    2D and 3D-Organized Cardiac Cells Shows Differences in Cellular Morphology, Adhesion Junctions, Presence of Myofibrils and Protein Expression

    Cardiac cells are organized in vivo in a complex tridimensional structural organization that is crucial for heart function. While in vitro studies can reveal details about cardiac cell biology, usually cells are grown on simplified two-dimensional (2D) environments. To address these differences, we established a cardiac cell culture composed of both 2D and three-dimensional (3D)-organized cells. Our results shows significant differences between the two culture contexts in relation to the overall morphology of the cells, contraction ability, proliferation rate, presence of intercellular adhesion structures, organization of myofibrils, mitochondria morphology, endoplasmic reticulum contents, cytoskeletal filaments and extracellular matrix distribution, and expression of markers of cardiac differentiation. Cardiac cells grown in 2D-context displayed a flattened and well spread shape, were mostly isolated and their cytoplasm was filled with a large network of microfilaments and microtubules. In contrast, 3D-cells were smaller in size, were always in close contact with each other with several cellular junctions, and displayed a less conspicuous cytoskeletal network. 3D-cells had more mitochondria and myofibrils and these cells contract spontaneously more often than 2D-cells. On the other hand, endoplasmic reticulum membranes were present in higher amounts in 2D-cells when compared to 3D-cells. The expression of desmin, cadherin and alpha-actinin was higher in 3D-aggregates compared to 2D-spread cells. These findings indicate that the tridimensional environment in which the cardiac cells are grown influence several aspects of cardiac differentiation, including cell adhesion, cell shape, myofibril assembly, mitochondria contents and protein expression. We suggest that the use of this cardiac culture model, with 2D and 3D-context cells, could be useful for studies on the effects of different drugs, or growth factors, giving valuable information on the biological response of cells grown in different spatial organizations

Para isso, utilizamos a análise de documentação de instituições públicas encarregadas de ações no âmbito do lazer, bem como um levantamento nos locais e equipamentos destinados ao mesmo uso. Esperamos fazer esse grande mapa da cidade apontando as ações que constituem a política de lazer do município, bem como fazer propostas de implementação de políticas de lazer para o governo municipal.Ministério do Esport

    SÍNTESE E AVALIAÇÃO DA ATIVIDADE CARRAPATICIDA DE DERIVADOS DE TIMOL, DO CARVACROL E DO EUGENOL

    O presente projeto procurou produzir produtos semi-sintéticos orgânicospreferencialmente que possuam atividade biológica, através de reações click,utilizando como composto de partida o timol, carvarol e eugenol. Os compostos estãopresentes nos óleos essenciais extraídos do orégano ( Origanum Vulgare ) tanto parao timol, quanto carvacrol e cravo da índia ( Eugenia caryophyllata ) para o eugenol. Osderivados foram produzidos via reações de cicloadição, utilizando o timol, o carvacrole o eugenol propargilados e benzilazidas. Seis compostos foram sintetizados comrendimentos entre 62 a 77%, caracterizados por técnicas espectroscópicas (IV, RMNde 1 H e de 13 C) e espectrométrica (EM), assim como, avaliados quato a sua atividadecarrapaticida, por meio de testes biológicos in vitro

    Aerobic exercise inhibits acute lung injury: from mouse to human evidence Exercise reduced lung injury markers in mouse and in cells

    Acute respiratory distress syndrome (ARDS) is defined as hypoxemic respiratory failure with intense pulmonary inflammation, involving hyperactivation of endothelial cells and neutrophils. Given the anti-inflammatory effects of aerobic exercise (AE), this study investigated whether AE performed daily for 5 weeks would inhibit extra-pulmonary LPS-induced ARDS. C57Bl/6 mice were distributed into Control, Exercise, LPS and Exercise+ LPS groups. AE was performed on a treadmill for 5x/week for four weeks before LPS administration. 24hours after the final AE physical test, animals received 100ug of LPS intra-peritoneally. In addition, whole blood cell culture, neutrophils and human endothelial cells were pre-incubated with IL-10, an anti-inflammatory cytokine induced by exercise. AE reduced total protein levels (p<0.01) and neutrophil accumulation in bronchoalveolar lavage (BAL) (p<0.01) and lung parenchyma (p<0.01). AE reduced BAL inflammatory cytokines IL-1 beta, IL-6 and GM-CSF (p<0.001), CXCL1/KC, IL-17, TNF-alpha and IGF-1 (p<0.01). Systemically, AE reduced IL-1 beta, IL-6 and IFN-gamma (p<0.001), CXCL1/KC (p<0.01) and TNF-alpha (p<0.05). AE increased IL-10 levels in serum (p<0.001) and BAL (p<0.001). Furthermore, AE increased superoxide dismutase SOD (p<0.01) and decreased superoxide anion accumulation in the lungs (p<0.01). Lastly, pre-incubation with IL-10 significantly reduced LPS-induced activation of whole blood cells, neutrophils and HUVECs, as observed by reduced production of IL-1 beta, IL-6, IL-8 and TNF-alpha.     Compreensão acerca dos fatores de risco de lesão por pressão em idosos internados em unidade de terapia intensiva

