    An Investigation into the Conversion of Non-Hazardous Medical Wastes into Biogas—A Case Study from the Health and Family Planning Sector in Bangladesh

    This study aims to investigate the potential scope of rural bioenergy production from the valorisation of non-hazardous waste, particularly from institutional health and family planning facilities. A crude bioenergy production potential will be determined, as well as relevant interrelationships with food security, equality and inclusivity, poverty reduction, and their contribution to achieving sustainable development goals (SDGs). Solid waste generation at LAMB Hospital in Bangladesh was assessed. Its energy yield capacity and replacement of conventional, inefficient fuel sources were estimated, analysed, and specified. Results showed that the biogas yield capacity of non-hazardous solid waste from the LAMB hospital is 62 m3, which is equivalent to the gas required to cook 31 family meals. From small-to-medium institutes, such as the LAMB hospital, derived waste has the capacity to meet the cooking energy demand of rural villages. The electrical yield capacity of this amount of gas is 124 kWh, and this amount of energy can replace the use of 41 tonnes of firewood that cause emissions, deforestation, and increases the propensity for flooding. The adoption of Anaerobic Digestion (AD) for biogas production also has an impact on waste management, stimulating improved governance and infrastructure and supporting agriculture through the production of nutrient-dense biofertiliser. Further analysis showed that biogas not only provides a clean and affordable fuel but, in turn, has benefits with regard to health issues. Emissions from traditional cooking fuels have deleterious impacts on health due to Indoor Air Pollution (IAP), which generally affects women to a greater extent. All of these factors contribute to achieving sustainable development goals. This study could help to reduce environmental pollution from the health sector and mitigate climate change impacts, and there is a potential for scaling up this study nationwide

    Determination of the potential impact of domestic Anaerobic Digester Systems: a community based research initiative in rural Bangladesh

    Abstract : This research examines the potential impact of domestic anaerobic digester (AD) systems adopted in Bangladesh and similar developing countries. Cattle dung and poultry litter feed stocks were specifically investigated, because these were freely available and plentiful to people living within agricultural areas of rural Bangladesh. Data was collected to ascertain whether these two representative AD facility types provide tangible social, economic and environmental impact that benefits homeowners. Primary quantitative and qualitative data was obtained by field data collection, and meeting with expert groups and stakeholders. Empirical analysis conducted revealed that variations were found in the biomass feedstocks available on different sites but also differences were apparent in terms of the operations and maintenance (O and M) systems of the biogas plants operated. The biogas and methane yield variation was also measured, and variations were found in the cattle dung and poultry litter AD yield capacity. Overall, 64% of feedstock was utilised, 91% of biogas plants remain underfed and energy yield efficiency was 57% from cattle smallholdings’ AD and 28% from poultry farms’ AD. These results showed that small scale AD can offer a significant impact upon rural lifestyles through augmented economics, improved social activities, relationship building with neighbours and improved lifestyle achieved via time savings accrued. These results could help rural entrepreneurs, AD equipment providers and government institutions to develop a road map to implement future AD installation on a much wider geographical scale

    Literature review on ability of agricultural crop residues and agro-industrial waste for treatment of wastewater

    Purpose Agricultural crop residues (ACR) and agro-industrial waste (AIW) are abundant in Indonesia and primarily used as substitutes for cattle feed or to be naturally decomposed in the nearby environment. This review attempts to examine the potential valorisation of ACR and AIW into biosorbent. This paper also provides the challenges and opportunities in applying wastewater biosorption treatment in Indonesia.Method A literature review from available literature was carried out to reveal and explore the ability and prospective application of ACR and AIW   for treatment of wastewaterResults The reviews show that ACR and AIW can be used for wastewater treatment in different forms including: filter media, activated carbon, biosorbent and biochar. Activated carbon has demonstrated its high ability and efficiency in removing organic pollutants in wastewater. This is due to its large porosity, internal surface area, and mechanical strength. ACR and AIW in wastewater biosorption can be applied in any small-scale agro-industries because of their simplicity procedures, technology, and low cost. Various options of wastewater technologies have also been investigated in recent years. Yet, various issues have been aroused surrounding this technology, including the biosorptive capacity, the performance-effectiveness, the design, and the high operation costs.Conclusion The study found that problems of a high cost of carbonation and activation process, the needs of regeneration treatment, and the up-scaling or commercialization might hinder the sustainable valorisation of ACR and AIW

    Quantification of resistant alleles in the β-tubulin gene of field strains of gastrointestinal nematodes and their relation with the faecal egg count reduction test

