    Expression of activin A in human keratinocytes at early stages of cultivation

    AbstractActivins are members of the TGF-β superfamily and are classified into 3 types: activin A, which consists of a homodimer of βA, activin B, which consists of a homodimer of βB, and activin AB, which consists of a heterodimer of βAβB. We studied the expression of activin mRNAs by RT-PCR in normal human epidermis, cultured keratinocytes, and DJM-1 cells (a squamous cell carcinoma line). We could detect only activin A mRNA (βA) in normal human epidermis. In cultured keratinocyte and DJM-1 cells, activin βA mRNA was observed at 4 h but not at 96 h after plating. Activin A activity was detected in the conditioned medium of DJM-1 cells within 48 h. In addition, although follistatin mRNA was not observed in human epidermis in situ, it was transiently expressed in cultured cells at 4 h after plating. These findings suggest that the expression of these molecules in keratinocytes is associated with cell proliferation. In an in vitro tissue injury model, activin A was observed at the wound edge, where cell migration and proliferation may be activated. In DJM-1 cells cultured for 92 h, βA mRNA was observed 4 h after injury treatment. These findings suggest that activin A acts as a potent inducer of proliferation in vitro, at least in keratinocytes

Dua senyawa, yaitu artelactokromen (1), suatu diprenil piranoflavon, dan B-resorsilaldehid (2) telah ditemukan untuk pertama kalinya pada kayu batang tumbuhan Artocarpus lanceifolius Roxb. (Moraceae), suatu tumbuhan langka yang endemic untuk Indonesia dan dikenal dengan nama Keledang. Struktur molekul kedua senyawa tersebut telah ditetapkan berdasarkan data fisika dan spektroskopi (MS, 1H dan 13C NMR).     Enhanced in vivo osteogenesis by nanocarrier-fused bone morphogenetic protein-4

    Purpose: Bone defects and nonunions are major clinical skeletal problems. Growth factors are commonly used to promote bone regeneration; however, the clinical impact is limited because the factors do not last long at a given site. The introduction of tissue engineering aimed to deter the diffusion of these factors is a promising therapeutic strategy. The purpose of the present study was to evaluate the in vivo osteogenic capability of an engineered bone morphogenetic protein-4 (BMP4) fusion protein. Methods: BMP4 was fused with a nanosized carrier, collagen-binding domain (CBD), derived from fibronectin. The stability of the CBD-BMP4 fusion protein was examined in vitro and in vivo. Osteogenic effects of CBD-BMP4 were evaluated by computer tomography after intramedullary injection without a collagen-sponge scaffold. Recombinant BMP-4, CBD, or vehicle were used as controls. Expressions of bone-related genes and growth factors were compared among the groups. Osteogenesis induced by CBD-BMP4, BMP4, and CBD was also assessed in a bone-defect model. Results: In vitro, CBD-BMP4 was retained in a collagen gel for at least 7 days while BMP4 alone was released within 3 hours. In vivo, CBD-BMP4 remained at the given site for at least 2 weeks, both with or without a collagen-sponge scaffold, while BMP4 disappeared from the site within 3 days after injection. CBD-BMP4 induced better bone formation than BMP4 did alone, CBD alone, and vehicle after the intramedullary injection into the mouse femur. -Bone-related genes and growth factors were expressed at higher levels in CBD-BMP4-treated mice than in all other groups, including BMP4-treated mice. Finally, CBD-BMP4 potentiated more bone formation than did controls, including BMP4 alone, when applied to cranial bone defects without a collagen scaffold. Conclusion: Altogether, nanocarrier-CBD enhanced the retention of BMP4 in the bone, thereby promoting augmented osteogenic responses in the absence of a scaffold. These results suggest that CBD-BMP4 may be clinically useful in facilitating bone formation

    A novel, visible light-induced, rapidly cross-linkable gelatin scaffold for osteochondral tissue engineering

