    Synthesizing realistic neural population activity patterns using generative adversarial networks

    The ability to synthesize realistic patterns of neural activity is crucial for studying neural information processing. Here we used the Generative Adversarial Networks (GANs) framework to simulate the concerted activity of a population of neurons. We adapted the Wasserstein-GAN variant to facilitate the generation of unconstrained neural population activity patterns while still benefiting from parameter sharing in the temporal domain. We demonstrate that our proposed GAN, which we termed Spike-GAN, generates spike trains that match accurately the first- and second-order statistics of datasets of tens of neurons and also approximates well their higher-order statistics. We applied Spike-GAN to a real dataset recorded from salamander retina and showed that it performs as well as state-of-the-art approaches based on the maximum entropy and the dichotomized Gaussian frameworks. Importantly, Spike-GAN does not require to specify a priori the statistics to be matched by the model, and so constitutes a more flexible method than these alternative approaches. Finally, we show how to exploit a trained Spike-GAN to construct’importance maps’ to detect the most relevant statistical structures present in a spike train. Spike-GAN provides a powerful, easy-to-use technique for generating realistic spiking neural activity and for describing the most relevant features of the large-scale neural population recordings studied in modern systems neuroscience

    Polycapillary optics for soft X-ray imaging and tomography

    Magnetic plasmas are extended volumetric sources of X-rays, and these emissions could reveal a lot of information about the processes occurring into the plasmas. Unfortunately, the constraints posed by these toroidal devices (high neutron flux, gamma and hard-X background, extremely high radiofrequency powers, high magnetic fields, optical limitations and so on) are very severe and limit strongly the possibility to install X-ray detectors directly into or close to the machine. Soft X-ray diagnostics are meant both as tomography and imaging. We started, therefore, to investigate the feasibility of using polycapillary optics for these purposes, in collaboration between Istituto Nazionale di Fisica Nucleare (INFN)- Frascati, Ente per le Nuove tecnologie, l’Energia e l’Ambiente (ENEA)-Frascati and the Commissariat de l’Energie Atomique (CEA)-Cadarache. The first tests were performed in order to characterize the polycapillary lenses (convergence, divergence, efficiency, spectral dispersion, etc.) for distances much larger than the optical focal length of the lenses, both for the detector and for the source. A silicon-based C-MOS imager (Medipix 2) has been used as a detector and the micro focus X-ray tubes as point-like sources.     Transcriptional profiling of zebrafish identifies host factors controlling susceptibility to Shigella flexneri

    Shigella flexneri is a human-adapted pathovar of Escherichia coli that can invade the intestinal epithelium, causing inflammation and bacillary dysentery. Although an important human pathogen, the host response to S. flexneri has not been fully described. Zebrafish larvae represent a valuable model to study human infections in vivo. Here we use a Shigella-zebrafish infection model to generate mRNA expression profiles of host response to Shigella infection at the whole animal level. Immune response-related processes dominate the signature of early Shigella infection (6 hours post-infection). Consistent with its clearance from the host, the signature of late Shigella infection (24 hours post-infection) is significantly changed, and only a small set of immune-related genes remain differentially expressed, including acod1 and gpr84. Using mutant lines generated by ENU, CRISPR mutagenesis and F0 Crispants, we show that acod1- and gpr84-deficient larvae are more susceptible to Shigella infection. Together, these results highlight the power of zebrafish to model infection by bacterial pathogens and reveal the mRNA expression of the early (acutely infected) and late (clearing) host response to Shigella infection

    Real-time identification of the current density profile in the JET Tokamak: method and validation

    International audienceThe real-time reconstruction of the plasma magnetic equilibrium in a Tokamak is a key point to access high performance regimes. Indeed, the shape of the plasma current density profile is a direct output of the reconstruction and has a leading effect for reaching a steady-state high performance regime of operation. In this paper we present the methodology followed to identify numerically the plasma current density in a Tokamak and its equilibrium. In order to meet the real-time requirements a C++ software has been developed using the combination of a finite element method, a nonlinear fixed point algorithm associated to a least square optimization procedure. The experimental measurements that enable the identification are the magnetics on the vacuum vessel, the interferometric and polarimetric measurements on several chords and the motional Stark effect. Details are given about the validation of the reconstruction on the JET tokamak, either by comparison with 'off-line' equilibrium codes or real time software computing global quantities

    Association Of Postalimentary Lipemia With Atherosclerotic Manifestations.

