    Launch Vehicle Production and Operations Cost Metrics

    Traditionally, launch vehicle cost has been evaluated based on $/Kg to orbit. This metric is calculated based on assumptions not typically met by a specific mission. These assumptions include the specified orbit whether Low Earth Orbit (LEO), Geostationary Earth Orbit (GEO), or both. The metric also assumes the payload utilizes the full lift mass of the launch vehicle, which is rarely true even with secondary payloads.1,2,3 Other approaches for cost metrics have been evaluated including unit cost of the launch vehicle and an approach to consider the full program production and operations costs.4 Unit cost considers the variable cost of the vehicle and the definition of variable costs are discussed. The full program production and operation costs include both the variable costs and the manufacturing base. This metric also distinguishes operations costs from production costs, including pre-flight operational testing. Operations costs also consider the costs of flight operations, including control center operation and maintenance. Each of these 3 cost metrics show different sensitivities to various aspects of launch vehicle cost drivers. The comparison of these metrics provides the strengths and weaknesses of each yielding an assessment useful for cost metric selection for launch vehicle programs

    Public questions spur the discovery of new bacterial species associated with lignin bioconversion of industrial waste

    A citizen science project found that the greenhouse camel cricket (Diestrammena asynamora) is common in North American homes. Public response was to wonder “what good are they anyway?” and ecology and evolution guided the search for potential benefit. We predicted that camel crickets and similar household species would likely host bacteria with the ability to degrade recalcitrant carbon compounds. Lignocellulose is particularly relevant as it is difficult to degrade yet is an important feedstock for pulp and paper, chemical, and biofuel industries. We screened gut bacteria of greenhouse camel crickets and another household insect, a hide beetle (Dermestes maculatus) for the ability to grow on and degrade lignocellulose components as well as the lignocellulose-derived industrial waste product black liquor. From three greenhouse camel crickets and three hide beetles, 14 bacterial strains were identified capable of growth on lignocellulosic components, including lignin. Cedecea lapagei was selected for further study due to growth on most lignocellulose components. The C. lapagei secretome was identified using LC/MS/MS analysis. This work demonstrates a novel source of lignocellulose-degrading bacteria and introduces an effective workflow to identify bacterial enzymes for transforming industrial waste into value-added products. More generally, our research suggests the value of ecologically-guided discovery of novel organisms

    Using remote substituents to control solution structure and anion binding in lanthanide complexes.

    A study of the anion-binding properties of three structurally related lanthanide complexes, which all contain chemically identical anion-binding motifs, has revealed dramatic differences in their anion affinity. These arise as a consequence of changes in the substitution pattern on the periphery of the molecule, at a substantial distance from the binding pocket. Herein, we explore these remote substituent effects and explain the observed behaviour through discussion of the way in which remote substituents can influence and control the global structure of a molecule through their demands upon conformational space. Peripheral modifications to a binuclear lanthanide motif derived from α,α′-bis(DO3 Ayl)-m-xylene are shown to result in dramatic changes to the binding constant for isophthalate. In this system, the parent compound displays considerable conformational flexibility, yet can be assumed to bind to isophthalate through a well-defined conformer. Addition of steric bulk remote from the binding site restricts conformational mobility, giving rise to an increase in binding constant on entropic grounds as long as the ideal binding conformation is not excluded from the available range of conformers

    Coordinated changes in mRNA turnover, translation, and RNA processing bodies in bronchial epithelial cells following inflammatory stimulation.

    Bronchial epithelial cells play a pivotal role in airway inflammation, but little is known about posttranscriptional regulation of mediator gene expression during the inflammatory response in these cells. Here, we show that activation of human bronchial epithelial BEAS-2B cells by proinflammatory cytokines interleukin-4 (IL-4) and tumor necrosis factor alpha (TNF-alpha) leads to an increase in the mRNA stability of the key chemokines monocyte chemotactic protein 1 and IL-8, an elevation of the global translation rate, an increase in the levels of several proteins critical for translation, and a reduction of microRNA-mediated translational repression. Moreover, using the BEAS-2B cell system and a mouse model, we found that RNA processing bodies (P bodies), cytoplasmic domains linked to storage and/or degradation of translationally silenced mRNAs, are significantly reduced in activated bronchial epithelial cells, suggesting a physiological role for P bodies in airway inflammation. Our study reveals an orchestrated change among posttranscriptional mechanisms, which help sustain high levels of inflammatory mediator production in bronchial epithelium during the pathogenesis of inflammatory airway diseases

