    Guru dalam penyampaian materi masih sering menggunakan slide pada PowerPoint dan buku. Di SMKN 1 Boyolangu pada mata pelajaran Dasar Desain Grafis masih menerapkan media pembelajaran berupa PowerPoint. Dimana para siswa terkadang belum terlalu memahami karena mereka tidak memiliki buku bahan ajar/modul dan hanya mengandalkan slide PowerPoint yang di terangkan oleh guru. Rumusan masalah dalam penelitian ini adalah “Adakah pengaruh media pembelajaran menggunakan  flipbook terhadap gaya belajar visual siswa kelas X TKI SMKN 1 Boyolanguâ€. Tujuan penelitian ini adalah mengetaui pengaruh media pembelajaran menggunakan  flipbook terhadap gaya belajar visual siswa kelas X TKI SMKN 1 Boyolangu.Jenis penelitian ini adalah penelitian kuantitatif. Metode pengumpulan data menggunakan angket kuesioner. Populasinya adalah kelas X TKI di SMKN 1 Boyolangu lalu sampelnya diambil dari kelas X TKI 1 dan 2 dengan jumlah 72 siswa.Dari penelitian yang telah dilaksanakan, penggunaan media pembelajaran flipbook terhadap gaya belajar visual siswa tidak dapat memberi pengaruh pada siswa kelas X TKI SMKN 1 Boyolangu. Kesimpulan ini berdasarkan hasil data yang diperoleh dengan nilai thitung = -1,514 dan ttabel = 1,691 dengan taraf signifikan α = 0,05, lalu di peroleh thitung ttabel atau -1,514 1,691 dan bisa di simpulkan bahwa Ho di terima. Kemudian dari kelas eksperimen di dapatkan hasil 13 siswa dengan gaya belajar visual mendapatkan rata-rata presentase sebesar dengan nilai 57,9%. Sementara dari kelas kontrol di dapatkan hasil 23 siswa dengan gaya belajar visual mendapatkan rata-rata presentase sebesar dengan nilai 58,1%. Yang artinya bahwa media pembelajaran menggunakan flipbook tidak mempengaruhi kemampuan gaya belajar visual siswa

    Tujuan penelitian yang ingin dicapai adalah untuk mengetahuibagaimana pelaksanaan dan hasil dari pelaksanaan layanan bimbingan klasikal dalam mengatasi perilaku prokrastinasi akademik peserta didik SMA Negeri 5 Metro Tahun Pelajaran 2019/2020. Jenis penelitian ini menggunakan rancangan pendekatandeskriptif kualitatif, yaitu penelitian yang menekankan pada data yang telah terkumpul berbentuk kata, gambar dan bukan angka. Penelitian ini membahas layanan bimbingan klasikal (X) dan prokrastinasi akademik (Y) tujuan utama dalam penelitian adalah mendapatkan data, dan merupakan pekerjaan yang penting dalam langkah penelitian tersebut. Metode pengumpulan data dalam penelitian ini adalah metode wawancara dan metode observasi. Berdasarkan hasil penelitian, hasil layanan bimbingan klasikal dalam mengurangi perilaku prokrastinasi peserta didik yaitu layanan klaikal yang diberikan guru Bimbingan dan Konseling memberikan pengaruh positif terhadap peserta didik yang mencerminkan perilaku prokrastinasi akademik

    Immune-mediated loss of transgene expression from virally transduced brain cells is irreversible, mediated by IFNγ, perforin, and TNFα, and due to the elimination of transduced cells