In the present study we compared the levels of resistance in Teladorsagia circumcincta and Trichostrongylus colubriformis by means of the faecal egg reduction test (FECRT) and the percentage of resistant alleles obtained after pyrosequencing. The study was conducted in 10 naturally infected sheep flocks. Each flock was divided into three groups: i) group treated with albendazole (ABZ); ii) group treated with ivermectin (IVM); iii) untreated group. The number of eggs excreted per gram of faeces was estimated at day 0 and 14 post-treatment. Results: Resistance to ABZ was observed in 12.5% (1/8) of the flocks and to IVM in 44.4% (4/9) of them. One flock was resistant to both drugs according to FECRT. Coprocultures were performed at the same dates to collect L3 for DNA extraction from pooled larvae and to determine the resistant allele frequencies by pyrosequencing analysis. In T. circumcincta, SNPs were not found at any of the three codons before treatment; after the administration of ABZ, SNPs were present only in two different flocks, one of them with a frequency of 23.8% at SNP 167, and the other 13.2% % at SNP 198. In relation to T. colubriformis, we found the SNP200 before treatment in 33.3% (3/9) of the flocks with values between 48.5 and 87.8%. After treatment with ABZ and IVM, the prevalence of this SNP increased to 75 and 100% of the flocks, with a mean frequency of 95.1% and 82.6%, respectively.     Preliminary results of the recombinase polymerase amplification technique for the detection of Haemonchus contortus from Hungarian field samples

    Haemonchus contortus is a parasitic nematode of small ruminants responsible for significant economic losses and animal health concerns globally. Detection of gastrointestinal nematode (GIN) infection in veterinary practice typically relies on microscopy-based methods such as the faecal egg count and morphological identification of larval culture. However, mixed co-infections are common and species-specific identification is typically time-consuming and expertise-intensive. Compounded by increasing anthelmintic resistance, there is an urgent need to implement the molecular diagnosis of GIN in the livestock industry, preferably in field settings. Advances in isothermal amplification techniques including recombinase polymerase amplification (RPA) assays could improve this. Yet, constraints in RPA kit availability and amplicon detection systems limit the use of this technology in point of care settings. In this study, we present an early-stage, proof-of-concept demonstration of RPA targeting the internal transcribed spacer (ITS2) region of H. contortus. Having tested against eight closely related nematodes and also against five farm isolates in Eastern Hungary, preliminary results derived from a comparative analysis of 3 primer sets showed the assay detects H. contortus DNA and has a limit of detection of 10 ng/Οl. We also tested an end-result naked eye detection system using various DNA binding dyes, of which EvaGreenŽ dye was successful for a qualitative RPA detection that could be adaptable at farm sites.     Persistence of apoptotic cells without autoimmune disease or inflammation in CD14−/− mice

    Interaction of macrophages with apoptotic cells involves multiple steps including recognition, tethering, phagocytosis, and anti-inflammatory macrophage responses. Defective apoptotic cell clearance is associated with pathogenesis of autoimmune disease. CD14 is a surface receptor that functions in vitro in the removal of apoptotic cells by human and murine macrophages, but its mechanism of action has not been defined. Here, we demonstrate that CD14 functions as a macrophage tethering receptor for apoptotic cells. Significantly, CD14−/− macrophages in vivo are defective in clearing apoptotic cells in multiple tissues, suggesting a broad role for CD14 in the clearance process. However, the resultant persistence of apoptotic cells does not lead to inflammation or increased autoantibody production, most likely because, as we show, CD14−/− macrophages retain the ability to generate anti-inflammatory signals in response to apoptotic cells. We conclude that CD14 plays a broad tethering role in apoptotic cell clearance in vivo and that apoptotic cells can persist in the absence of proinflammatory consequences

    Limited diversity associated with duplicated class II MHC-DRB genes in the red squirrel population in the United Kingdom compared with continental Europe

    The red squirrel (Sciurus vulgaris) population in the United Kingdom has declined over the last century and is now on the UK endangered species list. This is the result of competition from the eastern grey squirrel (S. carolinensis) which was introduced in the 19th century. However, recent evidence suggests that the rate of population decline is enhanced by squirrelpox disease, caused by a viral infection carried asymptomatically by grey squirrels but to which red squirrels are highly susceptible. Population genetic diversity provides some resilience to rapidly evolving or exotic pathogens. There is currently no data on genetic diversity of extant UK squirrel populations with respect to genes involved in disease resistance. Diversity is highest at loci involved in the immune response including genes clustered within the major histocompatibility complex (MHC). Using the class II DRB locus as a marker for diversity across the MHC region we genotyped 110 red squirrels from locations in the UK and continentalEurope. Twenty four Scvu-DRB alleles at two functional loci; Scvu-DRB1 and Scvu- DRB2, were identified. High levels of diversity were identified at both loci in the continental populations. In contrast, no diversity was observed at the Scvu-DRB2 locus in the mainland UK population while a high level of homozygosity was observed at the Scvu-DRB1 locus. The red squirrel population in the UK appears to lack the extensive MHC diversity associated with continental populations, a feature which may have contributed to their rapid decline
