BACKGROUND: While silver nanoparticles (AgNPs) are widely used in consumer and medical products, the mechanism by which AgNPs cause pulmonary cytotoxicity is not clear. AgNP agglomerates are found in endo-lysosomal structures within the cytoplasm of treated cells. In this study, the functional role of lysosomes in AgNP-induced cellular damage was examined in A549 human lung alveolar epithelial cells. We evaluated the intracellular distribution of AgNPs, lysosomal pH, cellular viability, Ag dissolution, and metallothionein (MT) mRNA levels in AgNP-exposed A549 cells that were treated with bafilomycin A1, the lysosomal acidification inhibitor. FINDINGS: Exposure of A549 cells to citrate-coated AgNPs (20 nm diameter) for 24 h induced cellular damage and cell death at 100 and 200 μg Ag/ml, respectively. Confocal laser microscopic examination of LysoTracker-stained cells showed that AgNPs colocalized with lysosomes and their agglomeration increased in a dose-dependent manner (50–200 μg Ag/ml). In addition, the fluorescence signals of LysoTracker were reduced following exposure to AgNPs, suggesting the elevation of lysosomal pH. Treatment of A549 cells with 200 nM bafilomycin A1 and AgNPs (50 μg Ag/ml) further reduced the fluorescence signals of LysoTracker. AgNP-induced cell death was also increased by bafilomycin A1 treatment. Finally, treatment with bafilomycin A1 suppressed the dissolution of Ag and decreased the mRNA expression levels of MT-I and MT-II following exposure to AgNPs. CONCLUSIONS: The perturbation of lysosomal pH by AgNP exposure may play a role in AgNP agglomeration and subsequent cellular damage in A549 cells

Effectiveness of the Vertical Gas Ventilation Pipes for Promoting Waste Stabilization in Post-Closure Phase

To make inside of the municipal solid waste (MSW) landfill aerobic as much as possible is thought to be preferable for promoting waste stabilization, reducing pollutant's load in leachate, minimizing greenhouse gas emission and shortening post-closure-care period. In Japan, installation of semi-aerobic landfill structure has widely spread in order to promote waste stabilization in MSW landfill from 1980s. In semi-aerobic landfill structure, outlet of main leachate collection pipe is opened to atmosphere. Heat generated by aerobic degradation of waste causes natural convection and natural aeration arises from the outlet of leachate collection pipe to the gas vents. It is so-called stack effect. This air flow is thought to be effective for purifying leachate flowing through drainage layer and leachate collection pipes. And it is also thought to be contributing to expanding aerobic region in waste layer in landfill. Recently, measures attempting the promotion of waste stabilization are taken at several landfills at where stabilization of waste delays, in which many vertical gas vents are newly installed and close structure to semi-aerobic landfill is created. However, in many cases, these gas vents are not connected to leachate collection pipes. Many vertical gas vents are just installed without scientific proof regarding whether they can contribute for waste stabilization. In this study, how such installation of gas vents is effective for waste stabilization and aerobization of waste layer was discussed by numerical analysis. In numerical analysis, heat transfer, gas movement by pressure, gas diffusion, biological degradation of organic matter, and heat generation by biodegradation were taken into account. Simulations were carried out by using the general purpose simulator of finite element method. Three types of landfill structure were assumed. As the results, the following information were obtained. In dig-down type landfill, installation of gas vents has no effect for changing air flow. On the other hand, in pile-up type landfill, installation of vertical gas vents can accelerate air invasion and significantly promote waste stabilization, if it has high permeable horizontal layer

Abstract

Background

We analyzed the causal relationship between LDL susceptibility to oxidation and the development of new carotid artery atherosclerosis over a period of 5 years. We previously described the determinants related to a risk of cardiovascular changes determined in a Japanese population participating in the Niigata Study, which is an ongoing epidemiological investigation of the prevention of cardiovascular diseases.

Methods

We selected 394 individuals (169 males and 225 females) who underwent a second carotid artery ultrasonographic examination in 2001 - 2002 for the present study. The susceptibility of LDL to oxidation was determined as the photometric absorbance and electrophoretic mobility of samples that had been collected in 1996 - 1997. The measurements were compared with ultrasonographic findings obtained in 2001 - 2002.

Results

The multivariate-adjusted model showed that age (odds ratio (OR), 1.034; 95% confidence interval (95%CI), 1.010 - 1.059), HbA1c (OR, 1.477; 95%CI, 0.980 - 2.225), and photometric O/N (OR, 2.012; 95%CI, 1.000 - 4.051) were significant variables that could independently predict the risk of new carotid artery atherosclerosis.

Conclusion

The susceptibility of LDL to oxidation was a significant parameter that could predict new carotid artery atherosclerosis over a 5-year period, and higher susceptibility was associated with a higher incidence of new carotid artery atherosclerosis.</p
