Lymphatic vasculature mediates macrophage reverse cholesterol transport in mice
Reverse cholesterol transport (RCT) refers to the mobilization of cholesterol on HDL particles (HDL-C) from extravascular tissues to plasma, ultimately for fecal excretion. Little is known about how HDL-C leaves peripheral tissues to reach plasma. We first used 2 models of disrupted lymphatic drainage from skin - 1 surgical and the other genetic - to quantitatively track RCT following injection of [H-3]-cholesterol-loaded macrophages upstream of blocked or absent lymphatic vessels. Macrophage RCT was markedly impaired in both models, even at sites with a leaky vasculature. Inhibited RCT was downstream of cholesterol efflux from macrophages, since macrophage efflux of a fluorescent cholesterol analog (BODIPY-cholesterol) was not altered by impaired lymphatic drainage. We next addressed whether RCT was mediated by lymphatic vessels from the aortic wall by loading the aortae of donor atherosclerotic Apoe-deficient mice with [H-2](6)-labeled cholesterol and surgically transplanting these aortae into recipient Apoe-deficient mice that were treated with anti-VEGFR3 antibody to block lymphatic regrowth or with control antibody to allow such regrowth. [H-2]-Cholesterol was retained in aortae of anti-VEGFR3-treated mice. Thus, the lymphatic vessel route is critical for RCT from multiple tissues, including the aortic wall. These results suggest that supporting lymphatic transport function may facilitate cholesterol clearance in therapies aimed at reversing atherosclerosis.LLC
Creating metamaterial building blocks with directed photochemical metallization of silver onto DNA origami templates
DNA origami can be used to create a variety of complex and geometrically unique nanostructures that can be further modified to produce building blocks for applications such as in optical metamaterials. We describe a method for creating metal-coated nanostructures using DNA origami templates and a photochemical metallization technique. Triangular DNA origami forms were fabricated and coated with a thin metal layer by photochemical silver reduction while in solution or supported on a surface. The DNA origami template serves as a localized photosensitizer to facilitate reduction of silver ions directly from solution onto the DNA surface. The metallizing process is shown to result in a conformal metal coating, which grows in height to a self-limiting value with increasing photoreduction steps. Although this coating process results in a slight decrease in the triangle dimensions, the overall template shape is retained. Notably, this coating method exhibits characteristics of self-limiting and defect-filling growth, which results in a metal nanostructure that maps the shape of the original DNA template with a continuous and uniform metal layer and stops growing once all available DNA sites are exhausted
A comparative evaluation of PDQ-Evidence
BACKGROUND: A strategy for minimising the time and obstacles to accessing systematic reviews of health system
evidence is to collect them in a freely available database and make them easy to find through a simple ‘Google-style’
search interface. PDQ-Evidence was developed in this way. The objective of this study was to compare PDQ-Evidence
to six other databases, namely Cochrane Library, EVIPNet VHL, Google Scholar, Health Systems Evidence, PubMed
and Trip.
METHODS: We recruited healthcare policy-makers, managers and health researchers in low-, middle- and highincome
countries. Participants selected one of six pre-determined questions. They searched for a systematic
review that addressed the chosen question and one question of their own in PDQ-Evidence and in two of the
other six databases which they would normally have searched. We randomly allocated participants to search
PDQ-Evidence first or to search the two other databases first. The primary outcomes were whether a systematic
review was found and the time taken to find it. Secondary outcomes were perceived ease of use and perceived
time spent searching. We asked open-ended questions about PDQ-Evidence, including likes, dislikes, challenges
and suggestions for improvements.
RESULTS: A total of 89 people from 21 countries completed the study; 83 were included in the primary analyses
and 6 were excluded because of data errors that could not be corrected. Most participants chose PubMed and
Cochrane Library as the other two databases. Participants were more likely to find a systematic review using
PDQ-Evidence than using Cochrane Library or PubMed for the pre-defined questions. For their own questions, this
difference was not found. Overall, it took slightly less time to find a systematic review using PDQ-Evidence. Participants
perceived that it took less time, and most participants perceived PDQ-Evidence to be slightly easier to use than the
two other databases. However, there were conflicting views about the design of PDQ-Evidence.
