The proposal of the Experimental Pedagogical School (eps) in the environmental field revisits several ideas of Gunter Pauli (2011), exposed in his book The Blue Economy. There are some similarities between the two proposals; they highlight the need to transform the traditional view of education (focused on contents so far), on the one hand, and to seek solutions to local problems, on the other. The objectives are: a) to transform the vicious circle that currently feeds the environmental crisis, b) to question the principles of the economy based on market transactions as a closed system, c) to go beyond the development of environmental actions linked to the production of biodegradable elements, and d) to build sustainable action frameworks based on our knowledge of natural systems, as well as on the dialogue between knowledges. In that sense, the commitment is to the formation of citizens who are responsible with the environment andare not mere consumers of it; for this effect, a profound transformation of the educational system is necessary, one which would value not only the need to think about the problems of context, but also the importance of educating citizens who question the processes of cultural and biological homogenization that prevail today. This article offers reflections and describes the experience of the eps, from the time it revisited the ideas of the blue economy to enrich its proposal and to consolidate nine environmental projects (through the teaching strategy of ata), which have resulted from the study of real problems present in the daily life of the school community. A proposta da Escola Pedagógica Experimental (epe) no âmbito ambiental retoma várias abordagens deGunter Pauli (2011), presentes no seu livro Economia azul. Somatic SNCA Copy Number Variants in Multiple System Atrophy Are Related to Pathology and Inclusions

    Get PDF
BACKGROUND: Somatic α-synuclein (SNCA) copy number variants (CNVs, specifically gains) occur in multiple system atrophy (MSA) and Parkinson's disease brains. OBJECTIVE: The aim was to compare somatic SNCA CNVs in MSA subtypes (striatonigral degeneration [SND] and olivopontocerebellar atrophy [OPCA]) and correlate with inclusions. METHODS: We combined fluorescent in situ hybridization with immunofluorescence for α-synuclein and in some cases oligodendrocyte marker tubulin polymerization promoting protein (TPPP). RESULTS: We analyzed one to three brain regions from 24 MSA cases (13 SND, 11 OPCA). In a region preferentially affected in one subtype (putamen in SND, cerebellum in OPCA), mosaicism was higher in that subtype, and cells with CNVs were 4.2 times more likely to have inclusions. In the substantia nigra, nonpigmented cells with CNVs and TPPP were about six times more likely to have inclusions. CONCLUSIONS: The correlation between SNCA CNVs and pathology (at a regional level) and inclusions (at a single-cell level) suggests a role for somatic SNCA CNVs in MSA pathogenesis. © 2022 The Authors. Diseño de un plan de mejora de cultura organizacional para la Empresa Punto Sabroso ubicada en la ciudad de Cali Valle del Cauca en el segundo semestre de 2021

    Get PDF
This degree project seeks to identify the type of organizational culture present in the selected company, in order to design an improvement plan to define the parameters that the organization's managers can consider applying to manage those social, personal and work elements that require be cared for at the organizational level, that allow meeting the needs that constitute well-being and job satisfaction in employees, and that represent productive results and consequently economic results for the company. For this reason, it is proposed to know the main weaknesses that the company presents in relation to the organizational culture, to know if the staff is satisfied or not with different aspects that frame the kind of culture that has been established, such as: the work environment , the incentive system, training program, communication effectiveness, leadership, teamwork, recognition, career plan, among others. To do this, a qualitative-quantitative analysis is proposed in which a survey was carried out in which 30 employees of the company Punto Sabroso participated, of which 23 are women and 7 are men. The results show us a general acceptance in what corresponds to the organizational culture adapted by the company, in the same way the population that presents a level of dissatisfaction is identified according to the categories into which the survey was divided, which correspond to commitment and support, work environment and recognition, finding between 6 to 10 dissatisfied people according to the parameters evaluated

Increased Matrix Metalloproteinase (MMPs) Levels Do Not Predict Disease Severity or Progression in Emphysema

