    Profiling the iron, copper and zinc content in primary neuron and astrocyte cultures by rapid online quantitative size exclusion chromatography-inductively coupled plasma-mass spectrometry

    Metals often determine the chemical reactivity of the proteins to which they are bound. Each cell in the body tightly maintains a unique metalloproteomic profile, mostly dependent on function. This paper describes an analytical online flow injection quantitative size exclusion chromatography-inductively coupled plasma-mass spectrometry (SEC-ICP-MS) method, which was applied to profiling the metal-binding proteins found in primary cultures of neurons and astrocytes. This method can be conducted using similar amounts of sample to those used for Western blotting (20-150 μg protein), and has a turnaround time of <15 minutes. Metalloprotein standards for Fe (as ferritin), Cu and Zn (as superoxide dismutase-1) were used to construct multi-point calibration curves for online quantification of metalloproteins by SEC-ICP-MS. Homogenates of primary neuron and astrocyte cultures were analysed by SEC-ICP-MS. Online quantification by external calibration with metalloprotein standards determined the mass of metal eluting from the column relative to time (as pg s-1). Total on-column Fe, Cu and Zn detection limits ranged from 0.825 ± 0.005 ng to 13.6 ± 0.7 pg. Neurons and astrocytes exhibited distinct metalloprotein profiles, featuring both ubiquitous and unique metalloprotein species. Separation and detection by SEC-ICP-MS allows appraisal of these metalloproteins in their native state, and online quantification was achieved using this relatively simple external calibration process. © 2013 The Royal Society of Chemistry

    The elastic constants of MgSiO3 perovskite at pressures and temperatures of the Earth's mantle

    The temperature anomalies in the Earth's mantle associated with thermal convection1 can be inferred from seismic tomography, provided that the elastic properties of mantle minerals are known as a function of temperature at mantle pressures. At present, however, such information is difficult to obtain directly through laboratory experiments. We have therefore taken advantage of recent advances in computer technology, and have performed finite-temperature ab initio molecular dynamics simulations of the elastic properties of MgSiO3 perovskite, the major mineral of the lower mantle, at relevant thermodynamic conditions. When combined with the results from tomographic images of the mantle, our results indicate that the lower mantle is either significantly anelastic or compositionally heterogeneous on large scales. We found the temperature contrast between the coldest and hottest regions of the mantle, at a given depth, to be about 800K at 1000 km, 1500K at 2000 km, and possibly over 2000K at the core-mantle boundary.Comment: Published in: Nature 411, 934-937 (2001

