    Untargeted Metabolomic Profiling and Antioxidant Capacities of Different Solvent Crude Extracts of Ephedra foeminea

    Ephedra foeminea is a traditional medicinal plant used in the Eastern Mediterranean region. This study aims to investigate the chemical profiles of different solvent extracts of E. foeminea via an untargeted metabolomics approach, alongside determining their antioxidant capacities. E. foeminea samples collected from Jordan were macerated in solvents of varying polarities; dichloromethane/methanol, methanol, ethanol, ethyl acetate, and acetone. The crude extracts were subjected to comprehensive chemical profiling and metabolomics study using Gas chromatography–Mass spectrometry (GC–MS), Liquid chromatography–Mass spectrometry (LC–MS), and Nuclear Magnetic Resonance (NMR). The obtained data were analyzed using Venn diagrams, Principle Component Analysis (PCA), and Metabolite Enrichment Set Analysis (MESA). ABTS assay was performed to measure the crude extracts’ antioxidant activity. MESA revealed the dominant chemical groups as amino acids, fatty acids, carboxylic acids, and carbohydrates. Results indicated that dichloromethane/methanol and methanolic extracts had the most distinct composition as well as the most unique compounds. The methanolic extract had the most potency (IC50 249.6 µg/mL) in the ABTS assay. However, no significant differences were found. In conclusion, solvents influenced the recovery of metabolites in E. foeminea and the antioxidant activity of the E. foeminea methanolic extract could be correlated to the abundant presence of diverse bioactive compounds

    Fabrication of Hollow Nanocones Membrane with an Extraordinary Surface Area as CO<sub>2</sub> Sucker

    Recently, more and more attention has been paid to the development of eco-friendly solid sorbents that are cost-effective, noncorrosive, have a high gas capacity, and have low renewable energy for CO2 capture. Here, we claimed the fabrication of a three-dimensional (3D) film of hollow nanocones with a large surface area (949.5 m2/g), a large contact angle of 136.3°, and high surface energy. The synthetic technique is based on an electrochemical polymerization process followed by a novel and simple strategy for pulling off the formed layers as a membrane. Although the polymer-coated substrates were reported previously, the membrane formation has not been reported elsewhere. The detachable capability of the manufactured layer as a membrane braked the previous boundaries and allows the membrane’s uses in a wide range of applications. This 3D hollow nanocones membrane offer advantages over conventional ones in that they combine a π-electron-rich (aromatic ring), hydrophobicity, a large surface area, multiple amino groups, and a large pore volume. These substantial features are vital for CO2 capturing and storage. Furthermore, the hydrophobicity characteristic and application of the formed polymer as a CO2 sucker were investigated. These results demonstrated the potential of the synthesized 3D hollow polymer to be used for CO2 capturing with a gas capacity of about 68 mg/g and regeneration ability without the need for heat up

    Characterization of persistent organic contaminants in the atmosphere of Gadani's ship breaking yards and its surrounding: Implications for sustainable ship recycling practices

    Gadani is internationally renowned for its extensive ship-breaking operations, positioning it as one of the globe's primary ship-breaking hubs. A comprehensive study was conducted to evaluate the presence of organic contaminants in the air within Gadani, encompassing the areas surrounding ship-breaking facilities, proximate residential settlements, and adjacent roadways. Passive air samplers were employed to collect a total of 30 air samples. The analytical results unveiled a notably elevated concentration of specific organic compounds, with a pronounced prevalence of polycyclic aromatic hydrocarbons (PAHs), short-chain chlorinated paraffins (SCCPs), and polychlorinated biphenyls (PCBs) at the ship-breaking yard. Notably, dichlorodiphenyltrichloroethanes (DDTs) and DDE were detected at substantially lower levels. In particular, sites in close proximity to ship-breaking activities exhibited elevated concentrations of PCBs (Σ7PCB 0.065429 to 7.345714 ng/sample), PAHs (Σ8PAH 2.44 to 134.23 ng/sample), and SCCPs (0.18 to 25.6 ng/sample). Conversely, DDTs and DDE demonstrated higher concentrations near residential settlements. The evaluation of Molecular Diagnostic Ratios for PAHs revealed anthracene/anthracene + phenanthrene ratios of 0.88, 0.69, and 0.5 for ship-breaking areas, roadside locations, and community surroundings, respectively. Furthermore, the benz[a]anthracene/benz-[a]anthracene + chrysene molecular ratios were measured at 0.77 (ship-breaking sites), 0.82 (roadside), and 0.83 (community), respectively. The molecular ratio of fluoranthene/fluoranthene + pyrene at ship-breaking sites was 0.23, while roadside and community ratios were 0.36 and 0.89, respectively. These findings underscore the significant contribution of ship-recycling activities to the atmospheric release of SCCPs, PCBs, and PAHs, emphasizing the global imperative for responsible ship recycling practices

