    Improved sequential injection method for phosphate quantification within a wide dynamic range with in-line pre-concentration to monitor soil leachates

    Phosphate is a well-known contaminant and its content is an indicator of water quality, so it is important to have an efficient monitoring of the soil leaching process. In this work, an automatic, low cost flow analysis method, capable of real-time monitoring of the soil leaching processes using spectrophotometric detection, based on the molybdenum blue reaction, was developed. The developed methodology for phosphate determination was based on the molybdenum blue reaction and includes an on-line solid phase extraction (SPE) step, involving an AG1-X8 anionic exchange sorbent. The described SI method enabled phosphate determination within a wide range, 1–80 µM, with a detection limit of 0.52 µM. It was successfully applied to leachates from laboratory scale soil columns (LSSC) and one sample analysis was carried out in triplicate, in less than 10 minutes.info:eu-repo/semantics/publishedVersio

    A review on sequential injection methods for water analysis

    The development of fast, automatic and less expensive methods of analysis has always been the main aim of flowmethodologies. The search for newprocedures that still maintain the reliability and accuracy of the reference procedures is an ever growing challenge. Newrequirements are continually added to analytical methodologies, such as lower consumption of samples and reagents, miniaturisation and portability of the equipment, computer interfaces for full decision systems and so on. Therefore, the development of flow methodologies meeting the extra requirements of water analysis is a challenging work. Sequential injection analysis (SIA) presents a set of characteristics that make it highly suitable for water analysis. With sequential injection analysis, most routine determinations in waters can be performed more quickly with much lower reagent consumption when compared to reference procedures. Estes podem ser classificados de acordo com as suas caraterísticas físicas e bioquímicas, tais como o tamanho, morfologia, genoma e tipo de replicação. Na presente monografia, são apresentados vários mecanismos de entrada viral, bem como exemplos de vírus que usam estes mecanismos, as patologias associadas a estes e estratégias terapêuticas direcionadas para a infeção dos mesmos. Serão abordados os mecanismos de Fusão, Macropinocitose, Lipid Rafts, Endocitose mediada por Caveolina e por Clatrina e a entrada viral através dos Exossomas. Para cada mecanismo viral, a título representativo, serão abordados os vírus Influenza, HIV, Semliki Forest Virus, Epstein-Barr, Papiloma Vírus Humano, Vírus Símio 40 e Vírus da Hepatite C, apresentando para cada um deles terapias atuais e emergentes. Viruses are obligate intracellular parasites that depend on the host cell so that their replication and infection occur. These can be classified according to their physical and biochemical characteristics such as size, morphology, genome and replication type. This monograph lists various viral entry mechanisms as well as examples of viruses that use these mechanisms, the pathologies associated and therapeutic strategies directed to infection thereof. The fusion mechanisms will be addressed as well as macropinocytosis, lipid rafts, endocytosis mediated by caveolin and clathrin and viral entry through exosomes. For each viral mechanism, will be discussed a few viruses like Influenza Virus, HIV, Semliki Forest Virus , Epstein- Barr, Human Papillomavirus , Simian Virus 40 and Hepatitis C Virus , presenting for each one, current and emerging therapies

