The presence of isolated or multiple gingival recessions in the mandibular anterior region is a challenge for the clinician, as they may be associated with a shallow vestibule, high frenum insertion and/or little or no attached gingiva. Only limited evidence is available on the use of the double lateral sliding bridge flap technique with connective tissue graft (CTG) technique for treating gingival recessions in the mandibular anterior region. The aim of this study was to describe and evaluate the clinical and esthetic outcomes of the double lateral sliding bridge flap technique with CTG on isolated and multiple gingival recessions at the level of the mandibular incisors. Nine patients required treatment of gingival recessions in the mandibular incisors at the University of Salamanca (Spain) (seven females, two males; mean age: 27.9 ± 6.9) with a total of 14 isolated (42.9%) and multiple (57.1%) Miller class II and III gingival recessions. After a mean follow-up of 36 months, the mean percentage of root coverage was 80.5% for all treated recessions. Statistically significant differences (p < 0.05), were observed for reduction in recession depth, increased width of keratinized tissue and increased gingival thickness, this being dependent on the Miller class. The esthetic outcome was acceptable, with a final mean esthetic score of 7.4 out of 10. The double lateral sliding bridge flap surgical technique with CTG is an effective procedure for the coverage of isolated and multiple gingival recessions in the anterior mandibular region, as it offers satisfactory esthetic results

The implementation of alley cropping in orchards has been suggested as a sustainable strategy to increase farmer revenues by crop diversification, enhance soil organic matter (SOM) and fertility, water retention, overall biodiversity, and contribute to climate change mitigation. Thus, the objective of this study was to assess if alley cropping with annual crops can contribute to i) mitigate soil greenhouse gas (GHG) emissions, ii) enhance C sequestration in a semiarid Mediterranean irrigated citrus orchard, and iii) increase land productivity. For this, two different treatments were established: i) conventional mandarin monoculture (MC) with no alley cropping; and ii) mandarin diversified with alley cropping of barley/vetch and fava bean (DIV). Measurements of soil CO2 and N2O emissions were periodically performed (every 7–20 days) during two years. Soil CO2 emission rates followed the soil moisture trend, and showed no significant differences between treatments. As an average, soil CO2 emission rates were 147 mg m-2 h-1 in MC and 196 mg m-2 h-1 in DIV. Soil N2O emission rates were not correlated to soil moisture nor temperature, and showed average values of 0.026 mg m-2 h-1 in MC and − 0.002 mg m-2 h-1 in DIV. Alley cropping did not contribute to significantly increase soil organic C and total nitrogen in two years’ time. With regard to production, mandarin yield showed no significant differences between treatments, but alley crops contributed to complementary commodities to the main cash crop, increasing overall land productivity. Thus, alley cropping in irrigated Mediterranean orchards has no significant effect on soil C sequestration and GHG emissions at short-term, with increased land productivity owing to new commodities grown in the alleys. These results confirm that under semiarid Mediterranean climate, long periods are needed to efficiently assess soil C sequestration potential of sustainable practices in orchards. This work was supported by the European Commission Horizon 2020 Project Diverfarming [Grant agreement 728003]. There is now sufficient evidence to support that vitamin D deficiency may predispose to SARS-CoV-2 infection and increase COVID-19 severity and mortality. It has been suggested that vitamin D3 supplementation may be used prophylactically as an affordable and safe strategy that could be added to the existing COVID-19 standard treatment. This multicenter, single-blinded, prospective randomized pilot clinical trial aimed to evaluate the safety, tolerability, and effectiveness of 10,000 IU/day in comparison with 2000 IU/day of cholecalciferol supplementation for 14 days to reduce the duration and severity of COVID-19 in 85 hospitalized individuals. The median age of the participants was 65 years (Interquartile range (IQR): 53-74), most of them (71%) were men and the mean baseline of 25-hydroxyvitamin D (25(OH)D) in serum was 15 ng/ml (standard deviation (SD):6). After 14 days of supplementation, serum 25(OH)D levels were significantly increased in the group who received 10,000IU/day (p < 0.0001) (n = 44) in comparison with the 2,000IU/day group (n = 41), especially in overweight and obese participants, and the higher dose was well tolerated. A fraction of the individuals in our cohort (10/85) developed acute respiratory distress syndrome (ARDS). The median length of hospital stay in these patients with ARDS was significantly different in the participants assigned to the 10,000IU/day group (n = 4; 7 days; IQR: 4-13) and the 2,000IU/day group (n = 6; 27 days; IQR: 12-45) (p = 0.04). Moreover, the inspired oxygen fraction was reduced 7.6-fold in the high dose group (p = 0.049). In terms of blood parameters, we did not identify overall significant improvements, although the platelet count showed a modest but significant difference in those patients who were supplemented with the higher dose (p = 0.0492). In conclusion, the administration of 10,000IU/day of vitamin D3 for 14 days in association with the standard clinical care during hospitalization for COVID-19 was safe, tolerable, and beneficial, thereby helping to improve the prognosis during the recovery process. This work was supported by Fundación Universidad Alfonso X el Sabio (FUAX, Madrid, Spain; ID Project: 1.012.010; ID Project EQA: 925.280); the Coordinated Research Activities at the National Center of Microbiology (Instituto de Salud Carlos III) (COV20_00679) to promote an integrated response against SARS-CoV-2 in Spain (Spanish Ministry of Science and Innovation) that is coordinated by Dr. Inmaculada Casas (WHO National Influenza Center of the CNM); a generous donation provided by Chiesi España, S.A.U. Trabajo presentado en la International Plant and Animal Genome Conference, celebrada en San Diego, CA (Estados Unidos), del 13 al 18 de enero de 2023Pistachio (Pistacia vera L., 2n =2x = 30) is one of the most economically important long-lived crops, including cashew and mango in the family Anacardiaceae. Although dioecious pistachio trees are xerophytic and highly resilient to abiotic stresses, they require a certain amount of winter rest period to exit bud dormancy and to flower synchronically between male and female trees. The `Kerman¿ (P. vera cv. Kerman) and `Peter¿ (P. vera cv. Peter) have been the dominant pair of female and male cultivars traditionally grown in California for more than 50 years, whereas the understanding of genomic diversity and approaches for pistachio breeding is limited without the reference-quality genome. Genetic diversity, such as structural and gene content variation, is a major source for crop improvement and breeding, which cannot be captured from a single reference genome. In the present study, we report haplotype-resolved chromosome-level reference genomes of six genetically diverse collections of pistachio (`Kerman¿, `Mateur¿, `Sirora¿, Napoletana¿, `Chaparrillo¿, and `T41¿). The diploid assemblies ranged from 568.55 to 608.21 Mb in size with contig N50 of 28 - 37 Mb. More than 98% of conserved orthologs (BUSCO) were represented for all genome assemblies. The dynamics of transposable elements (TEs) were characterized in the genomes where the TEs accounted for about 65 - 68% of the genomes. The gene annotation analyses using Iso-seq data generated from different five tissue types predicted about 38,000 protein-coding genes. Two haplotypes of each genome showed substantial sequence variation, suggesting high heterozygosity in pistachio genomes. Pan-genome construction and analyses characterized remarkable presence-absence gene content variation between cultivars. The reference genomes with fully annotated genes and pan-genome construction serve as rich resources for genomic breeding and improvement and comparative genomics of pistachio

