    Tracking of physical activity during adolescence: The 1993 Pelotas Birth Cohort, Brazil

    Objective: To analyze physical activity during adolescence in participants of the 1993 Pelotas Birth Cohort Study, Brazil. Methods: Data on leisure time physical activity at 11, 15, and 18 years of age were analyzed. At each visit, a cut-off point of 300 min/week was used to classify adolescents as active or inactive. A total of 3,736 participants provided data on physical activity at each of the three age points. Results: A significant decline in the proportion of active adolescents was observed from 11 to 18 years of age, particularly among girls (from 32.9% to 21.7%). The proportions of girls and boys who were active at all three age points were 28.0% and 55.1%, respectively. After adjustment for sex, economic status, and skin color, participants who were active at 11 and 15 years of age were 58.0% more likely to be active at 18 years of age compared with those who were inactive at 11 and 15 years of age. Conclusions: Physical activity declined during adolescence and inactivity tended to track over time. Our findings reinforce the need to promote physical activity at early stages of life, because active behavior established early tends to be maintained over time

    Longitudinal analysis of palatal volume in unilateral cleft lip and palate children

    Cleft lip and/or palate are among the most frequent congenital craniofacial defects, which every year affect one in 500-700 newborn worldwide (1,2). The aim of this study was to analyze the effect of orthopaedic and surgical treatments on the palatal size and shape of patients with unilateral cleft lip and palate (UCLP). Ninetysix palatal casts from 32 neonatal patients, attending the Fundacion Clinica de Noel (Colombia) were analyzed through a stereophotogrammetric system. The analysis was carried out before (mean age 10.5 days, SD 4.8) and after (mean age 83.3 days, SD 6.6) orthopaedic treatment (performed with acrylic plates) and after cheiloplasty (mean age 317.1 days, SD 44.2). Volumes of the greater and the minor alveolar segments were evaluated through a new measurement protocol. Intra and inter operator repeatability was evaluated using paired Student\u2019s t test. In order to investigate differences between alveolar segments and time, volume measurements were compared with a repeated two-way analysis of variance (ANOVA). No significant differences between repetitions, both intra and inter operator, were found (p>0.05). Random errors explained 3.7% of the sample variance. On the other hand, significant differences in volume measurements were found both in alveolar segment and time (p<0.01). Before orthopaedic treatment, the smaller palatal segment had a mean volume of 0.52 cm3 (SD 0.23), and the greater of 0.9 cm3 (SD 0.40); after orthopaedic treatment, the mean volumes were 0.58 cm3 (SD 0.25), and 1.09 cm3 (SD 0.43). After surgery, mean values of 0.73 cm3 (SD 0.28) and 1.31 cm3 (SD 0.52) were measured. Results suggest that a three-dimensional stereophotogrammetric system is a repeatable and reliable method to evaluate palatal casts of patients with UCLP. Obtained data offer a preliminary quantitative information about the changes occurring in maxillary arches of UCLP patients after orthopaedic treatment and surgery. Further investigation is required to increase the frequency of evaluations and the number of patients

    Distribution of Glycated Haemoglobin According to Early-Life and Contemporary Characteristics in Adolescents and Adults without Diabetes:The 1982 and 1993 Pelotas Birth Cohorts

    AIM:Glycated haemoglobin (HbA1c), a marker of glucose control in individuals with diabetes mellitus, is also related with the incidence of cardiometabolic risk in populations free of disease. The aim of this study was to describe the distribution of HbA1c levels according to early-life and contemporary factors in adolescents and adults without diabetes mellitus. METHODS:HbA1c was measured in adults aged 30 years and adolescents aged 18 years who are participants in the 1982 and 1993 Pelotas Birth Cohorts, respectively. Bivariate and multivariate analyses were performed to describe the HbA1c mean values according to early-life and contemporary characteristics collected prospectively since birth. RESULTS:The distribution of the HbA1c was approximately normal in both cohorts, with a mean (SD) 5.10% (0.43) in the 1982 cohort, and 4.89% (0.50) in the 1993 cohort. HbA1c mean levels were significantly higher in individuals self-reported as black/brown skin color compared to those self-reported as white in both cohorts. Parental history of diabetes was associated with higher HbA1c mean in adults, while stunting at one year old presented an inverse relation with the outcome in adolescents. No other early and contemporary factors were associated with HbA1c levels in adults or adolescents. CONCLUSIONS:We found a consistent relationship between HbA1c and skin color in both cohorts. Further research is needed to understand the role of genomic ancestry on levels of HbA1c concentrations which may inform policies and preventive actions for diabetes mellitus and cardiometabolic risk

    Imbalance of naive and memory T lymphocytes with sustained high cellular activation during the first year of life from uninfected children born to HIV-1-infected mothers on HAART