    The aim of this study is to identify and analyze the main risk factors for pressure injuries related to hospitalization in the Intensive Care Unit. This is a bibliographical review of exploratory, quantitative and qualitative scientific literature. To select the sample, a search was made of the electronic databases LILACS and BDENF, from which a careful analysis was carried out, enabling the selection of twenty articles to be used to identify the risks of pressure injury in the elderly in the intensive care unit. The data was collected between March and May of this year. It is understood that there is a high probability of PPI developing in elderly patients after 60 years of age. The average length of hospitalization and pathologies such as hypertension and diabetes mellitus are key factors in the appearance of PI. According to the publications researched, it can be concluded that PPLs are a problem in hospital units that care for critically ill patients due to the permanent or temporary reduction in motility that exists in many cases.El objetivo de este estudio es identificar y analizar los principales factores de riesgo de las lesiones por presión relacionadas con la hospitalización en la Unidad de Cuidados Intensivos. Se trata de una revisión bibliográfica de la literatura científica exploratoria, cuantitativa y cualitativa. Para seleccionar la muestra, se realizó una búsqueda en las bases de datos electrónicas LILACS y BDENF, a partir de la cual se llevó a cabo un cuidadoso análisis que permitió seleccionar veinte artículos para identificar los riesgos de lesiones por presión en ancianos en la Unidad de Cuidados Intensivos. Los datos se recogieron entre marzo y mayo de este año. Se entiende que existe una alta probabilidad de que se desarrolle una IPP en pacientes ancianos después de los 60 años. La duración media de la hospitalización y patologías como la hipertensión y la diabetes mellitus son factores clave en la aparición de la IPP. Según las publicaciones investigadas, se puede concluir que las IPP son un problema común en las unidades hospitalarias que atienden a pacientes críticos debido a la reducción permanente o temporal de la motilidad que existe en muchos casos.Este trabalho tem como objetivo identificar e analisar os principais fatores de risco para lesão por pressão, relacionados a internação em Unidade de Terapia Intensiva. Trata-se de uma revisão bibliográfica em literatura científica exploratória, quantitativa e qualitativa. Para a seleção da amostragem foi feita uma busca a partir dos bancos de dados eletrônicos LILACS e BDENF, a partir disso foi realizada uma análise criteriosa que possibilitou através de uma leitura a seleção de vinte artigos para serem utilizados na identificação dos riscos de lesão por pressão em idosos na unidade de terapia intensiva. Os dados foram coletados entre os meses março a maio deste ano. Entende-se uma alta probabilidade de crescimento de LPP em pacientes idosos após 60 anos de idade. O tempo médio de internação e as patologias como hipertensão arterial e diabetes mellitus apresenta-se como fator primordial ao aparecimento de LPP. De acordo com as publicações pesquisadas, conclui-se que as LPP são agravos presentes em unidades hospitalares que cuidam de pacientes críticos devido a redução permanente ou temporária de motilidade existente em muitos casos. &nbsp

    Effects of salinity on the physiology of Salvinia auriculata Aubl. (Salviniales, Pteridophyta)

