Cationic fluorescence probes in a polyanion-surfactant system.

V této bakalářské práci byly studovány vlastnosti vodných roztoků hyaluronanu a cetyltrimethylamonium bromidu pomocí fluorescenční sondy akridinové oranže. Nejprve byla zkoumána interakce akridinové oranže a hyaluronanu. Bylo sledováno emisní a absorpční spektrum akridinové oranže. Bylo zjištěno, že na karboxylových skupinách hyaluronanu docházelo k tvorbě dimerů akridinové oranže. Dále byla studována interakce cetyltrimethyl amonium bromidu se systémem akridinová oranž – hyaluronan. Bylo zjištěno, že kationaktivní tenzid vytlačoval dimery akridinové oranže a sám se vázal na karboxylové skupiny hyaluronanu. Tím došlo k rozpadu dimeru akridinové oranže na monomery. Důvodem, proč byla tato práce zaměřena na studování vlastností vodných roztoků hyaluronanu s kationaktivním tenzidem je ten, že systém hyaluronan – tenzid by mohl být přínosem pro výzkum nosičových systémů pro cílenou distribuci léčiv.In this bachelor thesis were studied properties of aqueous solutions of hyaluronan and celytrimethylamonnium bromid with help of acridine orange as fluorescence probe. At first, this thesis was focused on interactions of acridine orange and hyaluronan. Emision and excitation spectra of acridine orange were observed. Result of these measurement was bonding of acridine orange dimer to carboxylic group of hyaluronan. Next studied interaction was interaction of cetyltrimethyl amonium bromid with acridine orange – hyaluronan system. Result of these interaction was, that molecules of cationactive surfactant pushed away dimers of acridine orange and took place of dimer of acridine orange on carboxylic group of hyaluronan. Dimer of acridine orange was changed into monomers. The reason why this thesis was focused on study of aqueus solutions of hyaluronan with cationactive surfactant is benefit of hyaluronan – surfactant system in research of transport system for target distribution of drugs.

The EU Digital Single Market

Bakalářská práce definuje jednotný digitální trh EU a vysvětluje jeho význam. Identifikuje významné mezníky, kterých bylo dosaženo při vytváření jednotného digitálního trhu a hodnotí plnění vybraných cílů. Bakalářská práce je členěna na čtyři kapitoly. První kapitola definuje základní termíny. Druhá kapitola je zaměřena na charakteristiku procesu vytváření jednotného digitálního trhu. Třetí kapitola se zabývá plněním vybraných cílů JDT v EU a ČR. Poslední kapitola je zaměřena na zhodnocení významu JDT pro EU a ČR ve vybraných oblastech. Hodnocení pokroku v digitalizaci je provedeno pomocí indexů DESI a EDI. Z hodnocení vyplývá, že ČR má ještě mnoho prostoru k zlepšení, především v dostupnosti internetu a digitálního provozu veřejné správy.ObhájenoThe bachelor thesis defines the EU digital single market and explains its importance. It identifies important milestones that have been reached in the creation of the Digital Single Market and assesses the fulfillment of selected objectives. The bachelor thesis is divided into four chapters. The first chapter defines the basic terms. The second chapter focuses on the characteristics of the process of creating a digital single market. The third chapter deals with the fulfillment of selected goals of the digital single market in the EU and the Czech Republic. The last chapter focuses on the evaluation of the importance of the digital single market for the EU and the Czech Republic in selected areas. Evaluation of progress in digitization is performed using DESI and EDI indices. The evaluation shows that the Czech Republic still has a lot of room for improvement, especially in the availability of the Internet and digital public administration

Brasil: educador ou rentista? Um estudo sobre a dívida pública e a educação nos governos Lula e Dilma

Pedagogia.Trata-se de um estudo de como se configurou as prioridades da política econômica nos governos Lula (2003-2010) e Dilma (2010-2016), no que tange à dívida pública e o orçamento destinado à Educação, objetivando demonstrar o impacto da dívida pública no subfinanciamento da educação. Para isso, fizemos um apanhado histórico e levantamento de dados do orçamento da União e seus os desdobramos. Constatamos que os governos petistas deram sequência a priorização do pagamento da dívida pública como parte central da política orçamentária federal. Concluímos, também, que mesmo dentro do pequeno crescimento do orçamento educacional, se privilegiou a transferência de recursos para o setor privado, ao invés do real investimento no aparelho público.Este estudo se refiere a cómo se configuró las prioridades de la política económica de los gobiernos Lula (2003-2010) y Dilma (2010-2016), con respecto a la Deuda Pública y el presupuesto que les correspondía para la Educación, objetivando demostrar el impacto de la Deuda en el déficit de financiación de la educación. Para eso, nosotros hicimos un recordatorio histórico y levantamiento de los datos del presupuesto de la Unión y sus desdoblamientos. Constatamos que los gobiernos petistas dieran secuencia a la priorización del pago de la Deuda Pública como parte central de la política presupuestaria federal. Asimismo, concluimos que incluso en el crecimiento de lo presupuesto educacional, se ha privilegiado la transferencia de recursos para el sector privado, en lugar de hacer una real inversión en el aparato público

Cílem této práce je navrhnout levný a efektivní způsob, jak vytvořit webové stránky pro živnostníka a jeho zavedenou firmu na prodej obuvi v malém městě. V této práci jsou popsány některé metody současných nástrojů a aplikací pro tvorbu webových prezentací a užití těchto nástrojů pro konkrétní zhotovení internetové prezentace na základě vypracovaných analýz.The aim of this work is to design both cheap and efficient way how to create websites for a tradesman, and his/her established shoe shop in a small town. This work includes descriptions of some trendy tools methods, as well as webpresentations production applications, and theese tools use for particularinternet presentation creating, based on elaborated analyses.