    La mutación de la biblioteca en los inicios del siglo XXI

    El papel que las bibliotecas desempeñan en la actualidad ha sido cuestionado en numerosas ocasiones, a causa de la revolución tecnológica a la que se está asistiendo, la cual pondría en amenaza el papel básico que hasta ahora tenían éstas. Pero la realidad es que se observa una tendencia en la que cada vez se construyen más bibliotecas y más complejas. Por lo que se torna harto interesante descubrir hacia dónde camina la arquitectura en este sentido. A este respecto, se advierte un cambio de paradigma respecto al papel que juega la biblioteca en la actualidad, provocando a su vez una respuesta en cuanto a la arquitectura bibliotecaria. Los cambios sociales que se han producido de la mano del desarrollo de la tecnología, se han visto implicados de forma directa e indirecta en este cambio de paradigma, presentando los desafíos a los que se enfrenta la arquitectura actual. Debido a la incertidumbre que se presenta de cara a este nuevo periodo, en base a los constantes cambios que se manifiestan debido a la rápida evolución que sufren las nuevas tecnologías, y de cara a las demandas de esta nueva sociedad, se torna fundamental la aplicación del principio de la flexibilidad en los nuevos espacios bibliotecarios, pero se rehúye de la clásica solución espacial donde el espacio se vuelve uniforme, sin apenas expresión plástica ni variedad. Por otro lado, se presentan los temidos efectos que la globalización tiene sobre el planeta, a cualquier escala, y que parecen llevar todo hacia una tendencia universalista. Por lo que se hace interesante el estudio de la arquitectura bibliotecaria desde diferentes ópticas a nivel cultural, tomando como referencia la cultura occidental, debido a su importante papel de cara al origen de este proceso, así como la cultura oriental, definida prácticamente como la antítesis de ésta. Sobre la base de lo planteado, la presente investigación, a modo de primera toma de contacto, ha pretendido abrir un campo de estudio a través del cual se puedan identificar nuevas formas de aplicar la flexibilidad en los espacios bibliotecarios, de cara a que éstos no se vuelvan caducos en cuanto al diseño que presentan, además de poner el causa los procesos de globalización que pretenden reducir la realidad actual, en base a procesos de homogeneización; por lo que se busca identificar procesos de hibridación en la arquitectura actual, a través de los cuales poder identificar si se producen diferencias entre las distintas culturas, valorando en qué medida se incluyen las cuestiones de carácter local en las obras que representan cada una de estas culturas. A través de la metodología empleada, basada en los estudios de caso individual y en el análisis comparativo de éstos, se han podido evidenciar líneas que muestran esa evolución de la arquitectura actual, en la que se torna evidente la importancia que tiene la flexibilidad en el nuevo escenario, así como la posibilidad de aplicarla de diversa manera, además de reconocerse los mencionados procesos de hibridación, permitiendo establecer diferencias entre las dos culturas, desmontando así la ambiciosa visión de carácter unilateral que se tenía sobre la globalización.Abstract: The role played by libraries nowadays has been frequently questioned in view of the technological revolution currently underway, which would seem to threaten the basic function libraries had up until now. However, what we are actually seeing is a trend in which a growing number of increasingly complex libraries are being built. Therefore, it is of interest to take a look at where architecture in this sector is heading. The truth is, a change can be observed in what is considered to be the model role of libraries today, which in turn leads to a different response in terms of their architecture. The social changes that have taken place as a result of developments in technology have played both a direct and indirect role in such a change of model and represent the challenges facing current architecture. Given the uncertainty that exists these days due to the constant changes occurring as a result of the rapid evolution of new technologies and of society’s new demands, it has become essential to build a flexibility component into all new library design concepts, while shying away from the classic solution of uniform spaces with little plastic variety or expression. On the other hand, the effects of globalisation present at all scales of life on this planet inevitably arise and tend to drive everything towards a common universality. So it is interesting to study the architecture of libraries from different cultural perspectives, firstly in Western culture, given the important role it has played in the origin of this process, and subsequently from the viewpoint of Oriental culture, practically defined as the antithesis of the former. On the basis of the above premises, this paper seeks to stand as the initial contact in a line of research that identifies new ways of applying flexibility into the architecture of libraries so that they do not become obsolete in terms of their design, as well as identifying the globalisation processes that seek to reduce current reality through homogenisation procedures. Therefore, it aims to identify hybridisation processes in current architecture that serve to ascertain whether distinctions occur between different cultures and to assess the extent to which local issues find a place in iconic buildings representing each of those cultures. By means of the methodology used, based on individual case studies and comparative analysis of each, certain traits have been revealed that show an evolution in contemporary architecture, in which greater importance is given in this new scenario to flexibility and the ability to apply it in different ways, as well as acknowledging the aforementioned hybridization processes, enabling differences between the two cultures to be identified and thus any narrow-minded view of globalization as a unilateral phenomenon to be dismantled

    Theoretical and experimental evidence for a post-perovskite phase of MgSiO3 in Earth's D" layer

    The Earth's lower mantle is believed to be composed mainly of (Mg,Fe)SiO3 perovskite, with lesser amounts of (Mg,Fe)O and CaSiO3). But it has not been possible to explain many unusual properties of the lowermost 150 km of the mantle (the D" layer) with this mineralogy. Here, using ab initio simulations and high-pressure experiments, we show that at pressures and temperatures of the D" layer, MgSiO3 transforms from perovskite into a layered CaIrO3-type post-perovskite phase. The elastic properties of the post-perovskite phase and its stability field explain several observed puzzling properties of the D" layer: its seismic anisotropy, the strongly undulating shear-wave discontinuity at its top and possibly the anticorrelation between shear and bulk sound velocities.Comment: PUBLISHED IN Nature 430, 445-448 (2004

