    Representaciones del subsuelo

    El presente artículo trata sobre propuestas artísticas que giran en torno a uno de los espacios urbanos más desconocidos pero a la vez más imprescindibles de nuestra sociedad: la cloacas. Estas constituyen una compleja red de saneamiento, pero también tienen unos usos alternativos a los que originalmente se habían dispuesto. En un principio, la reestructuración de los sistemas de alcantarillado de finales del siglo XIX supuso para las ciudades occidentales un gran avance, sobre todo como sistema de salubridad e higiene. Sin embargo, a la vez que se erradicaban enfermedades mediante la eliminación de las heces, se alzaba también una industria paralela basada en la gestión de un común: los desechos. De esta manera surgieron unas problemáticas entorno a la administración y gestión de las aguas residuales, que afectaban directamente a los ecosistemas. Asimismo, toda esta infraestructura contribuyó a que se crearan distintas figuras obreros e ingenieros principalmente, pero también presos, indigentes o refugiados que pasarían a circular y poblar los túneles subterráneos. Por sí solos, estos habitantes constituyen una comunidad que es invisible para el resto de la ciudadanía. Aquí se analizan a partir de trabajos artísticos y cinematográficos que versan sobre el subsuelo las múltiples representaciones que se generan a partir de estos sujetos y de los canales que circulan o habitan

    In the centrosymmetric binuclear title compound, [Zn2Cl4(C20H17N3)2], the coordination geometry of the ZnII ion can be described as a distorted ZnN2Cl3 trigonal bipyramid (τ = 0.89), arising from the N,N′-bidentate ligand, a terminal chloride ion and two bridging chloride ions. The N atoms occupy one axial and one equatorial site and the terminal chloride ion occupies an equatorial site. The dihedral angle between the pyridine and pyrazole rings is 12.8 (2)°.     La nostra proposta inicial va ser la de fer evo lucionar les Trobades amb creadors, activitats que havíem realitzat al llarg dels darrers cursos, a través de dues activitats concretes: tallers (Workshops) i sessions expositives (Sessions en obert). Els tallers, en format workshops, havien de ser impartits per a professionals i anirien di- rigits a grups reduïts d’estudiants, es volia crear vincles més estrets entre el món acadè- mic i el professional. Les sessions expositives, en canvi, havien d’estar conformades per les conferències dels professionals (a manera de Trobades amb creadors) i taules rodones amb els estudiants participants dels workshops, obertes a un major nombre d’estudiants. Partint d’aquestes propostes inicials, el pro- jecte s’ha desenvolupat al ritme de diverses dificultats. Dues d’elles han sigut determinants en la redirecció del projecte: el no encaix com a proposta d’innovació (ja que parteix d’uns antecedents duts a terme els darrers anys) i la manca de finançament (per a dur a terme unes accions que recauen en la millora de la qualitat docent). Aquestes dues limitacions han generat un debat intens en- tre els diversos membres del grup que ens ha portat a organitzar parcialment les propostes inicials i a actuar en altres direccions. Finalment hem organitzat una nova edició de Trobades amb Creadors, de la qual hem extret valoracions molt positives, i al mateix temps hem iniciat una activitat nova que ha estat la visita als tallers dels artistes. Aquesta darrera activitat ha estat molt estimulant pels estudiants i ha incidit de manera directe en els treballs realitzats posteriorment als tallers de creació.Departament de Pintura de la Facultat de Belles Art

    Lack of synergistic effect of resveratrol and sigma-1 receptor agonist (PRE-084) in SOD1G?³A ALS mice: overlapping effects or limited therapeutic opportunity?

