    250-MHz synchronously pumped optical parametric oscillator at 2.25-2.6 {\mu}m and 4.1-4.9 {\mu}m

    A compact and versatile femtosecond mid-IR source is presented, based on an optical parametric oscillator (OPO) synchronously pumped by a commercial 250-MHz Er:fiber laser. The mid-IR spectrum can be tuned in the range 2.25-2.6 \mu m (signal) and 4.1-4.9 \mu m (idler), with average power from 20 to 60 mW. At 2.5 \mu m a minimum pulse duration of 110 fs and a power of 40 mW have been obtained. Active stabilization of the OPO cavity length has been achieved in the whole tuning range

    Role of D(-)-Lactic Acid in Prevention of Chlamydia trachomatis Infection in an In Vitro Model of HeLa Cells

    A vaginal microbiota dominated by certain Lactobacillus species may have a protective effect against Chlamydia trachomatis infection. One of the key antimicrobial compounds produced is lactic acid, which is believed to play a central role in host defense. Lactobacillus strains producing the D(-)-lactic acid isomer are known to exert stronger protection. However, the molecular mechanisms underlying this antimicrobial action are not well understood. The aim of this study was to investigate the role of D(-)-lactic acid isomer in the prevention of C. trachomatis infection in an in vitro HeLa cell model. We selected two strains of lactobacilli belonging to different species: a vaginal isolate of Lactobacillus crispatus that releases both D(-) and L(+) isomers and a strain of Lactobacillus reuteri that produces only the L(+) isomer. Initially, we demonstrated that L. crispatus was significantly more effective than L. reuteri in reducing C. trachomatis infectivity. A different pattern of histone acetylation and lactylation was observed when HeLa cells were pretreated for 24 h with supernatants of Lactobacillus crispatus or L. reuteri, resulting in different transcription of genes such as CCND1, CDKN1A, ITAG5 and HER-1. Similarly, distinct transcription patterns were found in HeLa cells treated with 10 mM D(-)- or L(+)-lactic acid isomers. Our findings suggest that D(-) lactic acid significantly affects two non-exclusive mechanisms involved in C. trachomatis infection: regulation of the cell cycle and expression of EGFR and α5ÎČ1-integrin

    [60]Fullerene\u2013porphyrin [n]pseudorotaxanes: self-assembly, photophysics and third-order NLO response

    By means of different spectroscopic techniques, we investigate a novel series of porphyrin derivatives (H2TPP), connected to dibenzo-24-crown-8 (DB24C8) moieties, which undergo self-assembly with different methano[60]fullerene units bearing dibenzylammonium (DBA) cations. The formation of both [2] and [3]pseudorotaxanes was proved by means of NMR, UV-Vis-NIR absorption and emission spectroscopies. With the support of molecular modelling studies, spectroscopic investigations showed the presence of a secondary interaction between the porphyrin and the C60 chromophores leading to the formation of different types of \u2018\u2018face-to-face\u2019\u2019 assemblies. Remarkably, investigations of the non-linear optical response of these supramolecular systems showed that individual porphyrin and fullerene derivatives exhibit significantly lower second hyperpolarizability values when compared to their pseudorotaxanes functionalised counterparts. This proves that this class of supramolecular materials possesses relevant NLO response, which strongly depends on the structural arrangement of the chromophores in solution. Introductio

    Solvent Molding of Organic Morphologies Made of Supramolecular Chiral Polymers

    12sihe self-assembly and self-organization behavior of uracil-conjugated enantiopure (R)- or (S)-1,1'-binaphthyl-2,2'-diol (BINOL) and a hydrophobic oligo(p-phenylene ethynylene) (OPE) chromophore exposing 2,6-di(acetylamino)pyridine termini are reported. Systematic spectroscopic (UVvis, CD, fluorescence, NMR, and SAXS) and microscopic studies (TEM and AFM) showed that BINOL and OPE compounds undergo triple H-bonding recognition, generating different organic nanostructures in solution. Depending on the solvophobic properties of the liquid media (toluene, CHCl3, CHCl3/CHX, and CHX/THF), spherical, rod-like, fibrous, and helical morphologies were obtained, with the latter being the only nanostructures expressing chirality at the microscopic level. SAXS analysis combined with molecular modeling simulations showed that the helical superstructures are composed of dimeric double-cable tape-like structures that, in turn, are supercoiled at the microscale.     Tunable 30 fs light pulses at 1 W power level from a Yb-pumped optical parametric oscillator

