    NiTi shape memory alloys (SMAs) are increasingly used in many engineering and medical applications, because they combine special functional properties, such as shape memory effect and pseudoelasticity, with good mechanical strength and biocompatibility.     IP6K3 and IPMK variations in LOAD and longevity: evidence for a multifaceted signaling network at the crossroad between neurodegeneration and survival

    Several studies reported that genetic variants predisposing to neurodegeneration were at higher frequencies in centenarians than in younger controls, suggesting they might favor also longevity. IP6K3 and IPMK regulate many crucial biological functions by mediating synthesis of inositol poly- and pyrophosphates and by acting non-enzymatically via proteinā€“protein interactions. Our previous studies suggested they affect Late Onset Alzheimer Disease (LOAD) and longevity, respectively. Here, in the same sample groups, we investigated whether variants of IP6K3 also affect longevity, and variants of IPMK also influence LOAD susceptibility. We found that: i) a SNP of IP6K3 previously associated with increased risk of LOAD increased the chance to become long-lived, ii) SNPs of IPMK, previously associated with decreased longevity, were protective factors for LOAD, as previously observed for UCP4. SNP-SNP interaction analysis, including our previous data, highlighted phenotype-specific interactions between sets of alleles. Moreover, linkage disequilibrium and eQTL data associated to analyzed variants suggested mitochondria as crossroad of interconnected pathways crucial for susceptibility to neurodegeneration and/or longevity. Overall, data support the view that in these traits interactions may be more important than single polymorphisms. This phenomenon may contribute to the non-additive heritability of neurodegeneration and longevity and be part of the missing heritability of these traits

    Exploring the Prognostic Role of Ki67 Proliferative Index in Merkel Cell Carcinoma of the Skin: Clinico-Pathologic Analysis of 84 Cases and Review of the Literature.

    The exact prediction of outcome of patients with Merkel cell carcinoma (MCC) of the skin is difficult to determine, although several attempts have been made to identify clinico-pathologic prognostic factors. The Ki67 proliferative index is a well-known marker routinely used to define the prognosis of patients with neuroendocrine neoplasms. However, its prognostic value has been poorly investigated in MCC, and available published results are often contradictory mainly because restricted to small series in the absence of standardized methods for Ki67 evaluation. For this reason, we explored the potential prognostic role of Ki67 proliferative index in a large series of MCCs using the WHO standardized method of counting positive cells in at least 500 tumor cells in hot spot areas on camera-captured printed images. In addition, since MCC may be considered as the cutaneous counterpart of digestive neuroendocrine carcinomas (NECs), we decided to stratify MCCs using the available and efficient Ki67 threshold of 55%, which was found prognostic in digestive NECs. This choice was also supported by the Youden index analysis. In addition, we analyzed the prognostic value of other clinico-pathologic parameters using both univariate and multivariate analysis.     Reentrant Spin-Peierls Transition in Mg-Doped CuGeO_3

    We report a synchrotron x-ray scattering study of the diluted spin-Peierls (SP) material Cu_{1-x}Mg_xGeO_3. In a recent paper we have shown that the SP dimerization attains long-range order only for x < x_c = 0.022(0.001). Here we report that the SP transition is reentrant in the vicinity of the critical concentration x_c. This is manifested by broadening of the SP dimerization superlattice peaks below the reentrance temperature, T_r, which may mean either the complete loss of the long-range SP order or the development of a short-range ordered component within the long-range ordered SP state. Marked hysteresis and very large relaxation times are found in the samples with Mg concentrations in the vicinity of x_c. The reentrant transition is likely related to the competing Neel transition which occurs at a temperature similar to T_r.     Predictor Analysis in Radiofrequency Ablation of Benign Thyroid Nodules: A Single Center Experience