    Background: amyotrophic lateral sclerosis (ALS) is an adult onset neurodegenerative disease characterized by the loss of motoneurons (MNs) in the spinal cord, brainstem and motor cortex, causing progressive paralysis and death. Nowadays, there is no effective therapy and most patients die 2-5 years after diagnosis. Sigma-1R is a transmembrane protein highly expressed in the CNS and specially enriched in MNs. Mutations on the Sigma-1R leading to frontotemporal lobar degeneration-ALS were recently described in human patients. We previously reported the therapeutic role of the selective sigma-1R agonist 2-(4-morpholi-nethyl)1-phenylcyclohexanecarboxylate (PRE-084) in SOD1G93A ALS mice, that promoted spinal MN preservation and extended animal survival by controlling NMDA receptor calcium influx. Resveratrol (RSV, trans-3,4',5-trihydroxystilbene) is a natural polyphenol with promising neuroprotective effects. We recently found that RSV administration to SOD1G93A mice preserves spinal MN function and increases mice survival. These beneficial effects were associated to activation of Sirtuin 1 (Sirt1) and AMP-activated protein kinase (AMPK) pathways, leading to the modulation of autophagy and an increase of mitochondrial biogenesis. The main goal of this work was to assess the effect of combined RSV and PRE-084 administration in SOD1G93A ALS mice.Methods: we determined the locomotor performance of the animals by rotarod test and evaluated spinal motoneuron function using electrophysiological tests.Results: RSV plus PRE-084 treatment from 8 weeks of age significantly improved locomotor performance and spinal MN function, accompanied by a significant reduction of MN degeneration and an extension of mice lifespan. In agreement with our previous findings, there was an induction of PKC-specific phosphorylation of the NMDA-NR1 subunit and an increased expression and activation of Sirt1 and AMPK in the ventral spinal cord of treated SOD1G93A animals.Conclusions: although combined PRE and RSV treatment significantly ameliorated SOD1G93A mice, it did not show a synergistic effect compared to RSV-only and PRE-084-only treated groups.<br/

    Nucleotide, Cytogenetic and Expression Impact of the Human Chromosome 8p23.1 Inversion Polymorphism

    Background: the human chromosome 8p23.1 region contains a 3.8-4.5 Mb segment which can be found in different orientations (defined as genomic inversion) among individuals. The identification of single nucleotide polymorphisms (SNPs) tightly linked to the genomic orientation of a given region should be useful to indirectly evaluate the genotypes of large genomic orientations in the individuals. Results: we have identified 16 SNPs, which are in linkage disequilibrium (LD) with the 8p23.1 inversion as detected by fluorescent in situ hybridization (FISH). The variability of the 8p23.1 orientation in 150 HapMap samples was predicted using this set of SNPs and was verified by FISH in a subset of samples. Four genes (NEIL2, MSRA, CTSB and BLK) were found differentially expressed (p<0.0005) according to the orientation of the 8p23.1 region. Finally, we have found variable levels of mosaicism for the orientation of the 8p23.1 as determined by FISH. Conclusion: by means of dense SNP genotyping of the region, haplotype-based computational analyses and FISH experiments we could infer and verify the orientation status of alleles in the 8p23.1 region by detecting two short haplotype stretches at both ends of the inverted region, which are likely the relic of the chromosome in which the original inversion occurred. Moreover, an impact of 8p23.1 inversion on gene expression levels cannot be ruled out, since four genes from this region have statistically significant different expression levels depending on the inversion status.     Gut epithelial barrier dysfunction in lupus triggers a differential humoral response against gut commensals

    Introduction: Systemic lupus erythematosus is an autoimmune disease with multisystemic involvement including intestinal inflammation. Lupus-associated intestinal inflammation may alter the mucosal barrier where millions of commensals have a dynamic and selective interaction with the host immune system. Here, we investigated the consequences of the intestinal inflammation in a TLR7-mediated lupus model. Methods: IgA humoral and cellular response in the gut was measured. The barrier function of the gut epithelial layer was characterised. Also, microbiota composition in the fecal matter was analysed as well as the systemic humoral response to differential commensals. Results: The lupus-associated intestinal inflammation modifies the IgA+ B cell response in the gut-associated lymphoid tissue in association with dysbiosis. Intestinal inflammation alters the tight junction protein distribution in the epithelial barrier, which correlated with increased permeability of the intestinal barrier and changes in the microbiota composition. This permeability resulted in a differential humoral response against intestinal commensals. Discussion: Lupus development can cause alterations in microbiota composition, allowing specific species to colonize only the lupus gut.     Treatment with anti-TNF alpha protects against the neuropathy induced by the proteasome inhibitor bortezomib in a mouse model

