    As opposed to an approach that stresses the consequences of urbanisation in terms of increasing insecurity, localism, violence, the author proposes a positive and action-oriented attitude, illustrated by a recent experience initiated by the City Council of Bologna. From the empirical observation of the existence of a new poverty linked with precarious work, even in a traditionally wealthy city, springs the ‘Zanardi houses’ project (‘case Zanardi’), that aims to promote new forms of mutual help and sharing, to counter individualism and strengthen the city’s social capital.Rispetto ad una visione della città contemporanea che evidenzia le conseguenze dell’urbanizzazione in termini di aumento dell’insicurezza, del localismo, della violenza, l’autrice propone un approccio positivo ed orientato all’intervento esemplificato da una recente iniziativa del Comune di Bologna. Dalla constatazione empirica dell’esistenza di una nuova povertà legata al precariato giovanile, anche in una città tradizionalmente benestante, nasce l’esperienza delle ‘case Zanardi’, che promuovono nuove forme di condivisione ed aiuto mutuo per combattere l’individualismo ed alimentare il capitale sociale della città

    CSF alpha-synuclein aggregates by seed amplification and clinical presentation of AD

    Introduction: Accumulating evidence suggests that α-synuclein (αSyn) can modulate Alzheimer's disease (AD) pathology. The aim of this study was to evaluate the prevalence and clinical features associated with cerebrospinal fluid (CSF) αSyn detected by seed amplification assay (SAA) in AD. Methods: Eighty AD patients with CSF AT(N) biomarker positivity (mean age 70.3 Â± 7.3 years) and 28 non-AD age-matched controls were included. All subjects underwent standardized clinical assessment; CSF αSyn aggregates were detected by SAA. Results: CSF was αSyn-SAA positive (αSyn+) in 36/80 AD patients (45%) and in 2/28 controls (7.1%). AD αSyn+ and αSyn- patients were comparable for age, disease severity, comorbidity profile, and CSF core biomarkers. AD αSyn+ presented a higher prevalence of atypical phenotypes and symptoms. Conclusions: Our findings demonstrate that concomitant CSF αSyn pathology is present in a significant proportion of AD patients starting in the early stages and can affect clinical presentation. Longitudinal studies are warranted to evaluate the significance for the disease course

    A Complex of Kif18b and MCAK Promotes Microtubule Depolymerization and Is Negatively Regulated by Aurora Kinases

    SummaryIntroductionSpindle assembly requires tight control of microtubule (MT) dynamics. This is dependent on a variety of MT binding proteins and their upstream regulators. The Aurora kinases have several well-described functions during cell division, but it remains unclear whether they control global spindle microtubule dynamics.ResultsHere, we find that simultaneous inhibition of Aurora A and B results in a dramatic decrease in spindle MT stability, and we identify the uncharacterized kinesin-8 Kif18b as a mediator of this effect. In interphase, Kif18b is nuclear, but upon nuclear envelope breakdown, Kif18b binds to astral MT plus ends through an interaction with EB1. Surprisingly, Kif18b also binds to the kinesin-13 motor MCAK, and this interaction is required for robust MT depolymerization. Furthermore, the Kif18b-MCAK interaction is negatively regulated by Aurora kinases through phosphorylation of MCAK, indicating that Aurora kinases regulate MT plus-end stability in mitosis through control of Kif18b-MCAK complex formation.ConclusionTogether, these results uncover a novel role for Aurora kinases in regulating spindle MT dynamics through Kif18b-MCAK and suggest that the Kif18b-MCAK complex constitutes the major MT plus-end depolymerizing activity in mitotic cells

    Discomfort relief after paracetamol administration in febrile children admitted to a third level paediatric emergency department

    Backgroundinternational guidelines recommend treating fever in children not at a predefined body temperature limit but based on the presence of discomfort. However few studies evaluated discomfort relief after administration of antipyretics in children.MethodsBetween 1st January and 30th September 2021 a single-center prospective observational study was performed in febrile children consecutively admitted to a pediatric emergency department and treated with paracetamol orally. For each child, body temperature, presence and severity of discomfort, defined using a previously published semiquantitative likert scale, were evaluated at baseline and 60 min after administration of paracetamol, and differences were analyzed.Results172 children (males: 91/172; 52.9%; median age: 41.7 months) were included. Significant reductions in body temperature (median body temperature at T0: 38.9 °C; IQR: 38.3–39.4, median body temperature at T60: 36.9 °C; IQR: 36.4–37.5; P Conclusionsparacetamol in febrile children is associated not only with significantly reduction in body temperature but also with discomfort relief.</p