    New bioactive metabolites from the elicited marine sponge-derived bacterium Actinokineospora spheciospongiae sp. nov.

    Abstract Several approaches have been dedicated to activate the cryptic gene clusters in the genomes of actinomycetes for the targeted discovery of new fascinating biomedical lead structures. In the current study, N-acetylglucosamine was used to maximize the chemical diversity of sponge-derived actinomycete Actinokineospora spheciospongiae sp. nov. HR–ESI–MS was employed for dereplication study and orthogonal partial least square-discriminant analysis was applied to evaluate the HR–ESI–MS data of the different fractions. As a result, two new fridamycins H (1) and I (2), along with three known compounds actinosporin C (3), D (4), and G (5) were isolated from the solid culture of sponge-associated actinomycete Actinokineospora spheciospongiae sp. nov., elicited with N-acetylglucosamine. Characterization of the isolated compounds was pursued using mass spectrometry and NMR spectral data. Fridamycin H (1) exhibited significant growth inhibitory activity towards Trypanosoma brucei strain TC221. These results highlight the potential of elicitation in sponge-associated actinomycetes as an effective strategy for the discovery of new anti-infective natural products

    Bedford-Type Palladacycle-Catalyzed Miyaura Borylation of Aryl Halides with Tetrahydroxydiboron in Water

    A mild aqueous protocol for palladium catalyzed Miyaura borylation of aryl iodides, aryl bromides and aryl chlorides with tetrahydroxydiboron (BBA) as a borylating agent is developed. The developed methodology requires low catalyst loading of Bedford-type palladacycle catalyst (0.05 mol %) and works best under mild reaction conditions at 40 °C in short time of 6 h in water. In addition, our studies show that for Miyaura borylation using BBA in aqueous condition, maintaining a neutral reaction pH is very important for reproducibility and higher yields of corresponding borylated products. Moreover, our protocol is applicable for a broad range of aryl halides, corresponding borylated products are obtained in excellent yields up to 93% with 29 examples demonstrating its broad utility and functional group tolerance

    Supplements of an aqueous combination of Justicia adhatoda and Ocimum tenuiflorum boost antioxidative effects and impede hyperlipidemia

    Background: Justicia adhatoda and Ocimum tenuiflorum, locally known as bashok and tulsi, are two ethnomedicinally important herbs that have been using as dietary supplements for several therapeutic applications. This study evaluated the combined effect of both the herbs as an antioxidative and antihyperlipidemic agent. Methods: Antihyperlipidemic effect was assessed in a high-fat diet-induced hyperlipidemic model in Wistar albino rats. The rats were treated orally with extracts of bashok (J adhatoda, 200 mg/kg bw), tulshi (O tenuiflorum, 200 mg/kg bw), and a combination of bashok and tulshi (50:50), as well as with a reference drug, atorvastatin (10 mg/kg/day), with or without high-fat diet for 14 days. The antioxidative effect was studied using established in vitro models. The studies were supported by experimentally testing the effects of the extracts on membrane stabilization and inhibition of protein denaturation. Results: The results showed that the serum lipid profile was significantly decreased in the different treatment groups, with bashok having the greatest effect. Body weights, total serum protein, LDH, and relative liver and adipose tissue weights were markedly restored towards baseline values, the lowest atherogenic index being achieved with the combined extract. The combination treatment significantly enhanced total phenolic content and antioxidative capacity and greatly potentiated membrane stabilization, but inhibition of protein denaturation was not significantly affected. Conclusion: The data demonstrate that a combination of Justicia adhatoda and Ocimum tenuiflorum could be developed as a food supplement with antioxidative and antihyperlipidemic benefits