    The adaptive immune response to viral vectors reduces vector-mediated transgene expression from the brain. It is unknown, however, whether this loss is caused by functional downregulation of transgene expression or death of transduced cells. Herein, we demonstrate that during the elimination of transgene expression, the brain becomes infiltrated with CD4 and CD8 T cells and that these T cells are necessary for transgene elimination. Further, the loss of transgene-expressing brain cells fails to occur in the absence of IFNγ, perforin, and TNFα receptor. Two methods to induce severe immune suppression in immunized animals also fail to restitute transgene expression, demonstrating the irreversibility of this process. The need for cytotoxic molecules and the irreversibility of the reduction in transgene expression suggested to us that elimination of transduced cells is responsible for the loss of transgene expression. A new experimental paradigm that discriminates between downregulation of transgene expression and the elimination of transduced cells demonstrates that transduced cells are lost from the brain upon the induction of a specific antiviral immune response.     Pengolahan Eceng Gondok (Eichornia crassipes) Menjadi Pupuk Organik di Kelurahan Panjisari Kecamatan Praya, Kabupaten Lombok Tengah

    Panjisari merupakan desa yang terletak di sekitar genangan dan daerah aliran sungai Bendungan Batujai. Di bendungan ini, gulma eceng gondok (Eichhornia crassipes) tumbuh pesat yang berdampak pada pemanfaatan perairan dan pendangkalan waduk. Salah satu upaya untuk mengatasi masalah tersebut adalah dengan memanfaatkan eceng gondok sebagai pupuk. Pupuk organik dari eceng gondok mampu memperbaiki struktur fisik tanah, meningkatkan ketersediaan unsur hara, dan pertumbuhan vegetatif. Tim KKN menjadikan kegiatan pembuatan pupuk ini sebagai program utama mereka. Program ini dilaksanakan dengan memberikan bantuan kepada kelompok masyarakat mulai Desember 2021 hingga Februari 2022 bertempat di rumah produksi. Proses pembuatannya dimulai dengan persiapan alat, pengambilan bahan (eceng gondok, kotoran sapi dan sekam), pencacahan dan pengeringan, pencampuran dan fermentasi, yang terakhir adalah pengemasan pupuk. Tim KKN berhasil membuat pupuk organik dengan bahan utama eceng gondok dan memberikan edukasi cara membuatnya kepada masyarakat. Selain itu, produk pupuk organik eceng gondok dapat dipasarkan secara luas, sehingga menciptakan peluang usaha baru untuk meningkatkan perekonomian masyarakat Panjisari

    Identification and Visualization of CD8+ T Cell Mediated IFN-γ Signaling in Target Cells during an Antiviral Immune Response in the Brain

    CD8+ T cells infiltrate the brain during an anti-viral immune response. Within the brain CD8+ T cells recognize cells expressing target antigens, become activated, and secrete IFNγ. However, there are no methods to recognize individual cells that respond to IFNγ. Using a model that studies the effects of the systemic anti-adenoviral immune response upon brain cells infected with an adenoviral vector in mice, we describe a method that identifies individual cells that respond to IFNγ. To identify individual mouse brain cells that respond to IFNγ we constructed a series of adenoviral vectors that contain a transcriptional response element that is selectively activated by IFNγ signaling, the gamma-activated site (GAS) promoter element; the GAS element drives expression of a transgene, Cre recombinase (Ad-GAS-Cre). Upon binding of IFNγ to its receptor, the intracellular signaling cascade activates the GAS promoter, which drives expression of the transgene Cre recombinase. We demonstrate that upon activation of a systemic immune response against adenovirus, CD8+ T cells infiltrate the brain, interact with target cells, and cause an increase in the number of cells expressing Cre recombinase. This method can be used to identify, study, and eventually determine the long term fate of infected brain cells that are specifically targeted by IFNγ. The significance of this method is that it will allow to characterize the networks in the brain that respond to the specific secretion of IFNγ by anti-viral CD8+ T cells that infiltrate the brain. This will allow novel insights into the cellular and molecular responses underlying brain immune responses

    A review on Numbered Head Together (NHT) learning model to improve biology learning outcomes

    This study aims to analyze student learning outcomes in the NHT model on respiratory system material. This research uses library data collection methods, reading and recording research materials (library research). Data collection used is through documentation study with data collection stages by editing and organizing. Data were analyzed using inductive and deductive techniques. The results of the study: 1) student learning outcomes showed an increase in student learning outcomes using the NHT model, 2) strategies in improving learning outcomes consisted of syntax, namely numbering, asking questions, thinking together and answering questions, 3) there were student learning outcomes before the model was applied. learning is low and after the application of the NHT model has increased, 4) the effectiveness of the NHT model is effective in improving student learning outcomes, 5) the use of the NHT model can provide opportunities for students to come up with ideas of knowledge in asking and arguing

