    An examination of nervous system revealed unexpected immunoreactivity of both secretory apparatus and excretory canals in plerocercoids of two broad tapeworms (Cestoda: Diphyllobothriidea)

    Dibothriocephalus ditremus and Dibothriocephalus latus are diphyllobothriidean tapeworms autochthonous to Europe. Their larval stages (plerocercoids) may seriously alter health of their intermediate fish hosts (D. ditremus) or cause intestinal diphyllobothriosis of the final human host (D. latus). Despite numerous data on the internal structure of broad tapeworms, many aspects of the morphology and physiology related to host–parasite co-existence remain unclear for these 2 species. The main objective of this work was to elucidate functional morphology of the frontal part (scolex) of plerocercoids, which is crucial for their establishment in fish tissues and for an early attachment in final hosts. The whole-mount specimens were labelled with different antibodies and examined by confocal microscope to capture their complex 3-dimensional microanatomy. Both species exhibited similar general pattern of immunofluorescent signal, although some differences were observed. In the nervous system, FMRF amide-like immunoreactivity (IR) occurred in the bi-lobed brain, 2 main nerve cords and surrounding nerve plexuses. Differences between the species were found in the structure of the brain commissures and the size of the sensilla. Synapsin IR examined in D. ditremus occurred mainly around FMRF amide-like IR brain lobes and main cords. The unexpected finding was an occurrence of FMRF amide-like IR in terminal reservoirs of secretory gland ducts and excretory canals, which has not been observed previously in any tapeworm species. This may indicate that secretory/excretory products, which play a key role in host–parasite relationships, are likely to contain FMRF amide-related peptide/s

    Českobudějovicko is the district situated in Southern Bohemia. Českobudějovicko can be found in the neighbourhood of town České Budějovice. The town was founded by King Přemysl Otakar II. in 1265 on the junction of Vltava and Malše rivers. This city lies in 381 metres above the sea. České Budějovice has population near one hundred thousand. It is a city full of history, magnificent monuments, but it can figure as a commercial, administrative and cultural centre. The České Budějovice district with its almost intact enviroment, its numerous ponds, forests and hills offers many opportunities for a pleasant holiday, hiking and cycling. Visitors can find here a lot of cykloturistic routes so when they decided to see the region from the bike, they are getting to know this region best. The best known sights of České Budějovice are castles, Black tower (it measures 72 meters) or historic Town Hall in the Přemysl Otakar II Square. This square is square-shaped and one hectare large. In the middle of the square is situated the baroque Samson{\crq}s Fountain from the 18th century decorated with a statue of Samson fighting with a lion. If tourists are hungry or thirsty they can wisit a restaurant. Most of today`s restaurants and hotels have a wide tradition, so it is a pleasure to sit down and taste some meal or beer and a half wich is real Bohemian product. The best-known beer in the world is certainly Budvar. When visitors want to know something they can use the tourist information center. The maps, guide books with description of the town or another souvenirs are sold there. People who live there say, that you may tour this district at any time of the year and you will allways find in it a new, still undiscovered, beauty

    v angličtině: This work is aimed at the study of sensor response of quartz microbalance towards biologically active substances such as glycerin and toxic substances, namely ethylene glycol, methanol and acetonitrile in liquid phase. These toxic organic compounds are widely used in chemical industry as solvents or anti-freeze components. Glycerin is commonly utilized in cosmetic industry as a moisturizer. As human can easily get into contact with these compounds, they should be easily detectable. If tested senzor proves to be suitable for detecting these substances, it will be possible to follow preliminary results and develop the device for practical industrial usage

    Import 04/07/2011Práce se zabývá stabilitním posouzením plošného založení železniční zastávky a výpočtu napěťo-deformačních stavů v tělese nového násypu. Vstupní parametry byly získány z geotechnického průzkumu pro konkrétní základové poměry (dynamická penetrace, laboratorní zkoušky). Pro řešení cílů práce je použit programový softwaru GEO5 společnosti FINE s.r.o. Byly využity moduly stabilita svahu a MKP (metoda konečných prvků). Pro ověření výsledků dosažených modelováním byl proveden ruční výpočet stability pro nejnebezpečnější smykovou plochu dle Pettersonovy proužkové metody. V závěru je provedeno vyhodnocení i s možným návrhem řešením kombinace plošného a hlubinného základu.The work deals with the stability of spread foundation the railway station and the calculation of stress-strain states in the body of the new embankment. Input parameters were obtained from the geotechnical exploration for the specific subsoil conditions (dynamic penetration, laboratory tests). To address the objectives of the work program is used by software GEO5 FINE Ltd. Modules were used in slope stability and FEM (finite element method). To verify the results obtained by modeling was performed hand calculation of stability for the most dangerous slip surface according to Petterson belt method.     This diploma work is devoted to the preparation and analytical testing of modified gold electrodes in QCM. The sensitivity and selectivity response towards selected aromatic analytes in aqueous phase was monitored. Studied analytes were 1-methylnaphthalene, 2-methylnaphthalene, naphthalene, o-xylene and toluene. These compounds are frequently used in chemistry industry and other industrial branches and, are known to be harmful to human organism and environment. Using QCM method it is possible to detect them rapidly and relatively easily

    Objective: Heparin-induced thrombocytopenia (HIT II) is an immune-mediated disorder observed in up to 0.5-5% of heparin-treated patients. Laboratory testing for the diagnosis of HIT II includes a 14C-serotonin release test, PF4/heparin enzyme-immunoassay, platelet aggregation and flow cytometric assay with various specificities or sensitivities for the diagnosis of HIT II. The aim of this work was to develop an HPLC method with electrochemical detection (EC) and prove the possibility of using a non-radioactive serotonin release test for the diagnosis of HIT II. Material and Methods: Two groups were tested. First group of patients suspect for HIT II, and second control group with or without heparin therapy. Serotonin release was analyzed by a newly developed HPLC-EC method. Results: Analytical performance of this method was satisfactory. The method was linear from 5 to 7000 nmol/l with a limit of detection of 1 nmol/l. The intra- and inter-assay coefficients of variation were 7.7% and 12%, respectively, for a concentration of serotonin 1460 nmol/l. Conclusion: In the present work we describe rapid, non-radioactive, specific and sensitive HPLC-ED methods to determine serotonin release from the donor platelets
