    The application of S isotopes and S/Se ratios in determining ore-forming processes of magmatic Niā€“Cuā€“PGE sulfide deposits: a cautionary case study from the northern Bushveld Complex

    The application of S/Se ratios and S isotopes in the study of magmatic Niā€“Cuā€“PGE sulfide deposits has long been used to trace the source of S and to constrain the role of crustal contamination in triggering sulfide saturation. However, both S/Se ratios and S isotopes are subject to syn- and post-magmatic processes that may alter their initial signatures. We present in situ mineral Ī“34S signatures and S/Se ratios combined with bulk S/Se ratios to investigate and assess their utility in constraining ore-forming processes and the source of S within magmatic sulfide deposits. Magmatic Niā€“Cuā€“PGE sulfide mineralization in the Grasvally Noriteā€“Pyroxeniteā€“Anorthosite (GNPA) member, northern Bushveld Complex was used as a case study based on well-defined constraints of sulfide paragenesis and local S isotope signatures. A crustal Ī“34S component is evident in the most primary sulfide assemblage regardless of footwall lithology, and is inferred that the parental magma(s) of the GNPA member was crustally contaminated and sulfide saturated at the time of emplacement. However, S/Se ratios of both the primary and in particular secondary sulfide assemblages record values within or below the mantle range, rather than high crustal S/Se ratios. In addition, there is a wide range of S/Se ratio for each sulfide mineral within individual assemblages that is not necessarily consistent with the bulk ratio. The initial crustal S/Se ratio is interpreted to have been significantly modified by syn-magmatic lowering of S/Se ratio by sulfide dissolution, and post-magmatic lowering of the S/Se ratio from hydrothermal S-loss, which also increases the PGE tenor of the sulfides. Trace element signatures and variations in Th/Yb and Nb/Th ratios support both an early pre-emplacement contamination event as seen by the S isotopes and S/Se ratios, but also a second contamination event resulting from the interaction of the GNPA magma with the local footwall country rocks at the time of emplacement; though this did not add any additional S. We are able to present an integrated emplacement and contamination model for the northern limb of the Bushveld Complex. Although the multitude of processes that affect variations in the Ī“34S signature and in particular S/Se ratio may be problematic in interpreting ore genesis, they can reveal a wealth of additional detail on a number of processes involved in the genetic history of a Niā€“Cuā€“PGE deposit in addition to crustal contamination. However, a prerequisite for being able to do this is to utilize other independent petrological and mineralogical techniques that provide constraints on both the timing and effect of various ore-forming and modifying processes. Utilizing both bulk and in situ methods in concert to determine the S/Se ratio allows for the assessment of multiple sulfide populations, the partitioning behaviour of Se during sulfide liquid fractionation and also the effects of low temperature fluid alteration. In comparison, S isotopes are relatively more robust and represent a more reliable indicator of the role of crustal S contamination. The addition of trace element data to the above makes for an incredibly powerful approach in assessing the role of crustal contamination in magmatic sulfide systems

    The ARBRE (ARable Biomass Renewable Energy) project, the first large-scale wood-fueled electricity generating plant in the U.K., represents a significant development in realising British and European policy objectives on renewable energy. The plant is fueled by a mix of wood from short rotation coppice (SRC) and forest residues. Where feasible, composted/conditioned sewage sludge is applied to coppice sites to increase yields and improve soil structure. In the Yorkshire Water region, typical total N:P:K composition of composted/conditioned sludge is 2.9:3.8:0.3, respectively. Sludge application is calculated on the basis of total nitrogen (N) content to achieve 750 kg N ha-1, for 3 yearsā€™ requirement. Willow coppice forms a dense, widely spaced, root network, which, with its long growing season, makes it an effective user of nutrients. This, in combination with willowā€™s use as a nonfood, nonfodder crop, makes it an attractive route for the recycling of sewage sludge in the absence of sea disposal, banned under the EC Urban Waste Water Treatment Directive (UWWTD). Further work is required on the nutritional requirements of SRC in order to understand better the quantities of sludge that can be applied to SRC without having a detrimental impact on the environment. This paper suggests the source of N rerouting under the UWWTD and suggests the likely expansion of SRC as an alternative recycling pathway

    Integrase-deficient lentiviral vectors mediate efficient gene transfer to human vascular smooth muscle cells with minimal genotoxic risk

    We have previously shown that injury-induced neointima formation was rescued by adenoviral-Nogo-B gene delivery. Integrase-competent lentiviral vectors (ICLV) are efficient at gene delivery to vascular cells but present a risk of insertional mutagenesis. Conversely, integrase-deficient lentiviral vectors (IDLV) offer additional benefits through reduced mutagenesis risk, but this has not been evaluated in the context of vascular gene transfer. Here, we have investigated the performance and genetic safety of both counterparts in primary human vascular smooth muscle cells (VSMC) and compared gene transfer efficiency and assessed the genotoxic potential of ICLVs and IDLVs based on their integration frequency and insertional profile in the human genome. Expression of enhanced green fluorescent protein (eGFP) mediated by IDLVs (IDLV-eGFP) demonstrated efficient transgene expression in VSMCs. IDLV gene transfer of Nogo-B mediated efficient overexpression of Nogo-B in VSMCs, leading to phenotypic effects on VSMC migration and proliferation, similar to its ICLV version and unlike its eGFP control and uninfected VSMCs. Large-scale integration site analyses in VSMCs indicated that IDLV-mediated gene transfer gave rise to a very low frequency of genomic integration compared to ICLVs, revealing a close-to-random genomic distribution in VSMCs. This study demonstrates for the first time the potential of IDLVs for safe and efficient vascular gene transfer