    Abstract: Research product development aims to develop physical fitness learning tools based on the articulate storyline application for the KKG PJOK SD in Purwosari District, Pasuruan Regency. This study uses the Research and Development (R&D) method or development research. The subjects for the small group trial involved 8 primary school Physical Education and Health teachers, and the large group trial subjects involved 26 Physical Education, Health and Sports teachers in Purwosari District. The results obtained are the percentage of learning experts 90%, elementary game experts 96%, PJOK experts 77%, media experts 96%, Physical fitness experts 78%, then 91% for trial I and 89% for trial II. The conclusion in this development research is that the articulate storyline application development product is feasible to be used as a support in learning physical fitness. Abstrak: Penelitian produk pengembangan yaitu bertujuan untuk mengembangkan perangkat pembelajaran kebugaran jasmani berbasis aplikasi articulate storyline untuk KKG PJOK SD di Kecamatan Purwosari, Kabupaten Pasuruan. Penelitian ini menggunakan metode Research and Development (R&D) atau penelitian pengembangan. Subjek untuk uji coba kelompok kecil melibatkan 8 guru Pendidikan Jasmani Olahraga dan Kesehatan sekolah dasar, dan subjek uji coba kelompok besar melibatkan 26 guru Pendidikan Jasmani, Kesehatan dan Olahraga di Kecamatan Purwosari.  Didapatkan hasil data yaitu dengan persentase ahli pembelajaran 90%, Ahli permainan SD 96%, Ahli PJOK 77%, Ahli Media 96%, Ahli Kebugaran Jasmani 78%, kemudian 91% untuk uji coba I dan 89% untuk uji coba II. Kesimpulan dalam penelitian pengembangan ini yaitu produk pengembangan aplikasi articulate storyline ini layak untuk dipergunakan sebagai penunjang dalam pembelajaran kebugaran jasmani

    Temozolomide Does Not Impair Gene Therapy-Mediated Antitumor Immunity In Syngeneic Brain Tumor Models

    PURPOSE: Glioblastoma multiforme is the most common primary brain cancer in adults. Chemotherapy with temozolomide (TMZ) significantly prolongs the survival of patients with glioblastoma multiforme. However, the three-year survival is still approximately 5%. Herein, we combined intratumoral administration of an adenoviral vector expressing Flt3L (Ad-Flt3L) with systemic temozolomide to assess its impact on therapeutic efficacy. EXPERIMENTAL DESIGN: Wild-type or immunodeficient mice bearing intracranial glioblastoma multiforme or metastatic melanoma were treated with an intratumoral injection of Ad-Flt3L alone or in combination with the conditionally cytotoxic enzyme thymidine kinase (Ad-TK), followed by systemic administration of ganciclovir and temozolomide. We monitored survival and measured the tumor-infiltrating immune cells. RESULTS: Although treatment with temozolomide alone led to a small improvement in median survival, when used in combination with gene therapy-mediated immunotherapy, it significantly increased the survival of tumor-bearing mice. The antitumor effect was further enhanced by concomitant intratumoral administration of Ad-TK, leading to 50% to 70% long-term survival in all tumor models. Although temozolomide reduced the content of T cells in the tumor, this did not affect the therapeutic efficacy. The antitumor effect of Ad-Flt3L+Ad-TK+TMZ required an intact immune system because the treatment failed when administered to knock out mice that lacked lymphocytes or dendritic cells. CONCLUSIONS: Our results challenge the notion that chemotherapy leads to a state of immune-suppression which impairs the ability of the immune system to mount an effective antitumor response. Our work indicates that temozolomide does not inhibit antitumor immunity and supports its clinical implementation in combination with immune-mediated therapies.Fil: Candolfi, Marianela.
