    The role of the Notch pathway in healthy and osteoarthritic articular cartilage: from experimental models to ex vivo studies

    Osteoarthritis is the most prevalent form of arthritis in the world. With the progressive ageing of the population, it is becoming a major public health problem. The involvement of certain signaling pathways, such as the Notch pathway, during cartilage pathology has been reported. In this review, we report on studies that investigated the expression pattern of the Notch family members in articular cartilage and the eventual involvement of this pathway in the modulation of the physiology and pathology of chondrocytes. Temporal and/or spatial modulation of this signaling pathway may help these cells to synthesize a new functional extracellular matrix and restore the functional properties of the articular cartilage

    Effects of acute and chronic exercise on immunological parameters in the elderly aged: can physical activity counteract the effects of aging?

    Immunosenescence is characterized by deterioration of the immune system caused by aging which induces changes to innate and adaptive immunity. Immunosenescence affects function and phenotype of immune cells, such as expression and function of receptors for immune cells which contributes to loss of immune function (chemotaxis, intracellular killing). Moreover, these alterations decrease the response to pathogens, which leads to several age-related diseases including cardiovascular disease, Alzheimer's disease, and diabetes in older individuals. Furthermore, increased risk of autoimmune disease and chronic infection is increased with an aging immune system, which is characterized by a pro-inflammatory environment, ultimately leading to accelerated biological aging. During the last century, sedentarism rose dramatically, with a concomitant increase in certain type of cancers (such as breast cancer, colon, or prostate cancer), and autoimmune disease. Numerous studies on physical activity and immunity, with focus on special populations (i.e., people with diabetes, HIV patients) demonstrate that chronic exercise enhances immunity. However, the majority of previous work has focused on either a pathological population or healthy young adults whilst research in elderly populations is scarce. Research conducted to date has primarily focused on aerobic and resistance exercise training and its effect on immunity. This review focuses on the potential for exercise training to affect the aging immune system. The concept is that some lifestyle strategies such as high-intensity exercise training may prevent disease through the attenuation of immunosenescence. In this context, we take a top-down approach and review the effect of exercise and training on immunological parameters in elderly at rest and during exercise in humans, and how they respond to different modes of training. We highlight the impact of these different exercise modes on immunological parameters, such as cytokine and lymphocyte concentration in elderly individuals

    Enhancing the motor learning of youth soccer players through preventing an internal focus of attention:The effect of shoes colour

    The purpose of this research was to assess how the motor learning skills in 7-years old soccer players can be improved by preventing an internal focus of attention via the use coloured shoes. We painted the classic black soccer shoes in six areas corresponding to six regions of the foot with which it is possible to interact with the ball. Thirty-four 7-years-old soccer players were randomized to two groups (Coloured n = 17 and Black, n = 17) to perform four basic football manoeuvres/tasks: reception (RECP), passing (PASS), ball management (MAGT), and shooting (SHOT). We found highly significant differences (P<0.001) in all four performance tests: mean(sd) RECP: 0.82(0.07) vs. 0.45(0.12); PASS: 0.85(0.07) vs. 0.47(0.09); MAGT: 0.91(0.09); SHOT: 1.00(1.00) vs. 0.44(0.16). Colored shoes appear to draw children's attention away from body centered cues without explicit verbal communications. We propose that this cognitive adaptation enhanced the technical gesture by preventing the negative processes associated with action constraining when adopting an internal focus attention (perhaps by allowing the foot to adapt to surfaces and movements more naturally than conditions that promote a focus on the body movement). Consequently, this type of coloured footwear could be used during childhood to allow children to enhance the performance of basic football exercises through preventing action constraining and promoting intuitive (non-body centered) action knowledge

    Impact of Moderate Physical Activity on Inflammatory Markers and Telomere Length in Sedentary and Moderately Active Individuals with Varied Insulin Sensitivity

    Introduction: Physical activity-associated immune response plays a crucial role in the aging process. This study aimed to determine the impact of short-term moderate physical activity on cytokine levels, oxidative stress markers, and telomere length in lean/ overweight young subjects. Methods: Fasting blood samples were collected from 368 participants at Qatar Biobank. Based on their homeostatic model assessment of insulin resistance (HOMA-IR), participants were categorized as insulin sensitive (IS) or insulin resistant (IR). Subsequently, they were divided into four groups: sedentary IS (n = 90), sedentary IR (n = 90), moderately active IS (n = 94), and moderately active IR (n = 94). Moderate physical activity was defined as walking at least two days per week for more than 150 minutes, as determined by physical activity questionnaires. Serum samples were analyzed for circulating inflammatory cytokines (IL-1β, IL-1RA, IL-6, IL-10, IL-22, MCP-1/CCL2, TNF-α), as well as antioxidant enzyme levels (SOD and catalase). Telomere lengths were measured in the respective DNA samples. Results: Moderately active IR participants exhibited significantly lower SOD activity, while catalase activity did not show significant differences. Moderately active IS participants had higher IL-6 and IL-10 levels compared to sedentary IS participants, with no significant differences observed in the IR counterparts. Telomere length did not significantly differ between the physically active and sedentary groups. Conclusion: This study highlights the potential anti-inflammatory and anti-oxidative stress effects of moderate physical activity in individuals with insulin sensitivity and insulin resistance. However, no significant changes in telomere length were observed, suggesting a complex relationship between physical activity and the aging process. Further research is needed to fully understand the underlying mechanisms and optimize the balance between anti-inflammation and anti-oxidation through exercise and lifestyle adjustments.</p

    The effect of acute and chronic exercise on steroid hormone fluctuations in young and middle-aged men

    The current study examine the effects of combined sprint and resistance training on serum total testosterone (TT), sex-hormone-binding globulin (SHBG) and cortisol (C), at rest, and in response to the Wingate Anaerobic-Test (WAnT) in 21 and 41years old men. Forty moderately-trained men were randomly assigned to a young trained (YT), young control (YC), middle-aged trained (MAT), and middle-aged control (MAC) group. Before (P1), and after (P2) training, blood samples were collected at rest and after exercise. At P1, higher C and lower TT was observed in middle-aged groups compared to younger ones (P0.05). Basal SHBG decreased significantly in YT at P2 (P0.05). Free-testosterone was significantly (P0.05). C post-WAnT increased significantly for MAT only (P0.05). The current study demonstrates that this training intervention may help increase steroids hormones in middle-aged men and counteract the negative effect of age on TT and free testosterone. Highlights: Total testosterone, cortisol and SHBG increase with the exercise intensity; Intensive training increase basal testosterone levels in middle­-aged men; Intensive training reduces age effect in testosterone levels in middle­-aged men; Intensive training does not alter basal SHBG and cortisol levels in middle­-aged men
