    The Micro-Shear Bond Strength of Various Resinous Restorative Materials to Aged Biodentine

    Introduction: The type of materials and application time of veneering restorations on calcium silicate cements are important factors which influence the interfacial properties. The aim of this study was to measure the micro-shear bond strength of a resin composite (RC) using several adhesive systems and a resin-modified glass ionomer cement (RM-GIC) to different aged Biodentine specimens. Methods and Materials: A total of 15 Biodentine blocks were prepared and assigned to three aging periods: 12 min, one week and one month. Then they were subdivided into five sub-groups to receive cylinders of resinous materials. RC was applied using different adhesive systems: A) no adhesive B) etch and rinse C) two-step self-etch and D) universal adhesive in self-etch mode and E) RM-GIC applied directly over Biodentine. Micro-shear bond strength was measured and the data were analyzed using one-way and two-way ANOVA. The level of significance was set at 0.05. Result: There was significant interaction between Biodentine aging periods and resinous materials (P<0.05). The highest value was obtained in group D bonded to the recently set Biodentine. Increasing the aging period to one week resulted in increased micro-shear bond strength in all groups expect for group D. One-month incubation time led to reduced shear bond strength in group A, C and D. Micro-shear bond strength values of group E increased to the longer aged Biodentine. Conclusion: Group D showed the highest bond strength to freshly mixed Biodentine.Keywords: Bond Strength; Composite Resin; Dental Adhesive; Glass Ionomer Cement; Tricalcium Silicat

    Home Financing Loans is an activity where an instution provides convience to consumers who wish to have a home without having to pay in cash, but by installments. Over time, emerged a contract program with the sharia system as cunducted by KPR Syariah Al Madani Village Karanganyar. Consumers order a house by making a down payment of 30% where the money is used to build a house. After payment is made both parties commit Istiṣnā contract. After the contract is done, the makers begin to make the process of making the house order and the consumer began to pay home installments. The formulation of the problem in this research is how the concept of Istiṣnā implementation of the contract on KPR Syariah Al Madani Village and wheter Istiṣnā contract applied is in accordance with the National Sharia Board Fatwa or not. This encourages researchers to examine in depth the “Istiṣnā contract Appraisal Analysis” on KPR Syariah Al Madani Village Karanganyar”. The purpose of this study is to know the concept of Istiṣnā application of contracts on this Syariah KPR and Istiṣnā contract confermance with the Fatwa DSN-MUI No. 06 of 2000 on Istiṣnā contract. The type of research used is field research with the qualitative descriptive approach. The method used in data collection is an interview and documentation. The analysis used is a deductive method. The result of this research can be conclude that KPR Syariah Al Madani Village has implemented Istiṣnā contract which is in accordance with the Fatwa of National Sharia Council of Indonesian Council of Ulama No. 06/ DSN-MUI/IV/2000 which explains Istiṣnā contract

    Improvement in symptoms and pulmonary function of asthmatic patients due to their treatment according to the Global Strategy for Asthma Management (GINA)

    <p>Abstract</p> <p>Background</p> <p>Global Initiative Strategy for Asthma Management (GINA) is poorly applied in undeveloped and developing countries. The current study examined the effects of applying GINA guidelines on treatment efficacy in asthmatic patients in Iran.</p> <p>Methods</p> <p>Twenty four asthmatic patients (usual care group) were treated as usual and 26 patients (intervention group) according to the GINA for 2 months. Asthma symptom score, asthma severity, frequency of symptoms/week and wheezing were recorded at the beginning (first visit), one month after treatment (second visit), and at the end of the study (third visit). Pulmonary function tests (PFTs) were performed by spirometry, and the patients' use of asthma drugs and their symptoms were evaluated, at each visit.</p> <p>Results</p> <p>Asthma symptoms, frequency of symptoms/week, chest wheezing, and PFT values were significantly improved in the intervention group at the second and third visits compared to first visit (p < 0.001 for all measures). In addition, exercise induced cough and wheeze were significant improved in the third visit compared to the second visit in this group (p < 0.01 for both measures). In the second and third visits all symptoms were significantly lower, and PFT values higher, in the intervention group compared to the usual care group (p < 0.005 to p < 0.001). In the usual care group, there were only small improvements in some parameters in just the second visit (p < 0.01 for all measures). The use of asthma drugs was unchanged in the usual care group and significantly reduced in the intervention group (p < 0.01) by the end of the study.</p> <p>Conclusion</p> <p>Adoption of GINA guidelines improves asthma symptoms and pulmonary function in asthmatic patients in Iran.</p

    PENDAMPINGAN PENINGKATAN KAPASITAS FASILITATOR DI FORUM PELAJAR SADAR HUKUM SMAN 27 BANDUNG MELALUI PENYUSUNAN MODUL STREET LAW