    Evidence of continued injecting drug use after attaining sustained treatment-induced clearance of the hepatitis C virus: implications for reinfection

    Background: People who inject drugs (PWID) are at the greatest risk of hepatitis C virus (HCV) infection, yet are often denied immediate treatment due to fears of on-going risk behaviour. Our principal objective was to examine evidence of continued injecting drug use among PWID following successful treatment for HCV and attainment of a sustained viral response (SVR). Methods: PWID who attained SVR between 1992 and June 2012 were selected from the National Scottish Hepatitis C Clinical Database. Hospitalisation and mortality records were sourced for these patients using record linkage techniques. Our primary outcome variable was any hospitalisation or death, which was indicative of injecting drugs post-SVR. Results: The cohort comprised 1170 PWID (mean age at SVR 39.6y; 76% male). The Kaplan Meier estimate of incurring the primary outcome after three years of SVR was 10.59% (95% CI, 8.75ā€“12.79) After adjusting for confounding, the risk of an injection related hospital episode or death post-SVR was significantly increased with advancing year of SVR: AHR:1.07 per year (95% CI, 1.01ā€“1.14), having a pre-SVR acute alcohol intoxication-related hospital episode: AHR:1.83 (95% CI, 1.29ā€“2.60), and having a pre-SVR opiate or injection-related hospital episode: AHR:2.59 (95% CI, 1.84ā€“3.64). Conclusion: Despite attaining the optimal treatment outcome, these data indicate that an increasing significant minority of PWID continue to inject post-SVR at an intensity which leads to either hospitalisation or death and increased risk of reinfection

    Stable carbon isotopes of dissolved inorganic carbon for a zonal transect across the subpolar North Atlantic Ocean in summer 2014

    The stable carbon isotope composition of dissolved inorganic carbon (Ī“13CDIC) in seawater was measured in samples collected during Juneā€“July 2014 in the subpolar North Atlantic. Sample collection was carried out on the RRS James Clark Ross cruise JR302, part of the ā€œRadiatively Active Gases from the North Atlantic Region and Climate Changeā€ (RAGNARoCC) research programme. The observed Ī“13CDIC values for cruise JR302 fall in a range from āˆ’0.07 to +1.95ā€Æā€°, relative to the Vienna Pee Dee Belemnite standard. From duplicate samples collected during the cruise, the 1Ļƒ precision for the 341 results is 0.08ā€Æā€°, which is similar to our previous work and other studies of this kind. We also performed a cross-over analysis using nearby historical Ī“13CDIC data, which indicated that there were no significant systematic offsets between our measurements and previously published results. We also included seawater reference material (RM) produced by A. G. Dickson (Scripps Institution of Oceanography, USA) in every batch of analysis, enabling us to improve upon the calibration and quality-control procedures from a previous study. The Ī“13CDIC is consistent within each RM batch, although its value is not certified. We report Ī“13CDIC values of 1.15ā€ÆĀ±ā€Æ0.03ā€Æā€° and 1.27ā€ÆĀ±ā€Æ0.05ā€Æā€° for batches 141 and 144 respectively. Our JR302 Ī“13CDIC data can be used ā€“ along with measurements of other biogeochemical variables ā€“ to constrain the processes that control DIC in the interior ocean, in particular the oceanic uptake of anthropogenic carbon dioxide and the biological carbon pump.     Ground-based lidar processing and simulator framework for comparing models and observations (ALCF 1.0)

    Automatic lidars and ceilometers (ALCs) provide valuable information on cloud and aerosols but have not been systematically used in the evaluation of general circulation models (GCMs) and numerical weather prediction (NWP) models. Obstacles associated with the diversity of instruments, a lack of standardisation of data products and open processing tools mean that the value of large ALC networks worldwide is not being realised. We discuss a tool, called the Automatic Lidar and Ceilometer Framework (ALCF), that overcomes these problems and also includes a ground-based lidar simulator, which calculates the radiative transfer of laser radiation and allows one-to-one comparison with models. Our ground-based lidar simulator is based on the Cloud Feedback Model Intercomparison Project (CFMIP) Observation Simulator Package (COSP), which has been extensively used for spaceborne lidar intercomparisons. The ALCF implements all steps needed to transform and calibrate raw ALC data and create simulated attenuated volume backscattering coefficient profiles for one-to-one comparison and complete statistical analysis of clouds. The framework supports multiple common commercial ALCs (Vaisala CL31, CL51, Lufft CHM 15k and Droplet Measurement Technologies MiniMPL), reanalyses (JRA-55, ERA5 and MERRA-2) and models (the Unified Model and AMPS - the Antarctic Mesoscale Prediction System). To demonstrate its capabilities, we present case studies evaluating cloud in the supported reanalyses and models using CL31, CL51, CHM 15k and MiniMPL observations at three sites in New Zealand. We show that the reanalyses and models generally underestimate cloud fraction. If sufficiently high-temporal-resolution model output is available (better than 6-hourly), a direct comparison of individual clouds is also possible. We demonstrate that the ALCF can be used as a generic evaluation tool to examine cloud occurrence and cloud properties in reanalyses, NWP models, and GCMs, potentially utilising the large amounts of ALC data already available. This tool is likely to be particularly useful for the analysis and improvement of low-level cloud simulations which are not well monitored from space. This has previously been identified as a critical deficiency in contemporary models, limiting the accuracy of weather forecasts and future climate projections. While the current focus of the framework is on clouds, support for aerosol in the lidar simulator is planned in the future

