    An antibody raised against a pathogenic serpin variant induces mutant-like behaviour in the wild-type protein

    A monoclonal antibody (mAb) that binds to a transient intermediate may act as a catalyst for the corresponding reaction; here we show this principle can extend on a macro molecular scale to the induction of mutant-like oligomerization in a wild-type protein. Using the common pathogenic E342K (Z) variant of α1-antitrypsin as antigen-whose native state is susceptible to the formation of a proto-oligomeric intermediate-we have produced a mAb (5E3) that increases the rate of oligomerization of the wild-type (M) variant. Employing ELISA, gel shift, thermal stability and FRET time-course experiments, we show that mAb5E3 does not bind to the native state of α1-antitrypsin, but recognizes a cryptic epitope in the vicinity of the post-helix A loop and strand 4C that is revealed upon transition to the polymerization intermediate, and which persists in the ensuing oligomer. This epitope is not shared by loop-inserted monomeric conformations. We show the increased amenity to polymerization by either the pathogenic E342K mutation or the binding of mAb5E3 occurs without affecting the energetic barrier to polymerization. As mAb5E3 also does not alter the relative stability of the monomer to intermediate, it acts in a manner similar to the E342K mutant, by facilitating the conformational interchange between these two states

    Exploring How Mindfulness May Enhance Perceived Value of Travel Experience

    This study aims to explore the effect of tourists’ mindfulness on the perceived value of travel experience (PVTE) through destination images (cognitive, affective, and conative) and tourist experiences. Data (n = 370) were gathered using a self-administered structured questionnaire distributed to travellers departing from Lisbon International airport towards Spain in July 2017. The results show the important role of mindfulness in shaping all dimensions of destination image. Tourist experience acts as a mediator between destination images and PVTE. However, perceived authenticity does not have a moderating, but rather a controlling effect on the relationship between tourist experience and PVTE. From a theoretical point of view, the study makes important contribution in conceptualising the influence of a tourist’s mindfulness on PVTE through destination image components and tourist experience.     Novel Microscopic Mechanism of Intermixing during Growth on Soft Metallic Substrates

    Generic computer simulations using empiric interatomic potentials suggest a new, collective mechanism that could be responsible for mixing at heteroepitaxial interfaces. Even if single adsorbate atoms diffuse by hopping on the substrate surface and do not mix at the terraces, two-dimensional islands formed by nucleation may become unstable above a certain critical size and explode upwards forming clusters of several atomic layers. This process is accompanied by strong distortions of the underlying atomic layers, and on soft materials it can result in surface etching and incorporation of substrate atoms into the islands.Fil: Gomez, Liliana Maria.     A DFT approach to the surface-enhanced raman scattering of 4-cyanopyridine adsorbed on silver nanoparticles

    A Surface-Enhanced Raman Scattering (SERS) spectrum of 4-cyanopyridine (4CNPy) was recorded on silver plasmonic nanoparticles and analyzed by using Density Functional Theory (DFT) calculations. Two simple molecular models of the metal\u20134CNPy surface complex with a single silver cation or with a neutral dimer (Ag+ \u20134CNPy, Ag2 \u20134CNPy), linked through the two possible interacting sites of 4CNPy (aromatic nitrogen, N, and nitrile group, CN), were considered. The calculated vibrational wavenumbers and intensities of the adsorbate and the isolated species are compared with the experimental Raman and SERS results. The analysis of the DFT predictions and the experimental data indicates that 4CNPy adsorbs preferentially on neutral/charged active sites of the silver nanoparticles through the nitrogen atom of the aromatic ring with a perpendicular orientation

    Propagation of gravity waves and spread F in the low-latitude ionosphere over Tucumán, Argentina, by continuous Doppler sounding: first results