    Perkembangan dunia secara global menimbulkan konsekuensi dan permasalahan hukum. Sayangnya, masyarakat Indonesia tidak seluruhnya memahami hukum, sedangkan dalam hukum dikenal friksi hukum yang menyatakan menganggap masyarakat mengetahui hukum. Pendidikan hukum masyarakat menjadi sebuah keharusan untuk memberdayakan pengetahuan dan kesadaran hukum masyarakat dengan menciptakan sebuah sistem pembelajaran yang berkelanjutan yang dilakukan oleh dan untuk masyarakat untuk menghasilkan efek bola salju pendidikan hukum. Salah satu upaya pemberdayaan pengetahuan hukum masyarakat yang sudah banyak dilakukan adalah dengan mengedepankan partisipasi masyarakat dalam proses pemberdayaan pengetahuan hukum itu sendiri, dimana masyarakat menjadi peer facilitator bagi kelompok masyarakat lainnya. Sejak 2013, Fakultas Hukum Universitas Pasundan menjadi pendamping kegiatan pemberdayaan pengetahuan hukum di SMAN 27 Bandung melalui Forum Pelajar Sadar Hukum yang dibentuk sekolah. Pada awalnya kegiatan pendampingan dan pembinaan dilakukan secara langsung dengan menerjunkan mahasiswa-mahasiswa hukum untuk mengajarkan hukum kepada para siswa yang terlibat dalam Forum Sadar Hukum SMAN 27 Bandung. Pada tahun-tahun berikutnya dimulai pemberdayaan siswa-siswa yang terlibat dalam Forum Sadar Hukum SMAN 27 untuk dipersiapkan menjadi peer facilitator bagi rekan-rekan siswa lainnya. Permasalahan yang dihadapi adalah bahwa masih perlu pendampingan terkait metode pembelajaran yang efektif untuk mengajarkan materi hukum selain itu materi-materi hukum yang sesuai dengan hukum yang berlaku, sehingga dirasa perlu adanya modul pembelajaran bagi fasilitator sebagai panduan dalam melakukan kegiatan pendidikan hukum bagi rekan-rekan lainnya. Kegiatan pengabdian ini akan berupa penyusunan modul pembelajaran hukum bagi fasilitator. Luaran yang ditargetkan dari kegiatan ini adalah modul pembelajaran bagi fasilitator

    ZIF-8 Modified Polypropylene Membrane: A Biomimetic Cell Culture Platform with a View to the Improvement of Guided Bone Regeneration.

    PurposeDespite the significant advances in modeling of biomechanical aspects of cell microenvironment, it remains a major challenge to precisely mimic the physiological condition of the particular cell niche. Here, the metal-organic frameworks (MOFs) have been introduced as a feasible platform for multifactorial control of cell-substrate interaction, given the wide range of physical and mechanical properties of MOF materials and their structural flexibility.ResultsIn situ crystallization of zeolitic imidazolate framework-8 (ZIF-8) on the polydopamine (PDA)-modified membrane significantly raised surface energy, wettability, roughness, and stiffness of the substrate. This modulation led to an almost twofold increment in the primary attachment of dental pulp stem cells (DPSCs) compare to conventional plastic culture dishes. The findings indicate that polypropylene (PP) membrane modified by PDA/ZIF-8 coating effectively supports the growth and proliferation of DPSCs at a substantial rate. Further analysis also displayed the exaggerated multilineage differentiation of DPSCs with amplified level of autocrine cell fate determination signals, like BSP1, BMP2, PPARG, FABP4, ACAN, and COL2A. Notably, osteogenic markers were dramatically overexpressed (more than 100-folds rather than tissue culture plate) in response to biomechanical characteristics of the ZIF-8 layer.ConclusionHence, surface modification of cell culture platforms with MOF nanostructures proposed as a powerful nanomedical approach for selectively guiding stem cells for tissue regeneration. In particular, PP/PDA/ZIF-8 membrane presented ideal characteristics for using as a barrier membrane for guided bone regeneration (GBR) in periodontal tissue engineering

    Isolation and differentiation of adipose-derived stem cells into odontoblast-like cells: a preliminary in vitro study

