    Fingolimod phosphate protection against mitochondrial damage in neuronal cells

    Background: Major role of oxidative stress in the pathogenesis of neurodegenerative diseases have been suggested, being mitochondria one of the main sources of ROS. Aim: In the present work, we have studied the antioxidant effect of fingolimod phosphate (FP) on neuronal mitochondrial function and morphology using a model of mitochondrial oxidative damage induced by menadione (Vitk3). Methods: SN4741 neuronal cells were grown (70-80% confluence) and used as control (non-treated cells) or treated cells with Vitk3 15 µM alone or in presence of FP 50 nM during 4 hours. Mitochondrial membrane potential (MMP), cytochrome c oxidase (COX) activity, mitochondrial oxygen consumption rate (OCR), mitochondrial distribution (MTG) and morphology (EM) were analysed. Statistical differences were determined using one-way ANOVA. Results: Vitk3 incubation produces a dramatical decrease in MMP compared to control (43.7 %); this can be almost totally reverted by the co-incubation of Vitk3 in presence of FP (p<0.05). A 20.7 % decrease in COX activity has been found after Vitk3 incubation, again this effect was counteracted when Vitk3 and FP are combined, restoring COX activity to control levels (p<0.05). Vitk3 incubation triggers initially an increase in OCR, decreasing dramatically (61%) after 4 hours. In experiments co-incubating Vitk3 in presence of FP, the OCR decrease found was reduced to only 17% (p<0.05). In experiments with MitoTracker™ Green, we found a change in the network pattern distribution after Vitk3 administration that partially disappears when co-incubated in presence of FP. Almost all the mitochondria treated with Vitk3 show ultrastructural alterations at the electron microscopy level while normal mitochondria can be found when Vitk3 and FP are combined. Conclusion: FP protects against the mitochondrial damage induced by Vitk3, as seen by the results obtained in mitochondrial functional markers, distribution and morphology.Universidad de Málaga.     Echinodermata present a high regenerative potential expressed at different anatomical levels in bothadults and larvae. Wound healing in the arm tip after amputation in the brittle star Ophioplocus januarii wasexamined in San José gulf, Argentina, to observe histological and morphological changes. The process of woundhealing is described and compared with other brittle stars species. This process in O. januarii involves the closureof the wound, the repair of the tissue damage and posterior formation of a presumptive stalked blastema byundifferentiated cells, sclerocytes and presumptive blastemic cells. The sclerocytes are the more abundant celltype due to this cells are crucial to the future skeleton regeneration.     Characterization and Agronomic Evaluation of Chia Germplasm in La Plata, Buenos Aires, Argentina

    Trabajo presentado a IV Conference Ia ValSe-Food CYTED and VII Symposium Chia-Link, La Plata y Jujuy, Argentina, 14–18 noviembre de 2022Chia (Salvia hispanica L.), an ancestral crop currently revalued for its nutritional properties, is one of the main sources of omega-3 fatty acids. It is a short-day plant and sensitive to frost. The objective of this study was to assess the possibility of growing chia in La Plata (Buenos Aires, Argentina) (34°54′29″ S, 58°2′25″ W) by analyzing interpopulation variability between four accessions of this species. Chia was sown on February 11 in a randomized complete block design (three replicates). Ten uniform and representative plants per plot were labeled and monitored throughout the crop cycle. Phenological stages and morphological characteristics (plant height, width of the main stem, number of pairs of folded leaves and number of pairs of side shoots, and length of central inflorescence) were recorded from seedling emergence to harvest. The emergence and the first pair of unfolded leaves were recorded 3 and 10 days after sowing, respectively. The beginning of verticillaster emergence was detected after 50 days and the beginning of flowering after 66 days of sowing. The highest growth rates were achieved in CMP and EMP. After 77 days of sowing, CMP presented the highest values for the width of the main stem (10.4 mm) and height (90.89 cm), which were statistically different from EMP. The lowest variability between populations was found for the number of pairs of unfolded leaves and side shoots. The observed variability is promising for plant breeding to obtain cultivars capable of completing their cycle at this latitude.EEA SaltaFil: Rodríguez, María Emilia.     Secretagogue stimulation of neurosecretory cells elicits filopodial extensions uncovering new functional release sites