    Paleoclimatic inferences for the late miocene of the angastaco basin based on the foliar physiognomic analysis: Palo pintado formation, Salta, Argentina

    Las rocas sedimentarias continentales que constituyen la FormaciĂłn Palo Pintado, de edad del Mioceno tardĂ­o, afloran en la provincia de Salta y suscitan un gran interĂ©s paleoclimĂĄtico, dadas las condiciones ambientales reinantes durante ese intervalo geocronolĂłgico. Los registros geolĂłgicos y paleobotĂĄnicos sugieren que durante la acumulaciĂłn de las rocas sedimentarias de la FormaciĂłn Palo Pintado (cuenca de Angastaco) habrĂ­an existido condiciones mĂĄs hĂșmedas en comparaciĂłn a las de otras formaciones cercanas y coetĂĄneas, por ejemplo, la AloformaciĂłn Playa del Zorro y la FormaciĂłn Chiquimil (Mioceno tardĂ­o, provincias de Catamarca y TucumĂĄn) y las formaciones Salicas y Toro Negro (Mioceno tardĂ­o, provincia de La Rioja). En este estudio se analizaron el margen y el ĂĄrea foliar de las hojas fĂłsiles contenidas en las rocas de la FormaciĂłn Palo Pintado, a fin de obtener la temperatura media anual (TMA) y la precipitaciĂłn media anual (PMA). Los valores resultantes fueron los siguientes: TMA=23,98 °C y PMA=330,8 mm. Estos resultados se condicen con la interpretaciĂłn de diferentes autores quienes consideran que la FormaciĂłn Palo Pintado se habrĂ­a depositado bajo un ambiente relativamente hĂșmedo, posiblemente como consecuencia de las lluvias caĂ­das localmente en la regiĂłn de la cuenca de Angastaco.The continental sedimentary rocks that constitute the Palo Pintado Formation of the late Miocene from Salta province, presents a great paleoclimatic interest due to the environmental conditions prevailing during this geochronologic interval. The geological and paleobotanical data suggest that during the sedimentary rocks accumulation of the Palo Pintado Formation (Angastaco Basin), wetter conditions would have existed comparing with other nearby and contemporary Formations, for example the Playa del Zorro Aloformation (late Miocene of Catamarca) and the Chiquimil (late Miocene of TucumĂĄn), Salicas and the Toro Negro Formations (both from the late Miocene of La Rioja). In this study, the margin and the foliar area of the leaves contained on rocks from the Palo Pintado Formation are analyzed, in order to obtain the mean annual temperature (MAT) and the mean annual precipitation (MAP). The resulting values were: 23.98 °C and 330.8 mm. These results are coincident by the interpretation of different authors, who consider that the Palo Pintado Formation would have been deposited under a relatively humid environment, possibly as a consequence of the rains that affected locally the Angastaco basin region.Fil: Robledo, Juan Manuel.     Localization of Magic-F1 Transgene, Involved in Muscular Hypertrophy, during Early Myogenesis

    We recently showed that Magic-F1 (Met-activating genetically improved chimeric factor 1), a human recombinant protein derived from hepatocyte growth factor/scatter factor (HGF/SF) induces muscle cell hypertrophy but not progenitor cell proliferation, both in vitro and in vivo. Here, we examined the temporal and spatial expression pattern of Magic-F1 in comparison with Pax3 (paired box gene 3) transcription factor during embryogenesis. Ranging from 9.5 to 17.5 dpc (days post coitum) mouse embryos were analyzed by in situ hybridization using whole mounts during early stages of development (9.5–10.5–11.5 dpc) and cryostat sections for later stages (11.5–13.5–15.5–17.5 dpc). We found that Magic-F1 is expressed in developing organs and tissues of mesenchymal origin, where Pax3 signal appears to be downregulated respect to the wt embryos. These data suggest that Magic-F1 could be responsible of muscular hypertrophy, cooperating with Pax3 signal pathway in skeletal muscle precursor cells