    Low pH immobilizes and kills human leukocytes and prevents transmission of cell-associated HIV in a mouse model

    BACKGROUND: Both cell-associated and cell-free HIV virions are present in semen and cervical secretions of HIV-infected individuals. Thus, topical microbicides may need to inactivate both cell-associated and cell-free HIV to prevent sexual transmission of HIV/AIDS. To determine if the mild acidity of the healthy vagina and acid buffering microbicides would prevent transmission by HIV-infected leukocytes, we measured the effect of pH on leukocyte motility, viability and intracellular pH and tested the ability of an acidic buffering microbicide (BufferGel(®)) to prevent the transmission of cell-associated HIV in a HuPBL-SCID mouse model. METHODS: Human lymphocyte, monocyte, and macrophage motilities were measured as a function of time and pH using various acidifying agents. Lymphocyte and macrophage motilities were measured using video microscopy. Monocyte motility was measured using video microscopy and chemotactic chambers. Peripheral blood mononuclear cell (PBMC) viability and intracellular pH were determined as a function of time and pH using fluorescent dyes. HuPBL-SCID mice were pretreated with BufferGel, saline, or a control gel and challenged with HIV-1-infected human PBMCs. RESULTS: Progressive motility was completely abolished in all cell types between pH 5.5 and 6.0. Concomitantly, at and below pH 5.5, the intracellular pH of PBMCs dropped precipitously to match the extracellular medium and did not recover. After acidification with hydrochloric acid to pH 4.5 for 60 min, although completely immotile, 58% of PBMCs excluded ethidium homodimer-1 (dead-cell dye). In contrast, when acidified to this pH with BufferGel, a microbicide designed to maintain vaginal acidity in the presence of semen, only 4% excluded dye at 10 min and none excluded dye after 30 min. BufferGel significantly reduced transmission of HIV-1 in HuPBL-SCID mice (1 of 12 infected) compared to saline (12 of 12 infected) and a control gel (5 of 7 infected). CONCLUSION: These results suggest that physiologic or microbicide-induced acid immobilization and killing of infected white blood cells may be effective in preventing sexual transmission of cell-associated HIV

    Detection of Usutu virus infection in wild birds in the United Kingdom, 2020

    In August 2020, as part of a long-term disease surveillance programme, Usutu virus was detected in five Eurasian blackbirds (Turdus merula) and one house sparrow (Passer domesticus) from Greater London, England. This was initially detected by reverse transcription-PCR and was confirmed by virus isolation and by immunohistochemical detection of flavivirus in tissues. Phylogenetic analysis identified Usutu virus African 3.2 lineage, which is prevalent in the Netherlands and Belgium, suggesting a potential incursion from mainland Europe

    Considering the role of cognitive control in expert performance

Dreyfus and Dreyfus’ (1986) influential phenomenological analysis of skill acquisition proposes that expert performance is guided by non-cognitive responses which are fast, effortless and apparently intuitive in nature. Although this model has been criticised (e.g., by Breivik Journal of Philosophy of Sport, 34, 116–134 2007, Journal of the Philosophy of Sport, 40, 85–106 2013; Eriksen 2010; Montero Inquiry:An interdisciplinary Journal of Philosophy, 53, 105–122 2010; Montero and Evans 2011) for over-emphasising the role that intuition plays in facilitating skilled performance, it does recognise that on occasions (e.g., when performance goes awry for some reason) a form of ‘detached deliberative rationality’ may be used by experts to improve their performance. However, Dreyfus and Dreyfus (1986) see no role for calculative problem solving or deliberation (i.e., drawing on rules or mental representations) when performance is going well. In the current paper, we draw on empirical evidence, insights from athletes, and phenomenological description to argue that ‘continuous improvement’ (i.e., the phenomenon whereby certain skilled performers appear to be capable of increasing their proficiency even though they are already experts; Toner and Moran 2014) among experts is mediated by cognitive (or executive) control in three distinct sporting situations (i.e., in training, during pre-performance routines, and while engaged in on-line skill execution). We conclude by arguing that Sutton et al. Journal of the British Society for Phenomenology, 42, 78–103 (2011) ‘applying intelligence to the reflexes’ (AIR) approach may help to elucidate the process by which expert performers achieve continuous improvement through analytical/mindful behaviour during training and competition