    PURPOSE: To confirm the efficacy of ultrasound (US) guided radiofrequency ablation (RFA) in the treatment of benign thyroid nodules, we evaluated as primary outcome the technical efficacy and clinical success in a single center dataset. The secondary outcome was to find a correlation between nodulesā€™ pre-treatment features and volume reduction rate (VRR) ā‰„75% at 12 months after RFA and during follow-up period. METHODS: This retrospective study included 119 consecutive patients (99 females, 20 males, 51.5 Ā± 14.4 years) with benign thyroid nodules treated in our hospital between October 2014 and December 2018 with a mean follow-up of 26.8 months (range 3ā€“48). Clinical and US features before and after RFA were evaluated by a US examination at 1, 3, 6, 12 months and annually thereafter up to 48 months. RESULTS: The median pre-treatment volume was 22.4Ā ml; after RFA we observed a statistically significant volume reduction from the first month (11.7Ā ml) to the last follow-up (p 22.4Ā ml (HR 0.54, p 0.036) were found to be independent positive and negative predictors of VRR ā‰„75% respectively. One-month post RFA VRR ā‰„50% represented the best positive predictor of technical success. CONCLUSIONS: This study confirmed the efficacy of RFA in the treatment of benign thyroid nodules. In particular we show that by selecting macrocystic nodules smaller than 22.4Ā ml better long-term response can be achieved, which is predicted by an early shrinkage of the nodule

    Intron 4ā€“5 hTERT DNA Hypermethylation in Merkel Cell Carcinoma: Frequency, Association with Other Clinico-pathological Features and Prognostic Relevance

    Merkel cell carcinoma (MCC) is an aggressive skin tumor with neuroendocrine differentiation, mainly affecting elderly population or immunocompromised individuals. As methylation of the human telomerase reverse transcriptase (mhTERT) has been shown to be a prognostic factor in different tumors, we investigated its role in MCC, in particular in intron 4ā€“5 where rs10069690 has been mapped and recognized as a cancer susceptibility locus. DNA methylation analysis of hTERT gene was assessed retrospectively in a cohort of 69 MCC patients from the University of Bologna, University of Turin and University of Insubria. Overall mortality was evaluated with Kaplan-Meier curves and multivariable Royston-Parmar models. High levels of mhTERT (mhTERThigh) (HR = 2.500, p = 0.015) and p63 (HR = 2.659, p = 0.016) were the only two clinico-pathological features significantly associated with a higher overall mortality at the multivariate analysis. We did not find different levels of mhTERT between MCPyV (+) and (āˆ’) cases (21 vs 14, p = 0.554); furthermore, mhTERThigh was strongly associated with older age (80.5 vs 72&nbsp;years, p = 0.026), no angioinvasion (40.7% vs 71.0%, p = 0.015), lower Ki67 (50 vs 70%, p = 0.005), and PD-L1 expressions in both tumor (0 vs 3%, p = 0.021) and immune cells (0 vs 10%, p = 0.002). mhTERT is a frequently involved epigenetic mechanism and a relevant prognostic factor in MCC. In addition, it belongs to the shared oncogenic pathways of MCC (MCPyV and UV-radiations) and it could be crucial, together with other epigenetic and genetic mechanisms as gene amplification, in determining the final levels of hTERT mRNA and telomerase activity in these patients

    Magnetic susceptibility, magnetization, specific heat and positive muon spin relaxation (\musr) measurements have been used to characterize the magnetic ground-state of the spinel compound CuGa2O4\rm CuGa_2O_4. We observe a spin-glass transition of the S=1/2 Cu2+\rm Cu^{2+} spins below Tf=2.5K\rm T_f=2.5K characterized by a cusp in the susceptibility curve which suppressed when a magnetic field is applied. We show that the magnetization of CuGa2O4\rm CuGa_2O_4 depends on the magnetic histo Well below Tf\rm T_f, the muon signal resembles the dynamical Kubo-Toyabe expression reflecting that the spin freezing process in CuGa2O4\rm CuGa_2O_4 results Gaussian distribution of the magnetic moments. By means of Monte-Carlo simulati we obtain the relevant exchange integrals between the Cu2+\rm Cu^{2+} spins in this compound.Comment: 6 pages, 16 figure

    Nonlinear magnetic susceptibility and aging phenomena in reentrant ferromagnet: Cu0.2_{0.2}Co0.8_{0.8}Cl2_{2}-FeCl3_{3} graphite bi-intercalation compound

