    Transport processes of particles in dilute suspensions in turbulent water flow—phase II

    This work was concerned with an improved and more practical understanding of particle motion in a turbulent fluid field. It is felt that such an increased understanding has been achieved both in an analytical and in an experimental sense. Analytically, a theory simplified by the restrictive constraints of isotropy and Stokesian drag has been developed. Assumption of a reasonable particle energy spectrum allowed calculation of various statistical quantities important in the determination of the particle's turbulent motion. By such an analysis three characterizing parameters were found. Two describe inertial effects due to the particle's size and density. They were found to be of prime importance in the determination of the ratio of the particle's fluctuating velocity variance to that of the fluid. The crossing of the particle from one region of correlated fluid turbulence to another by virtue of its free fall velocity is described by the third parameters. This parameter was shown to be of particular importance in limiting the particle's velocity correlation. The two effects, of inertia and of free fall velocity, together act to determine dispersion. A well parameterized particle experiment was undertaken. Analysis of the particles' behavior in the turbulent flow gave a three dimensional characterization of their motion. In particular calculation of velocity variances, autocorrelations, and energy spectra were made. These measurements in conjunction with measurements on the turbulent fluid field allowed comparison between theory and experiment. Comparison of theoretical prediction to experimental observation showed good agreement provided the underlying assumptions inherent in the theoretical derivation were valid. Agreement suffered when these assumptions proved less valid.     Understanding the basic mechanisms and predicting the behavior of particles suspended in turbulent fluid flow are essential to environmental conservation and to multiphase system design. Air and water pollution, sedimentation and erosion of river beds and coastal shorelines, and atmospheric fallout are some of the areas in which particle suspensions are of key importance. Detailed experimental measurements of dilute particle suspensions have been performed which examined the effects of particle size, shape and relative density on the statistical response of such particles in a turbulent fluid. Shape was found to be of minor importance for spheres, cubes and tetrahedrons. However, size was found to be important when the particle dimension was as large or larger than the fluid turbulence structure. Relative density influenced both free fall and inertial effects.     The solubility behavior of various sparingly soluble periodates was found to be anomalous as interpreted through the equilibrium constants for the dilute aqueous periodate system as reported in the literature. Thus it was necessary to investigate these equilibria as a possible reason for the anomaly. Reinterpretation of this solubility behavior in light of the results of this investigation will be reported at a later date

    A comparison of optical and radar measurements of mesospheric winds and tides

    Optical measurements of mesospheric winds by Fabry‐Perot spectrometers, FPSs, at Mawson, 67.6°S 62.9°E, and Davis, 68.6°S 78.0°E, Antarctica are compared with similar measurements obtained using a spaced‐antenna MF radar at Davis. The FPSs observed the OH emission. Different analysis procedures, used to determine the mean wind, and amplitude and phase of the semidiurnal tide, have been compared. At these latitudes the diurnal tide is weak and the semi‐diurnal tide, although highly variable in amplitude, is usually the dominant periodicity. When comparing the amplitude and phase of the semidiurnal tide good agreement is obtained between measurements by the two instruments

    Exposure to deltamethrin affects development of Plasmodium falciparum inside wild pyrethroid resistant Anopheles gambiae s.s. mosquitoes in Uganda.

    BACKGROUND: Pyrethroid resistance in African vector mosquitoes is a threat to malaria control. Resistant mosquitoes can survive insecticide doses that would normally be lethal. We studied effects of such doses on Plasmodium falciparum development inside kdr-resistant Anopheles gambiae s.s. in Uganda. METHODS: We collected An. gambiae s.s. homozygous for kdr-L1014S mutation, fed them on blood samples from 42 P. falciparum-infected local patients, then exposed them either to nets treated with sub-lethal doses of deltamethrin or to untreated nets. After seven days, we dissected 692 mosquitoes and examined their midguts for oocysts. Prevalence (proportion infected) and intensity of infection (number of oocysts per infected mosquito) were recorded for each group. RESULTS: Both prevalence and intensity of infection were significantly reduced in deltamethrin-exposed mosquitoes, compared to those exposed to untreated nets. With low doses (2.5-5.0 mg/m(2)), prevalence was reduced by 59% (95% CI = 22%-78%) and intensity by 41% (95% CI = 25%-54%). With high doses (10-16.7 mg/m(2)), prevalence was reduced by 80% (95% CI = 67%-88 %) and intensity by 34 % (95 % CI = 20%-46%). CONCLUSIONS: We showed that, with locally-sampled parasites and mosquitoes, doses of pyrethroids that are sub-lethal for resistant mosquitoes can interfere with parasite development inside mosquitoes. This mechanism could enable pyrethroid-treated nets to prevent malaria transmission despite increasing vector resistance

    p53 controls expression of the DNA deaminase APOBEC3B to limit its potential mutagenic activity in cancer cells.

    Cancer genome sequencing has implicated the cytosine deaminase activity of apolipoprotein B mRNA editing enzyme catalytic polypeptide-like (APOBEC) genes as an important source of mutations in diverse cancers, with APOBEC3B (A3B) expression especially correlated with such cancer mutations. To better understand the processes directing A3B over-expression in cancer, and possible therapeutic avenues for targeting A3B, we have investigated the regulation of A3B gene expression. Here, we show that A3B expression is inversely related to p53 status in different cancer types and demonstrate that this is due to a direct and pivotal role for p53 in repressing A3B expression. This occurs through the induction of p21 (CDKN1A) and the recruitment of the repressive DREAM complex to the A3B gene promoter, such that loss of p53 through mutation, or human papilloma virus-mediated inhibition, prevents recruitment of the complex, thereby causing elevated A3B expression and cytosine deaminase activity in cancer cells. As p53 is frequently mutated in cancer, our findings provide a mechanism by which p53 loss can promote cancer mutagenesis

    Epidemiology, prehospital care and outcomes of patients arriving by ambulance with dyspnoea: An observational study

    Background: This study aimed to determine epidemiology and outcome for patients presenting to emergency departments (ED) with shortness of breath who were transported by ambulance. Methods: This was a planned sub-study of a prospective, interrupted time series cohort study conducted at three time points in 2014 and which included consecutive adult patients presenting to the ED with dyspnoea as a main symptom. For this sub-study, additional inclusion criteria were presentation to an ED in Australia or New Zealand and transport by ambulance. The primary outcomes of interest are the epidemiology and outcome of these patients. Analysis was by descriptive statistics and comparisons of proportions. Results: One thousand seven patients met inclusion criteria. Median age was 74 years (IQR 61-68) and 46.1 % were male. There was a high rate of co-morbidity and chronic medication use. The most common ED diagnoses were lower respiratory tract infection (including pneumonia, 22.7 %), cardiac failure (20.5%) and exacerbation of chronic obstructive pulmonary disease (19.7 %). ED disposition was hospital admission (including ICU) for 76.4 %, ICU admission for 5.6 % and death in ED in 0.9 %. Overall in-hospital mortality among admitted patients was 6.5 %. Discussion: Patients transported by ambulance with shortness of breath make up a significant proportion of ambulance caseload and have high comorbidity and high hospital admission rate. In this study, >60 % were accounted for by patients with heart failure, lower respiratory tract infection or COPD, but there were a wide range of diagnoses. This has implications for service planning, models of care and paramedic training. Conclusion: This study shows that patients transported to hospital by ambulance with shortness of breath are a complex and seriously ill group with a broad range of diagnoses. Understanding the characteristics of these patients, the range of diagnoses and their outcome can help inform training and planning of services
