    Mimicking sarcolemmal damage in vitro: a contractile 3D model of skeletal muscle for drug testing in Duchenne muscular dystrophy

    Duchenne muscular dystrophy (DMD) is the most prevalent neuromuscular disease diagnosed in childhood. It is a progressive and wasting disease, characterized by a degeneration of skeletal and cardiac muscles caused by the lack of dystrophin protein. The absence of this crucial structural protein leads to sarcolemmal fragility, resulting in muscle fiber damage during contraction. Despite ongoing efforts, there is no cure available for DMD patients. One of the primary challenges is the limited efficacy of current preclinical tools, which fail in modeling the biological complexity of the disease. Human-based three-dimensional (3D) cell culture methods appear as a novel approach to accelerate preclinical research by enhancing the reproduction of pathophysiological processes in skeletal muscle. In this work, we developed a patient-derived functional 3D skeletal muscle model of DMD that reproduces the sarcolemmal damage found in the native DMD muscle. These bioengineered skeletal muscle tissues exhibit contractile functionality, as they responded to electrical pulse stimulation. Sustained contractile regimes induced the loss of myotube integrity, mirroring the pathological myotube breakdown inherent in DMD due to sarcolemmal instability. Moreover, damaged DMD tissues showed disease functional phenotypes, such as tetanic fatigue. We also evaluated the therapeutic effect of utrophin upregulator drug candidates on the functionality of the skeletal muscle tissues, thus providing deeper insight into the real impact of these treatments. Overall, our findings underscore the potential of bioengineered 3D skeletal muscle technology to advance DMD research and facilitate the development of novel therapies for DMD and related neuromuscular disorders

    Influenza vaccination among multiple sclerosis patients during the COVID-19 pandemic

    In the context of the COVID-19 pandemic, the co-circulation of influenza and SARS-CoV-2 viruses may have severe complications for vulnerable populations. For this reason, the World Health Organization pointed to the 2020–2021 anti-influenza campaign as being of special relevance. Our aim was to assess the 2020–2021 influenza vaccination coverage, and its associated factors, among patients in a Spanish multiple sclerosis (MS) unit. A cross–sectional study was conducted. People attending the MS unit of the Clinical Hospital of Zaragoza during 2020 were included. Variables were obtained by reviewing records. Associations with 2020–2021 influenza vaccination were analyzed using bivariate analysis and a multiple logistic regression model. A total of 302 patients were studied; 62.6% were women, whose mean age (standard deviation) was 47.3 (11.5) years. The 2020–2021 influenza vaccination coverage was 55.3% (59.8% in women and 47.8% in men). A total of 89.7% had at least one other indication for vaccination (e.g., immunosuppressive treatment in 225 patients). The variables associated with getting vaccinated were being female (adjusted odds ratio (95% confidence interval) (aOR (95%CI) = 2.12 (1.12–3.99)), having received the 2019–2020 influenza vaccine (aOR (95%CI) = 31.82 (14.71–68.86)) and being born in Spain (aOR (95%CI) = 12.91 (1.07–156.28)). Coverage is moderate compared to other countries. It is necessary to develop strategies to improve it, especially in men and those born outside Spain

    Influenza vaccination coverage among multiple sclerosis patients: evolution over time and associated factors

    Our objective was to determine the influenza vaccination rate in a Spanish cohort of multiple sclerosis (MS) patients. A retrospective cohort study was carried out. Patients who attended the MS unit of the Lozano Blesa Hospital of Zaragoza between January 2015 and 2020 were included. The variables were obtained by reviewing the specialized and primary care records. Associations between receiving the vaccine in each flu season and the other variables were analyzed using bivariate analysis and multiple logistic regression models. A total of 260 patients were studied, with a median age of 31 years at the time of diagnosis. A total of 62.3% (162/260) were women. Vaccination coverage ranged from 20.4% in the 2015–2016 and 2016–2017 seasons to 41.5% in the 2019–2020 season (p = 0.000). Having been vaccinated in the previous season (ORa: 16.47–390.22; p = 0.000) and receiving a vaccination recommendation from the hospital vaccination unit (ORa: 2.44–3.96; p < 0.009) were associated with being vaccinated. The coverage is in an intermediate position compared to other countries. It is necessary to improve the referral system of these patients to the hospital vaccination unit because the information obtained by this service contributed to higher vaccination rates

    We have tried to establish a relationship between the raw material and the mechanical properties of ceramic pieces subjected to different firing processes, so under the determinant of the thermal process we have done a follow up on the petrogenetic process that the manufacture of ceramics represents. This is evaluated in terms of the mechanical behaviour of the ceramic tile bodies, which have been normalised and submitted to tests for flexion. The principal objective is to be able to indicate what the optimum firing temperature for each sample is, and to determine what it is in the ceramic body which offers the greatest resistance when a specific force is applied. The study of the texture of the ceramic body will be considered mainly in relation to the existent porosity, as well as its distribution, which conditions its characteristics and specifications. The relation between porosity and the parameters that describe the porous texture of the ceramic tile pieces studied is complicated because of the fact that these have many pores with a complex and irregular spatial disposition, with a broad distribution of forms and sizes, and as such they do not fit into established empirical equations.Se ha tratado de establecer la relación entre materia prima y propiedades mecánicas de piezas cerámicas sometidas a procesos de cocción diversos. Bajo el condicionante del proceso térmico se realiza un seguimiento del proceso petrogenético que representa la fabricación cerámica y se evalúa a través del comportamiento mecánico de probetas cerámicas normalizadas y sometidas a ensayos de flexión. El objetivo principal es poder indicar cuál es la temperatura óptima de cocción para cada muestra y determinar cuál es aquélla en que la probeta cerámica ofrece una mayor resistencia al aplicarle una determinada carga. El estudio de la textura de la matriz cerámica es de gran interés en relación con la porosidad, asi como su distribución, que condiciona sus características y aplicaciones. La relación existente entre la porosidad y los parámetros relacionados con la textura porosa es muy compleja debido a la distribución irregular de formas y tamaños de poro, que no se ajusta a ninguna ecuación empírica establecida, pero aun así hay aproximaciones teóricas que pueden permitir prevenir la relación entre la porosidad y las propiedades mecánicas

    L'article exposa els resultats del projecte del mateix nom que va obtenir un ajut en el marc de la Convocatòria REDICE-10. El projecte s'ha proposat avançar en la implantació de la competència transversal de treball en equip en el Grau de Gestió i Administració Publica, després d'una experiència inicial en la qual s'havia adoptat una definició de la competència i s'havia planificat el seu desplegament de manera seqüencial. L'objectiu principal del projecte ha estat construir instruments d'avaluació de la competència i comprovar-ne l'efectivitat a partir de la seva aplicació en diferents assignatures del grau. L'article descriu el procés seguit per construir uns qüestionaris d'autoavaluació de la competència adreçats a l'alumnat i analitza la informació recollida amb el propòsit de valorar seva utilitat com a eina d'avaluació

    Theoretical modelling of quaternary GaInAsSb/GaAs self-assembled quantum dots

    Trabajo presentado al "Quantum Dot", celebrado en Nottingham (UK) del 26 al 30 de Abril de 2010.InAs/GaAs quantum dots exposed to Sb after growth exhibit spectral changes. We study in the present paper an idealized nanostructure consisting of a homogeneous distribution of the quaternary GaInAsSb surrounded by a barrier of GaAs. We nd that the valence band o set is a critical parameter in modelling its electronic structure. Depending on this value, we predict a transition from type-I to type-II band alignment at a di erent Sb concentration.