    Objective The aim of present study was to isolate and differentiate human adipose-derived stem cells (ASCs) into odontoblast-like cells. Materials and Methods In this experimental study, human adipose tissues were taken from the buccal fat pad of three individuals (mean age: 24.6 ± 2.1 years). The tissues were transferred to a laboratory in a sterile culture medium, divided into small pieces and digested by collagenase I (2 mg/mL, 60-90 minutes). ASCs were isolated by passing the cell suspension through cell strainers (70 and 40 µm), followed by incubation at 37ºC and 5% CO2in Dulbecco’s modified eagle medium (DMEM) supplemented with fetal bovine serum (FBS 5%) and penicillin/streptomycin (P/S). After three passages, the ASCs were harvested. Subsequently, flow cytometry and reverse transcriptase polymerase chain reaction (RT-PCR) were used to detect expression levels of NANOG and OCT4 to evaluate stemness. Then, a differentiation medium that included high-glucose DMEM supplemented with 10% FBS, dexamethasone (10 nM), sodium β-glycerophosphate (5 mM) and ascorbic acid (100 µM) was added. The cells were cultivated for four weeks, and the odontogenic medium was changed every two days. Cell differentiation was evaluated with Alizarin red staining and expressions of collagen I (COL1A1), dentin sialophosphoprotein (DSPP) and dentin matrix protein-1 (DMP1). Results The ASCs were effectively and easily isolated. They were negative for CD45 and positive for the CD105 and CD73 markers. The ASCs expressed OCT4 and NANOG. Differentiated cells highly expressed DSPP, COL1A1 and DMP1. Alizarin red staining revealed a positive reaction for calcium deposition. Conclusion ASCs were isolated successfully in high numbers from the buccal fat pad of human volunteers and were differentiated into odontoblast-like cells. These ASCs could be considered a new source of cells for use in regenerative endodontic treatments

    Comparative evaluation of the effects of three hydraulic calcium silicate cements on odontoblastic differentiation of human dental pulp stem cells: an in vitro study

    Objective: The study aimed to compare the response of human dental pulp stem cells (hDPSCs) towards three hydraulic calcium silicate cements (HCSCs) by measuring cytotoxicity and expression of dentinogenic genes. Methodology: Dental pulps of five impacted mandibular third molars were extirpated as a source for hDPSCs. Next to culturing, hDPSCs were subjected to fluorescence-activated cell sorting after the third passage to validate stemness of the cells. Human DPSCs were exposed to diluted supernatants of OrthoMTA (OMTA), Biodentine (BD) and Calcium-Enriched Mixture (CEM) at concentrations 10, 25, 50 and 100% at the first, third and fifth day of culture. Then, cells were exposed to 10% concentrations supernatant of HCSCs to determine DSPP and DMP1 gene expression, using a quantitative polymerase-chain reaction. Data were analyzed using one-way and three-way ANOVA, followed by Tukey post hoc statistical tests. Results: Optimal cell proliferation was observed in all groups, regardless of concentration and time-point. HCSC supernatants were non-cytotoxic to hDPSCs at all three time-points, except for 100% Biodentine on day five. On day seven, OMTA group significantly upregulated the expression of DSPP and DMP1 genes. On day 14, expression of DMP1 and DSPP genes were significantly higher in BD and OMTA groups, respectively. Conclusion: Biodentine significantly upregulated DMP1 gene expression over 14 days, whereas CEM was associated with only minimal expression of DSPP and DMP1 . &nbsp

    Takayasu's arteritis associated with Crohn's disease: a case report

    <p>Abstract</p> <p>Introduction</p> <p>The simultaneous presence of Takayasu's arteritis and Crohn's disease in a patient seems to be rare. To our knowledge, no patient with the combination of Crohn's disease and Takayasu's arteritis has been reported from our region.</p> <p>Case presentation</p> <p>Herein we present the case of a 22-year-old Iranian woman previously diagnosed as Crohn's disease and who had subsequently developed Takayasu's arteritis.</p> <p>Conclusion</p> <p>Clinical suspicion, proper imaging, and consideration of the differential diagnosis are important for the correct diagnosis and management of patients with this coincidence.</p

    X-Linked Immunodeficient Mice With No Functional Bruton's Tyrosine Kinase Are Protected From Sepsis-Induced Multiple Organ Failure

    We previously reported the Bruton's tyrosine kinase (BTK) inhibitors ibrutinib and acalabrutinib improve outcomes in a mouse model of polymicrobial sepsis. Now we show that genetic deficiency of the BTK gene alone in Xid mice confers protection against cardiac, renal, and liver injury in polymicrobial sepsis and reduces hyperimmune stimulation (“cytokine storm”) induced by an overwhelming bacterial infection. Protection is due in part to enhanced bacterial phagocytosis in vivo, changes in lipid metabolism and decreased activation of NF-κB and the NLRP3 inflammasome. The inactivation of BTK leads to reduced innate immune cell recruitment and a phenotypic switch from M1 to M2 macrophages, aiding in the resolution of sepsis. We have also found that BTK expression in humans is increased in the blood of septic non-survivors, while lower expression is associated with survival from sepsis. Importantly no further reduction in organ damage, cytokine production, or changes in plasma metabolites is seen in Xid mice treated with the BTK inhibitor ibrutinib, demonstrating that the protective effects of BTK inhibitors in polymicrobial sepsis are mediated solely by inhibition of BTK and not by off-target effects of this class of drugs
