    Louisa S. McCord's Caius Gracchus: A Transatlantic Southern Literary Response to 1848 European Revolutions

    Louisa S. McCord is the most important female intellectual in the antebellum South and one of its most recognized voices, even if her name rarely appears in studies not directly related to her region. McCord has a parallel, according to Mary Kelley, in Margaret Fuller, the relevant prewar Northern intellectual and essayist. From contrasting ideological positions, both left testimony of their interest in constructing a model of womanhood, capable of facing the contingencies of their times. Born in one of the most influential families of South Carolina, McCord produced a phenomenal synthesis of the conservative political, economic and religious arguments accepted in her times and managed to transform them into a coherent philosophy that stood as a firm foundation for a society based on slave labor and a rigid social hierarchy. With her political writings and especially with her tragedy Caius Gracchus, McCord shows that she was not only interested in Southern politics but, most important, in the possible consequences of European political upheavals, and specifically in the influence that the European Revolutions of 1848 could exert in American territories. McCord es la intelectual más destacada del sur de preguerra y una de sus voces más reconocidas, a pesar de que su nombre raras veces aparezca en estudios que no traten directamente de su región. Como explica la historiadora Mary Kelley, a McCord bien se la puede comparar con la otra gran intelectual y ensayista norteña de preguerra, Margaret Fuller. Desde posiciones antagónicas, ambas dejarían constancia en sus escritos de sus preocupaciones por construir un modelo de feminidad que hiciera frente a las contingencias históricas de su época. Nacida en una de las familias más influyentes de Carolina del Sur, McCord logró con sus escritos llegar a una síntesis de los argumentos políticos, económicos y religiosos aceptados en su época y transformarlos en una filosofía coherente, capaz de sustentar una sociedad basada en el trabajo de los esclavos y en una rígida jerarquía social. Con sus escritos y especialmente con su tragedia Caius Gracchus, McCord demuestra no solo haberse interesado por la vida política sureña sino también por las consecuencias que los sucesos europeos, en concreto, las revoluciones burguesas de 1848, ejercieron en los territorios norteamericanos

    Activity of cefiderocol against high-risk clones of multidrug-resistant Enterobacterales, Acinetobacter baumannii, Pseudomonas aeruginosa and Stenotrophomonas maltophilia

    BACKGROUND: Cefiderocol is a novel siderophore cephalosporin, developed for activity against MDR Gram-negative bacilli (MDR-GNB). OBJECTIVES: To assess the in vitro antibacterial activity of cefiderocol against a collection of MDR-GNB clinical isolates from hospitals in southern Spain. METHODS: Two hundred and thirty-one isolates of successful clones were tested: 125 Enterobacterales (121 ESBL- and/or carbapenemase-producing Klebsiella pneumoniae and 4 carbapenemase-producing Enterobacter cloacae), 80 Acinetobacter baumannii, 6 Pseudomonas aeruginosa and 20 Stenotrophomonas maltophilia. Ceftolozane/tazobactam, ceftazidime, ceftazidime/avibactam, cefepime, aztreonam, meropenem, amikacin, ciprofloxacin, colistin and tigecycline were used as comparators against Enterobacterales, P. aeruginosa and A. baumannii. Minocycline, levofloxacin and trimethoprim/sulfamethoxazole were studied against S. maltophilia instead of aztreonam, ciprofloxacin and cefepime. MICs were determined by broth microdilution according to CLSI guidelines. MIC determination was performed in CAMHB for all antimicrobials except cefiderocol, where iron-depleted CAMHB was used. RESULTS: Cefiderocol showed potent in vitro activity against the isolates analysed. MIC50 and MIC90 values were in the ranges 0.125-8 mg/L and 0.5-8 mg/L, respectively, and 98% of isolates were inhibited at ≤4 mg/L. Only five isolates showed cefiderocol MICs of >4 mg/L: three ST2/OXA-24/40-producing A. baumannii, one ST114/VIM-1-producing E. cloacae and one ST114/VIM-1 + OXA-48-producing E. cloacae. All KPC-3-producing K. pneumoniae were susceptible to cefiderocol, even those resistant to ceftazidime/avibactam. P. aeruginosa isolates showed cefiderocol MICs of <4 mg/L, including those resistant to ceftolozane/tazobactam. S. maltophilia isolates displayed cefiderocol MICs of <4 mg/L, including those resistant to levofloxacin and/or trimethoprim/sulfamethoxazole. CONCLUSIONS: Cefiderocol showed excellent activity against MDR-GNB, including carbapenem-resistant isolates, and was the most active antimicrobial tested against this collection

