    Oral bacterial microbiota and traumatic injuries of free-ranging Phrynops geoffroanus (Testudines, Chelidae) in southeastern Brazil

    During 2006 and 2007, we collected free-ranging Phrynops geoffroanus, from two anthropogenically altered rivers in southeastern Brazil. Oral microbiological samples were taken for isolation of aerobic and facultative anaerobic bacteria; a physical examination was performed;and we evaluated possible effects on the turtles’ health. Twenty-nine species of bacteria were isolated in Piracicaba River turtles (n=10), and twenty-four species in Piracicamirim stream turtles (n=8), most of them gram-negative. In both sites, potential pathogens for reptiles were: Escherichia coli, Klebsiella pneumoniae, Enterobacter agglomerans, Citrobacter freundii, and Bacillus sp. Although boatpropeller lesions were common on the carapace of the turtles, we have not found turtles with signs of clinical diseases. The oral bacterial microbiota of P. geoffroanus inhabiting the Piracicaba River basin are composed of a diverse microbe spectrum, and long-term studies of the effects of pollution and traumatic injuries on this population and its microbial flora are warranted

    Entre 2006 e 2007, coletamos amostras microbiológicas da cavidade oral de cágados Phrynops geoffroanus de vida livre, em dois rios antropizados da região sudeste do Brasil, para o isolamento de bactérias aeróbicas e anaeróbicas facultativas, e realizamos um exame físico e avaliamos possíveis efeitos sobre o estado de saúde dos animais. Vinte e nove espécies de bactérias foram isoladas em cágados do rio Piracicaba (n=10), e vinte e quatro espécies em animais do ribeirão Piracicamirim (n=8), a maioria delas gram-negativas. Em ambos os sítios estudados foram isolados patógenos potenciais para répteis, como Escherichia coli, Klebsiella pneumoniae, Enterobacter agglomerans, Citrobacter freundii e Bacillus sp. Embora lesões na carapaça dos cágados, causadas por hélices de barcos, foram um achado comum no exame físico, não encontramos animais com sinais clínicos de doenças.     The importance of acoustic background modelling in CNN-based detection of the neotropical White-lored Spinetail (Aves, Passeriformes, Furnaridae)

    Machine learning tools are widely used in support of bioacoustics studies, and there are numerous publications on the applicability of convolutional neural networks (CNNs) to the automated presence-absence detection of species. However, the relation between the merit of acoustic background modelling and the recognition performance needs to be better understood. In this study, we investigated the influence of acoustic background substance on the performance of the acoustic detector of the White-lored Spinetail (Synallaxis albilora). Two detector designs were evaluated: the 152-layer ResNet with transfer learning and a purposely created CNN. We experimented with acoustic background representations trained with season-specific (dry, wet, and all-season) data and without explicit modelling to evaluate its influence on the detection performance. The detector permits monitoring of the diel behaviour and breeding time of White-lored Spinetail solely based on the changes in the vocal activity patterns. We report an advantageous performance when background modelling is used, precisely when trained with all-season data. The highest classification accuracy (84.5%) was observed for the purposely created CNN model.     H1N1pdm Influenza Infection in Hospitalized Cancer Patients: Clinical Evolution and Viral Analysis

