    Toll-like receptors and NOD-like receptors in rheumatic diseases

    The past 10 years have seen the description of families of receptors that drive proinflammatory cytokine production in infection and tissue injury. Two major classes have been examined in the context of inflammatory joint disease - the Toll-like receptors (TLRs) and NOD-like receptors (NLRs). TLRs such as TLR2 and TLR4 are being implicated in the pathology of rheumatoid arthritis, ankylosing spondylitis, lyme arthritis and osteoarthritis. Nalp3 has been identified as a key NLR for IL-1β production and has been shown to have a particular role in gout. These findings present new therapeutic opportunities, possibly allowing for the replacement of biologics with small molecule inhibitors

    Natural and anthropogenic geohazards in Greater London observed from geological and ERS-1/2 and ENVISAT Persistent Scatterers ground motion data: results from the EC FP7-SPACE PanGeo Project

    We combine geological data and ground motion estimates from satellite ERS-1/2 and ENVISAT persistent scatterer interferometry (PSI) to delineate areas of observed natural and anthropogenic geohazards in the administrative area of Greater London (United Kingdom). This analysis was performed within the framework of the EC FP7-SPACE PanGeo project, and by conforming to the interpretation and geohazard mapping methodology extensively described in the Production Manual (cf. http://www.pangeoproject.eu). We discuss the results of the generation of the PanGeo digital geohazard mapping product for Greater London, and analyse the potential of PSI, geological data and the PanGeo methodology to identify areas of observed geohazards. Based on the analysis of PSI ground motion data sets for the years 1992–2000 and 2002–2010 and geology field campaigns, we identify 25 geohazard polygons, covering a total of ~650 km2. These include not only natural processes such as compaction of deposits on the River Thames flood plain and slope instability, but also anthropogenic instability due to groundwater management and changes in the Chalk aquifer, recent engineering works such as those for the Jubilee Line Extension project and electricity tunnelling in proximity to the River Thames, and the presence of made ground. In many instances, natural and anthropogenic observed geohazards overlap, therefore indicating interaction of different processes over the same areas. In terms of ground area covered, the dominant geohazard is anthropogenic land subsidence caused by groundwater abstraction for a total of ~300 km2, followed by natural compression of River Thames sediments over ~105 km2. Observed ground motions along the satellite line-of-sight are as high as +29.5 and −25.3 mm/year, and indicate a combination of land surface processes comprising ground subsidence and uplift, as well as downslope movements. Across the areas of observed geohazards, urban land cover types from the Copernicus (formerly GMES) EEA European Urban Atlas, e.g., continuous and discontinuous urban fabric and industrial units, show the highest average velocities away from the satellite sensor, and the smallest standard deviations (~0.7–1.0 mm/year). More rural land cover types such as agricultural, semi-natural and green areas reveal the highest spatial variability (up to ~4.4 mm/year), thus suggesting greater heterogeneity of observed motion rates within these land cover types. Areas of observed motion in the PSI data for which a geological interpretation cannot be found with sufficient degree of certainty are also identified, and their possible causes discussed. Although present in Greater London, some geohazard types such as shrink–swell clays and ground dissolution are not highlighted by the interpretation of PSI annual motion rates. Reasons for absence of evidence of the latter in the PSI data are discussed, together with difficulties related to the identification of good radar scatterers in landsliding areas

    A methodology to detect and characterize uplift phenomena in urban areas using Sentinel-1 data

    This paper presents a methodology to exploit the Persistent Scatterer Interferometry (PSI) time series acquired by Sentinel-1 sensors for the detection and characterization of uplift phenomena in urban areas. The methodology has been applied to the Tower Hamlets Council area of London (United Kingdom) using Sentinel-1 data covering the period 2015–2017. The test area is a representative high-urbanized site affected by geohazards due to natural processes such as compaction of recent deposits, and also anthropogenic causes due to groundwater management and engineering works. The methodology has allowed the detection and characterization of a 5 km2 area recording average uplift rates of 7 mm/year and a maximum rate of 18 mm/year in the period May 2015–March 2017. Furthermore, the analysis of the Sentinel-1 time series highlights that starting from August 2016 uplift rates began to decrease. A comparison between the uplift rates and urban developments as well as geological, geotechnical, and hydrogeological factors suggests that the ground displacements occur in a particular geological context and are mainly attributed to the swelling of clayey soils. The detected uplift could be attributed to a transient effect of the groundwater rebound after completion of dewatering works for the recent underground constructions

    Gymnosperms Demonstrate Patterns of Fine-root Trait Coordination Consistent with the Global Root Economics Space

    Gymnosperms encompass a diverse group of mostly woody plants with high ecological and economic value, yet little is known about the scope and organization of fine-root trait diversity among gymnosperms due to the undersampling of most gymnosperm families and the dominance of angiosperm groups in recent syntheses. New and existing data were compiled for morphological traits (root diameter, length, tissue density, specific root length [SRL] and specific root area [SRA]), the architectural trait branching ratio, root nitrogen content [N] and mycorrhizal colonization. We used phylogenetic least squares regression and principal component analysis to determine trait-trait relationships and coordination across 66 species, representing 11 of the 12 extant gymnosperm families from boreal, temperate, subtropical and tropical biomes. Finally, we compared the relationship between family divergence time and mean trait values to determine whether evolutionary history structured variation in fine-root traits within the gymnosperm phylogeny. Wide variation in gymnosperm root traits could be largely captured by two primary axes of variation defined by SRL and diameter, and root tissue density and root nitrogen, respectively. However, individual root length and SRA each had significant correlations with traits defining both main axes of variation. Neither mycorrhizal colonization nor root branching ratio were closely related to other traits. We did not observe a directional evolution of mean trait values from older to more recently diverged gymnosperm families. Synthesis. Despite their unique evolutionary history, gymnosperms display a root economic space similar to that identified in angiosperms, likely reflecting common constraints on plants adapting to diverse environments in both groups. These findings provide greater confidence that patterns observed in broad syntheses justly capture patterns of trait diversity among multiple, distinct lineages. Additionally, independence between root architecture and other traits may support greater diversity in below-ground resource acquisition strategies. Unlike angiosperms, there were no clear trends towards increasingly thin roots over evolutionary time, possibly because of lower diversification rates or unique biogeographic history among gymnosperms, though additional observations are needed to more richly test evolutionary trends among gymnosperms