    We report on a Yb-pumped optical parametric oscillator (OPO) that delivers 30 fs pulses with spectral coverage from 680 to 910 nm and an average output power of up to 1.1 W. The resulting peak power is Ăą\u88ÂŒ0.5 MW, which is, to the best of our knowledge, the highest ever demonstrated in a femtosecond OPO. The intensity noise remains at a level of 0.2% rms, and rapid wavelength tuning is obtained by simply scanning the resonator length. The performances of the OPO are promising for a variety of applications in nonlinear microscopy and ultrafast spectroscopy

    Improving Risk Communication Strategies through Public Awareness and Engagement: Insights from South Tyrol and Carinthia

    This chapter presents experiences and results from the INTERREG Italy-Austria Project RiKoST-Risk communication strategies. The project is a collaboration between partners from research and public authorities and aims at improving target-group-oriented risk communication in South Tyrol (Italy) and Carinthia (Austria). Risk communication plays an essential role for risk governance and may address different aspects and fulfill various purposes, from informing about natural hazards, generating acceptance and awareness for structural and non-structural measures, to triggering participation, increasing resilience, and supporting the development of a risk-competent society. To be effective, risk communication needs, firstly, to acknowledge the needs of different target groups and, secondly, to develop approaches, tools and contents that are most suitable to reach and involve them. This chapter describes the results from different activities carried out in the project: a population survey to better understand people’s risk perception and their knowledge about natural hazards, the information channels they use and trust; awareness raising activities in different municipalities; interactive lessons and a workshop in schools; stakeholder workshops. Our results show that that existing non-structural protection and prevention measures, especially Hazard Zone Plans, are little known among the population, that trust in the responsible authorities is high and that there is a need for a risk dialog through different risk communication activities at different stages to provide targeted information on how individual citizens can contribute to risk management. The chapter concludes on how the presented results can be used by public authorities and policy makers to innovate risk communication strategies and to initiate a risk dialog with the overall aim to improve risk governance at local level

    Italian natural history museums on the verge of collapse?

    The Italian natural history museums are facing a critical situation, due to the progressive loss of scientific relevance, decreasing economic investments, and scarcity of personnel. This is extremely alarming, especially for ensuring the long-term preservation of the precious collections they host. Moreover, a commitment in fieldwork to increase scientific collections and concurrent taxonomic research are rarely considered priorities, while most of the activities are addressed to public events with political payoffs, such as exhibits, didactic meetings, expositions, and talks. This is possibly due to the absence of a national museum that would have better steered research activities and overall concepts for collection management. We here propose that Italian natural history museums collaborate to instate a “metamuseum”, by establishing a reciprocal interaction network aimed at sharing budgetary and technical resources, which would assure better coordination of common long-term goals and scientific activities

    Association between polymorphisms of TAS2R16 and susceptibility to colorectal cancer

    Background: genetics plays an important role in the susceptibility to sporadic colorectal cancer (CRC). In the last 10 years genome-wide association studies (GWAS) have identified over 40 independent low penetrance polymorphic variants. However, these loci only explain around 1‑4% of CRC heritability, highlighting the dire need of identifying novel risk loci. In this study, we focused our attention on the genetic variability of the TAS2R16 gene, encoding for one of the bitter taste receptors that selectively binds to salicin, a natural antipyretic that resembles aspirin. Given the importance of inflammation in CRC, we tested whether polymorphic variants in this gene could affect the risk of developing this neoplasia hypothesizing a role of TAS2R16 in modulating chronic inflammation within the gut. Methods: we performed an association study using 6 tagging SNPs, (rs860170, rs978739, rs1357949, rs1525489, rs6466849, rs10268496) that cover all TAS2R16 genetic variability. The study was carried out on 1902 CRC cases and 1532 control individuals from four European countries. Results: we did not find any statistically significant association between risk of developing CRC and selected SNPs. However, after stratification by histology (colon vs. rectum) we found that rs1525489 was associated with increased risk of rectal cancer with a (Ptrend of = 0.0071). Conclusions: our data suggest that polymorphisms within TAS2R16 gene do not have a strong influence on colon cancer susceptibility, but a possible role in rectal cancer should be further evaluated in larger cohorts