    The Identification and Verification of Hazardous Convective Cells Over Oceans Using Visible and Infrared Satellite Observations

    Three algorithms based on geostationary visible and infrared (IR) observations are used to identify convective cells that do (or may) present a hazard to aviation over the oceans. The performance of these algorithms in detecting potentially hazardous cells is determined through verification with Tropical Rainfall Measuring Mission (TRMM) satellite observations of lightning and radar reflectivity, which provide internal information about the convective cells. The probability of detection of hazardous cells using the satellite algorithms can exceed 90% when lightning is used as a criterion for hazard, but the false-alarm ratio with all three algorithms is consistently large (40%), thereby exaggerating the presence of hazardous conditions. This shortcoming results in part from the algorithms’ dependence upon visible and IR observations, and can be traced to the widespread prevalence of deep cumulonimbi with weak updrafts but without lightning over tropical oceans, whose origin is attributed to significant entrainment during ascent

    Classifying LISA gravitational wave burst signals using Bayesian evidence

    We consider the problem of characterisation of burst sources detected with the Laser Interferometer Space Antenna (LISA) using the multi-modal nested sampling algorithm, MultiNest. We use MultiNest as a tool to search for modelled bursts from cosmic string cusps, and compute the Bayesian evidence associated with the cosmic string model. As an alternative burst model, we consider sine-Gaussian burst signals, and show how the evidence ratio can be used to choose between these two alternatives. We present results from an application of MultiNest to the last round of the Mock LISA Data Challenge, in which we were able to successfully detect and characterise all three of the cosmic string burst sources present in the release data set. We also present results of independent trials and show that MultiNest can detect cosmic string signals with signal-to-noise ratio (SNR) as low as ~7 and sine-Gaussian signals with SNR as low as ~8. In both cases, we show that the threshold at which the sources become detectable coincides with the SNR at which the evidence ratio begins to favour the correct model over the alternative.Comment: 21 pages, 11 figures, accepted by CQG; v2 has minor changes for consistency with accepted versio

    Role of A2B adenosine receptor signaling in adenosine-dependent pulmonary inflammation and injury.

    Adenosine has been implicated in the pathogenesis of chronic lung diseases such as asthma and chronic obstructive pulmonary disease. In vitro studies suggest that activation of the A2B adenosine receptor (A2BAR) results in proinflammatory and profibrotic effects relevant to the progression of lung diseases; however, in vivo data supporting these observations are lacking. Adenosine deaminase-deficient (ADA-deficient) mice develop pulmonary inflammation and injury that are dependent on increased lung adenosine levels. To investigate the role of the A2BAR in vivo, ADA-deficient mice were treated with the selective A2BAR antagonist CVT-6883, and pulmonary inflammation, fibrosis, and airspace integrity were assessed. Untreated and vehicle-treated ADA-deficient mice developed pulmonary inflammation, fibrosis, and enlargement of alveolar airspaces; conversely, CVT-6883-treated ADA-deficient mice showed less pulmonary inflammation, fibrosis, and alveolar airspace enlargement. A2BAR antagonism significantly reduced elevations in proinflammatory cytokines and chemokines as well as mediators of fibrosis and airway destruction. In addition, treatment with CVT-6883 attenuated pulmonary inflammation and fibrosis in wild-type mice subjected to bleomycin-induced lung injury. These findings suggest that A2BAR signaling influences pathways critical for pulmonary inflammation and injury in vivo. Thus in chronic lung diseases associated with increased adenosine, antagonism of A2BAR-mediated responses may prove to be a beneficial therapy

    Marx - From Hegel and Feuerbach to Adam Smith : a New Sysnthesis

    In The Economic and Philosophical Manuscripts of 1844 (completed in August 1844) Marx takes two crucial steps in the formation of his worldview. The first relates to his rejection of all 'old' materialism, including Feuerbach's, and the adoption of his own version, which he called 'communist', 'practical'. This view was later presented definitively in the first Thesis on Feuerbach (spring of 1845), and elaborated in The German Ideology (1845-46). The second step relates to Marx's synthesising of the philosophical standpoint that he had developed up to this point (around the spring of 1844) with political economy. Up to this point he had spoken of alienation in largely philosophical terms; now it is rooted in the process of production. On both counts Adam Smith was an important influence. This claim provides the focus of this paper. To make this point successfully I have found it necessary to briefly trace the development of Marx's philosophical standpoint up to the writing of the Manuscripts, and to distinguish between the two methodologically distinct aspects of Adam Smith's thought, the one that Marx accepted and the other that he rejected