    Osteochondral injuries remain difficult to repair. We developed a novel photo-cross-linkable furfurylamine-conjugated gelatin (gelatin-FA). Gelatin-FA was rapidly cross-linked by visible light with Rose Bengal, a light sensitizer, and was kept gelled for 3 weeks submerged in saline at 37 degrees C. When bone marrow-derived stromal cells (BMSCs) were suspended in gelatin-FA with 0.05% Rose Bengal, approximately 87% of the cells were viable in the hydrogel at 24 h after photo-cross-linking, and the chondrogenic differentiation of BMSCs was maintained for up to 3 weeks. BMP4 fusion protein with a collagen binding domain (CBD) was retained in the hydrogels at higher levels than unmodified BMP4. Gelatin-FA was subsequently employed as a scaffold for BMSCs and CBD-BMP4 in a rabbit osteochondral defect model. In both cases, the defect was repaired with articular cartilage-like tissue and regenerated subchondral bone. This novel, photo-cross-linkable gelatin appears to be a promising scaffold for the treatment of osteochondral injury

    The stress that is one of these causes becomes a big social problem; the reason is that stress gives bad influence for mind and body of children. Research on various types of stress is being conducted from a wide range of perspectives, and there are many reports dealing with students in school education settings. The aim of this study, therefore, was to analyze health reactions and tendencies in lifestyle habits of students in different faculties and departments at a general women’s university, to identify the causes of the results, and obtain basic data required for individual physical and psychological health promotion. Analysis of the results showed a tendency for health levels and lifestyle habits to differ according to department. Physical activity waned, and it was predicted that when specific female physical complaints were added, the situation of stress would become long drawn out, further fostering feelings of oppression and fatigue. It is important for the maintenance of physical and mental health that students themselves look at the causes of their stress, revise their own lifestyle habits, and endeavor to overcome the biological reaction to stress. Furthermore, on the psychological front, it is necessary to heighten emotional self-control and to try to ensure smooth human relations with the support of specialist. That is, it seems that the important factors in maintaining physical and mental health are being aware of one’s own stress and achieving and maintaining improved lifestyle habits

It aims at mitigation of an ache and at urging the braking effect. Recently, many sports taping aiming at improvement in athletic performance, such as titanium and kinesio taping, is also used. Painkilling and a stillness action are urged to titanium according to the thermotherapy effect. And titanium is said to adjust biological current within the body and produce a relaxing effect. On the other hand, kinesio taping has the effect of increasing our natural ability to heal ourselves and has an effect in relief and medical treatment of an obstacle. The last research examines the physical effects of titanium. It results demonstrate that in the highly reliable group titanium was contributed to improving athletic performance. In this last research, the placebo effect is considered to have acted mentally. In the field of pharmacological, it is said that the placebo effect is based on cognitive evaluation of the cerebral cortex, influences the function of a living body and induces a difference to a vital reaction. In the field of clinical research of psychosomatics medicine, it is said that placebo can improve 30 - 40 percent of a patient\u27s subjective and objective clinical symptoms in wide range diseases, such as sharp pain, asthma, and high blood pressure. Furthermore, in the field of research of sports, it is said that cognitive evaluation to the placebo effect relates to Individual difference by the personality types. However, the quality of the objective data which supports this knowledge is not evaluated strictly. Accordingly, the present research, kinesio taping is shown as it is titanium, it examined whether the athletic performance was raised like titanium. That is, this research examined the justification of the placebo effect. As for kinesio taping, the usage differs from titanium. For the reason, it is thought that there is little influence by the effect of original of kinesio taping. It is surmised that kinesio taping plays the role of a placebo to cognitive evaluation of the placebo effect. The purpose of the present research is the elucidation of the relation of individual difference of the cognitive evaluation by the reliability of titanium and the placebo effect using the kinesio taping. In the first place, drum\u27s temperature, skin\u27s temperature, and brain waves were measured from the physiological side. To the second, the athletic performance was measured from the physical side. To the third, it inquired by using STAI from the psychological side. Our results demonstrate that in the highly reliable group titanium was received favorably and contributed to improving athletic performance
