    Soil Microbial Communities and Antibiotic Resistance in Cattle Farms in the United States: The Importance of Soils in Protecting Environmental Health

Hayes Graduate Research Forum)Cattle farming is a large and evolving industry in the United States (US) with potential health effects in workers, consumers, and individuals living in areas surrounding these operations. Because cattle farming inherently generates a large amount of manure, it is considered a major contributor to antibiotic resistant (AR) genes and bacteria in nearby environments. This study sought to quantify AR genes and pathogens in cattle-associated soils, as well as describe the associated microbial communities. Microbial communities and AR genes were compared in soils within and outside of cattle pens. Soil was sampled from seven cattle farms from different states in the United States. From each farm, one soil sample was taken from within the cattle pens and one sample was taken from outside of the pens. Following DNA extraction, bacterial communities were analyzed via 16S rRNA sequencing. Droplet Digital PCR and real-time PCR were used to quantify three enteric pathogens, three AR genes, and three host-specific microbial source tracking (MST) markers, which were used to identify sources of fecal pollution. The family Peptostreptococcaceae was more abundant in pen soils, and Rhodanobacteraceae was more abundant in non-pen soils. The marker for ruminant fecal contamination (Rum2Bac) was abundant both in pen and non-pen soils, with no significant difference. However, the concentration of the human fecal contamination marker (HF183) was lower in pen soils than non-pen soils. Soils from within cattle pens had higher levels of the pathogens Campylobacter and Salmonella (p<0.05). Cattle pen soils also had higher concentrations of two AR genes (p<0.05), Klebsiella pneumonia Carbapenemase (KPC) and sul1, which offers resistance to sulfonamide antibiotics. This is the first study identifying KPC in livestock-associated soils in the US, an AR gene of concern because it is clinically relevant and spreads easily between bacteria. This study supports increased hazard associated with livestock soils for animal and human health, as well as environmental quality.A one-year embargo was granted for this item

    Long-Period Giant Companions to Three Compact, Multiplanet Systems

    Understanding the relationship between long-period giant planets and multiple smaller short-period planets is critical for formulating a complete picture of planet formation. This work characterizes three such systems. We present Kepler-65, a system with an eccentric (e = 0.28 ± 0.07) giant planet companion discovered via radial velocities (RVs) exterior to a compact, multiply transiting system of sub-Neptune planets. We also use precision RVs to improve mass and radius constraints on two other systems with similar architectures, Kepler-25 and Kepler-68. In Kepler-68 we propose a second exterior giant planet candidate. Finally, we consider the implications of these systems for planet formation models, particularly that the moderate eccentricity in Kepler-65\u27s exterior giant planet did not disrupt its inner system

    Recycling and Composing at the Appalachian Center for Craft

    In summarizing our project, the 2017-2018 ESS Capstone students would like to revamp the existing recycling bins in the Appalachian Center for Craft campus along with introducing composting to the campus. After evaluating the existing recycling bins on the campus, it was decided to offer a new system of color coded bins that the Office of Sustainability could easily provide. This would replace the old bins and offer more of them to residents and other areas of the campus to provide ease of access and more incentive to recycle. Composting was introduced to the Craft Center for their on-site restaurant, The Blue Water Grille, to begin recycling their waste for future use of soil on their own grounds. This would educate residents and staff on composting and provide the campus the opportunity to be more environmentally friendly. Both projects would provide the campus with environmental benefits for almost no cost due to the Office of Sustainability providing all recycling and composting bins

    Phenome-wide Mendelian randomisation analysis of 378,142 cases reveals risk factors for eight common cancers

    For many cancers there are only a few well-established risk factors. Here, we use summary data from genome-wide association studies (GWAS) in a Mendelian randomisation (MR) phenome-wide association study (PheWAS) to identify potentially causal relationships for over 3,000 traits. Our outcome datasets comprise 378,142 cases across breast, prostate, colorectal, lung, endometrial, oesophageal, renal, and ovarian cancers, as well as 485,715 controls. We complement this analysis by systematically mining the literature space for supporting evidence. In addition to providing supporting evidence for well-established risk factors (smoking, alcohol, obesity, lack of physical activity), we also find sex steroid hormones, plasma lipids, and telomere length as determinants of cancer risk. A number of the molecular factors we identify may prove to be potential biomarkers. Our analysis, which highlights aetiological similarities and differences in common cancers, should aid public health prevention strategies to reduce cancer burden.     Targeting melanoma growth and viability reveals dualistic functionality of the phosphonothionate analogue of carba cyclic phosphatidic acid

