Atomic Force Microscopy-Based Screening of Drug-Excipient Miscibility and Stability of Solid Dispersions
PURPOSE: Development of a method to assess the drug/polymer miscibility and stability of solid dispersions using a melt-based mixing method. METHODS: Amorphous fractured films are prepared and characterized with Raman Microscopy in combination with Atomic Force Microscopy to discriminate between homogenously and heterogeneously mixed drug/polymer combinations. The homogenous combinations are analyzed further for physical stability under stress conditions, such as increased humidity or temperature. RESULTS: Combinations that have the potential to form a molecular disperse mixture are identified. Their potential to phase separate is determined through imaging at molecular length scales, which results in short observation time. De-mixing is quantified by phase separation analysis, and the drug/polymer combinations are ranked to identify the most stable combinations. CONCLUSIONS: The presented results demonstrate that drug/polymer miscibility and stability of solid dispersions, with many mechanistic details, can be analyzed with Atomic Force Microscopy. The assay allows to identify well-miscible and stable combinations within hours or a few days

5\. Literaturverzeichnis 79Natriumtetraphenylporphyrinphosphonat kristallisiert auf Glimmer in Form
starrer, segregierter und molekular dünner Porphyrinstäbchen (d = 2,8 nm). Die
Kristallisation konnte mit einem Rasterkraftmikrokop induziert, beobachtet und
gesteuert werden. Die Amplituden-Abstandsbeziehung indizierte eine
FlĂĽssigkeitsbrĂĽcke, die sich ab einer Sonden-Proben Distanz von unter r < 20
nm bilden kann. Die Sonde konnte die Oberfläche daher in zwei
unterschiedlichen Weisen kontaktieren, bei großem Abstand gewöhnlich (nicht
verbrückt) und in geringem Abstand durch Kapillarkräfte (verbrückt). Die
Oberfläche konnte so wahlweise topologisch korrekt oder in einem invertiertem
topologischem Kontrast abgebildet werden. Auf gleiche Weise konnte die mit
Porphyrinen bedeckte Oberfläche entweder gemessen oder vergleichbar mit der
kraftmikroskopischen Nanolithografie manipuliert werden. Eine partielle
Protonierung des Porphyrins hob die Segregation teilweise auf. Bei pH = 11, 5
bildeten sich molekular dĂĽnne Platten (d = 2,8 nm). Auf Graphit entstanden
auĂźerdem separierte Porphyrinreihen, deren molekulare Struktur im dynamischen
Modus aufgelöst wurde. Auf defektreichen Au(111) Terrassen wurden bei pH = 13
einheitlich über die Oberfläche verteilte, senkrecht orientierte und
segregierte Porphyrinstapel von h = 5 - 10 nm Höhe dargestellt. Durch Einsatz
unkonventioneller Präparationstechniken, die auf der Akkumulation
nanoskopischer Gasblasen oder Umkristallisation in Gegenwart von Feuchtigkeit
basierten, wurden segregierte und senkrecht stehende Säulen von bis zu 1500 nm
Höhe sowie die Oberfläche überragende Stäbe dargestellt. Nach Bestimmung der
Sondengeometrie (TEM) und geometrische Entfaltung konnte gezeigt werden, dass
die Säulen einheitlich sind und einen Durchmesser von d < 6 nm haben. Es
fanden sich zahlreiche Hinweise darauf, dass die Säulen nur einen Durchmesser
von d = 3 nm haben und wahrscheinlich mit den auf Glimmer dargestellten
Porphyrinstäben identisch sind.Meso-Tetra(phenyl-p-phosphonate) porphyrin forms rigid and well-separated rods
of monomolecular thickness (d = 2.8 nm) and lengths of several micrometers on
mica at pH = 13 (octasodium salt). The formation of these rods could be
directley observed and controlled by tapping mode Scanning Force Microscopy
and was induced by capillary forces. Amplitude-distance curves indicated that
a stable meniscus was formed on hydrophilic surface areas below a tip-sample
separation of r <20 nm. Alternatively, normal height images or images with a
topographical inversion could be observed depending on the distance of the SFM
tip. The meniscus let the original nanorods appear as ditches in the mica
surface and enabled rearrangements. A partly protonated form of the same
porphyrin (pH 11.5) gave separated rows of flat-lying porphyrins on graphite,
which appear with molecular resolution in SFM images as well as two-
dimensional platelets of monomolecular thickness. Self-Assembley of the same
porphyrin (pH = 13) on ill-defined Au(111) surface led to well separated and
upright-standing porphyrin stacks ( h = 5 - 10 nm). Recrystallisation in
dampness or the accumulation of nanoscopic gasbubbles by an unconventional
preparation let to high (up to 1500 nm) and steep columns as well as
overhanging rods. Indirect evidence suggests, that the width of these columns
was less than 6 nm and probably identical to the 2,8 nm porphyrinrods on mica
