G protein beta gamma subunits synthesized in Sf9 cells. Functional characterization and the significance of prenylation of gamma
Heterotrimeric guanine nucleotide-binding regulatory proteins (G proteins) consist of a nucleotide-binding alpha subunit and a high- affinity complex of beta and gamma subunits. There is molecular heterogeneity of beta and gamma, but the significance of this diversity is poorly understood. Different G protein beta and gamma subunits have been expressed both singly and in combinations in Sf9 cells. Although expression of individual subunits is achieved in all cases, beta gamma subunit activity (support of pertussis toxin-catalyzed ADP-ribosylation of rGi alpha 1) is detected only when beta and gamma are expressed concurrently. Of the six combinations of beta gamma tested (beta 1 or beta 2 with gamma 1, gamma 2, or gamma 3), only one, beta 2 gamma 1, failed to generate a functional complex. Each of the other five complexes has been purified by subunit exchange chromatography using Go alpha-agarose as the chromatographic matrix. We have detected differences in the abilities of the purified proteins to support ADP- ribosylation of Gi alpha 1; these differences are attributable to the gamma component of the complex. When assayed for their ability to inhibit calmodulin-stimulated type-I adenylylcyclase activity or to potentiate Gs alpha-stimulated type-II adenylylcyclase, recombinant beta 1 gamma 1 and transducin beta gamma are approximately 10 and 20 times less potent, respectively, than the other complexes examined. Prenylation and/or further carboxyl-terminal processing of gamma are not required for assembly of the beta gamma subunit complex but are indispensable for high affinity interactions of beta gamma with either G protein alpha subunits or adenylylcyclases
The Physics Inside Topological Quantum Field Theories
We show that the equations of motion defined over a specific field space are
realizable as operator conditions in the physical sector of a generalized Floer
theory defined over that field space. The ghosts associated with such a
construction are found not to be dynamical. This construction is applied to
gravity on a four dimensional manifold, ; whereupon, we obtain Einstein's
equations via surgery, along , in a five-dimensional topological quantum

Co-expression of Gbeta 5 Enhances the Function of Two Ggamma Subunit-like Domain-containing Regulators of G Protein Signaling Proteins
Regulators of G protein signaling (RGS) stimulate the GTPase activity of G protein Galpha subunits and probably play additional roles. Some RGS proteins contain a Ggamma subunit-like (GGL) domain, which mediates a specific interaction with Gbeta 5. The role of such interactions in RGS function is unclear. RGS proteins can accelerate the kinetics of coupling of G protein-coupled receptors to G-protein-gated inwardly rectifying K+ (GIRK) channels. Therefore, we coupled m2-muscarinic acetylcholine receptors to GIRK channels in Xenopus oocytes to evaluate the effect of Gbeta 5 on RGS function. Co-expression of either RGS7 or RGS9 modestly accelerated GIRK channel kinetics. When Gbeta 5 was co-expressed with either RGS7 or RGS9, the acceleration of GIRK channel kinetics was strongly increased over that produced by RGS7 or RGS9 alone. RGS function was not enhanced by co-expression of Gbeta 1, and co-expression of Gbeta 5 alone had no effect on GIRK channel kinetics. Gbeta 5 did not modulate the function either of RGS4, an RGS protein that lacks a GGL domain, or of a functional RGS7 construct in which the GGL domain was omitted. Enhancement of RGS7 function by Gbeta 5 was not a consequence of an increase in the amount of plasma membrane or cytosolic RGS7 protein
NAFTA Numbers Don\u27t Add Up
On November 17,1993, the U.S. Congress passed the North American Free Trade Agreement, which became the law of the land in Canada, Mexico, and the United States on January 1, 1994. Despite the passage of over a year, there has been precious little discussion of the economic results of the free trade agreement. In fact, until the recent devaluation of the Mexican peso, proponents of NAFTA had claimed that the matter was settled and not worthy of further discussion or investigation. It was a fait accompli, they said; time to move on to other, more pressing social and political issues. The irony of this thinly disguised attempt to stifle public inquiry is that only now—one year after the passage of NAFTA—can its impact be tested in the field and reality substituted for rhetoric
Radiationless Travelling Waves In Saturable Nonlinear Schr\"odinger Lattices
The longstanding problem of moving discrete solitary waves in nonlinear
Schr{\"o}dinger lattices is revisited. The context is photorefractive crystal
lattices with saturable nonlinearity whose grand-canonical energy barrier
vanishes for isolated coupling strength values. {\em Genuinely localised
travelling waves} are computed as a function of the system parameters {\it for
the first time}. The relevant solutions exist only for finite velocities.Comment: 5 pages, 4 figure
Gravitating Fluxbranes
We consider the effect that gravity has when one tries to set up a constant
background form field. We find that in analogy with the Melvin solution, where
magnetic field lines self-gravitate to form a flux-tube, the self-gravity of
the form field creates fluxbranes. Several exact solutions are found
corresponding to different transverse spaces and world-volumes, a dilaton

Soluble models in 2d dilaton gravity
A one-parameter class of simple models of two-dimensional dilaton gravity,
which can be exactly solved including back-reaction effects, is investigated at
both classical and quantum levels. This family contains the RST model as a
special case, and it continuously interpolates between models having a flat
(Rindler) geometry and a constant curvature metric with a non-trivial dilaton
field. The processes of formation of black hole singularities from collapsing
matter and Hawking evaporation are considered in detail. Various physical
aspects of these geometries are discussed, including the cosmological