CONCLUSIONS: PDQ-Evidence is at least as efficient as other databases for finding health system evidence. However,
using PDQ-Evidence is not intuitive for some people
Evidence of a causal relationship between body mass index and psoriasis:A mendelian randomization study
Background:
Psoriasis is a common inflammatory skin disease that has been reported to be associated with obesity. We aimed to investigate a possible causal relationship between body mass index (BMI) and psoriasis.
Methods and Findings:
Following a review of published epidemiological evidence of the association between obesity and psoriasis, mendelian randomization (MR) was used to test for a causal relationship with BMI. We used a genetic instrument comprising 97 single-nucleotide polymorphisms (SNPs) associated with BMI as a proxy for BMI (expected to be much less confounded than measured BMI). One-sample MR was conducted using individual-level data (396,495 individuals) from the UK Biobank and the Nord-Trøndelag Health Study (HUNT), Norway. Two-sample MR was performed with summary-level data (356,926 individuals) from published BMI and psoriasis genome-wide association studies (GWASs). The one-sample and two-sample MR estimates were meta-analysed using a fixed-effect model. To test for a potential reverse causal effect, MR analysis with genetic instruments comprising variants from recent genome-wide analyses for psoriasis were used to test whether genetic risk for this skin disease has a causal effect on BMI. Published observational data showed an association of higher BMI with psoriasis. A mean difference in BMI of 1.26 kg/m2 (95% CI 1.02-1.51) between psoriasis cases and controls was observed in adults, while a 1.55 kg/m2 mean difference (95% CI 1.13-1.98) was observed in children. The observational association was confirmed in UK Biobank and HUNT data sets. Overall, a 1 kg/m2 increase in BMI was associated with 4% higher odds of psoriasis (meta-analysis odds ratio [OR] = 1.04; 95% CI 1.03-1.04; P = 1.73 × 10-60). MR analyses provided evidence that higher BMI causally increases the odds of psoriasis (by 9% per 1 unit increase in BMI; OR = 1.09 (1.06-1.12) per 1 kg/m2; P = 4.67 × 10-9). In contrast, MR estimates gave little support to a possible causal effect of psoriasis genetic risk on BMI (0.004 kg/m2 change in BMI per doubling odds of psoriasis (-0.003 to 0.011). Limitations of our study include possible misreporting of psoriasis by patients, as well as potential misdiagnosis by clinicians. In addition, there is also limited ethnic variation in the cohorts studied.
Conclusions:
Our study, using genetic variants as instrumental variables for BMI, provides evidence that higher BMI leads to a higher risk of psoriasis. This supports the prioritization of therapies and lifestyle interventions aimed at controlling weight for the prevention or treatment of this common skin disease. Mechanistic studies are required to improve understanding of this relationship
Ice geometry and thermal regime of Lyngmarksbræen Ice Cap, West Greenland
Observations remain sparse for peripheral glaciers and ice caps in Greenland. Here, we present the results of a multi-frequency radar survey of Lyngmarksbræen Ice Cap in West Greenland conducted in April 2017. Radar measurements show thick ice of up to ~120 m in subglacial valleys associated with the largest outlet glaciers, while relatively thin ice cover the upper plateau ice divides, suggesting future vulnerability to ice cap fragmentation. At the time of the radar survey, Lyngmarksbræen Ice Cap had a total volume of 0.82 ± 0.1 km3. Measurements show a 1.5–2 m thick end-of-winter snowpack, and that firn is largely absent, signifying a prolonged period of negative mass balance for most of the ice cap. The thermal regime of Lyngmarksbræen Ice Cap is investigated through analysis of scattering observed along radar profiles. Results show that the ice cap is largely below the pressure melting point, but that temperate ice exists both in deep basal pockets and in shallow zones that some places extend from ~15 m depth and to the ice base. The distribution of shallow temperate ice appears unrelated to variations in ice thickness; instead we find a strong correlation to the presence of nearby surface crevasses.publishedVersio