Protein Identification and Haplotype Description of Homozygote Mutation Causing Congenital Plasminogen Deficiency

    Get PDF
Severe type I Plasminogen (PLG) deficiency was clinically diagnosed after hyaline-positive periodic acid Schiff material was detected in the histologic study of superior tarsal conjunctiva and vulvar pseudomembrane of the patient. Direct immunofluorescence also confirmed multiple deposits of fibrinogen in the dermis. Plasma plasminogen activity was calculated in a <5% value (reference values, 75% to 150%) and sequencing of the PLG gene evidenced the homozygous mutation in c.2377T/A (p.Tyr793Asn), confirming the molecular diagnosis of congenital deficiency of plasminogen type 1. Genotype-Phenotype correlation among family members evidenced the recessive hereditary pattern of clinical manifestations of chronic inflammatory disease of the mucous membranes due to PLG deficiency, but co-dominance effect to present a decreased plasma plasminogen activity (46%) among heterozygous asymptomatic individuals. SNPs/CNVs whole genome array hybridization analysis in the patient, detected long Loss of Heterozygosity regions (LOH) and demonstrated the consanguinity in the family. Proteomic analysis identified impaired secretion of mutant PLG tissue specific proteins, as definitive molecular etiopathogenesis of the type I PLG deficiency in the patient

Siglo Cero. 43 (243): 7-28.)
