    Consistent distribution-free KK-sample and independence tests for univariate random variables

    A popular approach for testing if two univariate random variables are statistically independent consists of partitioning the sample space into bins, and evaluating a test statistic on the binned data. The partition size matters, and the optimal partition size is data dependent. While for detecting simple relationships coarse partitions may be best, for detecting complex relationships a great gain in power can be achieved by considering finer partitions. We suggest novel consistent distribution-free tests that are based on summation or maximization aggregation of scores over all partitions of a fixed size. We show that our test statistics based on summation can serve as good estimators of the mutual information. Moreover, we suggest regularized tests that aggregate over all partition sizes, and prove those are consistent too. We provide polynomial-time algorithms, which are critical for computing the suggested test statistics efficiently. We show that the power of the regularized tests is excellent compared to existing tests, and almost as powerful as the tests based on the optimal (yet unknown in practice) partition size, in simulations as well as on a real data example.Comment: arXiv admin note: substantial text overlap with arXiv:1308.155

    Making Talk Work: Exploring the Teaching of Collaborative Talk

    This thesis is the outcome of a PhD CASE Studentship funded by the ESRC and British Telecom. It presents an exploration into the teaching of collaborative talk. The study was conducted in three phases: exploratory, development and implementation. During the exploratory phase, observations and interviews were conducted in authentic workplace settings to gain an understanding of workplace collaboration and collaborative talk. During the development phase, a teaching unit for the teaching of collaborative talk at GCSE was devised, informed by understandings gleaned during the preceding phase. During the implementation phase, the teaching unit was taught by two teachers in their secondary English classrooms. Both participating classes were arranged into groups of 4: 8 groups in School 1 and 7 in School 2. For the duration of the 3 week teaching unit, groups were recorded via camera and audio recorder, and the data later synchronised. Both teachers wore an audio recorder to capture interactions with groups and the whole class. To complement the core data set, students were interviewed for their views on their learning. Student booklets provided a means of collecting both group and individual reflections and evaluative comments. The data was analysed to explore the development of students’ collaborative talk. The role of the teacher in implementing the teaching unit and supporting students’ development was also examined. The findings provide an insight into the realities of implementing successful collaborative talk in the ‘real’ secondary classroom. It contributes to conceptualisations of collaborative talk and its development. It makes links between the role of emotional engagement and dialogic interactions in supporting that development. It proposes teaching strategies which challenge perceived notions of ‘good’ talk and encourages the development of meta-language to support self-evaluation and the development of collaborative talk.ESRC and British Teleco

    Comprehensive Rare Variant Analysis via Whole-Genome Sequencing to Determine the Molecular Pathology of Inherited Retinal Disease

    Inherited retinal disease is a common cause of visual impairment and represents a highly heterogeneous group of conditions. Here, we present findings from a cohort of 722 individuals with inherited retinal disease, who have had whole-genome sequencing (n = 605), whole-exome sequencing (n = 72), or both (n = 45) performed, as part of the NIHR-BioResource Rare Diseases research study. We identified pathogenic variants (single-nucleotide variants, indels, or structural variants) for 404/722 (56%) individuals. Whole-genome sequencing gives unprecedented power to detect three categories of pathogenic variants in particular: structural variants, variants in GC-rich regions, which have significantly improved coverage compared to whole-exome sequencing, and variants in non-coding regulatory regions. In addition to previously reported pathogenic regulatory variants, we have identified a previously unreported pathogenic intronic variant in CHM\textit{CHM} in two males with choroideremia. We have also identified 19 genes not previously known to be associated with inherited retinal disease, which harbor biallelic predicted protein-truncating variants in unsolved cases. Whole-genome sequencing is an increasingly important comprehensive method with which to investigate the genetic causes of inherited retinal disease.This work was supported by The National Institute for Health Research England (NIHR) for the NIHR BioResource – Rare Diseases project (grant number RG65966).     La priorización de nuevos requerimientos a implementar en un software propietario es un punto fundamental para su mantenimiento, la conservación de la calidad, observación de las reglas de negocio y los estándares de la empresa. Aunque existen herramientas de priorización basadas en técnicas probadas y reconocidas, las mismas requieren una calificación previa de cada requerimiento. Si la empresa cuenta con solicitudes provenientes de varios clientes de un mismo producto, aumentan los factores que afectan a la empresa, las herramientas disponibles no contemplan estos aspectos y hacen mucho más compleja la tarea de calificación. Este trabajo de investigación abarca la realización de un relevamiento de los métodos de priorización y selección de nuevos requerimientos utilizados por empresas de la zona de Rosario, y la definición de una metodología para la selección un nuevo requerimiento, que implica el análisis y evaluación de todas las implicaciones sobre el producto de software y la empresa, respetando sus reglas de negocio.     Effect of angiotensin-converting enzyme inhibitor and angiotensin receptor blocker initiation on organ support-free days in patients hospitalized with COVID-19

