    Effect of metabolic syndrome and individual components on colon cancer characteristics and prognosis

    Metabolic syndrome (MS) is recognized as a risk factor for colon cancer (CC). However, whether the cluster of metabolic changes that define MS also influence CC prognosis remains unclear. Thus, our aim was to investigate whether the presence of MS or any of the MS individual components could provide prognostic information on tumor phenotype and survival outcomes. Clinical and pathological data from patients with CC (n = 300) who underwent surgical resection at a single tertiary hospital were retrospectively collected to evaluate presence of MS components and diagnostic criteria, CC phenotype and disease outcomes. Patients were allocated into two groups according to the presence or absence of MS (n = 85 MS vs n = 83 non-MS). The overall prevalence of MS individual components was 82.7% for increased waist-circumference (WC), 61.3% for high blood pressure (BP), 48.8% for low HDL-cholesterol, 39.9% for high fasting glucose, and 33.9% for hypertriglyceridemia. Patients in the MS group presented smaller tumors (p = 0.006) with lower T-stage (p = 0.002). High BP (p = 0.029) and hypertriglyceridemia (p = 0.044) were associated with a smaller tumor size, while low-HDL (p = 0.008) was associated with lower T-stage. After propensity score matching using age, tumor size and staging as covariates high-BP (p = 0.020) and WC (p = 0.003) were found to influence disease-free survival, but not overall survival. In conclusion, despite MS being an established risk factor for CC, our data does not support the hypothesis that MS components have a negative impact on disease extension or prognosis.     Toxoplasma gondii Tissue Cyst: Cyst Wall Incorporation Activity and Matrix Cytoskeleton Proteins Paving the Way to Nutrient Acquisition

    Toxoplasma gondii is an intracellular parasite that causes chronic infection by the development of bradyzoites housed in tissue cysts, preferably in the muscles and central nervous system. The composition and the function of the cyst wall are still not fully understood. Are T. gondii cysts able to incorporate nutrients through its wall? If so, how would these nutrients be traversed to cross the cyst matrix to reach the bradyzoite forms? Herein, we tested the uptake capacity of the Toxoplasma tissue cyst wall by employing some fluid-phase endocytosis tracers as peroxidase (HRP) and bovine serum albumin (BSA). Fluorescence images revealed these molecules on the cyst wall as well as in the cyst matrix. The subcellular localization of the tracer was confirmed by ultrastructural analysis showing numerous labeled vesicles and tubules distributed within the cyst matrix in close association with intracystic bradyzoite membrane, suggesting the cyst wall as a route of nutrient uptake. Furthermore, we confirmed the presence of cytoskeleton proteins, such as tubulin, actin, and myosin, in the tissue cyst matrix that may explain the nutrient input mechanism through the cyst wall. A better understanding of the nutrient acquisition process by the cyst might potentially contribute to the development of new therapeutic targets against chronic toxoplasmosis

    Ultrasonography Reliability in the Detection of Inflammatory and Structural Abnormalities: An Exercise in Multiple Joints

    Background Ultrasonography is an image technique that allows rheumatologists to visualize structural and inflammatory changes within a joint. The objective of this study was to assess the interobserver and intraobserver reliability of musculoskeletal ultrasound (US) in the detection of inflammatory and destructive joint changes in patients with polyarthritis. Methods A Delphi exercise was undertaken to standardize and adapt the EULAR-OMERACT elementary US definitions of inflammatory lesions (effusion, synovial hypertrophy, power Doppler, bone erosions, and synovitis) for each joint. Fifteen patients were analyzed, and video clips of 600 joints were collected. Each joint was scored for the presence of each elementary component, on 2 separate occasions, by 6 examiners. Interobserver and intraobserver agreement analysis was assessed through Fleiss κ coefficient (κ). Results Considering all patients and all joints, the interobserver values were highest for erosions and lowest for effusion (κ = 0.7314 and κ = 0.6044, respectively). When analyzing different regions, the highest interobserver agreement was for tibiotalar joint (κ = 0.8043) and the lowest for wrist (κ = 0.6767). Intraobserver reliability was excellent for each and all elementary components and anatomical region. Conclusions The present study showed either a good or excellent US interobserver and intraobserver reliability in elementary elements and anatomical region. This kind of US reliability exercises are important for standardization of exploration in everyday practice by reducing the variability associated with this imaging technique, and ensuring a greater degree of homogeneity and future comparability in the assessment of disease activity in polyarthritis patients.info:eu-repo/semantics/publishedVersio

    The social vulnerability associated with the COVID-19 Pandemic: a study with citizens who resorted to a social emergency service in Portugal

