Enzymatic activity and immunoreactivity of Aca s 4, an alpha-amylase allergen from the storage mite Acarus siro

<p>Abstract</p> <p>Background</p> <p>Enzymatic allergens of storage mites that contaminate stored food products are poorly characterized. We describe biochemical and immunological properties of the native alpha-amylase allergen Aca s 4 from <it>Acarus siro</it>, a medically important storage mite.</p> <p>Results</p> <p><it>A. siro </it>produced a high level of alpha-amylase activity attributed to Aca s 4. This enzyme was purified and identified by protein sequencing and LC-MS/MS analysis. Aca s 4 showed a distinct inhibition pattern and an unusual alpha-amylolytic activity with low sensitivity to activation by chloride ions. Homology modeling of Aca s 4 revealed a structural change in the chloride-binding site that may account for this activation pattern. Aca s 4 was recognized by IgE from house dust mite-sensitive patients, and potential epitopes for cross-reactivity with house dust mite group 4 allergens were found.</p> <p>Conclusions</p> <p>We present the first protein-level characterization of a group 4 allergen from storage mites. Due to its high production and IgE reactivity, Aca s 4 is potentially relevant to allergic hypersensitivity.</p

Spatial pattern of the Walkability Index, Walk Score and Walk Score modification for elderly

Contemporary cities require excellent walking conditions to support human physical activity, increase humans' well-being, reduce traffic, and create a healthy urban environment. Various indicators and metrics exist to evaluate walking conditions. To evaluate the spatial pattern of objective-based indicators, two popular indices were selected-the Walkability Index (WAI), representing environmental-based indicators, and Walk Score (WS), which applies an accessibility-based approach. Both indicators were evaluated using adequate spatial units (circle buffers with radii from 400 m to 2414 m) in two Czech cities. A new software tool was developed for the calculation of WS using OSM data and freely available network services. The new variant of WS was specifically designed for the elderly. Differing gait speeds, and variable settings of targets and their weights enabled the adaptation of WS to local conditions and personal needs. WAI and WS demonstrated different spatial pattern where WAI is better used for smaller radii (up to approx. 800 m) and WS for larger radii (starting from 800 m). The assessment of WS for both cities indicates that approx. 40% of inhabitants live in unsatisfactory walking conditions. A sensitivity analysis discovered the major influences of gait speed and the beta coefficient on the walkability assessment.Web of Science115art. no. 27

SmSP2: A serine protease secreted by the blood fluke pathogen Schistosoma mansoni with anti-hemostatic properties.

BackgroundSerine proteases are important virulence factors for many pathogens. Recently, we discovered a group of trypsin-like serine proteases with domain organization unique to flatworm parasites and containing a thrombospondin type 1 repeat (TSR-1). These proteases are recognized as antigens during host infection and may prove useful as anthelminthic vaccines, however their molecular characteristics are under-studied. Here, we characterize the structural and proteolytic attributes of serine protease 2 (SmSP2) from Schistosoma mansoni, one of the major species responsible for the tropical infectious disease, schistosomiasis.Methodology/principal findingsSmSP2 comprises three domains: a histidine stretch, TSR-1 and a serine protease domain. The cleavage specificity of recombinant SmSP2 was determined using positional scanning and multiplex combinatorial libraries and the determinants of specificity were identified with 3D homology models, demonstrating a trypsin-like endopeptidase mode of action. SmSP2 displayed restricted proteolysis on protein substrates. It activated tissue plasminogen activator and plasminogen as key components of the fibrinolytic system, and released the vasoregulatory peptide, kinin, from kininogen. SmSP2 was detected in the surface tegument, esophageal glands and reproductive organs of the adult parasite by immunofluorescence microscopy, and in the excretory/secretory products by immunoblotting.Conclusions/significanceThe data suggest that SmSP2 is secreted, functions at the host-parasite interface and contributes to the survival of the parasite by manipulating host vasodilatation and fibrinolysis. SmSP2 may be, therefore, a potential target for anti-schistosomal therapy

Byla provedena analýza firmy pomocí analýzy SWOT, finanční analýzy, personální analýzy, PEST analýzy českého a německého trhu a analýzy konkurence. Na základě výsledků analýz byla vytvořena strategie firmy. S ohledem na požadavky majitele byly vytvořeny nabídky v českém a německém jazyce, logo firmy v elektronické podobě a webové stránky společnosti. Firma byla zanesena do databází firem na serverech Seznam, Atlas a Centrum. V rámci strategie čerpání dotací a řízení lidských zdrojů byly naznačeny možné způsoby řešení

Vaccination against the papilomavirus infections at female students 8th and 9 elementary school classes in the region Písek and their awareness of the issues

This dissertation thesis touches very current and among public and experts often discussed topic. It concerns particularly matter of papillomaviral infections and vaccination against them. The research proved vaccination-coverage level of respondents and also their knowledge level. The vaccination-coverage level of girls is very high (almost 80%). A lot of girls also used possibility of full vaccination in the age of 13. This dissertation thesis should serve as a source and summary information about HPV viruses, diseases caused by them, about ways of transmission and epidemiological measures. Furthermore it also informs about cervical cancer, risk factors, diagnostical possibilities, therapy and at last but not lest about prevention of cervical carcinoma

Comparison of physically active and inactive middle-aged women in terms of quality of life and overall body composition

Bakalářská práce se zabývá posouzením vnímané kvality života a celkového tělesného složení žen ve středním věku v závislosti na míře vykonávané pohybové aktivity. Cílem práce je porovnat rozdíly v tělesném složení a kvalitě života u skupiny pohybově aktivních a neaktivních žen středního věku. Výzkumu se účastnilo 38 žen ve věku 35-55 let. Na skupinu pohybově aktivních a neaktivních žen byly probandky rozděleny podle stanoveného kritéria na základě týdenního záznamu pohybové aktivity, který každá z žen před zahájením výzkumu vyplnila. Tělesné složení bylo diagnostikováno pomocí přístroje InBody370. Posouzení kvality života bylo provedeno na základě standardizovaného dotazníku WHOQOL-BREF.ObhájenoThe bachelor thesis deals with the assessment of the perceived quality of life and overall body composition of middle-aged women, depending on the level of physical activity. The aim of this work is to compare the differences in body composition and quality of life in a group of physically active and inactive middle-aged women. The study involved 38 women aged 35-55. The probands were divided into a group of physically active and inactive women according to a set criterion on the basis of a weekly record of physical activity, which each of the women filled in before the start of the research. Body composition was diagnosed using an InBody370. The quality of life assessment was performed on the basis of a standardized WHOQOL-BREF questionnaire