    BACKGROUND: The novel influenza A pandemic virus (H1N1pdm) caused considerable morbidity and mortality worldwide in 2009. The aim of the present study was to evaluate the clinical course, duration of viral shedding, H1N1pdm evolution and emergence of antiviral resistance in hospitalized cancer patients with severe H1N1pdm infections during the winter of 2009 in Brazil. METHODS: We performed a prospective single-center cohort study in a cancer center in Rio de Janeiro, Brazil. Hospitalized patients with cancer and a confirmed diagnosis of influenza A H1N1pdm were evaluated. The main outcome measures in this study were in-hospital mortality, duration of viral shedding, viral persistence and both functional and molecular analyses of H1N1pdm susceptibility to oseltamivir. RESULTS: A total of 44 hospitalized patients with suspected influenza-like illness were screened. A total of 24 had diagnosed H1N1pdm infections. The overall hospital mortality in our cohort was 21%. Thirteen (54%) patients required intensive care. The median age of the studied cohort was 14.5 years (3-69 years). Eighteen (75%) patients had received chemotherapy in the previous month, and 14 were neutropenic at the onset of influenza. A total of 10 patients were evaluated for their duration of viral shedding, and 5 (50%) displayed prolonged viral shedding (median 23, range=11-63 days); however, this was not associated with the emergence of a resistant H1N1pdm virus. Viral evolution was observed in sequentially collected samples. CONCLUSIONS: Prolonged influenza A H1N1pdm shedding was observed in cancer patients. However, oseltamivir resistance was not detected. Taken together, our data suggest that severely ill cancer patients may constitute a pandemic virus reservoir with major implications for viral propagation

    Iron Deficiency Generates Oxidative Stress and Activation of the SOS Response in Caulobacter crescentus

    In C. crescentus, iron metabolism is mainly controlled by the transcription factor Fur (ferric uptake regulator). Iron-bound Fur represses genes related to iron uptake and can directly activate the expression of genes for iron-containing proteins. In this work, we used total RNA sequencing (RNA-seq) of wild type C. crescentus growing in minimal medium under iron limitation and a fur mutant strain to expand the known Fur regulon, and to identify novel iron-regulated genes. The RNA-seq of cultures treated with the iron chelator 2-2-dypiridyl (DP) allowed identifying 256 upregulated genes and 236 downregulated genes, being 176 and 204 newly identified, respectively. Sixteen transcription factors and seven sRNAs were upregulated in iron limitation, suggesting that the response to low iron triggers a complex regulatory network. Notably, lexA along with most of its target genes were upregulated, suggesting that DP treatment caused DNA damage, and the SOS DNA repair response was activated in a RecA-dependent manner, as confirmed by RT-qPCR. Fluorescence microscopy assays using an oxidation-sensitive dye showed that wild type cells in iron limitation and the fur mutant were under endogenous oxidative stress, and a direct measurement of cellular H2O2 showed that cells in iron-limited media present a higher amount of endogenous H2O2. A mutagenesis assay using the rpoB gene as a reporter showed that iron limitation led to an increase in the mutagenesis rate. These results showed that iron deficiency causes C. crescentus cells to suffer oxidative stress and to activate the SOS response, indicating an increase in DNA damage

