have extended this strategy for minor QTL identification by eliminating QTLs in both parent strains and repeating the QTL mapping with pooled-segregant whole-genome sequence analysis. We first mapped multiple QTLs responsible for high thermotolerance in a natural yeast strain, MUCL28177, compared to the laboratory strain, BY4742. Using single and bulk reciprocal hemizygosity analysis we identified MKT1 and PRP42 as causative genes in QTLs linked to the superior and inferior parent, respectively. We subsequently downgraded both parents by replacing their superior allele with the inferior allele of the other parent. QTL mapping using pooled-segregant whole-genome sequence analysis with the segregants from the cross of the downgraded parents, revealed several new QTLs. We validated the two most-strongly linked new QTLs by identifying NCS2 and SMD2 as causative genes linked to the superior downgraded parent and we found an allele-specific epistatic interaction between PRP42 and SMD2. Interestingly, the related function of PRP42 and SMD2 suggests an important role for RNA processing in high thermotolerance and underscores the relevance of analyzing minor QTLs. Our results show that identification of minor QTLs involved in complex traits can be successfully accomplished by crossing parent strains that have both been downgraded for a single QTL. This novel approach has the advantage of maintaining all relevant genetic diversity as well as enough phenotypic difference between the parent strains for the trait-of-interest and thus maximizes the chances of successfully identifying additional minor QTLs that are relevant for the phenotypic difference between the original parents

    Validation of suitable internal control genes for expression studies in aging.

    Quantitative data from experiments of gene expression are often normalized through levels of housekeeping genes transcription by assuming that expression of these genes is highly uniform. This practice is being questioned as it becomes increasingly clear that the level of housekeeping genes expression may vary considerably in certain biological samples. To date, the validation of reference genes in aging has received little attention and suitable reference genes have not yet been defined. Our aim was to evaluate the expression stability of frequently used reference genes in human peripheral blood mononuclear cells with respect to aging. Using quantitative RT-PCR, we carried out an extensive evaluation of five housekeeping genes, i.e. 18s rRNA, ACTB, GAPDH, HPRT1 and GUSB, for stability of expression in samples from donors in the age range 35-74 years. The consistency in the expression stability was quantified on the basis of the coefficient of variation and two algorithms termed geNorm and NormFinder. Our results indicated GUSB be the most suitable transcript and 18s the least for accurate normalization in PBMCs. We also demonstrated that aging is a confounding factor with respect to stability of 18s, HPRT1 and ACTB expression, which were particularly prone to variability in aged donors
