    Spectrum of topics for world congresses and other activities of the International Society for Physical and Rehabilitation Medicine (ISPRM) : a first proposal

    Background: One of the objectives of the International Society for Physical and Rehabilitation Medicine is to improve the continuity of World Congresses. This requires the development of an abstract topic list for use in congress announcements and abstract submissions. Methods: An abstract topic list was developed on the basis of the definitions of human functioning and rehabilitation research, which define 5 main areas of research (biosciences in rehabilitation, biomedical rehabilitation sciences and engineering, clinical Physical and Rehabilitation Medicine (PRM) sciences, integrative rehabilitation sciences, and human functioning sciences). For the abstract topic list, these research areas were grouped according to the proposals of congress streams. In a second step, the first version of the list was systematically compared with the topics of the 2003 ISPRM World Congress. Results: The resulting comprehensive abstract topic list contains 5 chapters according to the definition of human functioning and rehabilitation research. Due to the high significance of clinical research, clinical PRM sciences were placed at the top of the list, comprising all relevant health conditions treated in PRM services. For congress announcements a short topic list was derived. Discussion: The ISPRM topic list is sustainable and covers a full range of topics. It may be useful for congresses and elsewhere in structuring research in PRM

    Duodenal intraepithelial lymphocytes of children with cow milk allergy preferentially bind the glycan-binding protein galectin-3

    A breakdown in intestinal homeostasis results in inflammatory bowel diseases including coeliac disease and allergy. Galectins, evolutionarily conserved beta-galactoside-binding proteins, can modulate immune-epithelial cell interactions by influencing immune cell fate and cytokine secretion. In this study we investigated the glycosylation signature, as well as the regulated expression of galectin-1 and -3 in human duodenal samples of allergic and non-allergic children. Whereas galectin-1 was predominantly localized in the epithelial compartment (epithelial cells and intraepithelial lymphocytes) and the underlying lamina propria (T cells, macrophages and plasma cells), galectin-3 was mainly expressed by crypt epithelial cells and macrophages in the lamina propria. Remarkably, expression of these galectins was not significantly altered in allergic versus non-allergic patients. Investigation of the glycophenotype of the duodenal inflammatory microenvironment revealed substantial alpha2-6-linked sialic acid bound to galactose in lamina propria plasma cells, macrophages and intraepithelial lymphocytes and significant levels of asialo core 1 O-glycans in CD68+ macrophages and enterocytes. Galectin-1 preferentially bound to neutrophils, plasma cells and enterocytes, while galectin-3 binding sites were mainly distributed on macrophages and intraepithelial lymphocytes. Notably, galectin-3, but not galectin-1 binding, was substantially increased in intraepithelial gut lymphocytes of allergic patients compared to non-allergic subjects, suggesting a potential role of galectin-3-glycan interactions in shaping epithelial-immune cell connections during allergic inflammatory processes.Fil: Mercer, Natalia.     Comunicación oralLa infancia y adolescencia se considera un momento clave en el que invertir esfuerzos en prevención de posibles dificultades. Para promover la salud en la infancia se plantea la necesidad del desarrollo de recursos que ayuden al abordaje de temas como la violencia desde un lenguaje que los menores manejen y que sirvan de forma lúdica para informar y sensibilizar a toda la población. Las herramientas basadas en la metodología creativa para conectar con los niños y su entorno han demostrado su importancia, especialmente cuando se incluye a los adultos. Estos recursos pretenden dotar a los menores y sus adultos de medios e información para poder abordar diferentes temas a través de la serie de animación que presentamos a través de un lenguaje visual, artístico y atractivo. Los docentes y los adultos responsables, apoyados por este material pueden abordar diferentes temas y promover el bienestar de forma adecuada y con metodologías lúdicas y participativas. Los recursos que se presentan tratan temas relacionados con la violencia y con el uso de las propias estrategias y fortalezas de los menores. Han sido desarrollados por expertos de la psicología y de las artes escénicas, contando con profesionales de guion, ilustración, doblaje, música y psicología. Actualmente, se cuenta con un total de tres cortos ya elaborados y/o en desarrollo que han contado con el apoyo de diferentes instituciones y organismos. La serie se desarrolla en el contexto siempre de una pandilla de amigos, de entre 8 y 10 años, tres niños y tres niñas que se reúnen en torno a un árbol madre, donde siempre hay un búho y una perrita, las situaciones que vivirán les ayudan a enfrentar distintos retos y dificultades, y a través de sus aventuras podrán ayudar a comprender a otros niños lo que está pasando y qué recursos poner en marcha.Universidad de Málaga.     DES-TECER O ESTADO COLONIAL