    Holospiniferoside: A New Antitumor Cerebroside from The Red Sea Cucumber Holothuria spinifera: In Vitro and In Silico Studies

    Chemical investigation of the methanolic extract of the Red Sea cucumber Holothuria spinifera led to the isolation of a new cerebroside, holospiniferoside (1), together with thymidine (2), methyl-α-d-glucopyranoside (3), a new triacylglycerol (4), and cholesterol (5). Their chemical structures were established by NMR and mass spectrometric analysis, including gas chromatography–mass spectrometry (GC–MS) and high-resolution mass spectrometry (HRMS). All the isolated compounds are reported in this species for the first time. Moreover, compound 1 exhibited promising in vitro antiproliferative effect on the human breast cancer cell line (MCF-7) with IC50_{50} of 20.6 µM compared to the IC50 of 15.3 µM for the drug cisplatin. To predict the possible mechanism underlying the cytotoxicity of compound 1, a docking study was performed to elucidate its binding interactions with the active site of the protein Mdm2–p53. Compound 1 displayed an apoptotic activity via strong interaction with the active site of the target protein. This study highlights the importance of marine natural products in the design of new anticancer agents

    Organic extracts of Asian plants potentially support thrombolysis in varied BMI groups

    A number of plant-derived drugs have been prescribed for treating atherothrombotic disorders such as myocardial or cerebral infarction. Therefore, this study investigated the individual and combined antithrombotic effects of six Asian plants on individuals with various BMIs. Venous blood drawn from healthy volunteers was allowed to form clots and treated with six organic fractions of Ophiorrhiza harrisiana Heyne, Haldina cordifolia (Roxb.) Ridsdale, Eclipta alba (L.) Hassk, Syzygium operculatum L., Lantana camara L., and Acanthus ilicifolius Linn. using the clot-lysis method. The weight of the clot before and after treatment provided the percentage of clot lysis. Variations in clot lysis in different BMI groups were assessed. A. ilicifolius induced the most thrombolytic activity, whereas L. camara showed slightly lower thrombolytic activity. The maximum clot-lysis activity was primarily observed in different fractions of O. harrisana, H. cordifolia, E. alba, S. operculatum, and A. ilicifolius. However, chloroform fractions of all plants except O. harrisana and L. camara showed maximum activity in individual plants. O. harrisana, H. cordifolia, E. alba, S. operculatum, L. camara, and A. ilicifolius showed the highest significant (P < 0.5 and P < 0.001) clotlysis activity viz., 46.60±0.79% (ethyl acetate), 55.40±1.023% (chloroform), 48.86±1.35% (CCl4 ), 44.89±1.76% (chloroform), 29.17±1.12% (ethanol), and 56.318±1.64% (chloroform). Obtained results were verified by ligandreceptor interaction through in silico approach. In conclusion, H. cordifolia, E. alba, and A. ilicifolius demonstrated promising thrombolytic effects; therefore, it is recommended that they be studied further for their therapeutic potential

    Global economic burden of unmet surgical need for appendicitis

    Background There is a substantial gap in provision of adequate surgical care in many low- and middle-income countries. This study aimed to identify the economic burden of unmet surgical need for the common condition of appendicitis. Methods Data on the incidence of appendicitis from 170 countries and two different approaches were used to estimate numbers of patients who do not receive surgery: as a fixed proportion of the total unmet surgical need per country (approach 1); and based on country income status (approach 2). Indirect costs with current levels of access and local quality, and those if quality were at the standards of high-income countries, were estimated. A human capital approach was applied, focusing on the economic burden resulting from premature death and absenteeism. Results Excess mortality was 4185 per 100 000 cases of appendicitis using approach 1 and 3448 per 100 000 using approach 2. The economic burden of continuing current levels of access and local quality was US 92492millionusingapproach1and92 492 million using approach 1 and 73 141 million using approach 2. The economic burden of not providing surgical care to the standards of high-income countries was 95004millionusingapproach1and95 004 million using approach 1 and 75 666 million using approach 2. The largest share of these costs resulted from premature death (97.7 per cent) and lack of access (97.0 per cent) in contrast to lack of quality. Conclusion For a comparatively non-complex emergency condition such as appendicitis, increasing access to care should be prioritized. Although improving quality of care should not be neglected, increasing provision of care at current standards could reduce societal costs substantially