    <p>Abstract</p> <p>Background</p> <p>Although the incidence of melanoma in the U.S. is rising faster than any other cancer, the FDA-approved chemotherapies lack efficacy for advanced disease, which results in poor overall survival. Lysophosphatidic acid (LPA), autotaxin (ATX), the enzyme that produces LPA, and the LPA receptors represent an emerging group of therapeutic targets in cancer, although it is not known which of these is most effective.</p> <p>Results</p> <p>Herein we demonstrate that thio-ccPA 18:1, a stabilized phosphonothionate analogue of carba cyclic phosphatidic acid, ATX inhibitor and LPA1/3 receptor antagonist, induced a marked reduction in the viability of B16F10 metastatic melanoma cells compared with PBS-treated control by 80-100%. Exogenous LPA 18:1 or D-sn-1-O-oleoyl-2-O-methylglyceryl-3-phosphothioate did not reverse the effect of thio-ccPA 18:1. The reduction in viability mediated by thio-ccPA 18:1 was also observed in A375 and MeWo melanoma cell lines, suggesting that the effects are generalizable. Interestingly, siRNA to LPA3 (siLPA3) but not other LPA receptors recapitulated the effects of thio-ccPA 18:1 on viability, suggesting that inhibition of the LPA3 receptor is an important dualistic function of the compound. In addition, siLPA3 reduced proliferation, plasma membrane integrity and altered morphology of A375 cells. Another experimental compound designed to antagonize the LPA1/3 receptors significantly reduced viability in MeWo cells, which predominantly express the LPA3 receptor.</p> <p>Conclusions</p> <p>Thus the ability of thio-ccPA 18:1 to inhibit the LPA3 receptor and ATX are key to its molecular mechanism, particularly in melanoma cells that predominantly express the LPA3 receptor. These observations necessitate further exploration and exploitation of these targets in melanoma.</p

    Radial Velocity Discovery of an Eccentric Jovian World Orbiting at 18 au

    Based on two decades of radial velocity (RV) observations using Keck/High Resolution Echelle Spectrometer (HIRES) and McDonald/Tull, and more recent observations using the Automated Planet Finder, we found that the nearby star HR 5183 (HD 120066) hosts a 3 minimum mass planet with an orbital period of yr. The orbit is highly eccentric (e ≃ 0.84), shuttling the planet from within the orbit of Jupiter to beyond the orbit of Neptune. Our careful survey design enabled high cadence observations before, during, and after the planet\u27s periastron passage, yielding precise orbital parameter constraints. We searched for stellar or planetary companions that could have excited the planet\u27s eccentricity, but found no candidates, potentially implying that the perturber was ejected from the system. We did identify a bound stellar companion more than 15,000 au from the primary, but reasoned that it is currently too widely separated to have an appreciable effect on HR 5183 b. Because HR 5183 b\u27s wide orbit takes it more than 30 au (1\u27\u27) from its star, we also explored the potential of complimentary studies with direct imaging or stellar astrometry. We found that a Gaia detection is very likely, and that imaging at 10 μm is a promising avenue. This discovery highlights the value of long-baseline RV surveys for discovering and characterizing long-period, eccentric Jovian planets. This population may offer important insights into the dynamical evolution of planetary systems containing multiple massive planets

    Detailed Molecular and Immune Marker Profiling of Archival Prostate Cancer Samples Reveals an Inverse Association between TMPRSS2:ERG Fusion Status and Immune Cell Infiltration

    Prostate cancer is a significant global health issue and limitations to current patient management pathways often result in over- or under-treatment. New ways to stratify patients are urgently needed. We conducted a feasibility study of such novel assessments looking for associations between genomic changes and lymphocyte infiltration. An innovative workflow utilizing an in-house targeted sequencing panel, immune cell profiling using an image analysis pipeline, RNA-Seq, and exome sequencing in select cases was tested. Gene fusions were profiled by RNA-seq in 27/27 cases and a significantly higher TIL count was noted in tumors without a TMPRSS2:ERG fusion compared to those with the fusion (P = 0.01). Although this finding was not replicated in a larger validation set (n=436) of The Cancer Genome Atlas images, there was a trend in the same direction. Differential expression analysis of TIL-High and TIL-Low tumors revealed the enrichment of both innate and adaptive immune response pathways. Mutations in mismatch repair genes (MLH1 and MSH6 mutations in 1/27 cases) were identified. We describe a potential immune escape mechanism in TMPRSS2:ERG fusion positive tumors. Detailed profiling, as shown here, can provide novel insights into tumor biology. Likely differences with findings with other cohorts are related to methods used to define region of interest, but this warrants further study in a larger cohort

    Physical Parameters of the Multiplanet Systems HD 106315 and GJ 9827

    HD 106315 and GJ 9827 are two bright, nearby stars that host multiple super-Earths and sub-Neptunes discovered by K2 that are well suited for atmospheric characterization. We refined the planets' ephemerides through Spitzer transits, enabling accurate transit prediction required for future atmospheric characterization through transmission spectroscopy. Through a multiyear high-cadence observing campaign with Keck/High Resolution Echelle Spectrometer and Magellan/Planet Finder Spectrograph, we improved the planets' mass measurements in anticipation of Hubble Space Telescope transmission spectroscopy. For GJ 9827, we modeled activity-induced radial velocity signals with a Gaussian process informed by the Calcium II H&K lines in order to more accurately model the effect of stellar noise on our data. We measured planet masses of M_b = 4.87 ± 0.37 M_⊕, M_c = 1.92 ± 0.49 M_⊕, and M_d = 3.42 ± 0.62 M_⊕. For HD 106315, we found that such activity radial velocity decorrelation was not effective due to the reduced presence of spots and speculate that this may extend to other hot stars as well (T_(eff) > 6200 K). We measured planet masses of M_b = 10.5 ± 3.1 M_⊕ and M_c = 12.0 ± 3.8 M_⊕. We investigated all of the planets' compositions through comparison of their masses and radii to a range of interior models. GJ 9827 b and GJ 9827 c are both consistent with a 50/50 rock-iron composition, GJ 9827 d and HD 106315 b both require additional volatiles and are consistent with moderate amounts of water or hydrogen/helium, and HD 106315 c is consistent with a ~10% hydrogen/helium envelope surrounding an Earth-like rock and iron core