    EEste trabalho tem como objetivo refletir acerca de como a mudança do antigo Estado Uninacional para o atual Estado Plurinacional, na Bolívia, corrobora as práticas descoloniais que os corpos bolivianos vêm praticando ao longo dos séculos. A metodologia utilizada para o desenvolvimento desta pesquisa é de caráter eminentemente bibliográfico, justificando-se na constatação sobre como o termo Plurinacionalidade, dentro de nosso lócus, América do Sul, é pautado no reconhecimento de nossa realidade fronteiriça em que nacionalidades indígenas, negras, mestiças e brancas existem e convivem. Assim, ainda que isso não seja oficialmente reconhecido, praticamente todos os países da América do Sul são plurinacionais. Como resultados, observamos que a ideia de nação, como conceito do Estado colonial moderno, ou seja, do Estado uninacional, é invertida e passa a ser entendida, a partir de alterações em sua estrutura e nos moldes de desenvolvimento, com base em epistemologias advindas dos ancestrais indígenas. O Estado reivindicado por indígenas e/ou afrodescendentes é um horizonte que preza a pluriversalidade ante um abstrato universal empregado. A partir de teorizações que des-tecem os resquícios coloniais dentro do antigo Estado Colonial presente na Bolívia e re-tecem garantias e leis estabelecidas pela Constituição do Estado Plurinacional, buscamos refletir sobre como essa mudança alterou a estrutura étnica racial e o convívio dos corpos em meio às diferenças sem ignorá-las, rastreando suas consequências.     Uncovering the genetic basis for quality traits in the Mediterranean old wheat germplasm and phenotypic and genomic prediction assessment by cross-validation test

    The release of new wheat varieties is based on two main characteristics, grain yield and quality, to meet the consumer’s demand. Identifying the genetic architecture for yield and key quality traits has wide attention for genetic improvement to meet the global requirement. In this sense, the use of landraces represents an impressive source of natural allelic variation. In this study, a genome-wide association analysis (GWAS) with PCA and kinship matrix was performed to detect QTLs in bread wheat for fifteen quality and agronomic traits using 170 diverse landraces from 24 Mediterranean countries in two years of field trials. A total of 53 QTL hotspots containing 165 significant marker-trait associations (MTAs) were located across the genome for quality and agronomical traits except for chromosome 2D. The major specific QTL hotspots for quality traits were QTL_3B.3 (13 MTAs with a mean PVE of 8.2%) and QTL_4A.3 (15 MTAs, mean PVE of 11.0%), and for yield-related traits were QTL_2B.1 (8 MTAs, mean PVE of 7.4%) and QTL_4B.2 (5 MTAs, mean PVE of 10.0%). A search for candidate genes (CG) identified 807 gene models within the QTL hotspots. Ten of these CGs were expressed specifically in grain supporting the role of identified QTLs in Landraces, associated to bread wheat quality traits and grain formation. A cross-validation approach within the collection was performed to calculate the accuracies of genomic prediction for quality and agronomical traits, ranging from -0.03 to 0.64 for quality and 0.46 to 0.65 for agronomic traits. In addition, five prediction equations using the phenotypic data were developed to predict bread loaf volume in landraces. The prediction ability varied from 0.67 to 0.82 depending on the complexity of the traits considered to predict loaf volume

    Identification of a Molecularly-Defined Subset of Breast and Ovarian Cancer Models that Respond to WEE1 or ATR Inhibition, Overcoming PARP Inhibitor Resistance

    Cáncer de mama y de ovario; Inhibición WEE1Càncer de mama i d'ovari; Inhibició WEE1Breast and ovarian cancer; WEE1 inhibitionPurpose: PARP inhibitors (PARPi) induce synthetic lethality in homologous recombination repair (HRR)-deficient tumors and are used to treat breast, ovarian, pancreatic, and prostate cancers. Multiple PARPi resistance mechanisms exist, most resulting in restoration of HRR and protection of stalled replication forks. ATR inhibition was highlighted as a unique approach to reverse both aspects of resistance. Recently, however, a PARPi/WEE1 inhibitor (WEE1i) combination demonstrated enhanced antitumor activity associated with the induction of replication stress, suggesting another approach to tackling PARPi resistance. Experimental Design: We analyzed breast and ovarian patient-derived xenoimplant models resistant to PARPi to quantify WEE1i and ATR inhibitor (ATRi) responses as single agents and in combination with PARPi. Biomarker analysis was conducted at the genetic and protein level. Metabolite analysis by mass spectrometry and nucleoside rescue experiments ex vivo were also conducted in patient-derived models. Results: Although WEE1i response was linked to markers of replication stress, including STK11/RB1 and phospho-RPA, ATRi response associated with ATM mutation. When combined with olaparib, WEE1i could be differentiated from the ATRi/olaparib combination, providing distinct therapeutic strategies to overcome PARPi resistance by targeting the replication stress response. Mechanistically, WEE1i sensitivity was associated with shortage of the dNTP pool and a concomitant increase in replication stress. Conclusions: Targeting the replication stress response is a valid therapeutic option to overcome PARPi resistance including tumors without an underlying HRR deficiency.     Antioxidants and Oxidative Stress: Focus in Obese Pregnancies

    The prevalence of obesity in women of childbearing age around the globe has dramatically increased in the last decades. Obesity is characterized by a low-state chronic inflammation, metabolism impairment and oxidative stress, among other pathological changes. Getting pregnant in this situation involves that gestation will occur in an unhealthy environment, that can potentially jeopardize both maternal and fetal health. In this review, we analyze the role of maternal obesity-induced oxidative stress as a risk factor to develop adverse outcomes during gestation, including reduced fertility, spontaneous abortion, teratogenesis, preeclampsia, and intrauterine growth restriction. Evidences of macromolecule oxidation increase in reactive oxygen species generation and antioxidant defense alterations are commonly described in maternal and fetal tissues. Thus, antioxidant supplementation become an interesting prophylactic and therapeutic tool, that yields positive results in cellular, and animal models. However, the results from most meta-analysis studying the effect of these therapies in complicated gestations in humans are not really encouraging. It is still to be analyzed whether these therapies could work if applied to cohorts of patients at a high risk, such as those with low concentration of antioxidants or obese pregnant women
