    Asymmetric organocatalytic Strecker-type reactions of aliphatic N,N-dialkylhydrazones

    The enantioselective organocatalytic Strecker-type reaction of aliphatic N,N-dialkylhydrazones is presented. Using trimethylsilyl cyanide (TMSCN) as the cyanide source, the reaction can be efficiently catalyzed by a tert-leucine-derived bifunctional thiourea to afford the corresponding hydrazino nitriles in good to excellent yields (50-96%) and moderate to good enantioselectivities, up to 86% ee. Further transformations of the nitrile functionality allow access to useful protected hydrazino acids and imidazolidinones.     El diario de prácticas clínicas en la unidad de cuidados intensivos

    El objetivo de este trabajo se centra en conocer la experiencia clínica vivida por los alumnos de 4º curso de enfermería en las unidades de cuidados especiales. Para la obtención de los datos se empleará la utilización de una herramienta específica como es el diario de prácticas clínicas elaborado por los porpios alumnos donde se recojan las vivencias en las unidades especiales. Por otro lado se realizarán sesiones con grupos focales para debatir procesualmente, el problema de investigación a la luz de las narrativas vertidas en los diarios. Los resultados obtenidos ponen de manifiesto las siguientes categorías:-el miedo y la angustia vivido por los alumnos al comienzo en este tipo de unidades. La relevancia de la figura del tutor en este entorno. La labor desarrollada por un el equipo de soporte de la enseñanza clínica. La influencia de la tecnificación sin desatender los cuidados básicos y por último el aumento de la seguridad y confianza con el entorno con el paso de los día

    Conocer la experiencia clínica vivida por los alumnos de 4º curso de enfermería en las unidades de cuidados intensivos y reanimación. Potenciar el diario de prácticas clínicas como herramienta de reflexión durante los procesos de enseñanza aprendizaje clínico en las unidades de cuidados especiales. Introducir al alumno de prácticas clínicas en la rehumanización de los cuidados en unidades especiales (contextos clínicos cerrados y de alta incidencia tecnológica). Metodológicamente se emplearán, por una parte, los diarios de prácticas clínicas enfocados al problema planteado en esta investigación; por otra, se realizarán sesiones con grupos focales para debatir procesualmente, el problema de investigación a la luz de las narrativas vertidas en los diarios. Los principales resultados obtenidos ponen de manifiesto las siguientes categorías:-el miedo y la angustia vivido por los alumnos al comienzo en este tipo de unidades. La relevancia del tutor en este entorno. La importante labor desarrollada por un equipo multidisciplinar. La influencia de la tecnificación sin desatender los cuidados básicos.     Prognostic Factors in Patients with Rocky Mountain Spotted Fever

    Background: Rocky Mountain Spotted Fever (RMSF) is a disease with a high mortality rate, caused by Rickettsia rickettsii, a bacteria transmitted to humans by infected ticks. In 2008 there was a Rocky Mountain Spotted Fever (RMSF) outbreak in the city of Mexicali, México, resulting in an increased mortality rate amongst the area population. Methods: Case-series study of patients admitted to the General Hospital of Mexicali between 2014 and 2019 with a confirmed diagnosis of RMSF. Mortality was compared dividing the population on those ?20 and younger than ?21 years of age. Results: A total of 129 patients’ records during a 5-year period whose diagnosis was RMSF confirmed with PCR were included. Mortality was compared among patients admitted who were younger than ?20 years of age with that among patients who were older than ?20 years of age (61 versus 68 respectively), the latter being higher with an OR 4.2 (p<0.0001). Conclusion: RMSF in hospitalized patients has a high mortality rate in spite of early treatment in all age groups, without showing any predominance in gender. However, patients older than 20 years of age had a higher mortality rate than those younger than 20 years, without any predominance in gender

    Approximately 87.5% of flowering plant species are pollinated by animals, however, the magnitude of the impact on plant pollination by rodents, a group to which squirrels belong, has been underestimated due to the lack of information. For the first time, we documented the consumption of inflorescences, nectar or pollen of maguey (Agave sp.) By the Peters squirrel (Parasciurus oculatus). It is necessary to continue monitoring the pollinators of the magueys, in order to determine the role that squirrels play in the pollination or predation processes of these plants.Aproximadamente el 87,5 % de las especies de plantas con flores son polinizadas por animales, sin embargo, la magnitud del impacto en la polinización de plantas por parte de los roedores, grupo al que pertenecen las ardillas, ha sido subestimada por la falta de información. Documentamos por primera ocasión consumo de las inflorescencias, néctar o polen de maguey (Agave sp.) por la ardilla de Peters (Parasciurus oculatus). Es necesario continuar el monitoreo de los polinizadores de los magueyes, para determinar el papel que juegan las ardillas en los procesos de polinización o depredación de estas plantas

