    Endostatin genetically engineered placental mesenchymal stromal cells carrying mesoporous silica nanoparticles for combined chemo- and antiangiogenic therapy

    Combination therapies constitute a powerful tool for cancer treatment. By combining drugs with different mechanisms of action, the limitations of each individual agent can be overcome, while increasing therapeutic benefit. Here, we propose employing tumor-migrating decidua-derived mesenchymal stromal cells as therapeutic agents combining antiangiogenic therapy and chemotherapy. First, a plasmid encoding the antiangiogenic protein endostatin was transfected into these cells by nucleofection, confirming its expression by ELISA and its biological effect in an ex ovo chick embryo model. Second, doxorubicin-loaded mesoporous silica nanoparticles were introduced into the cells, which would act as vehicles for the drug being released. The effect of the drug was evaluated in a coculture in vitro model with mammary cancer cells. Third, the combination of endostatin transfection and doxorubicin-nanoparticle loading was carried out with the decidua mesenchymal stromal cells. This final cell platform was shown to retain its tumor-migration capacity in vitro, and the combined in vitro therapeutic efficacy was confirmed through a 3D spheroid coculture model using both cancer and endothelial cells. The results presented here show great potential for the development of combination therapies based on genetically-engineered cells that can simultaneously act as cellular vehicles for drug-loaded nanoparticles

    Rombencefalite aguda por Listeria monocytogenes: relato de caso

    Rhombencephalitis is inflammation of the brainstem and cerebellum; one of the causes of this entity is Listeria monocytogenes, although it is an unusual presentation. The case is a 70-year-old woman with high blood pressure, obesity, bronchial asthma, and hypothyroidism. She began a 6-day clinical picture with nausea, vomiting, and headache, then dizziness and right lateropulsion were added, and a nuclear magnetic resonance was performed, diagnosing ischemic stroke in the medulla oblongata. She is hospitalized, she has a fever of 38.5 °C, compromised level of consciousness, and neck stiffness. Listeria monocytogenes infection was confirmed by CSF culture, she received meropenem, vancomycin, ampicillin, and dexamethasone, then the patient fully recovered on the fifth day of antibiotic treatment, without neurological sequela. The presence of posterior fossa symptoms on a CNS inflammatory background should lead to suspicion of rhombencephalitis due to Listeria; early antibiotic treatment limits neurological sequela.La rombencefalitis es la inflamación del tronco encefálico y cerebelo; una de las causas de esta entidad es la Listeria monocytogenes, aunque es una presentación inusual. El caso es en una mujer de 70 años, con hipertensión arterial, obesidad, asma bronquial, e hipotiroidismo. Inicia cuadro de 6 días con náuseas, vómitos y cefalea, luego se agregan mareos y lateropulsión derecha, se le realiza una resonancia magnética nuclear diagnosticando accidente cerebrovascular isquémico en bulbo raquídeo. Es hospitalizada, presenta fiebre de 38,5 °C, compromiso del nivel de conciencia y rigidez de nuca. Se confirmó infección por Listeria monocytogenes por cultivo de LCR, recibe meropenem, vancomicina, ampicilina y dexametasona, luego la paciente se recupera completamente al quinto día de tratamiento antibiótico, sin secuelas neurológicas. La presencia de síntomas de fosa posterior sobre un fondo inflamatorio de SNC debe hacer sospechar de rombencefalitis por Listeria; el tratamiento antibiótico precoz limita las secuelas neurológicas.A rombencefalite é a inflamação do tronco cerebral e do cerebelo; uma das causas desta entidade é a Listeria monocytogenes, embora seja uma apresentação incomum. O caso é de uma mulher de 70 anos com hipertensão arterial, obesidade, asma brônquica e hipotireoidismo. Um quadro clínico de 6 dias inicia-se com náuseas, vômitos e cefaleia, depois se somam tontura e lateropulsão direita, é realizada ressonância nuclear magnética, diagnosticando acidente vascular cerebral isquêmico na medula oblonga. Está internada, apresenta febre de 38,5 °C, nível de consciência comprometido e rigidez de nuca. A infecção por Listeria monocytogenes foi confirmada por cultura do LCR, ela recebeu meropenem, vancomicina, ampicilina e dexametasona, então a paciente se recuperou totalmente no quinto dia de antibioticoterapia, sem sequelas neurológicas. A presença de sintomas da fossa posterior em um fundo inflamatório do SNC deve levar a suspeitar rombencefalite por Listeria; o tratamento antibiótico precoce limita as sequelas neurológicas