    Regulated exocytosis in neurosecretory cells relies on the timely fusion of secretory granules (SGs) with the plasma membrane. Secretagogue stimulation leads to an enlargement of the cell footprint (surface area in contact with the coverslip), an effect previously attributed to exocytic fusion of SGs with the plasma membrane. Using total internal reflection fluorescence microscopy, we reveal the formation of filopodia-like structures in bovine chromaffin and PC12 cells driving the footprint expansion, suggesting the involvement of cortical actin network remodeling in this process. Using exocytosis-incompetent PC12 cells, we demonstrate that footprint enlargement is largely independent of SG fusion, suggesting that vesicular exocytic fusion plays a relatively minor role in filopodial expansion. The footprint periphery, including filopodia, undergoes extensive F-actin remodeling, an effect abolished by the actomyosin inhibitors cytochalasin D and blebbistatin. Imaging of both Lifeact-GFP and the SG marker protein neuropeptide Y-mCherry reveals that SGs actively translocate along newly forming actin tracks before undergoing fusion. Together, these data demonstrate that neurosecretory cells regulate the number of SGs undergoing exocytosis during sustained stimulation by controlling vesicular mobilization and translocation to the plasma membrane through actin remodeling.     Evolution of Andean structures in the Chos Malal fold and thrust belt: Interaction between the basement and the sedimentary cover of the Neuquén Basin

    La faja corrida y plegada de Chos Malal se localiza al noroeste de la provincia de Neuquén e involucra más de 4.500 metros de sedimentos los cuales fueron depositados en la cuenca Neuquina y posteriormente deformados durante la orogenia Andina. En base a un detallado trabajo de campo se reconstruyó la geometría de las estructuras a lo largo de un perfil E-O en el cual se diferencian dos sectores: uno con la participación del basamento en la deformación y otro de deformación de la cubierta sedimentaria. La cordillera del Viento es una gran cuña que involucra el basamento paleozoico y a los Grupos Cuyo y Lotena, hasta alcanzar los niveles de yeso de la Formación Auquilco, los cuales constituyen el primer nivel de despegue. Este despegue, ubicado en un nivel más profundo que lo propuesto por otros autores, se evidencia por el cabalgamiento de parte de la Formación Tordillo sobre la Formación Vaca Muerta a través del retrocorrimiento El Alamito. Hacia el este de la cordillera del Viento, se reconocen en el perfil estructural cinco anticlinales, interpretados como pliegues por flexión y propagación de falla, con vergencia predominante hacia el este y subsidiaria hacia el oeste, los cuales presentan frecuentemente sus limbos frontales invertidos. Los anticlinales de mayor amplitud involucran a la Formación Tordillo y tienen como nivel de despegue las evaporitas de la Formación Auquilco. La restitución de las estructuras desarrolladas en la cubierta sedimentaria permitió calcular un acortamiento de 10 km (26%).The Chos Malal fold and thrust belt is located in northwestern Neuquén province and includes more than 4,500 meters of sediments that belong to the Neuquén Basin that were deformed during the Andean orogeny. Based in detailed field work, we reconstruct the geometry of the structures along an E-W cross-section which allow us to recognize two different sectors: one with the basement participation in the deformation and the other with deformation in the sedimentary cover. The cordillera del Viento is a huge basement wedge that involves paleozoics rocks and the Lotena and Cuyo Groups and reaches the levels Auquilco Formation, which represents the first significant detachment level in the cover. This detachment, localized on a more deep level than those proposed by other authors, is evidenced by the thrusting of Tordillo over Vaca Muerta Formation along the El Alamito backthrust. To the east of cordillera del Viento, five anticlines are recognized on the structural cross-section. They are interpreted as fault-bend and fault-propagation folds with predominantly vergence toward the east and subsidiary to the west, which have frequently frontals overturned limbs. The anticlines of greater amplitude involve the Tordillo Formation and have a detachment level along the evaporites of the Auquilco Formation. The restitution of the structures developed in the sedimentary cover allowed us to calculate of 10 km of shortening (26 %).Fil: Sánchez, Natalia Paola.     Institutional logics of service provision: the national and urban governance of activation policies in three European countries

    Labour market activation policies aimed at those with multiple barriers to employment require inter-agency collaboration between local organisations providing social and employment services. This local collaborative context is shaped by different institutional logics (state, markets, professions and community) that are crucial for policy implementation. Using case studies of nine German, UK and Swedish cities, it is argued that different institutional logics help shape the form and operation of local collaboration between different agencies and actors and thus the concrete support for service users at the local level. Although all three countries are characterised by centralised employment policies, each city shows a prevalence, but not exclusivity, of certain institutional logics over others. These differences, partly reflecting the local context, are likely to lead to local variations in understandings of solidarity, policy implementation, inter-agency collaboration and networks of actors and national-local tensions. The paper suggests that an institutional logics approach provides a useful framework for helping to understand local variations and potential national-local policy conflicts

    Intrapartum and perinatal results associated with different degrees of staining of meconium stained amniotic fluid