    O objetivo do estudo foi caracterizar a situação familiar, socioeconómica e de apoio social de cidadãos assistidos numa resposta de emergência social durante a pandemia por COVID-19, analisando os processos de vulnerabilização social. Participaram 230 sujeitos que recorreram ao Serviço Social de uma instituição no Norte de Portugal, na sua maioria mulheres (77,8%; n = 179), com idades entre os 20 e os 85 anos. O estudo, observacional e correlacional, usou um inquérito para avaliar as variáveis sociodemográficas, socioeconómicas e apoio institucional, assim como a Escala Breve de Redes Sociais de Lubben e a Escala Breve de Coping Resiliente. Os resultados apontam para um agravamento da situação socioeconómica dos participantes, verificando-se que após o Estado de Emergência existiu uma maior probabilidade de os inquiridos estarem desempregados e procurarem apoio social formal. A maioria apresentava uma situação de privação material severa (97.4%), associada a um baixo nível de suporte informal. Houve associação entre apoio social informal e coping resiliente. / The present study aims to characterize the family, socioeconomic, and social support situation of citizens who used social emergency services during the COVID-19 pandemic. The participants were 230 individuals that resort to Social Work of a Northern Portuguese institution. The participants were mostly women (77,8%; n = 179), aged between 20 and 85 years old. It was conducted an observational and correlational study, through an inquiry used to assess sociodemographic, socioeconomic, and institutional support variables, as well as Lubben’s Brief Social Network Scale and Brief Resilient Coping Scale. The results evidence an aggravation of the socioeconomic situation of the participants with respondents more likely to be unemployed and to seek formal social support after the State of Emergency. Most presented a situation of severe material deprivation (97.4%), associated with a low level of informal support. There was an association between informal social support and resilient coping

    Antioxidant, allelopathic and toxic activity of Ochna serrulata

    Ochna serrulata (Ochnaceae) is an ornamental plant introduced into Brazil from Asia and Africa. Species of the Ochna genus are rich in phenolic compounds, mainly flavonoids. The biological screening of extracts and fractions showed that this plant exhibited a significant antioxidant activity, when evaluated by the DPPH and reducing potential assays. Ochna serrulata also demonstrated slight toxic activity against Artemia salina and a potential inhibitory allelopathic activity, when evaluated using the Lactuca sativa seed germination test. The ethyl acetate fraction, the most active one, was partitioned on a silica gel column to obtain epicatechin, which showed potential antioxidant activity.Colegio de Farmacéuticos de la Provincia de Buenos Aire

    European Portuguese Adaptation of Glasgow Content of Thoughts Inventory (GCTI): Psychometric Characterization in Higher Education Students

    Persistent cognitive activity is an important factor in disturbing sleep-onset both during bedtime and when attempting to get back to sleep after nocturnal awakenings. One of the most specific self-report measures designed to assess this feature is the Glasgow Content of Thoughts Inventory (GCTI). In this study, we investigated the preliminary psychometric properties of GCTI in a large sample of higher education European Portuguese students (N = 2995). Our results suggest that there is evidence of good internal consistency (α = 0.93) and validity indicators. Moreover, we found an interpretable factorial structure comprising 5 correlated factors that needs to be confirmed in future studies. The European Portuguese version of the GCTI appears to be a reliable and valid instrument for measurement of sleep-onset disturbing cognitions

    Analysis of the biofilm proteome of Xylella fastidiosa

    <p>Abstract</p> <p>Background</p> <p><it>Xylella fastidiosa </it>is limited to the xylem of the plant host and the foregut of insect vectors (sharpshooters). The mechanism of pathogenicity of this bacterium differs from other plant pathogens, since it does not present typical genes that confer specific interactions between plant and pathogens (avr and/or hrp). The bacterium is injected directly into the xylem vessels where it adheres and colonizes. The whole process leads to the formation of biofilms, which are considered the main mechanism of pathogenicity. Cells in biofilms are metabolically and phenotypically different from their planktonic condition. The mature biofilm stage (phase of higher cell density) presents high virulence and resistance to toxic substances such as antibiotics and detergents. Here we performed proteomic analysis of proteins expressed exclusively in the mature biofilm of <it>X. fastidiosa </it>strain 9a5c, in comparison to planktonic growth condition.</p> <p>Results</p> <p>We found a total of 456 proteins expressed in the biofilm condition, which correspond to approximately 10% of total protein in the genome. The biofilm showed 37% (or 144 proteins) different protein than we found in the planktonic growth condition. The large difference in protein pattern in the biofilm condition may be responsible for the physiological changes of the cells in the biofilm of <it>X. fastidiosa</it>. Mass spectrometry was used to identify these proteins, while real-time quantitative polymerase chain reaction monitored expression of genes encoding them. Most of proteins expressed in the mature biofilm growth were associated with metabolism, adhesion, pathogenicity and stress conditions. Even though the biofilm cells in this work were not submitted to any stress condition, some stress related proteins were expressed only in the biofilm condition, suggesting that the biofilm cells would constitutively express proteins in different adverse environments.</p> <p>Conclusions</p> <p>We observed overexpression of proteins related to quorum sensing, proving the existence of communication between cells, and thus the development of structuring the biofilm (mature biofilm) leading to obstruction of vessels and development of disease. This paper reports a first proteomic analysis of mature biofilm of <it>X. fastidiosa</it>, opening new perspectives for understanding the biochemistry of mature biofilm growth in a plant pathogen.</p