    Rhenium enrichment in the Muratdere Cu-Mo (Au-Re) porphyry deposit, Turkey: evidence from stable isotope analyses (Ī“34S, Ī“18O, Ī“D) and laser ablation-inductively coupled plasma-mass spectrometry analysis of sulfides

    The Muratdere Cu-Mo (Au) porphyry deposit in western Turkey contains elevated levels of rhenium and is hosted within granodioritic intrusions into an ophiolitic mĆ©lange sequence in the Anatolian belt. The deposit contains several stages of mineralization: early microfracture-hosted molybdenite and chalcopyrite, followed by a quartz-pyrite-chalcopyrite vein set associated with Cu-Au grade, a quartz-chalcopyrite-pyrite-molybdenite vein set associated with Cu-Mo-Re grade, and a later polymetallic quartz-barite-sphalerite-galena-pyrite vein set. The rhenium in Muratdere is hosted within two generations of molybdenite: early microfracture-hosted molybdenite and later vein-hosted molybdenite. In situ laser ablation-inductively coupled plasma-mass spectrometry analysis of sulfides shows that the later molybdenite has significantly higher concentrations of Re (average 1,124 ppm, Ļƒ = 730 ppm, n = 43) than the early microfracture-hosted molybdenite (average 566 ppm, Ļƒ = 423 ppm, n = 28). Pyrite crystals associated with the Re-rich molybdenite have higher Co and As concentrations than those in other vein sets, with Au associated with As. The microfracture-hosted sulfides have Ī“34S values between āˆ’2.2ā€° and +4.6ā€°, consistent with a magmatic source. The vein-hosted sulfides associated with the high-Re molybdenite have a Ī“34S signature of 5.6ā€° to 8.8ā€°, similar to values found in peridotite lenses in the Anatolian belt. The later enrichment in Re and Ī“34S-enriched S may be sourced from the surrounding ophiolitic country rock or may be the result of changing redox conditions during deposit formation

    The risk of hepatocellular carcinoma in cirrhotic patients with hepatitis C and sustained viral response:role of the treatment regimen

    Background & Aims: Previous studies have reported a high frequency of hepatocellular carcinoma (HCC) occurrence in patients with advanced liver disease, after receipt of interferon (IFN)-free therapy for hepatitis C virus (HCV) infection. Our objective was to verify and account for this phenomenon using data from the Scottish HCV clinical database. Methods: We identified HCC-naĆÆve individuals with liver cirrhosis receiving a course of antiviral therapy in Scotland from 1997ā€“2016 resulting in a sustained virologic response. Patients were followed-up from their treatment start date to the earliest of: date of death, date of HCC occurrence, or 31 January 2017. We used Cox regression to compare the risk of HCC occurrence according to treatment regimen after adjusting for relevant co-factors (including: demographic factors; baseline liver disease stage; comorbidities/health behaviours, virology, and previous treatment experience). HCC occurrence was ascertained through both the HCV clinical database and medical chart review. For our main analysis, treatment regimen was defined as IFN-free vs. IFN-containing. Results: A total of 857 patients met the study criteria, of whom 31.7% received an IFN-free regimen. Individuals receiving IFN-free therapy were more likely to be: older; of white ethnicity, Child-Turcotte-Pugh B/C vs. Child-Turcotte-Pugh A; thrombocytopenic; non-genotype 3; and treatment experienced. HCC occurrence was observed in 46 individuals during follow-up. In univariate analysis, IFN-free therapy was associated with a significantly increased risk of HCC (HR: 2.48; pā€Æ=ā€Æ0.021). However, after multivariate adjustment for baseline factors, no significant risk attributable to IFN-free therapy persisted (aHR: 1.15, pā€Æ=ā€Æ0.744). Conclusion: These findings suggest that the higher incidence of HCC following sustained virologic response with IFN-free therapy relates to baseline risk factors/patient selection, and not the use of IFN-free therapy per se. Lay summary: We examined the risk of liver cancer in 857 patients with cirrhosis in Scotland who received hepatitis C antiviral therapy and achieved a cure. We compared the risk of first-time liver cancer in patients treated with the newest interferon-free regimens, to patients treated with interferon. After accounting for the different characteristics of these two treatment groups, we found no evidence that interferon-free therapy is associated with a higher risk of liver cancer
