    L'Adenocarcinoma Polmonare Ovino (OPAC) o Adenomatosi Polmonare, è un tumore contagioso dei piccoli ruminanti a lento sviluppo e basso indice di metastatizzazione, il cui agente eziologico è un retrovirus tipo-D denominato Jaagsiekte Sheep Retrovirus (JSRV). Nel il 10% circa dei casi, la patologia risulta associata a pneumopatie interstiziali croniche causate da Maedi Visna Virus (MVV), lentivirus tipo-B responsabile anche di lesioni encefaliche, mammarie e articolari (1). L'esistenza e la qualità di eventuali sinergie fra i due virus è stata più volte sospettata ma non ha ancora trovato convincenti conferme sperimentali. Con l'obiettivo di verificare la distribuzione cellulare di JSRV in corso di OPAC associato a Maedi, abbiamo condotto prove ibridocitochimiche ed immunoistochimiche su polmoni ovini naturalmente infetti dai due virus e su controlli negativi e di reagente. Sono stati utilizzati a tal scopo: i) un protocollo di PCR-in situ specifico per DNA provirale di JSRV esogeno (Sanna et al, 2001); ii) un anticorpo policlonale sviluppato contro il major capsid antigen (MCA) di JSRV e gentilmente messo a disposizione dal Dr. Chris Cousens del Moredun Research Institute, Edinburgh (UK)

    3D model generation using oblique images acquired by UAV

    In recent years, many studies revealed the advantages of using airborne oblique images for obtaining improved 3D city models (including façades and building footprints). Here the acquisition and use of oblique images from a low cost and open source Unmanned Aerial Vehicle (UAV) for the 3D high-level-of-detail reconstruction of historical architectures is evaluated. The critical issues of such acquisitions (flight planning strategies, ground control points distribution, etc.) are described. Several problems should be considered in the flight planning: best approach to cover the whole object with the minimum time of flight; visibility of vertical structures; occlusions due to the context; acquisition of all the parts of the objects (the closest and the farthest) with similar resolution; suitable camera inclination, and so on. In this paper a solution is proposed in order to acquire oblique images with one only flight. The data processing was realized using Structure-from-Motion-based approach for point cloud generation using dense image-matching algorithms implemented in an open source software. The achieved results are analysed considering some check points and some reference LiDAR data. The system was tested for surveying a historical architectonical complex: the “Sacro Monte di Varallo Sesia” in north-west of Italy. This study demonstrates that the use of oblique images acquired from a low cost UAV system and processed through an open source software is an effective methodology to survey cultural heritage, characterized by limited accessibility, need for detail and rapidity of the acquisition phase, and often reduced budgets

    Fires in Amazonian Blackwater Floodplain Forests: Causes, Human Dimension, and Implications for Conservation

    The Amazon basin is being increasingly affected by anthropogenic fires, however, most studies focus on the impact of fires on terrestrial upland forests and do not consider the vast, annually inundated floodplains along the large rivers. Among these, the nutrient-poor, blackwater floodplain forests (igapós) have been shown to be particularly susceptible to fires. In this study we analyzed a 35-year time series (1982/1983–2016/2017) of Landsat Thematic Mapper from the Jaú National Park (Central Amazonia) and its surroundings. Our overall objective was to identify and delineate fire scars in the igapó floodplains and relate the resulting time series of annual burned area to the presence of human populations and interannual variability of regional hydroclimatic factors. We estimated hydroclimatic parameters for the study region using ground-based instrumental data (maximum monthly temperature–Tmax_{max}, precipitation–P, maximum cumulative water deficit–MCWD, baseflow index–BFI, minimum water level–WLmin90_{min90} of the major rivers) and large-scale climate anomalies (Oceanic Niño Index–ONI), considering the potential dry season of the non-flooded period of the igapó floodplains from September to February. Using a wetland mask, we identified 518,135 ha of igapó floodplains in the study region, out of which 17,524 ha (3.4%) burned within the study period, distributed across 254 fire scars. About 79% of the fires occurred close to human settlements (<10 km distance), suggesting that human activities are the main source of ignition. Over 92.4% of the burned area is associated with El Niño events. Non-linear regression models indicate highly significant relationships (p < 0.001) with hydroclimatic parameters, positive with Tmax_{max} (R2^{2}adj. = 0.83) and the ONI (R2^{2}adj. = 0.74) and negative with P (R2^{2}adj. = 0.88), MCWD (R2^{2}adj. = 0.90), WLmin90_{min90} (R2^{2}adj. = 0.61) and BFI (R2^{2}adj. = 0.80). Hydroclimatic conditions were of outstanding magnitude in particular during the El Niño event in 2015/2016, which was responsible for 42.8% of the total burned floodplain area. We discuss these results under a historical background of El Niño occurrences and a political, demographic, and socioeconomic panorama of the study region considering the past 400 years, suggesting that disturbance of igapós by fires is not a recent phenomenon. Concluding remarks focus on current demands to increase the conservation to prevent and mitigate the impacts of fire in this vulnerable ecosystem