Rationale: Though matrix metalloproteinases (MMPs) are critical in the pathogenesis of COPD, their utility as a disease biomarker remains uncertain. This study aimed to determine whether bronchoalveolar lavage (BALF) or plasma MMP measurements correlated with disease severity or functional decline in emphysema. Methods: Enzyme-linked immunosorbent assay and luminex assays measured MMP-1, -9, -12 and tissue inhibitor of matrix metalloproteinase-1 in the BALF and plasma of non-smokers, smokers with normal lung function and moderate-to-severe emphysema subjects. In the cohort of 101 emphysema subjects correlative analyses were done to determine if MMP or TIMP-1 levels were associated with key disease parameters or change in lung function over an 18-month time period. Main Results: Compared to non-smoking controls, MMP and TIMP-1 BALF levels were significantly elevated in the emphysema cohort. Though MMP-1 was elevated in both the normal smoker and emphysema groups, collagenase activity was only increased in the emphysema subjects. In contrast to BALF, plasma MMP-9 and TIMP-1 levels were actually decreased in the emphysema cohort compared to the control groups. Both in the BALF and plasma, MMP and TIMP-1 measurements in the emphysema subjects did not correlate with important disease parameters and were not predictive of subsequent functional decline. Conclusions: MMPs are altered in the BALF and plasma of emphysema; however, the changes in MMPs correlate poorly with parameters of disease intensity or progression. Though MMPs are pivotal in the pathogenesis of COPD, these findings suggest that measuring MMPs will have limited utility as a prognostic marker in this disease. © 2013 D'Armiento et al

Discordance between patient and physician global assessment of disease activity in Behçet's syndrome: a multicenter study cohort

    Get PDF
Background: To compare the patients' and physician's global assessment of disease activity in Behçet's syndrome (BS) and investigate the frequency, magnitude, and determinants of potential discordance. Methods: A total of 226 adult BS patients with a median (IQR) age of 46.9 (35.6-55.2) years were enrolled across Italy, Greece, Portugal, and Spain. Demographic, clinical, and therapeutic variables, as well as the patient reported outcomes, were collected at the recruitment visit. The physical (PCS) and mental (MCS) component summary scores of the Short Form Questionnaire 36 (SF-36) and the Behçet's syndrome Overall Damage Index (BODI) were calculated. Disease activity was assessed by the patients' (PtGA) and physician's global assessment (PGA) in a 10-cm visual analog scale, as well as the Behçet Disease Current Activity Form (BDCAF). Discordance (∆) was calculated by subtracting the PGA from the PtGA and defined as positive (PtGA>PGA) and negative (PtGA 80%) of disagreements were due to patients rating higher their disease activity. Higher values of BDCAF were associated to increased rate of positive discordance. When BDCAF = 0, the median (IQR) values of PtGA and PGA were 0.2 (0-2) and 0 (0-1), respectively. PCS (adjusted odds ratio (adjOR) 0.96 per unit, 95% CI 0.93-0.98, p = 0.006) and MCS (adjOR 0.96 per unit, 95% CI 0.93-0.99, p = 0.003) were independently associated with positive discordance using both cutoffs. Active ocular involvement emerged as a potential determinant of negative discordance (adjOR 5.88, 95% CI 1.48-23.30, p = 0.012). Conclusions: PtGA and PGA should be considered as complementary measures in BS, as patients and physicians may be influenced by different factors when assessing active disease manifestations. Particularly, PtGA may be a useful tool in the assessment of BS disease activity, as it carries a low risk to misclassify an inactive disease, and may allow to capture aspects of the patient's health that negatively affect his well-being and the treatment

Engineering DNA-Grafted Quatsomes as Stable Nucleic Acid-Responsive Fluorescent Nanovesicles

    Get PDF
The development of artificial vesicles into responsive architectures capable of sensing the biological environment and simultaneously signaling the presence of a specific target molecule is a key challenge in a range of biomedical applications from drug delivery to diagnostic tools. Herein, the rational design of biomimetic DNA-grafted quatsome (QS) nanovesicles capable of translating the binding of a target molecule to amphiphilic DNA probes into an optical output is presented. QSs are synthetic lipid-based nanovesicles able to confine multiple organic dyes at the nanoscale, resulting in ultra-bright soft materials with attractiveness for sensing applications. Dye-loaded QS nanovesicles of different composition and surface charge are grafted with fluorescent amphiphilic nucleic acid-based probes to produce programmable FRET-active nanovesicles that operate as highly sensitive signal transducers. The photophysical properties of the DNA-grafted nanovesicles are characterized and the highly selective, ratiometric detection of clinically relevant microRNAs with sensitivity in the low nanomolar range are demonstrated. The potential applications of responsive QS nanovesicles for biosensing applications but also as functional nanodevices for targeted biomedical applications is envisaged. This work was financially supported by the European Union’s Horizon 2020 research and innovation programme under the Marie Skłodowska- Curie grant agreement “Nano-Oligo Med” (No 778133), Ministry of Science and Innovation (MINECO), Spain, through the “MOL4BIO” project (PID2019-105622RB-I00) and by Instituto de Salud Carlos III (DTS20/00018), Italian Ministry of University and Research (Project of National Interest, PRIN, 2017Y2PAB8_004 through the project “Cutting Edge Analytical Chemistry Methodologies and Bio-Tools to Boost Precision Medicine in Hormone-Related Diseases”.