    Bortezomib (BTZ), a proteasome inhibitor, is an effective anti-neoplastic drug used in the treatment of multiple myeloma and mantle cell lymphoma. However, it can induce a reversible peripheral neuropathy that may lead to treatment discontinuation. The mechanism through which BTZ exerts toxic effects in peripheral neurons is not clear. Release of proinflammatory cytokines after nerve damage can induce neurodegeneration, but the effects of BTZ on cytokine expression in neurons are unknown, although BTZ modulates the expression of cytokines, such as TNF-α and IL-6, in tumor cells. The aim of this study was to evaluate the expression and the role of these cytokines on the course of BTZ induced neuropathy in mice. IL-6, TNF-α, TGF-β1 and IL-1β were up-regulated in dorsal root ganglia but TNF-α and IL-6 increased faster and higher. Then, we studied the potential neuroprotective effect of selective antibodies anti-TNF-α and anti-IL-6 on the evolution of the neuropathy. Treatment with anti-TNF-α but not with anti-IL-6 significantly prevented the decrease of sensory nerve action potentials amplitude and the loss of myelinated and unmyelinated fibers. We conclude that monoclonal antibodies directed against TNF-α may be a suitable neuroprotective therapy against the neurotoxicity induced by BTZ. © 2013 Elsevier Inc

    Persistence of Asthmatic Response after Ammonium Persulfate-Induced Occupational Asthma in Mice

    Since persulfate salts are an important cause of occupational asthma (OA), we aimed to study the persistence of respiratory symptoms after a single exposure to ammonium persulfate (AP) in AP-sensitized mice. BALB/c mice received dermal applications of AP or dimethylsulfoxide (DMSO) on days 1 and 8. On day 15, they received a single nasal instillation of AP or saline. Airway hyperresponsiveness (AHR) was assessed using methacholine provocation, while pulmonary inflammation was evaluated in bronchoalveolar lavage (BAL), and total serum immunoglobulin E (IgE), IgG1 and IgG2a were measured in blood at 1, 4, 8, 24 hours and 4, 8, 15 days after the single exposure to the causal agent. Histological studies of lungs were assessed. AP-treated mice showed a sustained increase in AHR, lasting up to 4 days after the challenge. There was a significant increase in the percentage of neutrophils 8 hours after the challenge, which persisted for 24 hours in AP-treated mice. The extent of airway inflammation was also seen in the histological analysis of the lungs from challenged mice. Slight increases in total serum IgE 4 days after the challenge were found, while IgG gradually increased further 4 to 15 days after the AP challenge in AP-sensitized mice. In AP-sensitized mice, an Ig-independent response is induced after AP challenge. AHR appears immediately, but airway neutrophil inflammation appears later. This response decreases in time; at early stages only respiratory and inflammatory responses decrease, but later on immunological response decreases as well

    Identification of Aquifer Recharge Sources as the Origin of Emerging Contaminants in Intensive Agricultural Areas. La Plana de Castellón, Spain

    In urban, industrial, and agricultural areas, a vast array of contaminants may be found because they are introduced into the aquifers by different recharge sources. The emerging contaminants (ECs) correspond to unregulated contaminants, which may be candidates for future regulation depending on the results of research into their potential effects on health and on monitoring data regarding their occurrence. ECs frequently found in wastewater, such as acetaminophen, carbamazepine, primidone, and sulfamethoxazole, may be good indicators of the introduction of the reclaimed water to the aquifers. The resistance of the ECs to removal in wastewater treatment plants (WWTPs) causes them to be appropriate sewage markers. Plana de Castellón (Spain) is a coastal area that has been characterized by intensive citrus agriculture since the 1970s. Traditionally, in the southern sector of Plana de Castellón, 100% of irrigation water comes from groundwater. In recent years, local farmers have been using a mixture of groundwater and reclaimed water from wastewater treatment plants (WWTPs) to irrigate the citrus. The aims of the present study were: (i) to assess the occurrences, spatial distributions, and concentrations of selected ECs, including 32 antibiotics, 8 UV filters, and 2 nonsteroidal anti-inflammatory drugs, in groundwater in a common agricultural context; (ii) to identify the recharge (pollution) sources acting as the origin of the ECs, and (iii) to suggest ECs as indicators of reclaimed water arrival in detrital heterogeneous aquifers. The obtained data provided relevant information for the management of water resources and elucidated the fate and behavior of emerging contaminants in similar contexts