    A success story: New rules and fewer injuries in traditional

    Objective: To analyse the effectiveness of a change in the rules on the incidence of injuries in traditional Leonese Wrestling (LW). Material and methods: Cohort study, retrospective (2006---2007) and prospective (2008---2012). The population studied comprised all the male wrestlers over 16 years of age who took part in Winter League competitions in the seasons 2006---2012. Data collected included the number, location and severity of injuries. Classifying them according to location and severity, the incidence of injuries by bouts and falls before and after the change was calculated. The formula for calculating effectiveness was: 1 --- relative risk. Results: Over the 7 seasons, a total of 34 injuries were reported ---- 9 in the seasons before the change and 25 afterwards. Since the introduction of the new regulations in 2008, no serious upper limb injuries have been reported. Before the change in rules there was an incidence of 10.4 per 1000 bouts and 3.9 for every 1000 falls, the differences being statistically significant (P = .002 and P = .006, respectively). The effectiveness in reducing the incidence of serious injuries was 94% by bouts, and 91% by falls. Conclusions: The results of this study confirm the importance of a methodological approach for the prevention of injuries; in this case a change in the rules. © 2012 Consell Català de l’Esport.     1,3-diphenylpropan-1-ones as allosteric modulators of α7 nACh receptors with analgesic and antioxidant properties

    Nicotine acethylcholine receptors (nAChRs) play critical roles in cognitive processes, neuroprotection and inflammation. Results: According to their substituents, 1,3-diphenylpropan-1-one derivatives act as α7 nAChRs negative allosteric modulators (NAM, OMe) or Type I positive allosteric modulators (PAMs, OH). Compounds 7 and 31 were the most effective (989 and 666% enhancement of ACh-induced currents) and potent (EC: 12.9 and 6.85 μM) PAMs. They exhibited strong radical scavenging values. Compound 31, selective over other neuronal nAChR subtypes and with acceptable pharmacokinetic profile, showed antinociceptive effects in a model of inflammatory pain. Conclusion: Compound 31 is a novel, potent and selective α7 nAChR PAM, displaying antioxidant and analgesic activities.     Una historia de éxito: nuevas reglas y menos lesiones en la Lucha Leonesa (2006-2012)

    Introducción El objetivo de nuestro estudio es analizar la efectividad de un cambio de reglamento en la incidencia de lesiones de Lucha Leonesa. Material y métodos Estudio de cohorte retrospectivo (2006-2007) y prospectivo (2008-2012). La población estudiada comprendió todos los luchadores masculinos de más de 16 años que participaron en las Ligas de Invierno en las temporadas del 2006-2012. Se recogieron datos sobre el número, la localización y la severidad de las lesiones, clasificándolos según su localización y su gravedad. Se calculó la incidencia de lesiones por combates y por caídas antes y después del cambio de reglamento. La fórmula para calcular la efectividad fue: 1 – Riesgo Relativo. Resultados En las 7 temporadas, se registraron un total de 34 lesiones, 9 antes del cambio de reglamento y 25 después. Desde la introducción de las nuevas reglas en 2008 no se produjo ninguna lesión grave en los miembros superiores, mientras que antes del cambio de reglamento la incidencia de lesiones fue de 10,4 por cada 1000 combates y de 3,9 por cada 1000 caídas, siendo estadísticamente significativa (p= 0,002 y p=0,006). La efectividad de la reducción de la incidencia de lesiones graves fue del 94% por combates y del 91% por caídas. Conclusiones Los resultados de este estudio confirman la importancia de un enfoque metodológico para la prevención de lesiones; en este caso, los cambios de reglamento

    Conflictos sociales y mediación universitaria en el Espacio Europeo de Educación Superior

    Este artículo expone los principales datos y conclusiones obtenidas en el Workshop realizado en el año 2016 entre la Universidad de Málaga y Leuven (Bélgica). La líneas de investigación se centra en los conflictos sociales que se producen en el ámbito universitario.El presente trabajo, trata de analizar los conflictos que se producen en el ámbito universitario relacionados con la implementación del Espacio Europeo de Educación Superior y la nueva metodología docente derivada del mismo, que provocan disyuntivas sociales específicas. Estos conflictos, se hacen plausibles en la conciliación, el aprendizaje y el desarrollo del proceso educativo. Por un lado, con la asistencia obligatoria y la dedicación de tiempo que requiere al estudiante en la Universidad y de trabajo en casa. Por otro, por la situación devenida de los recortes en becas universitarias y la necesidad que impone esta reforma educativa de acreditación del B1 como lengua extranjera para la obtención del Título de Graduado. Estas circunstancias, construyen realidades relativas a los significados que el ámbito universitario tiene para las personas, e inciden en las oportunidades del alumnado en condiciones de igualdad en función de su situación social y personal.     Analysis of the pathogenic potential of nosocomial Pseudomonas putida strains

    Pseudomonas putida strains are ubiquitous in soil and water but have also been reported as opportunistic human pathogens capable of causing nosocomial infections. In this study we describe the multilocus sequence typing of four P. putida strains (HB13667, HB8234, HB4184 and HB3267) isolated from in-patients at the Besançon Hospital (France). The four isolates (in particular HB3267) were resistant to a number of antibiotics. The pathogenicity and virulence potential of the strains was tested ex vivo and in vivo using different biological models: human tissue culture, mammalian tissues and insect larvae. Our results showed a significant variability in the ability of the four strains to damage the host; HB13667 did not exhibit any pathogenic traits, HB4184 caused damage only ex vivo in human tissue cultures, and HB8234 had a deleterious effect in tissue culture and in vivo on rat skin, but not in insect larvae. Interestingly, strain HB3267 caused damage in all the model systems studied. The putative evolution of these strains in medical environments is discussed