    Trabalho de Conclusão de Curso (graduação)—Universidade de Brasília, Faculdade de Comunicação, 2019.Este trabalho conta a história de criação do produto curta documentário “O Sentido do Like”. A produção que tem por objetivo oferecer, ao público alvo, subsídios para uma reflexão quanto a sobrevalorização da interação positiva nas redes sociais e sua influência na auto percepção de meninas adolescentes. Essa reflexão surge a partir de uma experiência de pesquisa, sobre a importância e significado dos likes do Instagram para meninas adolescentes. A construção desse projeto passou por um longo período de pré-produção, onde foi realizada uma pesquisa no Centro Educacional do Lago, com o objetivo de embasar as reflexões propostas, e também uma oficina com adolescentes que corroboraram para reflexão e construção deste produto. Após esse período, foi realizada a produção, com gravações realizadas dentro da escola com as alunas que participaram da oficina, e também no estúdio de gravação da Estação Brasileira. O período de pós produção e edição do curta documentário foi mais um período de criação buscando criar pontos de conexão com o público alvo. Toda reflexão suscitada trouxe o entendimento de que os Likes da rede social Instagram, tanto na perspectiva de quem recebe o like, quanto na de quem oferta, possui forte vínculo com a aceitação estética, o que impacta a auto percepção e a segurança especialmente de meninas adolescentes.This work is about the history behind the creation of the short documentary titled “O Sentido do Like”. The project’s objective is to offer, to the target audience, allowance for a reflection of how overvalued are the positive interactions on the social media and how these interactions can influence the self- perception of teenagers. This reflection actually arises from a research with respect to the relevance of likes on Instagram for a selected group of adolescent girls. For that matter, the project’s construction has passed across a long stage of pre-production at the school Centro Educacional do Lago, where the research was actually conducted. The objectives of all this pre-production were to substantiate certain reflections and to devise a work-shop with the group of adolescents. In the aftermath of the preparation period, the production effectively started with video recordings inside the school with the selected group of adolescents mentioned before and at the recording studio of Estação Brasileira as well. The period of pos-production and editions of the short-documentary was driven to find a good connection with the target audience. Lastly, the conclusion highlights the understanding that the “likes” of the Instagram social media are closely linked with aesthetical acceptance, either from the person that receive the “likes” or the person that gives it. Which, in the end, strongly impacts the self-percepction and the security especially of adolescent girls

    Identidades e Economia : valorizando o papel de subjetividades não-hegemônicas na pesquisa acadêmica e nas vivências cotidianas de trabalhadoras lésbicas e bissexuais

    Dissertação (mestrado)—Universidade de Brasília, Faculdade de Economia, Administração e Contabilidade, Departamento de Economia, Programa de Pós-Graduação em Ciências Econômicas, 2020.O objetivo deste trabalho é discutir o papel das identidades na pesquisa acadêmica e nas relações econômicas. Para isso, é preciso levar as identidades a sério, sendo necessário, como pré-requisito, rejeitar a objetividade da Economia Positiva. Para fazê-lo, este trabalho discute as duas metodologias consideradas pelo mainstream como vetores da objetividade: o refutacionismo popperiano e o formalismo matemático. Defende-se que a metodologia popperiana se assenta sobre a epistemologia do mesmo autor, a qual é apresentada no segundo capítulo. Já o formalismo é trabalhado no quarto capítulo, em que se questiona o seu uso retórico, usando como base teórica McCloskey (1994). Em substituição à filosofia modernista da Economia, adota-se uma abordagem Pós-Moderna, valendo-se, principalmente, do pensamento de Lyotard (2013 [1979]) e Foucault (1984). Com isso, a ideia de que existe uma referencial de verdade estável é questionada em nome de verdades contexto-relativas. Se há diversas verdades, perspectivas não-ortodoxas são igualmente legítimas, o que leva à valorização do Feminismo e da Teoria Queer na Economia. Para ilustrar empiricamente o uso da abordagem Pós-Moderna na Economia, foram feitas uma pesquisa quantitativa e outra qualitativa, ambas abordando a temática da homofobia no mercado de trabalho. Na parte quantitativa, foram feitas regressões econométricas usando o Método de Mínimos Quadrados (MQO) e o Modelo de Dois Estágios de Heckman de correção de viés de seleção a partir de dados do Censo 2010. Já na parte qualitativa, é usada a abordagem de Gergen e Gergen (1988) para análise de narrativas. Essas narrativas foram coletadas por meio do método de entrevista em profundidade mesclado com a técnica de grupo focal. Pela parte qualitativa do trabalho, conclui-se que o PósModernismo contribui não só teórica, mas, sobretudo, empiricamente para o estudo de problemas econômicosCoordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) e Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq).This dissertation aims to discuss identities’ role on academic research and on economic interaction in daily lives. To accomplish that, we must take identities seriously, and, in order to do that, first of all, it is quite essential to reject the assumption of Positive Economics’ objectivity. To make that discussion, the research presents two methodologies considered by the mainstream as the main vectors of objectivity on Economics: the Popperian refutationism and the mathematical formalism. Here, it is advocated Popper’s methodology is based on the epistemology of the same philosopher, which is presented on the second chapter. On the other hand, the mathematical formalism is presented on the fourth chapter, where it is questioned his rhetorical purpose, using as theorical support McCloskey (1994). Instead of modernist philosophy on Economics, it is adopted a Postmodern approach, based, mainly, on Lyotard’s (2013 [1979]) and Foucault’s (1984) theories. Therefore, the assumption that exist a stable truth referential is questioned on behalf of contextual truths. If there are many truths, instead of only one, nonorthodox perspectives become as legitimate as the orthodox one. Thereby, Feminism and Queer Theory on Economics can be revalorized. In order to illustrate the Postmodern approach on Applied Economics, on the last chapter, it is presented two studies about homophobia on labor market: the first of them, it is based on quantitative method; the second, it is based on qualitative method. On the quantitative part, it was used the Brazilian Census 2010 to estimate two regressions using, respectively, Ordinary Least Squares (OLS) and Heckman’s Two Stages Method of Selection Bias Correction. On the qualitative section, it has been used Gergen & Gergen’s (1988) self-narratives method to analyze interviews from lesbian and bisexual female workers, collected using deep interview and focal group techniques. Based on the qualitative section of the applied research, it is possible to conclude that Postmodernism contributed both theoretically and, foremost, empirically to economic issues investigations