    We identified different lipemic and metabolic responses after the ingestion of a standardized meal by healthy adults and related them to atherosclerotic markers. Samples from 60 normolipidemic adults were collected before and after a liquid meal (40 g fat/m² body surface) at 0, 2, 4, 6, and 8 h for measurements of lipids, free fatty acids (FFA), insulin, cholesteryl ester transfer protein (CETP), autoantibodies to epitopes of oxidized LDL (oxLDL Ab), lipolytic activities, and apolipoprotein E polymorphism. Mean carotid intima-media thickness (cIMT) was determined by Doppler ultrasound. The volunteers were classified into early (N = 39) and late (N = 31) triacylglycerol (TAG) responders to the test meal. Late responders showed lower HDL cholesterol concentration at fasting and in the TAG peak, lower insulin and higher FFA concentrations compared to early responders. Multivariate regression analyses showed that mean cIMT was associated with gender (male) and age in early responders and by cholesterol levels at the 6th hour in late responders. oxLDL Ab were explained by lipoprotein lipase and negatively by hepatic lipase and oxLDL Ab (fasting period) by CETP (negative) and FFA (positive). This study is the first to identify a postalimentary insulin resistance state, combined with a reduced CETP response exclusively among late responders, and the identification of the regulators of postalimentary atherogenicity. Further research is required to determine the metabolic mechanisms described in the different postalimentary phenotypes observed in this study, as well as in different pathological states, as currently investigated in our laboratory.451086-9

The primary response to T. gondii was supported by IgM, IgG and IgA seroconversion.     3rd harmonic ECRH absorption enhancement by 2nd harmonic heating at the same frequency in a tokamak

    The fundamental mechanisms responsible for the interplay and synergy between the absorption dynamics of extraordinary-mode electron cyclotron waves at two different harmonic resonances (the 2nd and 3rd) are investigated in the TCV tokamak. An enhanced 3rd harmonic absorption in the presence of suprathermal electrons generated by 2nd harmonic heating is predicted by Fokker-Planck simulations, subject to complex alignment requirements in both physical space and momentum space. The experimental signature for the 2nd/3rd harmonic synergy is sought through the suprathermal bremsstrahlung emission in the hard x-ray range of photon energy. Using a synthetic diagnostic, the emission variation due to synergy is calculated as a function of the injected power and of the radial transport of suprathermal electrons. It is concluded that in the present experimental setup a synergy signature has not been unambiguously detected. The detectability of the synergy is then discussed with respect to variations and uncertainties in the plasma density and effective charge in view of future optimized experiments

    Pre-harvest herbicide application reduces the Italian ryegrass seed viability.

    Italian ryegrass (Lolium multiflorum Lam.) is a troublesome weed found in temperate agricultural fields, wherein the future infestations depend on the soil seed bank. The aim of this research was to evaluate Italian ryegrass seed viability subjected to herbicide application in different phenological stages of development. Experiments were carried out in a completely randomized experimental design with four repetitions. Paraquat, ammonium-glufosinate, iodosulfuron-methyl and clethodim herbicides were applied in three development stages (pre-spike, anthesis, and grain filling) and compared to the control. The effects were evaluated by percentage of control at seven, 14, and 28 days after application (DAA), seeds per plant, 1000 seeds weight, and percentage of germination, abnormal seedlings, dead seeds, viable and non-viable dormant seeds. Control was greater than 97% at 28 DAA, with a decrease of more 90% for seed per plant, and strong reduction of 1000 seeds weight for the herbicides applied up to the anthesis stage. At the grain filling stage, clethodim reduced seed production by 18% and 1000-seeds weight by 36% compared to the control. Ammonium-glufosinate and paraquat herbicides applied up to the anthesis stage reduced seed viability by more than 98%. Overall, herbicide application reduces seed production, seed weight and viability, especially when applied before the grain filling stage. Highlighted Conclusions 1. Herbicide application up to the anthesis developmental stages reduces seed production, 1000 seeds weight, and seed viability. 2. Paraquat was the best herbicide to reduce seed production per plant, 1000-seeds weight and, viability of Italian ryegrass seeds, regardless of development stages. 3. Reducing future infestations of Italian ryegrass in cropping systems depends on herbicide application before the grain filling
