    AMPK-α1 or AMPK-α2 Deletion in Smooth Muscles Does Not Affect the Hypoxic Ventilatory Response or Systemic Arterial Blood Pressure Regulation During Hypoxia

    The hypoxic ventilatory response (HVR) is markedly attenuated by AMPK-α1 deletion conditional on the expression of Cre-recombinase in tyrosine hydroxylase (TH) expressing cells, precipitating marked increases in apnea frequency and duration. It was concluded that ventilatory dysfunction caused by AMPK deficiency was driven by neurogenic mechanisms. However, TH is transiently expressed in other cell types during development, and it is evident that central respiratory depression can also be triggered by myogenic mechanisms that impact blood supply to the brain. We therefore assessed the effect on the HVR and systemic arterial blood pressure of AMPK deletion in vascular smooth muscles. There was no difference in minute ventilation during normoxia. However, increases in minute ventilation during severe hypoxia (8% O2) were, if affected at all, augmented by AMPK-α1 and AMPK-α2 deletion in smooth muscles; despite the fact that hypoxia (8% O2) evoked falls in arterial SpO2 comparable with controls. Surprisingly, these mice exhibited no difference in systolic, diastolic or mean arterial blood pressure during normoxia or hypoxia. We conclude that neither AMPK-α1 nor AMPK-α2 are required in smooth muscle for the regulation of systemic arterial blood pressure during hypoxia, and that AMPK-α1 deficiency does not impact the HVR by myogenic mechanisms

    Allogeneic hematopoietic cell transplantation (HCT) remains the only proven curative therapy for the hematologic manifestation of Fanconi anemia (FA). Over the past 2 decades, major advances have been made such that transplant outcomes have markedly improved. With the development of in vitro fertilization and preimplantation genetic diagnosis, HLA-matched sibling donor umbilical blood transplantation may be an option for more patients with FA. Recently, the use of pluripotent stem cells has been explored as a novel approach to model the hematopoietic developmental defects in FA, and to provide a potential source of autologous stem cells that can be genetically manipulated and used to generate corrected hematopoietic progenitors

    Shoulder muscle EMG activity during push up variations on and off a Swiss ball

    BACKGROUND: Surface instability is a common addition to traditional rehabilitation and strength exercises with the aim of increasing muscle activity, increasing exercise difficulty and improving joint proprioception. The aim of the current study was to determine if performing upper body closed kinetic chain exercises on a labile surface (Swiss ball) influences myoelectric amplitude when compared with a stable surface. METHODS: Thirteen males were recruited from a convenience sample of college students. Surface electromyograms were recorded from the triceps, pectoralis major, latissimus dorsi, rectus abdominis and external oblique while performing push up exercises with the feet or hands placed on a bench and separately on a Swiss ball. A push up plus exercise was also evaluated with hands on the support surface. RESULTS AND DISCUSSION: Not all muscles responded with an increase in muscle activity. The pectoralis major muscle was not influenced by surface stability. The triceps and rectus abdominis muscles showed increases in muscle activity only when the hands were on the unstable surface. The external oblique muscle was only influenced by surface stability during the performance of the push up plus exercise. No muscle showed a change in activation level when the legs were supported by the Swiss ball instead of the bench. CONCLUSION: Muscle activity can be influenced by the addition of surface instability however an increase in muscle activity does not influence all muscles in all conditions. The relationship between the participant's center of mass, the location of the unstable surface and the body part contacting the Swiss ball may be important factors in determining the muscle activation changes following changes in surface stability

    Contemporary Pastoralism in the Dhofar Mountains of Oman

    In the Dhofar Mountains of Oman stakeholders are concerned about the social and ecological sustainability of pastoralism. In this study we used interviews with pastoralists to examine the prevailing drivers of pastoralism and how they are changing. We find that people are committed to pastoralism for sociocultural reasons, but also that this commitment is under pressure because of husbandry costs and changing values. We find that capital investment in feedstuff enables pastoralists to overcome the density-dependent regulation of livestock populations. However, high production costs deter investment in marketing and commercialization, and there is little offtake of local livestock. Our study reveals how pastoral values, passed down within households, motivate pastoralism in the face of high husbandry costs, modernization and social change

    Concentration‐dependent optical‐absorption coefficient in n‐type GaAs

    The doping-dependent, near-band-edge optical-absorption coefficient CY(h v) was deduced from optical transmission measurements in n-type GaAs thin films. The selenium-doped films were grown by metalorganic chemical-vapor deposition and do ed to produce room-temperature electron concentrations from 1.3 x 10” to 3.8X 1018 cm- P . The transmission measurements covered photon energies between 1.35 and 1.7 eV and were performed on double heterostructures with the substrate removed by selective etching. The results show good qualitative agreement with previous studies and good quantitative agreement, except for the heavily doped samples. For na=3.8 X 10” cme3, a( 1.42 eV\u3e is approximately four times that reported by previous workers. Secondary-ion-mass spectrometry measurements on flms grown under differing conditions demonstrate that a(hv) is sensitive to electrically inactive dopants and supports the hypothesis that precipitates or compensation influenced previous measurements. These comprehensive results on high-quality, uncompensated material should prove useful for fundamental studies of optical transitions in n-type GaAs as well as for modeling optoelectronic devices

    Microsecond Lifetimes and Low Interface Recombination Velocities in Moderately Doped n-GaAs Thin Films

    We have observed lifetimes greater than 1 ps in moderately doped, thin film, n-GaAs/A1a,Gae,As double heterostructure membranes formed by etching away the substrate. We attribute these ultralong lifetimes to enhanced photon recycling caused by the removal of the substrate. Nonradiative recombination in the bulk and at the interfaces is very low; the upper limit of the interface recombination velocity is 25 cm/S.-Such long lifetimes in GaAs doped at N,= 1.3 X 10” cme3 suggest that thin-film solar cells offer a potential option for achieving very high efficiencies

    Nonspherical similarity solutions for dark halo formation

    We carry out fully 3-dimensional simulations of evolution from self-similar, spherically symmetric linear perturbations of a Cold Dark Matter dominated Einstein-de Sitter universe. As a result of the radial orbit instability, the haloes which grow from such initial conditions are triaxial with major-to-minor axis ratios of order 3:1. They nevertheless grow approximately self-similarly in time. In all cases they have power-law density profiles and near-constant velocity anisotropy in their inner regions. Both the power-law index and the value of the velocity anisotropy depend on the similarity index of the initial conditions, the former as expected from simple scaling arguments. Halo structure is thus not "universal" but remembers the initial conditions. On larger scales the density and anisotropy profiles show two characteristic scales, corresponding to particles at first pericentre and at first apocentre after infall. They are well approximated by the NFW model only for one value of the similarity index. In contrast, at all radii within the outer caustic the pseudo phase-space density can be fit by a single power law with an index which depends only very weakly on the similarity index of the initial conditions. This behaviour is very similar to that found for haloes formed from LCDM initial conditions and so can be considered approximately universal.Comment: 8 pages, 7 figures, submitted to MNRA