    Systematic review of the effectiveness of preventing and treating Staphylococcus aureus carriage in reducing peritoneal catheter-related infections

    Objectives: To determine the clinical effectiveness and cost-effectiveness of (1) alternative strategies for the prevention of Staphylococcus aureus carriage in patients on peritoneal dialysis (PD) and (2) alternative strategies for the eradication of S. aureus carriage in patients on PD. Data sources: Major electronic databases were searched up to December 2005 (MEDLINE Extra up to 6 January 2006). Review methods: Electronic searches were undertaken to identify published and unpublished reports of randomised controlled trials and systematic reviews evaluating the effectiveness of preventing and treating S. aureus carriage on peritoneal catheterrelated infections. The quality of the included studies was assessed and data synthesised. Where data were not sufficient for formal meta-analysis, a qualitative narrative review looking for consistency between studies was performed. Results: Twenty-two relevant trials were found. These fell into several groups: the first split is between prophylactic trials, aiming to prevent carriage, and trials which aimed to eradicate carriage in those who already had it; the second split is between antiseptics and antibiotics; and the third split is between those that included patients having the catheter inserted before dialysis started and people already on dialysis. Many of the trials were small or short-term. The quality was often not good by today’s standards. The body of evidence suggested a reduction in exit-site infections, but this did not seem to lead to a significant reduction in peritonitis, although to some extent this reflected insufficient power in the studies and a low incidence of peritonitis in them. The costs of interventions to prevent or treat S. aureus carriage are relatively modest. For example, the annual cost of antibiotic treatment of S. aureus carriage per identified carrier of S. aureus was estimated at £179 (£73 screening and £106 cost of antibiotic). However, without better data on the effectiveness of the interventions, it is not clear whether such costs are offset by the cost of treating infections and averting changes from peritoneal dialysis to haemodialysis. Although treatment is not expensive, the lack of convincing evidence of clinical effectiveness made cost-effectiveness analysis unrewarding at present. However, consideration was given to the factors needed in a hypothetical model describing patient pathways from methods to prevent S. aureus carriage, its detection and treatment and the detection and treatment of the consequences of S. aureus (e.g. catheter infections and peritonitis). Had data been available, the model would have compared the costeffectiveness of alternative interventions from the perspective of the UK NHS, but as such it helped identify what future research would be needed to fill the gaps. Conclusions: The importance of peritonitis isnot in doubt. It is the main cause of people having to switch from peritoneal dialysis to haemodialysis, which then leads to reduced quality of life for patients and increased costs to the NHS. Unfortunately, the present evidence base for the prevention of peritonitis is disappointing; it suggests that the interventions reduce exit-site infections, but not peritonitis, although this may be due to trials being in too small numbers for too short periods.     On the persistence of Cenococcum geophilum ectomycorrhizas and its implications for forest carbon and nutrient cycles

    a b s t r a c t The turnover of ectomycorrhizal (EM) fungal biomass represents an important litter input into forest biogeochemical cycles. Cenococcum geophilum is a nearly ubiquitous and often abundant EM fungus, making the turnover dynamics of this species relevant and important across forest ecosystems. To better understand the turnover dynamics of C. geophilum ectomycorrhizas we examined their persistence using minirhizotron imaging and vitality status using a fluorescein diacetate (FDA) stain and contrasted these results with ectomycorrhizas of other EM fungi. Ectomycorrhizas formed by C. geophilum persisted 4e10 times longer and exhibited contrasting seasonal patterns of vitality compared to ectomycorrhizas of other EM fungi. Together, this suggests that litter resulting from the death of C. geophilum ectomycorrhizas is relatively recalcitrant to decay and may disproportionately influence forest biogeochemical cycles by retarding the rate at which carbon and nutrients are cycled

    The Cool Farm Biodiversity metric: an evidence-based online tool to report and improve management of biodiversity at farm scale

    Halting biodiversity loss and achieving food security are both aims of the United Nations 2030 Agenda for Sustainable Development, but there is complex interplay between them. Agriculture drives biodiversity loss, but biodiversity provides benefits to agriculture. There is substantial potential to develop ‘win-win’ solutions for biodiversity and people within productive farmland, by boosting wildlife that can be supported, whilst maintaining yield and other services. To achieve this, farmers need to be able to assess the impacts of their management on biodiversity at farm scale. While suitable tools exist to drive improvement in biodiversity management, none incorporates evidence on the effectiveness of specific management practices. In this study we present the Cool Farm Biodiversity metric, which generates a farm-scale action-based biodiversity management assessment, scored using expert judgements and expert assessment of experimental evidence. The metric is designed to be biome-specific, so it responds to conservation aims, ecosystem processes and farming systems in particular biomes. To demonstrate that the metric is responsive to changes in farm management, we present an example of use on a large arable farm from the temperate forest biome