    A cascade of 24 histatins (histatin 3 fragments) in human saliva. Suggestions for a pre-secretory sequential cleavage pathway

    The systematic search by tandem mass spectrometry of human saliva from four different subjects, of 136 possible fragments originated from histatin 3, allowed the detection of 24 different peptides. They include, with the exception of histatin 4, all the known histatin 3 fragments, namely histatins 5-12 and the peptides corresponding to 15-24, 26-32, 29-32 residues, and 13 new fragments corresponding to 1-11, 1-12, 1-13, 5-13, 6-11, 6-13, 7-11, 7-12, 7-13, 14-24, 14-25, 15-25, and 28-32 residues of histatin 3. On the contrary, none of 119 possible fragments of histatin 1, including histatin 2, was detected. The results suggest that the genesis of histatin 3-related peptides, being under the principal action of trypsin-like activities, is probably not a random process but rather follows a sequential fragmentation pathway. Lack of detection of C-terminal fragments, with the exception of 26-32, 28-32, and 29-32 fragments, suggested that arginine 25 should be the first cleavage site, generating histatin 6 and 26-32 fragments. The genesis of 28-32 and 29-32 fragments and histatin 5 should implicate a subsequent exo-protease action. Similarly, lack of detection of fragments having Lys-5 and Arg-6 at the N terminus and Arg-25 at the C terminus strongly suggested that sequences KRKF (11-14 residues) and AKR (4-6 residues) should be the second and the third cleavage sites, respectively. Lys-17 and Arg-22 are not cleaved at all

    Peripheral inflammation preceeding ischemia impairs neuronal survival through mechanisms involving miR‐127 in aged animals

    Envelliment; Inflamació; MicroARNEnvejecimiento; Inflamación; MicroARNAging; Inflammation; MicroRNAIschemic stroke, the third leading cause of death in the Western world, affects mainly the elderly and is strongly associated with comorbid conditions such as atherosclerosis or diabetes, which are pathologically characterized by increased inflammation and are known to influence the outcome of stroke. Stroke incidence peaks during influenza seasons, and patients suffering from infections such as pneumonia prior to stroke exhibit a worse stroke outcome. Earlier studies have shown that comorbidities aggravate the outcome of stroke, yet the mediators of this phenomenon remain obscure. Here, we show that acute peripheral inflammation aggravates stroke‐induced neuronal damage and motor deficits specifically in aged mice. This is associated with increased levels of plasma proinflammatory cytokines, rather than with an increase of inflammatory mediators in the affected brain parenchyma. Nascent transcriptomics data with mature microRNA sequencing were used to identify the neuron‐specific miRNome, in order to decipher dysregulated miRNAs in the brains of aged animals with stroke and co‐existing inflammation. We pinpoint a previously uninvestigated miRNA in the brain, miR‐127, that is highly neuronal, to be associated with increased cell death in the aged, LPS‐injected ischemic mice. Target prediction tools indicate that miR‐127 interacts with several basally expressed neuronal genes, and of these we verify miR‐127 binding to Psmd3. Finally, we report reduced expression of miR‐127 in human stroke brains. Our results underline the impact of peripheral inflammation on the outcome of stroke in aged subjects and pinpoint molecular targets for restoring endogenous neuronal capacity to combat ischemic stroke.This study was supported by Emil Aaltonen Foundation, Academy of Finland and Finnish Cultural Foundation