Time-resolved fluorescence of system polymer-surfactant

V diplomové práci byla studována časově-rozlišená fluorescence v systému polymer tenzid. Nejdříve byla studována agregační čísla vybraného kationtového (cetyltrimethylamonium bromid), aniontového (dodecylsíran sodný) a neionogenního (Triton X-100) tenzidu pomocí steady-state a časově rozlišené fluorescence a tyto dvě metody byly srovnány. Bylo zjištěno, že steady-state metoda dává vždy nižší hodnoty agregačních čísel než časově-rozlišená metoda a že steady-state metoda není použitelná pro přesné stanovení agregačních čísel o vysokých hodnotách a ve vzorcích obsahujících rozpuštěný kyslík. Dále byl studován vliv přídavku nativního hyaluronanu k těmto tenzidům. Byla sledována změna agregačního čísla tenzidů a vliv hyaluronanu na procentuální zastoupení dynamického a statického zhášení. Hyaluronan ovlivňoval agregační číslo tenzidů cetyltrimethylamonium bromidu a Tritonu X-100. Dále bylo zjištěno, že hyaluronan zvýšil procentuální zastoupení dynamického zhášení u cetyltrimethylamonium bromidu a u Tritonu X-100. V případě dodecylsíranu sodného byl pyren zhášen efektivní sférou a zastoupení dynamického zhášení bylo v tomto systému zanedbatelné. Hyaluronan neměl žádný vliv na zhášení v dodecylsíranu sodném. Dále byla studována relativní pozice pyrenu v micelách cetyltrimethylamonium bromidu, dodecylsíranu sodného a Tritonu X-100. Bylo zjištěno, že se pozice pyrenu mění s nábojem a strukturou micely. Také bylo zkoumáno, jak se mění procentuální zastoupení dynamického zhášení v micelách s různým nábojem při zhášení jodovými deriváty. Ve všech případech bylo vypočteno většinové zastoupení dynamického zhášení v systému.In this diploma thesis was studied time-resolved fluorescence in polymer-surfactant system. At first aggregation numbers of cationic (cetyltrimethylammonium bromide), anionic (sodium dodecylsulfate) and nonionic (Triton X-100) surfactants were studied by steady-state and time-resolved fluorescence spectroscopy. These two methods were compared. Aggregation numbers by steady-state method were always lower than aggregation numbers measured by time-resolved method. Steady-state method of determination aggregation numbers is useless for surfactants with high aggregation number and for aerated samples. Addition of hyaluronan to surfactant system was studied. There was observed change in aggregation number after addition of hyaluronan and change in percentage of dynamic quenching after addition of hyaluronan. Hyaluronan affected aggregation number of cetyltrimethylammonium bromide and Triton X-100. Hyaluronan increased percentage of dynamic quenching in cetyltrimethylammonium bromide and in Triton X-100. Pyren in sodium dodecylsulfate was quenched by sphere of action with negligible percentage of dynamic quenching and addition of hyaluronan had no effect on quenching. As next goal of this thesis, the determination of the position of fluorescence probe pyrene in cetyltrimethylammonium bromide, sodium dodecylsulfate and Triton X-100 micelles was chosen. Position of pyrene changed with charge and structure of micelles. Next was studied how percentage of dynamic quenching by iodide compounds changes with different charge of micelle. In all cases majority of dynamic quenching was calculated.

The spectrum of surgical procedures in the conditions of pre-hospital emergency care

The bachelor's thesis is focused on surgical procedures in pre-hospital emergency care within solving reversible causes. The bachelor's thesis consists of a theoretical and research part. At the beginning of theoretical part, I deal with surgical procedures that belong to the environment of pre-hospital emergency care and describe their division, indications, risks and practical procedure for their application. I also describe traumatic circulatory arrest and extended cardiopulmonary resuscitation. In other chapters of the work, I deal with patient monitoring and examination, which is important for revealing reversible causes. I discuss the need for analgesia in pre-hospital care, and the last chapter focuses on the importance of maintaining an aseptic environment and risk sepsis. The research part covers qualitative research in the form of a semi-structured interview, while the research team consisted of 9 paramedics from the South Bohemian Region. The aim of the research was to map the indication and risks of surgical procedures in the conditions of pre-hospital emergency care. Another aim was to find out what awareness and experience the paramedics of the South Bohemian Region have in this issue. All research questions were answered and the aims of the thesis were fulfilled. The research found that the experience and knowledge of the paramedics at the various sites are very similar, the paramedics are sufficiently familiar with the issue, while the station in Jindřichův Hradec is objectively slightly better. This work could be used as study material in the education of paramedic students or for the self-education of paramedics
