    An order effect in English infants’ discrimination of an Urdu affricate contrast

    An order effect was found in English infants’ discrimination of an Urdu contrast. In Experiment 1 7- and 11-month-old English infants were tested on the Urdu contrast between the affricates /tʃʰ/ and /tʃ/. The order of presentation was counterbalanced: At each age half the infants were habituated to the aspirated and tested on the unaspirated affricate, the other half habituated to the unaspirated and tested on the aspirated. As expected, younger infants discriminated the contrast whereas older infants did not, showing the expected decline in discrimination. Interestingly, order of presentation seemed to affect the older infants’ response. Experiment 2 tested the order effect directly. The results showed no asymmetry in the performance of 7-month olds but clear asymmetry in that of 11-month-olds, who discriminated the contrast only when the non-English-like aspirated affricate was presented first. Experiment 3 tested adult native-speakers of both Urdu and English. Although the English listeners showed a reduced sensitivity to the contrast, there was no effect due to order of presentation of the stimuli in either adult group. The finding of an asymmetry in the infants suggests that infants’ perceptual narrowing for speech sounds may be a more complex phenomenon than has generally been assumed

    Background The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd. Methods We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background. Results First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001). Conclusions In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival

    Effects of four different modality training programs on IQ and reading readiness performance in the lower socio-economic level kindergarten child

    Four perceptual training programs were incorporated into an individualized kindergarten curriculum for disadvantaged children. One program stressed general auditory and visual perceptual skills, a second auditory skills specific to decoding, a third visual skills specific to decoding, and a fourth auditory and visual skills specific to decoding. Each of the four treatment groups were taught by the same teachers. Post-tests included IQ, auditory and visual measures of reading readiness, and two learning rate tests, the Murphy-Durrell learning rate test which uses a look-say instructional method, and a parallel learning rate test constructed by the investigator using a linguistic instructional approach. Although there was a significant difference between groups at the end of the treatment period, no group or groups outperformed the others on all post-test measures. The auditory visual specific skill group registered significantly greater gains than the other three groups on IQ, and scored significantly higher than the auditory group on visual but not auditory reading post-tests. There was no interaction effect between treatment group and IQ level, auditory perception level or visual perception level as measured by the pre-tests. The investigator hypothesized that groups trained in specific visual skills would outperform the others on the Murphy-Durrell Learning rate test, and that the groups trained in specific auditory skills would outperform the others on the Linguistic Learning rate test. This hypothesis was not supported. A trend in the direction of the hypothesis suggested that a longer training period may have produced more definitive results

    Infant Recognition of Hebrew Vocalic Word Patterns

    The ability of infants to recognize phonotactic patterns in their native language is widely acknowledged. However, the specific ability of infants to recognize patterns created by nonadjacent vowels in words has seldom been investigated. In Semitic languages such as Hebrew, groups of multisyllabic words are identical in their nonadjacent vowel sequences and stress position but differ in the consonants interposed between the vowels. The goals of this study were to assess whether infants learning Hebrew show a preference for (1) a nonadjacent vocalic pattern or template, common in Hebrew nouns (CéCeC), over a nonattested nonadjacent vocalic pattern (CóCoC), and (2) a nonadjacent vocalic pattern common in Hebrew words (CaCóC) over an existing but less common pattern (CaCéC). Twenty Hebrew-learning infants aged 8 to 11 months were presented with lists of nonsense words featuring the first two patterns (Experiment 1), and 20 were presented with nonsense words featuring the second two patterns (Experiment 2). The results showed longer listening to CéCeC than to CóCoC lists and to CaCóC than to CaCéC lists, suggesting that infants recognized the common nonadjacent vocalic patterns in both cases. The study thus demonstrates that Hebrew-learning infants are able to disregard the intervening consonants within words and generalize their vocalic pattern to previously unheard nonwords, whether this pattern includes identical or different vowels and regardless of the rhythmic pattern of the word (trochaic or iambic). Analysis of the occurrence of the relevant vowel patterns in input speech in three Hebrew corpora (two addressed to children and one to adults) suggests that exposure to these patterns in words underlies the infants' preferences