    Leaders' voices on curriculum and curricular flexibility

    The responsibilities of teachers as curriculum managers are today a challenge given the curricular flexibility. These challenges stem from the discourse of autonomy and the capacity for collaborative deliberation of teachers, which refers to decisions at the level of educational policy, which can be operated in the transformation of schools oriented to the success and meaningful learning of students. This article aims to present a part of a more comprehensive project entitled "Beliefs, knowledge, and practices of teachers". It analyzes the voice of the leaders of eleven groups of schools of the Intermunicipal Community of Lands of Trás-os-Montes (CIM-TTM), Portugal. The study follows a qualitative methodology, using group interviews with school principals, department coordinators, and class directors as a data collection technique. From the content analysis on the school, category emerged the following subcategories: meanings of curriculum and curriculum management. The data reveal that participants associate the concept of the curriculum with knowledge, disciplinary content, experience, learning, and school. The influence of the hidden curriculum in the teaching and learning process was perceived, and its political connotation and utopian sense. Regarding the management of the curriculum, the teacher emerged in the participant's conceptions as an executor, a builder, and a decision-maker. The curriculum was specifically recognized as guidance and control over compliance with the program. In the context of curricular flexibility, the: (i) the adequacy to specific contexts was emphasized; (ii) the integration of the regional or local component of the curriculum; (iii) hope for curricular flexibility: recognition of potentialities and limitations; and (iv) the discrepancy between design and implementation.     Gas diffusion sequential injection system for the spectrophotometric determination of free chlorine with o-dianisidine

    A gas diffusion sequential injection system for spectrophotometric determination of free chlorine is described. The detection is based in the colorimetric reaction between free chlorine and a low toxicity reagent o-dianisidine. A gas diffusion unit is used to isolate free chlorine from the sample in order to avoid possible interferences. This feature results from the conversion of free chlorine to molecular chlorine (gaseous) with sample acidification. With minor changes in the operating conditions, two different dynamic ranges were obtained enhancing the application both to water samples and bleaches. The results obtained with the developed system were compared to the reference method, iodometric titration and proved not to be statistically different. A detection limit of 0.6 mg ClO−/L was achieved. Repeatability was evaluated from 10 consecutive determinations being the results better than 2%. The two dynamic ranges presented different determination rates: 15 h−1 for 0.6–4.8 mg ClO−/L (water samples) and 30 h−1 for 0.047–0.188 g ClO−/L (bleaches).info:eu-repo/semantics/acceptedVersio