    IMPORTANCE Overactivation of the renin-angiotensin system (RAS) may contribute to poor clinical outcomes in patients with COVID-19. Objective To determine whether angiotensin-converting enzyme (ACE) inhibitor or angiotensin receptor blocker (ARB) initiation improves outcomes in patients hospitalized for COVID-19. DESIGN, SETTING, AND PARTICIPANTS In an ongoing, adaptive platform randomized clinical trial, 721 critically ill and 58 non–critically ill hospitalized adults were randomized to receive an RAS inhibitor or control between March 16, 2021, and February 25, 2022, at 69 sites in 7 countries (final follow-up on June 1, 2022). INTERVENTIONS Patients were randomized to receive open-label initiation of an ACE inhibitor (n = 257), ARB (n = 248), ARB in combination with DMX-200 (a chemokine receptor-2 inhibitor; n = 10), or no RAS inhibitor (control; n = 264) for up to 10 days. MAIN OUTCOMES AND MEASURES The primary outcome was organ support–free days, a composite of hospital survival and days alive without cardiovascular or respiratory organ support through 21 days. The primary analysis was a bayesian cumulative logistic model. Odds ratios (ORs) greater than 1 represent improved outcomes. RESULTS On February 25, 2022, enrollment was discontinued due to safety concerns. Among 679 critically ill patients with available primary outcome data, the median age was 56 years and 239 participants (35.2%) were women. Median (IQR) organ support–free days among critically ill patients was 10 (–1 to 16) in the ACE inhibitor group (n = 231), 8 (–1 to 17) in the ARB group (n = 217), and 12 (0 to 17) in the control group (n = 231) (median adjusted odds ratios of 0.77 [95% bayesian credible interval, 0.58-1.06] for improvement for ACE inhibitor and 0.76 [95% credible interval, 0.56-1.05] for ARB compared with control). The posterior probabilities that ACE inhibitors and ARBs worsened organ support–free days compared with control were 94.9% and 95.4%, respectively. Hospital survival occurred in 166 of 231 critically ill participants (71.9%) in the ACE inhibitor group, 152 of 217 (70.0%) in the ARB group, and 182 of 231 (78.8%) in the control group (posterior probabilities that ACE inhibitor and ARB worsened hospital survival compared with control were 95.3% and 98.1%, respectively). CONCLUSIONS AND RELEVANCE In this trial, among critically ill adults with COVID-19, initiation of an ACE inhibitor or ARB did not improve, and likely worsened, clinical outcomes.     Since its publication, André Gide's works mainly generated biographical analyses. As Gide's texts are first-person narratives and as the stories written recall the author's life experience, especially as far as homosexuality is concerned, it is tempting to think that each book he wrote is somehow autobiographical. The point of this thesis is to question those analyses which seem to forget an essential dimension: fiction. Instead of thinking that each text is a mirror reflecting Gide's image, we are going to see how the author set himself in his fiction. Gide takes a posture: through his literary work, he doesn't show himself as he is, neither as he thinks he is, but rather as he would likely to be seen. The author-function established correspond to the author's invention of his own character: the author is created through fiction. The role Gide would like to endorse consists in defending pederasty. He is the one who creates gidian immoralism, a new ethics both hedonist and antimoralist. As Gide writes against social conventions and protestant moral, he establishes himself the prophet of a new religion. The posture Gide takes frees him from the guilt he inherited from his protestant upbringing for being a homosexual, trying to persuade not only his readership but also himself that he is right of doing so.Depuis sa parution, l'œuvre d'André Gide a principalement suscité des analyses biographiques. Comme les textes gidiens sont écrits à la première personne et que les diégèses présentent des similitudes avec la vie de l'auteur, notamment en ce qui concerne l'homosexualité, il est tentant de penser que chaque ouvrage relève d'une forme d'autobiographie. L'objectif de ce mémoire est de montrer les limites de ces théories en s'appuyant sur une dimension qu'elles oublient, à savoir la fiction. Au lieu de voir dans chaque texte une projection de soi, il s'agit d'y déceler une mise en fiction de soi. Gide prend une posture : grâce à son écriture, il se donne à lire à son lectorat non pas tel qu'il est ni tel qu'il pense être, mais tel qu'il aimerait être perçu. La fonction auteur mise en place par Gide correspond à l'invention de son propre personnage : l'auteur est créé par la fiction. Le rôle que Gide cherche à endosser est celui du défenseur de la pédérastie. Il est celui qui établit l'immoralisme gidien, une nouvelle éthique hédoniste et antimoraliste. Se dressant contre les conventions sociales et la morale protestante, s'établissant prophète d'une nouvelle religion, la posture que prend Gide lui permet de se débarrasser de toute la culpabilité que lui a laissée son éducation puritaine en regard de l'homosexualité, à la fois aux yeux du lecteur et à ses propres yeux