    GSK-3 is an RNA polymerase II phospho-CTD kinase

    We have previously found that UV-induced DNA damage causes hyperphosphorylation of the carboxy terminal domain (CTD) of RNA polymerase II (RNAPII), inhibition of transcriptional elongation and changes in alternative splicing (AS) due to kinetic coupling between transcription and splicing. In an unbiased search for protein kinases involved in the AS response to DNA damage, we have identified glycogen synthase kinase 3 (GSK-3) as an unforeseen participant. Unlike Cdk9 inhibition, GSK-3 inhibition only prevents CTD hyperphosphorylation triggered by UV but not basal phosphorylation. This effect is not due to differential degradation of the phospho-CTD isoforms and can be reproduced, at the AS level, by overexpression of a kinase-dead GSK-3 dominant negative mutant. GSK-3 inhibition abrogates both the reduction in RNAPII elongation and changes in AS elicited by UV. We show that GSK-3 phosphorylates the CTD in vitro, but preferentially when the substrate is previously phosphorylated, consistently with the requirement of a priming phosphorylation reported for GSK-3 efficacy. In line with a role for GSK-3 in the response to DNA damage, GSK-3 inhibition prevents UV-induced apoptosis. In summary, we uncover a novel role for a widely studied kinase in key steps of eukaryotic transcription and pre-mRNA processing.Fil: Nieto Moreno, Nicolás.     As mais antigas marmotas da Europa: Estudo métrico dos fósseis de Marmota do Plistocénico Inferior e Medio das jazidas de Sierra de Atapuerca (Burgos, Espanha)

    ABSTRACT: The oldest European remains of marmots (Genus Marmota) are 0.8 my old and come from the site of Gran Dolina, Atapuerca. Dental measurements from the specimens recovered at Gran Dolina are compared with other Early Pleistocene fossil marmots from Croatia; as well as a set of Middle and Late Pleistocene marmots from France and Italy and Middle Pleistocene fossils from other sites at Sierra de Atapuerca. These fossils are also compared with four species of modern marmots, including? an extensive sample of Marmota marmota. Plotting the length of each dental piece of Gran Dolina versus its width, less than 25% of the Early Pleistocene specimens fall into the variability of Marmota marmota, meanwhile the Middle Pleistocene fossils fall within (or extremely close) to its variability. These Early Pleistocene marmots cannot be metrically assigned to the extant alpine marmot.RESUMO: Os vestígios europeus mais antigos de marmotas (género Marmota) têm 0.8 milhões de anos e provêm da Gran Dolina, Atapuerca. As medições realizadas em peças dentárias de marmotas fósseis encontrados em Gran Dolina são comparadas com exemplares de marmotas provenientes do Pleistocénico inferior da Croácia e do Pleistocénico Médio e Superior de França e Itália, bem como com exemplares encontrados em outras localidades do Pleistocénico médio da Serra de Atapuerca. Estes fósseis também são comparados com quatro espécies de marmotas modernas, incluindo uma extensa amostra de Marmota marmota. Ao plotar o comprimento de cada peça dentária de Gran Dolina face a sua largura, menos do 25% dos exemplares do Pleistocénico Inferior caem dentro da variabilidade existente em Marmota marmota. Os exemplares do Pleistocénico Médio estão dentro (ou extremamente próximos) da variabilidade de Marmota marmota.     FIREMAP: Cloud-based software to automate the estimation of wildfire-induced ecological impacts and recovery processes using remote sensing techniques

    [EN] The formulation and planning of integrated fire management strategies must be strengthened by decision support systems about fire-induced ecological impacts and ecosystem recovery processes, particularly in the context of extreme wildfire events that challenge land management initiatives. Wildfire data collection and analysis through remote sensing earth observations is of utmost importance for this purpose. However, the needs of land managers are not always met because the exploitation of the full potential of remote sensing techniques requires a high level of technical expertise. In addition, data acquisition and storage, database management, networking, and computing requirements may present technical difficulties. Here, we present FIREMAP software, which leverages the potential of Google Earth Engine (GEE) cloud-based platform, an intuitive graphical user interface (GUI), and the European Forest Fire Information System (EFFIS) wildfire database for wildfire analyses through remote sensing techniques and data collections. FIREMAP software allows automatic computing of (i) machine learning-based burned area (BA) detection algorithms to facilitate the mapping of (historical) fire perimeters, (ii) fire severity spectral indices, and (iii) post-fire recovery trajectories through the inversion of physically-based radiative transfer models.
