    Development of New Placental and Fetal Expressed Sequence Tags (EST) for Gene Discovery in Pig Reproduction

    One major problem that has high economic impact on pig reproduction is the unexplained loss of potential porcine conceptuses during the first month of gestation. To better understand when and how these losses occur, it is imperative to investigate the underlying genetic regulatory mechanisms. We have recently initiated a large-scale cDNA sequencing project to provide molecular information regarding the genes expressed in female reproductive tissues. cDNA libraries are planned for ovary, hypothalamus, pituitary, placenta, uterus, and several stages of embryonic development. Sequence information will also be highly useful in developing sequence-tagged sites for physical mapping and developing comparative links between the human, mouse, and pig genome maps. We have previously reported the creation of two cDNA libraries, porcine fetal (day 20), and conceptus (day 17). Sequencing of these libraries produced 220 Expressed Sequence Tags (ESTs), with 180 sequences analyzed by clustering algorithms, and 139 clusters identified within these sequences. We now report the creation of two more libraries from porcine fetal (day 45) and placental tissues. The day 45 fetal library has 971,150 independent clones (average insert: 1.4 kb), whereas the placental library has 1,320,000 independent clones. Initial sequencing of the fetal library has produced 119 ESTs (81 clusters), whereas we have obtained 1411 ESTs (1056 clusters) from the placental library. After clustering all sequences thus far obtained, we have identified 1,233 unique clusters. Sequences obtained in this project will be deposited into Genbank dbEST, and all comparative homolog

    Hypothyroidism takes one of the main places in the structure of endocrine diseases. The urgency of the problem of people with disabilities due to hypothyroidism is emphasized by the growth not only of the primary but also the acquired disability.Purpose: studying the peculiarities of the dynamics of disability due to hypothyroidism in Ternopil region for 2013–2015.Materials and Methods. The medical-expert documentation of 692 patients and invalids due to hypothyroidism was researched, 474 of them – as a result of postoperative hypothyroidism and 218 – as a result of idiopathic. The analysis of ofcial statistical data of disability due to hypothyroidism of the adult working population was conducted.Results and Discussion. The dynamics of the primary disability index among patients and invalids with postoperative hypothyroidism has considerably improved. Indicators of primary disability of idiopathic hypothyroidism have a negative dynamics. In addition, the category of disabled persons I and II groups grew signicantly.Conclusions. Taking into account that the indicators of full and partial rehabilitation in idiopathic hypothyroidism remain low, the main principle of action with regard to persons with disabilities is their rehabilitation. It is recommended to introduce into the work of medical and preventive institutions a complex of rehabilitation measures, remedial treatment in rehabilitation centers and sanatorium and resort conditions.Гіпотиреоз займає одне з основних місць в структурі ендокринних захворювань. Актуальність проблеми інвалідів внаслідок гіпотиреозу підкреслюється зростанням не тільки первинної, але й набутої інвалідності.Мета. Вивчити особливості динаміки інвалідності внаслідок гіпотиреозу у Тернопільській області за 2013-2015 рр.Матеріали і методи: Досліджувалась медико-експертна документація 692 хворих та інвалідів внаслідок гіпотиреозу, з них 474 – внаслідок післяопераційного гіпотиреозу та 218 – внаслідок ідіопатичного. Проведено аналіз офіційних статистичних даних інвалідності внаслідок гіпотиреозу дорослого працездатного населення.Результати та їх обговорення: Динаміка показника первинної інвалідності серед хворих та інвалідів з післяопераційним гіпотиреозом вагомо покращилась. Показники первинної інвалідності ідіопатичного гіпотиреозу мають негативну динаміку. Крім того вагомо зросла категорія інвалідів І та ІІ груп.Висновки:Враховуючи, що показники повної та часткової реабілітації при ідіопатичному гіпотиреозі залишаються низькими, основним принципом дії щодо інвалідів є їх реабілітація. Рекомендується впровадити в роботу лікувально-профілактичних закладів комплекс реабілітаційних заходів, відновне лікування в реабілітаційних центрах і санаторно-курортних умовах

    Patients with the lung abscess and gangrene demonstrate quantitative disproportion of the polymorphnonuclear and mononuclear phagocytes and disorders in their oxygen-dependent metabolic, procoagulant and proteolytic activities in BAL fluid from the injured lung. The phagocytes’ reaction to activators and inhibitors can be different depending on the initial status of these cells. The correction of the phagocytes’ function requires a prior assessment of the changes in the leukocytes’ metabolic activity provided by activators and inhibitors. Sometimes the correction could be performed by extracorporal methods only followed by transposition of autologous mononuclear phagocytes into the injured lung.У больных с абсцессами и гангреной легких выявлены количественные изменения соотношения полиморфно-ядерных и мононуклеарных фагоцитов в лаважной жидкости из пораженного легкого, а также нарушения кислородзависимой метаболической, прокоагулянтной и протеолитической активности этих клеток. Реакция фагоцитов на активаторы и ингибиторы зависит от исходного состояния клеток и может быть разнонаправленной. Коррекция функций фагоцитов требует предварительной оценки характера изменений метаболической активности лейкоцитов под влиянием активаторов и ингибиторов. В ряде случаев коррекция может быть выполнена только экстракорпорально с последующей транспозицией аутологичных мононуклеарных фагоцитов в пораженное легкое