    Bacterial and viral investigations combined with determination of phytoplankton and algal biotoxins in mussels and water from a Mediterranean coastal lagoon (Sardinia, Italy).

    Calich Lagoon is a Mediterranean coastal lagoon located along the northwestern coast of Sardinia (Italy). The connection to marine and fresh water determines the high productivity of this coastal lagoon. Despite its great potential and the presence of natural beds of bivalve mollusks (Mytilus galloprovincialis), the lagoon has not yet been classified for shellfish production. In this study, through a multidisciplinary approach, the presence of several bacterial pathogens (Escherichia coli, Salmonella spp., and Vibrio spp.) and viral pathogens (hepatitis A virus and norovirus genogroups I and II) was evaluated from March 2017 to February 2018. In addition, phytoplankton composition in lagoon waters and associated algal biotoxins (paralytic and diarrhetic shellfish poisoning) in mussels were also monitored. The aim of this study was to provide useful data to improve knowledge about their seasonal presence and to assess the potential risk for public health, as well as to provide input for future conservation and management strategies. In mussels, Salmonella spp. were found in spring, along with E. coli, but Salmonella spp. were not found in autumn or winter, even though E. coli was detected in these seasons. Vibrio parahaemolyticus was found in autumn and winter, but not in spring. Norovirus genogroups I and II were found in winter samples. None of the bacteria were found in summer. Algal biotoxins have never been detected in mussel samples. Among potentially harmful phytoplankton, only Pseudo-nitzschia spp. were present, mainly in summer. The results showed that a possible bacterial and viral contamination, together with the presence of potentially toxic microalgae, is a real problem. Therefore, the development of natural resource management strategies is necessary to ensure the good quality of waters and guarantee the protection of consumers

    Effective Neutralizing Antibody Response Against SARS-CoV-2 Virus and Its Omicron BA.1 Variant in Fully Vaccinated Hematological Patients

    SARS-CoV-2 and its variants cause CoronaVIrus Disease 19 (COVID-19), a pandemic disease. Hematological malignancies increase susceptibility to severe COVID-19 due to immunosuppression. Anti-SARS-CoV-2 neutralizing antibodies protect against severe COVID-19. This retrospective real-life study aimed to evaluate seropositivity and neutralizing antibody rates against SARS-CoV-2 and its Omicron BA.1 variant in hematological patients. A total of 106 patients with different hematologic malignancies, who have mostly received three or more vaccine doses (73%), were included in this study. Serum was collected between May and June 2022. The primary endpoint was anti-SARS-CoV-2 antibody response against ancestral (wild type; wt) and Omicron BA.1 virus, defined as a neutralizing antibody titer ≥ 1:10. Adequate neutralizing antibody response was observed in 75 (71%) and 87 (82%) of patients for wt and Omicron BA.1 variants, respectively.However, patients with B-cell lymphoproliferative disorders and/or those treated with anti-CD20 monoclonal antibodies in the prior 12&nbsp;months showed a lower seropositivity rate compared to other patients against both Omicron BA.1 variant (73% vs 91%; P = 0.02) and wt virus (64% vs 78%; P = 0.16). Our real-life experience confirmed that full vaccination against SARS-CoV-2 induces adequate neutralizing antibody protection for both the wt virus and Omicron BA.1 variants, even in hematological frail patients. However, protective measures should be maintained in hematological patients, especially those with B-cell lymphoproliferative diseases treated with anti-CD20 monoclonal antibodies, because these subjects could have a reduced neutralizing antibody production