    Results of systematic analysis of propagation directions and horizontal velocities of gravity waves (GWs) and spread F structures in low-latitude ionosphere (magnetic inclination ~27°) in Tucumán region, Argentina, are presented. Measurements were carried out by multipoint continuous Doppler system during 1 year from December 2012 to November 2013. It was found that meridian propagation of GWs dominated and that southward propagation prevailed in the local summer. Oblique spread structures observed in Doppler shift spectrograms and associated with spread F propagated roughly eastward at velocities from ~70 to ~180 m/s and were observed at night from ~ September to ~ March. The velocities were computed for 182 events and the azimuths for 64 events. Continuous Doppler sounding makes it possible to analyze more events compared to optical observations often used for propagation studies since the measurements do not depend on weather.Fil: Chum, J..     Occurrence of mite Ophionyssus natricis (Acari: Macronyssidae) on captive snakes from Panama

    We report for the first time the presence of the snake mite, Ophionyssus natricis (Gervais) (Mesostigmata: Macronyssidae) on captive snakes kept at Panama City, Panama. This occurrence adds a new record to the geographical distribution of O. natricis as well as confirms its high prevalence on captive reptiles. Several Boidae species, Boa constrictor, Epicrates maurus, Corallus ruschenbergerii, Corallus caninus and a Pythonidae, Python regius were found infested with intensities varying from 10 to 2200 mites each. These findings represent the second record of O. natricis for Central America

    Beatus Manuscripts under the microscope: The Alcobaça Beatus and the Iberian Cistercian Tradition revisited

    SFRH/BPD/70305/2010The Alcobaça Beatus (Lisbon, National Library, Alc. 247) is one of the few extant unillustrated copies of the eighth-century Commentary on the Apocalypse attributed to the Iberian monk Beatus of Liébana. It has traditionally been considered that this Beatus was produced at the Cistercian Portuguese abbey of Santa Maria de Alcobaça and that its model was the Lorvão Beatus (Lisbon, Arquivo Nacional da Torre do Tombo, Ordem de Cister, Mosteiro de Santa Maria de Lorvão, Liv. 44). This article provides the first in-depth examination of the Alcobaça Beatus, carried out by a multidisciplinary team using combined methods from the natural and human sciences. The technical data provided by the analysis of the decoration, molecular palette and bookbinding support the theory of its production at the scriptorium of the Portuguese monastery of Alcobaça, whereas the analysis of the textual structure and content reveals that the Alcobaça Beatus and the Lorvão Beatus show a different sequence of texts and that the Alcobaça copy contains later notes that evidence its use in the monastic reading practice, especially the liturgical readings of the Divine Office. This set of results turns this manuscript, which had received minimal scholarly attention to date, into a significant case study since it reveals itself as the basis to further investigate the corpus of extant Beatus copies and question the importance of such a work within the Cistercian order, thus provoking new questions within the debate on Cistercians and their identity around the year 1200.authorsversionpublishe

Publisher Copyright: © 2023 by the authors.The illuminated manuscripts at the Gulbenkian Museum were produced in the 17th century, in scriptoria of the Armenian diaspora. In this work, we selected analytical methods that can be used in situ to study the colors of the illuminations. Scientific analysis based on fiber-optics reflectance spectroscopy in the visible and Raman spectroscopy has shown the use of a medieval palette based on inorganic pigments such as lapis lazuli, minium, vermilion, orpiment, indigo, two different greens (vergaut and malachite), lead white and carbon black. More importantly, in this context, it showed that the very important reds and pinks are possibly based on carminic acid. The painting technique is, however, different, as are the ways of painting the faces, hands, and vestments. The range of colors in the Bible and the three Gospel Books, enhanced by lapis lazuli blue and organic reds and pinks, demonstrates a desire to create exceptional illuminated manuscripts.publishersversionpublishe

    Naltrexone Reverses Ethanol-Induced Rat Hippocampal and Serum Oxidative Damage

    Naltrexone, an antagonist of �-opioid receptors, is clinically used as adjuvant therapy of alcohol dishabituation. The aim of the present work was to test the effect of 1 mg/kg body weight of naltrexone to revert oxidative stress-related biochemical alterations, in the hippocampus and serum of chronic alcoholic adult rats. Malondialdehyde concentration was increased and glutathione peroxidase activity was decreased in hippocampus and serum of alcohol-treated rats. Naltrexone treatment restored these alterations. The in vitro antioxidant ability of Ntx could not justify these effects considering the doses used.