    Inorganic polyphosphate as an energy source in tumorigenesis

    Indexación ScopusCancer cells have high demands for energy to maintain their exceedingly proliferative growth. However, the mechanism of energy expenditure in cancer is not well understood. We hypothesize that cancer cells might utilize energy-rich inorganic polyphosphate (polyP), as energetic reserve. PolyP is comprised of orthophosphates linked by phosphoanhydride bonds, as in ATP. Here, we show that polyP is highly abundant in several types of cancer cells, including brain tumor-initiating cells (BTICs), i.e., stem-like cells derived from a mouse brain tumor model that we have previously described. The polymer is avidly consumed during starvation of the BTICs. Depletion of ATP by inhibiting glycolysis and mitochondrial ATP-synthase (OXPHOS) further decreases the levels of polyP and alters morphology of the cells. Moreover, enzymatic hydrolysis of the polymer impairs the viability of cancer cells and significantly deprives ATP stores. These results suggest that polyP might be utilized as a source of phosphate energy in cancer. While the role of polyP as an energy source is established for bacteria, this finding is the first demonstration that polyP may play a similar role in the metabolism of cancer cells.     Microenvironment alters epigenetic and gene expression profiles in Swarm rat chondrosarcoma tumors

    <p>Abstract</p> <p>Background</p> <p>Chondrosarcomas are malignant cartilage tumors that do not respond to traditional chemotherapy or radiation. The 5-year survival rate of histologic grade III chondrosarcoma is less than 30%. An animal model of chondrosarcoma has been established - namely, the Swarm Rat Chondrosarcoma (SRC) - and shown to resemble the human disease. Previous studies with this model revealed that tumor microenvironment could significantly influence chondrosarcoma malignancy.</p> <p>Methods</p> <p>To examine the effect of the microenvironment, SRC tumors were initiated at different transplantation sites. Pyrosequencing assays were utilized to assess the DNA methylation of the tumors, and SAGE libraries were constructed and sequenced to determine the gene expression profiles of the tumors. Based on the gene expression analysis, subsequent functional assays were designed to determine the relevancy of the specific genes in the development and progression of the SRC.</p> <p>Results</p> <p>The site of transplantation had a significant impact on the epigenetic and gene expression profiles of SRC tumors. Our analyses revealed that SRC tumors were hypomethylated compared to control tissue, and that tumors at each transplantation site had a unique expression profile. Subsequent functional analysis of differentially expressed genes, albeit preliminary, provided some insight into the role that thymosin-β4, c-fos, and CTGF may play in chondrosarcoma development and progression.</p> <p>Conclusion</p> <p>This report describes the first global molecular characterization of the SRC model, and it demonstrates that the tumor microenvironment can induce epigenetic alterations and changes in gene expression in the SRC tumors. We documented changes in gene expression that accompany changes in tumor phenotype, and these gene expression changes provide insight into the pathways that may play a role in the development and progression of chondrosarcoma. Furthermore, specific functional analysis indicates that thymosin-β4 may have a role in chondrosarcoma metastasis.</p

    Photometric multi-site campaign on the open cluster NGC 884 I. Detection of the variable stars

    CONTEXT: Recent progress in the seismic interpretation of field beta Cep stars has resulted in improvements of the physics in the stellar structure and evolution models of massive stars. Further asteroseismic constraints can be obtained from studying ensembles of stars in a young open cluster, which all have similar age, distance and chemical composition. AIMS: To improve our comprehension of the beta Cep stars, we studied the young open cluster NGC 884 to discover new B-type pulsators, besides the two known beta Cep stars, and other variable stars. METHODS: An extensive multi-site campaign was set up to gather accurate CCD photometry time series in four filters (U, B, V, I) of a field of NGC884. Fifteen different instruments collected almost 77500 CCD images in 1286 hours. The images were calibrated and reduced to transform the CCD frames into interpretable differential light curves. Various variability indicators and frequency analyses were applied to detect variable stars in the field. Absolute photometry was taken to deduce some general cluster and stellar properties. RESULTS: We achieved an accuracy for the brightest stars of 5.7 mmag in V, 6.9 mmag in B, 5.0 mmag in I and 5.3 mmag in U. The noise level in the amplitude spectra is 50 micromag in the V band. Our campaign confirms the previously known pulsators, and we report more than one hundred new multi- and mono-periodic B-, A- and F-type stars. Their interpretation in terms of classical instability domains is not straightforward, pointing to imperfections in theoretical instability computations. In addition, we have discovered six new eclipsing binaries and four candidates as well as other irregular variable stars in the observed field.Comment: Accepted for publication in Astronomy and Astrophysics, 21 pages, 14 figures, 4 tables.