    Salvinia auriculata Aubl. is reported to occur in different zones of the Capibaribe River, Pernambuco State, Brazil. This river varies in salinity in different areas. This study evaluated the growth, photosynthesis and pigment contents of S. auriculata at different salinity levels. Plant sections were collected in the Cursaí Reservoir, located in the municipality of Paudalho, Pernambuco, and were brought to a greenhouse, where they were put in glass flasks filled with 250 mL of liquid, placed on benches. The plants were exposed for 40 h to salinity levels of 0, 17 and 34, obtained with reservoir freshwater, 1:1 freshwater:seawater and pure seawater, respectively. At the end of the experimental period, the plants in salt water showed color changes, with brownish leaves. In addition, plant growth rates decreased. Salinity and time had a negative influence on photosynthetic responses such as Fv/Fm, ETRmax and ETR, which showed reductions under the highest salinity treatment. Response patterns may help to explain S. auriculata occurrence, and its distribution can be regulated by salinity

    MAMMALS IN PORTUGAL : A data set of terrestrial, volant, and marine mammal occurrences in P ortugal

    Mammals are threatened worldwide, with 26% of all species being includedin the IUCN threatened categories. This overall pattern is primarily associatedwith habitat loss or degradation, and human persecution for terrestrial mam-mals, and pollution, open net fishing, climate change, and prey depletion formarine mammals. Mammals play a key role in maintaining ecosystems func-tionality and resilience, and therefore information on their distribution is cru-cial to delineate and support conservation actions. MAMMALS INPORTUGAL is a publicly available data set compiling unpublishedgeoreferenced occurrence records of 92 terrestrial, volant, and marine mam-mals in mainland Portugal and archipelagos of the Azores and Madeira thatincludes 105,026 data entries between 1873 and 2021 (72% of the data occur-ring in 2000 and 2021). The methods used to collect the data were: live obser-vations/captures (43%), sign surveys (35%), camera trapping (16%),bioacoustics surveys (4%) and radiotracking, and inquiries that represent lessthan 1% of the records. The data set includes 13 types of records: (1) burrowsjsoil moundsjtunnel, (2) capture, (3) colony, (4) dead animaljhairjskullsjjaws, (5) genetic confirmation, (6) inquiries, (7) observation of live animal (8),observation in shelters, (9) photo trappingjvideo, (10) predators dietjpelletsjpine cones/nuts, (11) scatjtrackjditch, (12) telemetry and (13) vocalizationjecholocation. The spatial uncertainty of most records ranges between 0 and100 m (76%). Rodentia (n=31,573) has the highest number of records followedby Chiroptera (n=18,857), Carnivora (n=18,594), Lagomorpha (n=17,496),Cetartiodactyla (n=11,568) and Eulipotyphla (n=7008). The data setincludes records of species classified by the IUCN as threatened(e.g.,Oryctolagus cuniculus[n=12,159],Monachus monachus[n=1,512],andLynx pardinus[n=197]). We believe that this data set may stimulate thepublication of other European countries data sets that would certainly contrib-ute to ecology and conservation-related research, and therefore assisting onthe development of more accurate and tailored conservation managementstrategies for each species.     Iconografia tropical: motivos locais na arte colonial brasileira

    Este artigo estuda a representação visual da natureza tropical na arte sacra do período colonial brasileiro, entre os séculos XVI e XVIII, época em que as artes visuais do país se desenvolveram no contexto do barroco introduzido pelos missionários católicos. Foi na decoração das igrejas que apareceram algumas das primeiras representações artísticas de elementos da natureza local, notadamente as frutas tropicais, produzindo novas combinações junto à tradicional ornamentação fitomórfica europeia, constituída de folhas de acantos e vinhas. Após um levantamento das ocorrências dessas manifestações da temática local na decoração dos templos presentes nas regiões nordeste e sudeste do país, este trabalho aborda, nos textos dos viajantes e missionários produzidos no período, as interpretações cristãs da natureza tropical que permitiram o aproveitamento desses motivos como parte da estratégia de pregação e conversão católica por meio da alegorização moral e religiosa da natureza do Novo Mundo.This paper studies the visual representation of local nature in the sacred art developed during the colonial period of Brazilian history. In this period, between the XVIth and the XVIIIth centuries, the visual arts in the country evolved in the context of the Baroque introduced by Catholic missionaries. It was in the decoration of the churches in which the first representations of aspects of local nature, mostly the tropical fruits, appeared in Brazilian visual arts, producing new combinations together with the traditional European phytomorphic ornamentation of acanthus leaves and grapes. This research draws upon texts written by travellers and missionaries during the period to demonstrate how the Europeans interpreted and represented tropical nature and used these representations as part of the Catholic preaching strategy by means of moral and religious allegorization of the New World nature