    Anti-HIV-1 activity of anionic polymers: a comparative study of candidate microbicides

    BACKGROUND: Cellulose acetate phthalate (CAP) in soluble form blocks coreceptor binding sites on the virus envelope glycoprotein gp120 and elicits gp41 six-helix bundle formation, processes involved in virus inactivation. CAP is not soluble at pH < 5.5, normal for microbicide target sites. Therefore, the interaction between insoluble micronized CAP and HIV-1 was studied. Carbomer 974P/BufferGel; carrageenan; cellulose sulfate; dextran/dextrin sulfate, poly(napthalene sulfonate) and poly(styrene-4-sulfonate) are also being considered as anti-HIV-1 microbicides, and their antiviral properties were compared with those of CAP. METHODS: Enzyme linked immunosorbent assays (ELISA) were used to (1) study HIV-1 IIIB and BaL binding to micronized CAP; (2) detect virus disintegration; and (3) measure gp41 six-helix bundle formation. Cells containing integrated HIV-1 LTR linked to the β-gal gene and expressing CD4 and coreceptors CXCR4 or CCR5 were used to measure virus infectivity. RESULTS: 1) HIV-1 IIIB and BaL, respectively, effectively bound to micronized CAP. 2) The interaction between HIV-1 and micronized CAP led to: (a) gp41 six-helix bundle formation; (b) virus disintegration and shedding of envelope glycoproteins; and (c) rapid loss of infectivity. Polymers other than CAP, except Carbomer 974P, elicited gp41 six-helix bundle formation in HIV-1 IIIB but only poly(napthalene sulfonate), in addition to CAP, had this effect on HIV-1 BaL. These polymers differed with respect to their virucidal activities, the differences being more pronounced for HIV-1 BaL. CONCLUSIONS: Micronized CAP is the only candidate topical microbicide with the capacity to remove rapidly by adsorption from physiological fluids HIV-1 of both the X4 and R5 biotypes and is likely to prevent virus contact with target cells. The interaction between micronized CAP and HIV-1 leads to rapid virus inactivation. Among other anionic polymers, cellulose sulfate, BufferGel and aryl sulfonates appear most effective in this respect

    Assessment of the proliferative, apoptotic and cellular renovation indices of the human mammary epithelium during the follicular and luteal phases of the menstrual cycle

    Introduction During the menstrual cycle, the mammary gland goes through sequential waves of proliferation and apoptosis. in mammary epithelial cells, hormonal and non-hormonal factors regulate apoptosis. To determine the cyclical effects of gonadal steroids on breast homeostasis, we evaluated the apoptotic index ( AI) determined by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling ( TUNEL) staining in human mammary epithelial cells during the spontaneous menstrual cycle and correlated it with cellular proliferation as determined by the expression of Ki-67 during the same period.Methods Normal breast tissue samples were obtained from 42 randomly selected patients in the proliferative ( n = 21) and luteal ( n = 21) phases. Menstrual cycle phase characterization was based on the date of the last and subsequent menses, and on progesterone serum levels obtained at the time of biopsy.Results the proliferation index ( PI), defined as the number of Ki-67-positive nuclei per 1,000 epithelial cells, was significantly larger in the luteal phase (30.46) than in the follicular phase (13.45; P = 0.0033). the AI was defined as the number of TUNEL-positive cells per 1,000 epithelial cells. the average AI values in both phases of the menstrual cycle were not statistically significant ( P = 0.21). However, the cell renewal index ( CRI = PI/AI) was significantly higher in the luteal phase ( P = 0.033). A significant cyclical variation of PI, AI and CRI was observed.
