Kombinierte organokatalytische und elektrosynthetische Reaktionen in einem neu entwickelten Elektrolyseaufbau und Vergleich von Elektrosynthese und Photoredoxkatalyse
In diesem Promotionsprojekt wurden kombinierte organokatalytische und elektrosynthetische Reaktionen durchgefĂŒhrt. HierfĂŒr wurde ein Elektrolyseaufbau entwickelt, mit dem galvanostatische Elektrolysen in ca. 2 ml durchgefĂŒhrt werden können. Zudem wurde eine Methode zur schnellen Ermittlung der produktiven Stromdichten solcher Reaktionen gefunden.
AbschlieĂend wurde anhand eines Beispiels ein Vergleich zwischen Elektrosynthese und Photoredoxkatalyse durchgefĂŒhrt
Interactions between gravity waves and cirrus clouds: asymptotic modeling of wave induced ice nucleation
We present an asymptotic approach for the systematic investigation of the
effect of gravity waves (GW) on ice clouds formed through homogeneous
nucleation. In particular, we consider high- and mid-frequency GW in the
tropopause region driving the formation of ice clouds, modeled with a
double-moment bulk ice microphysics scheme. The asymptotic approach allows for
identifying reduced equations for self-consistent description of the ice
dynamics forced by GW including the effects of diffusional growth and
nucleation of ice crystals. Further, corresponding analytical solutions for a
monochromatic GW are derived under a single-parcel approximation. It is
demonstrated that the asymptotic solutions capture the dynamics of the full ice
model and provide a simple expression for the nucleated number of ice crystals.
The present approach is extended to allow for superposition of GW, as well as,
for variable mean mass in the ice crystal distribution. Implications of the
results for an improved representation of GW variability in cirrus
parameterizations are discussed
Li-Fraumeni-like syndrome associated with a large BRCA1 intragenic deletion
Background: Li-Fraumeni (LFS) and Li-Fraumeni-like (LFL) syndromes are associated to germline TP53 mutations, and are characterized by the development of central nervous system tumors, sarcomas, adrenocortical carcinomas, and other early-onset tumors. Due to the high frequency of breast cancer in LFS/LFL families, these syndromes clinically overlap with hereditary breast cancer (HBC). Germline point mutations in BRCA1, BRCA2, and TP53 genes are associated with high risk of breast cancer. Large rearrangements involving these genes are also implicated in the HBC phenotype. Methods: We have screened DNA copy number changes by MLPA on BRCA1, BRCA2, and TP53 genes in 23 breast cancer patients with a clinical diagnosis consistent with LFS/LFL; most of these families also met the clinical criteria for other HBC syndromes. Results: We found no DNA copy number alterations in the BRCA2 and TP53 genes, but we detected in one patient a 36.4 Kb BRCA1 microdeletion, confirmed and further mapped by array-CGH, encompassing exons 9-19. Breakpoints sequencing analysis suggests that this rearrangement was mediated by flanking Alu sequences. Conclusion: This is the first description of a germline intragenic BRCA1 deletion in a breast cancer patient with a family history consistent with both LFL and HBC syndromes. The germline mutational landscape of BRCA1 and BRCA2 in Brazil
The detection of germline mutations in BRCA1 and BRCA2 is essential to the formulation of clinical management strategies, and in Brazil, there is limited access to these services, mainly due to the costs/availability of genetic testing. Aiming at the identification of recurrent mutations that could be included in a low-cost mutation panel, used as a first screening approach, we compiled the testing reports of 649 probands with pathogenic/likely pathogenic variants referred to 28 public and private health care centers distributed across 11 Brazilian States. Overall, 126 and 103 distinct mutations were identified in BRCA1 and BRCA2, respectively. Twenty-six novel variants were reported from both genes, and BRCA2 showed higher mutational heterogeneity. Some recurrent mutations were reported exclusively in certain geographic regions, suggesting a founder effect. Our findings confirm that there is significant molecular heterogeneity in these genes among Brazilian carriers, while also suggesting that this heterogeneity precludes the use of screening protocols that include recurrent mutation testing only. This is the first study to show that profiles of recurrent mutations may be unique to different Brazilian regions. Reappraising the appropriate calculation of the potential temperature
The potential temperature is a widely used quantity in atmospheric science since it corresponds to the entropy and is conserved for adiabatic changes of dry air. As such, it is routinely employed in applications ranging from atmospheric dynamics to transport modeling. The common formula to compute the potential temperature is based on the assumption of a constant specific heat capacity for the dry air, even though the latter is known to vary with temperature. We re-derive the (dry air) potential temperature for a recent temperature-dependent formulation of the specific heat capacity of dry air. The result is expected to provide values which are much closer at the true entropy value (expressed as a temperature) and hence serves as the reference potential temperature. However, its computation is less straightforward compared to the classical one, motivating the development of efficient approximations. Moreover, similarities and differences are discussed between the newly derived reference potential temperature and the classical one based on a constant specific heat capacity. The new reference shows different values and vertical gradients, in particular in the stratosphere and above. Applications of the new reference potential temperature are discussed in the context of common computations in the atmospheric sciences, including the potential vorticity or diabatic heating rates.</p

International audienceUltracold neutrons (UCNs) are formidable probes in precision tests of gravity. With their negligible electric charge, dielectric moment, and polarizability they naturally evade some of the problems plaguing gravity experiments with atomic or macroscopic test bodies. Taking advantage of this fact, the qBounce collaboration has developed a technique â gravity resonance spectroscopy (GRS) â to study bound quantum states of UCN in the gravity field of the Earth. This technique is used as a high-precision tool to search for hypothetical Non-Newtonian gravity on the micrometer scale. In the present article, we describe the recently commissioned Ramsey-type GRS setup, give an unambiguous proof of principle, and discuss possible measurements that will be performed

IgE-mediated activation of mast cells and basophils contributes to protective immunity against helminths but also causes allergic responses. The development and persistence of IgE responses are poorly understood, which is in part due to the low number of IgE-producing cells. Here, we used next generation sequencing to uncover a striking overlap between the IgE and IgG1 repertoires in helminth-infected or OVA/alum-immunized wild-type BALB/c mice. The memory IgE response after secondary infection induced a strong increase of IgE+ plasma cells in spleen and lymph nodes. In contrast, germinal center B cells did not increase during secondary infection. Unexpectedly, the memory IgE response was lost in mice where the extracellular part of IgG1 had been replaced with IgE sequences. Adoptive transfer studies revealed that IgG1+ B cells were required and sufficient to constitute the memory IgE response in recipient mice. T cell-derived IL-4/IL-13 was required for the memory IgE response but not for expansion of B cells from memory mice. Together, our results reveal a close relationship between the IgE and IgG1 repertoires in vivo and demonstrate that the memory IgE response is mainly conserved at the level of memory IgG1+ B cells. Therefore, targeting the generation and survival of allergen-specific IgG1+ B cells could lead to development of new therapeutic strategies to treat chronic allergic disorders