    PROTAGONISMO DO ENFERMEIRO NO PROCESSO DE CICATRIZAÇÃO DAS FERIDAS CRÔNICAS: UM ENSAIO DA LITERATURA

    Introduction: Chronic wounds are a type of lesion that has its healing process stagnated for a period of six weeks or more, despite adequate treatment. Objectives: to understand the role of nurses in the assessment and treatment of chronic wounds and to describe their performance in the face of the systematization of nursing care in the care process. Methodology: This is a literature search with a qualitative approach. The data were collected in a virtual database. For this purpose, the Virtual Health Library (VHL) was used, in the following information base: LILACS; BDENF and MEDLINE in the period from March to April 2021. After the association of all descriptors, 85 articles were found, 70 were excluded and 15 articles were selected. Results and Discussion: After reflective reading and repetition of the themes developed in the articles found, two categories emerged: the role of the nurse in the assessment and treatment of chronic wounds and the role of the nurse in the systematization of nursing care in the care process. Conclusion: It is concluded that it is of great relevance that the dermatologist nurse has theoretical and scientific knowledge about chronic wounds for implementation in practice and, thus, to meet the demands arising from the patient, considering that, care for the patient with a chronic wound needs to be holistically targeted.Introducción: Las heridas crónicas son un tipo de lesión que tiene su proceso de cicatrización estancado por un período de seis semanas o más, a pesar de un tratamiento adecuado. Objetivos: comprender el papel del enfermero en la evaluación y tratamiento de las heridas crónicas y describir su desempeño frente a la sistematización del cuidado de enfermería en el proceso asistencial. Metodología: Se trata de una búsqueda bibliográfica con enfoque cualitativo. Los datos se recopilaron en una base de datos virtual. Para ello se utilizó la Biblioteca Virtual en Salud (BVS), en la siguiente base de información: LILACS; BDENF y MEDLINE en el período de marzo a abril de 2021. Tras la asociación de todos los descriptores, se encontraron 85 artículos, se excluyeron 70 y se seleccionaron 15 artículos. Resultados y Discusión: Luego de la lectura reflexiva y repetición de los temas desarrollados en los artículos encontrados, surgieron dos categorías: el rol del enfermero en la evaluación y tratamiento de las heridas crónicas y el rol del enfermero en la sistematización del cuidado de enfermería en el cuidado. proceso. Conclusión: Se concluye que es de gran relevancia que el enfermero dermatólogo cuente con conocimientos teóricos y científicos sobre las heridas crónicas para su implementación en la práctica y, así, atender las demandas que surjan del paciente, considerando que, el cuidado del paciente con una enfermedad crónica.     Avaliação dos efeitos miorelaxante, antiespasmódico e antinociceptivo do extrato aquoso da Phoradendron piperoides (Kunt.) Trel. (Viscaceae)

    Foram investigados os efeitos miorelaxante, antiespasmódico e antinociceptivo do extrato aquoso liofi lizado das folhas da Phoradendron piperoides. A toxicidade aguda também foi avaliada. No íleo isolado de cobaio, o extrato aquoso da P. piperoides (0,05 - 2,0 mg/mL) produziu relaxamento de forma concentração-dependente (IC50 = 0,114 mg/mL) e, na concentração de 1,5 mg/mL, reduziu a amplitude das contrações induzidas por carbacol (2 μM), histamina (2 μM) e BaCl2 (0,03 M) em 46,6; 38,6 e 55,3% (p < 0,001), respectivamente. Em camundongos, o extrato aquoso liofi lizado (100-400 mg/kg) não reduziu de forma signifi cativa as contorções abdominais induzidas por ácido acético, não modifi cou o tempo de reação dos animais no teste da formalina e não aumentou o tempo de latência ao calor no teste da placa quente. No ensaio de toxicidade aguda utilizado, não foi detectada a morte de nenhum animal após tratamento com doses de até 5 g/kg (p.o.) do extrato. Em conclusão, os resultados obtidos indicam que o extrato aquoso da P. piperoides apresenta efeito antiespasmódico e baixa toxicidade aguda. O extrato, no entanto, não possui efeito antinociceptivo

    SARS-CoV-2 uses CD4 to infect T helper lymphocytes

    The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the agent of a major global outbreak of respiratory tract disease known as Coronavirus Disease 2019 (COVID-19). SARS-CoV-2 infects mainly lungs and may cause several immune-related complications, such as lymphocytopenia and cytokine storm, which are associated with the severity of the disease and predict mortality. The mechanism by which SARS-CoV-2 infection may result in immune system dysfunction is still not fully understood. Here, we show that SARS-CoV-2 infects human CD4+ T helper cells, but not CD8+ T cells, and is present in blood and bronchoalveolar lavage T helper cells of severe COVID-19 patients. We demonstrated that SARS-CoV-2 spike glycoprotein (S) directly binds to the CD4 molecule, which in turn mediates the entry of SARS-CoV-2 in T helper cells. This leads to impaired CD4 T cell function and may cause cell death. SARS-CoV-2-infected T helper cells express higher levels of IL-10, which is associated with viral persistence and disease severity. Thus, CD4-mediated SARS-CoV-2 infection of T helper cells may contribute to a poor immune response in COVID-19 patients.</p