    Mycobacterium tuberculosis is a major human pathogen and the causative agent for the pulmonary disease, tuberculosis (TB). Current treatment programs to combat TB are under threat due to the emergence of multi-drug and extensively-drug resistant TB. As part of our efforts towards the discovery of new anti-tubercular leads, a number of potent tetrahydropyrazolo[1,5-a]pyrimidine-3-ca​rboxamide(THPP) and N-benzyl-6′,7′-dihydrospiro[piperidine-4,​4′-thieno[3,2-c]pyran](Spiro) analogues were recently identified against Mycobacterium tuberculosis and Mycobacterium bovis BCG through a high-throughput whole-cell screening campaign. Herein, we describe the attractive in vitro and in vivo anti-tubercular profiles of both lead series. The generation of M. tuberculosis spontaneous mutants and subsequent whole genome sequencing of several resistant mutants identified single mutations in the essential mmpL3 gene. This ‘genetic phenotype’ was further confirmed by a ‘chemical phenotype’, whereby M. bovis BCG treated with both the THPP and Spiro series resulted in the accumulation of trehalose monomycolate. In vivo efficacy evaluation of two optimized THPP and Spiro leads showed how the compounds were able to reduce >2 logs bacterial cfu counts in the lungs of infected mice

    Identification of clinical variants beyond the exome in inborn errors of metabolism

    Inborn errors of metabolism (IEM) constitute a huge group of rare diseases affecting 1 in every 1000 newborns. Next-generation sequencing has transformed the diagnosis of IEM, leading to its proposed use as a second-tier technology for confirming cases detected by clinical/biochemical studies or newborn screening. The diagnosis rate is, however, still not 100%. This paper reports the use of a personalized multi-omics (metabolomic, genomic and transcriptomic) pipeline plus functional genomics to aid in the genetic diagnosis of six unsolved cases, with a clinical and/or biochemical diagnosis of galactosemia, mucopolysaccharidosis type I (MPS I), maple syrup urine disease (MSUD), hyperphenylalaninemia (HPA), citrullinemia, or urea cycle deficiency. Eight novel variants in six genes were identified: six (four of them deep intronic) located in GALE, IDUA, PTS, ASS1 and OTC, all affecting the splicing process, and two located in the promoters of IDUA and PTS, thus affecting these genes’ expression. All the new variants were subjected to functional analysis to verify their pathogenic effects.     Cianobacterias y cianotoxinas en ecosistemas límnicos de Uruguay = Cyanobacteria and cyanotoxins in freshwaters of Uruguay

    Las floraciones de cianobacterias en cuerpos de agua dulce constituyen un problema mundial, asociado a la eutrofización (enriquecimiento de nutrientes) y cambios hidrológicos de los ecosistemas. En este estudio se generó una base de datos históricos (n= 3061 para 64 ecosistemas, de 1980 a 2014), elaborada por un grupo de trabajo interinstitucional (OSE, DINAMA, IM, Universidad de la República e IIBCE), y se analizó la distribución de cianobacterias planctónicas y de cianotoxinas (microcistina, saxitoxina y cilindrospermopsina) en Uruguay. Los valores de clorofila ay nutrientes totales indicaron procesos de eutrofización en diversos ecosistemas. En dos mapas georreferenciados se visualiza la distribución de cianobacterias en el país, según indicadores cuantitativos globales, y de la microcistina (cianotoxina más frecuente). En dichos mapas se destacan los embalses (Río Uruguay y Río Negro) y las playas del Río de la Plata como las zonas de mayor riesgo de exposición a cianobacterias según las categorías de la Organización Mundial de la Salud para aguas recreacionales. Además, se analizaron muestras de floraciones (espuma) y se cuantificó 20 mg L-1de microcistinas totales y la presencia de genes que sugieren diferentes variedades de microcistinas.     In vivo cholinergic basal forebrain degeneration and cognition in Parkinson's disease: Imaging results from the COPPADIS study

    COPPADIS Study Group.[Introduction] We aimed to assess associations between multimodal neuroimaging measures of cholinergic basal forebrain (CBF) integrity and cognition in Parkinson's disease (PD) without dementia.[Methods] The study included a total of 180 non-demented PD patients and 45 healthy controls, who underwent structural MRI acquisitions and standardized neurocognitive assessment through the PD-Cognitive Rating Scale (PD-CRS) within the multicentric COPPADIS-2015 study. A subset of 73 patients also had Diffusion Tensor Imaging (DTI) acquisitions. Volumetric and microstructural (mean diffusivity, MD) indices of CBF degeneration were automatically extracted using a stereotactic CBF atlas. For comparison, we also assessed multimodal indices of hippocampal degeneration. Associations between imaging measures and cognitive performance were assessed using linear models.[Results] Compared to controls, CBF volume was not significantly reduced in PD patients as a group. However, across PD patients lower CBF volume was significantly associated with lower global cognition (PD-CRStotal: r = 0.37, p < 0.001), and this association remained significant after controlling for several potential confounding variables (p = 0.004). Analysis of individual item scores showed that this association spanned executive and memory domains. No analogue cognition associations were observed for CBF MD. In covariate-controlled models, hippocampal volume was not associated with cognition in PD, but there was a significant association for hippocampal MD (p = 0.02).[Conclusions] Early cognitive deficits in PD without dementia are more closely related to structural MRI measures of CBF degeneration than hippocampal degeneration. In our multicentric imaging acquisitions, DTI-based diffusion measures in the CBF were inferior to standard volumetric assessments for capturing cognition-relevant changes in non-demented PD.This work was supported by the Alzheimer Forschung Initiative e.V.