    OBJECTIVE: To determine the intrapartum and perinatal results associated with different degrees of staining of meconium stained amniotic fluid (MSAF). STUDY DESIGN: In a retrospective cohort study of all singleton deliveries over a period of one year (2015) in a tertiary hospital, we compared different degrees of MSAF (yellow, green and thick) to clear amniotic fluids, and analysed in each group maternal, intrapartum and neonatal variables as well as umbilical cord blood gas analysis. RESULTS: Of the 3590 deliveries included, 503 (14%) had MSAF. The incidence of MSAF rises with gestational age at delivery, reaching 20.7% in gestations above 41 weeks compared to 4.3% below 37 weeks. As the amniotic fluid staining progresses we found a higher proportion of intrapartum fevers (p  0.05). We have observed a similar distribution of umbilical artery pH ranges in all groups (p > 0.05). CONCLUSIONS: MSAF was associated with an increase in the rate of pathological fetal heart rate patterns, intrapartum fevers, operative vaginal and cesarean section deliveries, need for neonatal resuscitation, low Apgar scores and higher fetal-neonatal mortality. Moreover, we found that the risks increase as the staining and consistency of the amniotic fluid evolves so it should alert the obstetrician and paediatrician to the potential adverse outcomes

    Mutações em JAK2, MPL e CALR em neoplasias mieloproliferativas: análise de dissociação em alta resolução

    La policitemia vera (PV), la trombocitemia esencial (TE) y la mielofibrosisidiopática (MI) constituyen las Neoplasias Mieloproliferativas cromosomaFiladelfia negativas (NMP Ph-neg). La mutación V617F en el exón 14 delgen JAK2 ha sido descripta en un 90% de los casos de PV y en un 50%de TE y MI. Recientemente, se identificaron mutaciones en el exón 10del gen MPL y en el exón 9 del gen CALR, presentes en un 5 y 73% depacientes con TE y MI sin mutaciones en JAK2, respectivamente. En el presentetrabajo se estudió la detección de dichas mutaciones en 52 pacientescon NMP, mediante amplificaciones por PCR en Tiempo Real con posterioranálisis por High Resolution Melting (HRM) y secuenciación. La mutaciónV617F en JAK2 fue registrada en un 83,3% de pacientes con PV y 42,8%con TE y MI. Un 6,25% y 56,25% de pacientes con TE y MI JAK2 negativosresultaron positivos para mutaciones en el exón 10 de gen del receptorde la trombopoyetina (MPL) y el exón 9 de gen de la calreticulina (CALR).El análisis por HRM puede ser considerado como herramienta diagnósticaeficaz para las NMP debido a su alta sensibilidad, bajo costo y tiempo deprocesado, teniendo en cuenta el impacto clínico que podría tener en los pacientes la detección temprana de dichas mutaciones.Polycythemia vera (PV), essential thrombocythemia (TE) and idiopathic myelofibrosis (MI) are Philadelphia chromosome–negative myeloproliferative neoplasms (MPN-Ph. Neg). The presence of the V617F mutation in exon 14 of the JAK2 gene has been described in 90% of cases of PV and 50% of MI and TE. Recently, mutations in exon 10 of the MPL gene and exon 9 of CALR gene have been identified, which are present in 5 to 73% of patients with TE and MI without mutations in JAK2, respectively. In this work, the detection of these mutations was studied in 52 patients with NMP, using real time PCR amplifications with subsequent High Resolution Melting (HRM) analysis and sequencing. A total of 83.3% of patients with PV and 42.8% with MI and TE were recorded as positive for the V617F mutation in JAK2. A total of 6.25% and 56.25% of the patients with MI and TE with non-mutated JAK2 were positive for mutations in MPL exon 10 and CALR exon 9.     The passage of the Farallon-Aluk spreading ridge along the Andean margin

    Plate reconstructions show that the Farallon-Aluk mid-ocean ridge interacted with the Andean marginat 30°S by ~80 Ma and then moved southwards. This process influenced the marginproducing contrasts in the magmatic evolution. At 35º30?S, the Late Cretaceous-Paleocene LosAngeles Unit (67 Ma; Fennell et al., in press), developed with a tholeiitic to alkalinesignature, and Sr-Nd isotopic trends similar to OIB magmatism (Iannelli et al., 2018). The coevalPlan de Los Yeuques Formation (34º30?S, Muñoz et al., 2018) and Naunauco Group (37-38ºS,Zamora Valcarce et al., 2006) show calc-alkaline signatures with arc-like trends. By Eocenetimes the Farallon-Aluk ridge reached the 42°S. Its influence is seen when comparing arc-likePVNM (37-38°S; Llambías and Rapela 1989) with the alkaline-like Pilcaniyeu Belt (40-42°S;Aragón et al. 2011). The segmented configuration of this spreading ridge would have favored thedevelopment of diachronic slab windows and the upwelling of more enriched mantle. This couldexplain the geochemical signatures and distribution of magmatism.