    The immune consequences of in utero HIV exposure to uninfected children whose mothers were submitted to highly active antiretroviral therapy (HAART) during gestation are not well defined. We evaluated 45 HIV-exposed uninfected (ENI) neonates and 45 healthy unexposed control (CT) neonates. All HIV-infected mothers received HAART during pregnancy, and the viral load at delivery was <50 copies/mL for 56.8%. Twenty-three ENI neonates were further evaluated after 12 months and compared to 23 unexposed healthy age-matched infants. Immunophenotyping was performed by flow cytometry in cord and peripheral blood. Cord blood lymphocyte numbers did not differ between groups. However, ENI neonates had a lower percentage of naive T cells than CT neonates (CD4+, 76.6 vs 83.1%, P < 0.001; CD8+, 70.9 vs 79.6%, P = 0.003) and higher percentages of central memory T cells than CT neonates (CD4+, 13.9 vs 8.7%, P < 0.001; CD8+, 8.6 vs 4.8%, P = 0.001). CD38 mean fluorescence intensity of T cells was higher in ENI neonates (CD4+, 62.2 vs 52.1, P = 0.007; CD8+, 47.7 vs 35.3, P < 0.001). At 12 months, ENI infants still had higher mean fluorescence intensity of CD38 on T cells (CD4+, 34.2 vs 23.3, P < 0.001; CD8+, 26.8 vs 19.4, P = 0.035). Despite effective maternal virologic control at delivery, HIV-exposed uninfected children were born with lower levels of naive T cells. Immune activation was present at birth and remained until at least 12 months of age, suggesting that in utero exposure to HIV causes subtle immune abnormalities.FAPESPCoordenacao de Aperfeicoamento de Pessoal de Nivel Superior (CAPES

    Influence of the scrotum bipartite on the reproductive parameters in goats

    Avaliou-se a influência do grau de bipartição escrotal sobre os parâmetros reprodutivos em caprinos. Foram utilizados 30 animais, distribuídos em três grupos: Grupo I, animais que apresentavam escroto sem bipartição (n =10); Grupo II, animais com bipartição escrotal até 50% do comprimento dos testículos (n=10); e Grupo III, caprinos com bipartição escrotal acima de 50% do comprimento testicular (n=10). Os parâmetros avaliados foram peso corporal, perímetro escrotal, comprimento testicular, características físicas e morfológicas do sêmen, concentração plasmática de testosterona e libido dos animais, comparando caprinos com escroto bipartido e não bipartido. Os animais que apresentavam bipartição escrotal expressaram maior peso corporal e comprimento testicular, quando comparados com os animais do grupo sem escroto bipartido, sendo que estes apresentaram maior número de células espermáticas defeituosas. Conclui-se que a bipartição escrotal influencia positivamente alguns parâmetros biométricos bem como a qualidade do sêmen em caprinos.The influence of scrotum division on the reproductive parameters in goats was evaluated. Thirty animals were divided into three groups of 10 animals each as follows. Group I: animals with simple scrotum (n=10); Group II: animals with 50% of scrotum bipartite (n=10); and Group III: animals with more than 50% of scrotum bipartite (n=10). It was evaluated: body weight, scrotum perimeter and the testicular length, the seminal characteristic, plasmatic concentration of testosterone, and the libido of the animals with different degrees of scrotum division. Animals with divided scrotum had higher weight and testicular length when compared with animals from the first group that had a higher percentage of spermatic defects. It is possible to conclude that there was a positive influence of scrotal bipartition on the biometric parameters and the sperm quality in goats

    Chemopreventive targeted treatment of head and neck precancer by Wee1 inhibition

    HPV-negative head and neck squamous cell carcinomas (HNSCCs) develop in precancerous changes in the mucosal lining of the upper-aerodigestive tract. These precancerous cells contain cancer-associated genomic changes and cause primary tumors and local relapses. Therapeutic strategies to eradicate these precancerous cells are very limited. Using functional genomic screens, we identified the therapeutic vulnerabilities of premalignant mucosal cells, which are shared with fully malignant HNSCC cells. We screened 319 previously identified tumor-lethal siRNAs on a panel of cancer and precancerous cell lines as well as primary fibroblasts. In total we identified 147 tumor-essential genes including 34 druggable candidates. Of these 34, 13 were also essential in premalignant cells. We investigated the variable molecular basis of the vulnerabilities in tumor and premalignant cell lines and found indications of collateral lethality. Wee1-like kinase (WEE1) was amongst the most promising targets for both tumor and precancerous cells. All four precancerous cell lines were highly sensitive to Wee1 inhibition by Adavosertib (AZD1775), while primary keratinocytes tolerated this inhibitor. Wee1 inhibition caused induction of DNA damage during S-phase followed by mitotic failure in (pre)cancer cells. In conclusion, we uncovered Wee1 inhibition as a promising chemopreventive strategy for precancerous cells, with comparable responses as fully transformed HNSCC cells