    Linear and nonlinear dynamic properties of a reentrant ferromagnet Cu0.2_{0.2}Co0.8_{0.8}Cl2_{2}-FeCl3_{3} graphite bi-intercalation compound are studied using AC and DC magnetic susceptibility. This compound undergoes successive phase transitions at the transition temperatures ThT_{h} (= 16 K), TcT_{c} (= 9.7 K), and TRSGT_{RSG} (= 3.5 K). The static and dynamic behaviors of the reentrant spin glass phase below TRSGT_{RSG} are characterized by those of normal spin glass phase with critical exponent Ī²\beta = 0.57 Ā±\pm 0.10, a dynamic critical exponent xx = 8.5 Ā±\pm 1.8, and an exponent pp (= 1.55 Ā±\pm 0.13) for the de Almeida -Thouless line. A prominent nonlinear susceptibility is observed between TRSGT_{RSG} and TcT_{c} and around ThT_{h}, suggesting a chaotic nature of the ferromagnetic phase (TRSGā‰¤Tā‰¤TcT_{RSG} \leq T \leq T_{c}) and the helical spin ordered phase (Tcā‰¤Tā‰¤ThT_{c} \leq T \leq T_{h}). The aging phenomena are observed both in the RSG and FM phases, with the same qualitative features as in normal spin glasses. The aging of zero-field cooled magnetization indicates a drastic change of relaxation mechanism below and above TRSGT_{RSG}. The time dependence of the absorption Ļ‡ā€²ā€²\chi^{\prime \prime} is described by a power law form (ā‰ˆtāˆ’bā€²ā€²\approx t^{-b^{\prime \prime}}) in the ferromagnetic phase, where bā€²ā€²ā‰ˆ0.074Ā±0.016b^{\prime \prime} \approx 0.074 \pm 0.016 at ff = 0.05 Hz and TT = 7 K. No Ļ‰t\omega t-scaling law for Ļ‡ā€²ā€²\chi^{\prime \prime} [ā‰ˆ(Ļ‰t)āˆ’bā€²ā€²\approx (\omega t)^{-b^{\prime \prime}}] is observed.Comment: 14 pages, 16 figures, and 2 table

    Comparison of perioperative outcomes between standard laparoscopic and robot-assisted approach in patients with rectosigmoid endometriosis

    Introduction: Robot-assisted laparoscopic surgery (RALS) has gained widespread application in several surgical specialties. Previous studies on the feasibility and safety of RALS vs standard laparoscopy (S-LPS) for rectosigmoid endometriosis are limited and reported conflicting data. This study aims to compare S-LPS and RALS in patients with rectosigmoid endometriosis in terms of perioperative surgical and clinical data. Material and methods: This is a multicentric, observational, prospective cohort study including 44 patients affected by rectosigmoid endometriosis referred to two tertiary referral centers for endometriosis from September 2018 to September 2019. Patients were divided into two groups: 22 patients underwent S-LPS, and 22 underwent RALS. Our primary outcome was to compare operative time (from skin incision to suture) between the two groups. Secondary outcomes included: operative room time (patient entry into operative room and patient out), estimated blood loss, laparotomic conversion rate, length of hospital stay, perioperative complications, and evaluation of endometriosis-related symptoms at 12-month follow up. Results: The two groups were comparable regarding preoperative and surgical data, except for higher rates of hysterectomies and bilateral uterosacral ligament removal procedures in the RALS group. Also after adjusting for these discrepancies, operative time was similar between S-LPS and RALS. Operative room time was statistically longer in the RALS group compared with that of S-LPS. No statistically significant difference was found concerning other study outcomes. Pain and bowel symptoms improved in both groups at 12-month follow up. Conclusions: If performed by expert teams, RALS provides similar perioperative outcomes compared with S-LPS in rectosigmoid endometriosis surgical treatment, except for longer operative room time

    A Genome-Wide Screening and SNPs-to-Genes Approach to Identify Novel Genetic Risk Factors Associated with Frontotemporal Dementia

    Frontotemporal dementia (FTD) is the second most prevalent form of early onset dementia after Alzheimerā€™s disease (AD). We performed a case-control association study in an Italian FTD cohort (n = 530) followed by the novel SNPs-to-genes approach and functional annotation analysis. We identified two novel potential loci for FTD. Suggestive SNPs reached p-values ~10-7 and OR > 2.5 (2p16.3) and 1.5 (17q25.3). Suggestive alleles at 17q25.3 identified a disease-associated haplotype causing decreased expression of -cis genes such as RFNG and AATK involved in neuronal genesis and differentiation, and axon outgrowth, respectively. We replicated this locus through the SNPs-to-genes approach. Our functional annotation analysis indicated significant enrichment for functions of the brain (neuronal genesis, differentiation and maturation), the synapse (neurotransmission and synapse plasticity), and elements of the immune system, the latter supporting our recent international FTD-GWAS. This is the largest genome-wide study in Italian FTD to date. Although our results are not conclusive, we set the basis for future replication studies and identification of susceptible molecular mechanisms involved in FTD pathogenesis
